BACKGROUND AND OBJECTIVES: Long-term pathological myocardial hypertrophy (MH) seriously affects the normal function of the heart. Dronedarone was reported to attenuate left ventricular hypertrophy of mice. However, the molecular regulatory mechanism of dronedarone in MH is unclear. METHODS: Angiotensin II (Ang II) was used to induce cell hypertrophy of H9C2 cells. Transverse aortic constriction (TAC) surgery was performed to establish a rat model of MH. Cell size was evaluated using crystal violet staining and rhodamine phalloidin staining. Reverse transcription quantitative polymerase chain reaction and western blot were performed to detect the mRNA and protein expressions of genes. JASPAR and luciferase activity were conducted to predict and validate interaction between forkhead box O3 (FOXO3) and protein kinase inhibitor alpha (PKIA) promoter. RESULTS: Ang II treatment induced cell hypertrophy and inhibited sirtuin 1 (SIRT1) expression, which were reversed by dronedarone. SIRT1 overexpression or PKIA overexpression enhanced dronedarone-mediated suppression of cell hypertrophy in Ang II-induced H9C2 cells. Mechanistically, SIRT1 elevated FOXO3 expression through SIRT1-mediated deacetylation of FOXO3 and FOXO3 upregulated PKIA expression through interacting with PKIA promoter. Moreover, SIRT1 silencing compromised dronedarone-mediated suppression of cell hypertrophy, while PKIA upregulation abolished the influences of SIRT1 silencing. More importantly, dronedarone improved TAC surgery-induced MH and impairment of cardiac function of rats via affecting SIRT1/FOXO3/PKIA axis. CONCLUSIONS: Dronedarone alleviated MH through mediating SIRT1/FOXO3/PKIA axis, which provide more evidences for dronedarone against MH.

Scientific assessment of landscape ecological risk in ecologically fragile areas of the upper reaches of the Yangtze River is of great significance to regional ecological regulation and construction of the Yangtze River ecological security barrier. With the dry-hot valley area of Jinsha River in Yunnan Province as the research area, we constructed a landscape ecological risk evaluation model, and analyzed the spatial and temporal variations of regional landscape ecological risk. The results showed that the average values of landscape ecological risk index (LER) in the study area were 0.414, 0.398, and 0.462 in 2000, 2010 and 2020, respectively. The LER value of the whole region had reached a higher risk level by 2020. In 2000 and 2010, the landscape ecological risk zones of each level were staggered, and the high-risk zones showed a centralized distribution in 2020. During the two decades, the average LER of each section in the study area was around 0.42, which was close to the high risk level, indicating high landscape ecological risk level. The area of middle and low risk zones had decreased, while the area of high risk zone had significantly increased. The area of high risk zone in the western and middle sections was much higher than that in the eastern section. The area with significant changes of landscape ecological risk accounted for about 55% of the total study area, with obvious spatial agglomeration characteristics of significant increase and decrease of risk. The competition between government-led ecological management policies and measures and market-led land use activities was the main cause of landscape ecological risk variations in this region. In the future, the driving mechanism of climate change coupled with human activities on global and local landscape ecological risk changes in the study area should be uncovered to effectively cope with regional ecological risks.

Objective: To investigate the association between muscle mass and quality of life in adults in Shaanxi adults. Methods: The data in this analysis were part of the baseline survey of the Regional Ethnic Cohort Study in Northwest China from June 2018 to May 2019 in Shaanxi Province. The participants' quality of life, including physical component summary (PCS) and mental component summary (MCS), was assessed by the 12-Item Short Form Survey, and the Body Fat Determination System measured muscle mass. A logistic regression model with adjustment for confounding factors was established to analyze the association between muscle mass and quality of life in different genders. Further, sensitivity and subgroup analyses were conducted to explore its stability. Finally, a restricted cubic spline was employed to investigate the dose-response relationship between muscle mass and quality of life in different genders. Results: A total of 20 595 participants were included, with an average age of 55.0, and 33.4% were male. After controlling for potential confounders, compared with the Q1 group, the risk of low PCS was reduced by 20.6% (OR=0.794, 95%CI: 0.681-0.925) and the risk of low MCS was lower reduced by 20.1% (OR=0.799, 95%CI: 0.689-0.926) in female Q5 groups. Compared with the Q1 group, the risk of low PCS was reduced by 24.4% (OR=0.756, 95%CI: 0.644-0.888) in the male Q2 group. However, no significant association between muscle mass and MCS in males has been found. In females, restricted cubic spline analysis showed a significant linear dose-response relationship between muscle mass and PCS and MCS. Conclusions: There is a positive association between muscle mass and quality of life in Shaanxi adults, especially females. With the increase in muscle mass, the physical and mental functions of the population continue to improve.

@#Objective To investigate the effect of repetitive transcranial magnetic stimulation (rTMS) combined with eye-acupuncture on cognitive dysfunction on cognitive dysfunction in patients with traumatic brain injury. Methods From May, 2014 to June, 2016, 90 trau-matic brain injury patients with cognitive impairment were randomly divided into eye-acupuncture group (n=30), rTMS group (n=30) and combination group (n=30), who accepted eye-acupuncture, rTMS and combination of eye-acupuncture and rTMS, repectively, for eight weeks. They were assessed with Loewenstein Occupational Therapy Cognitive Assessment (LOTCA) before and after treatment. Results There was no significant difference in scores of LOTCA among three groups before treatment (F<0.523, P>0.05). The scores of LOTCA were the best in the combination group after treatment (F>3.789, P<0.05). Conclusion The combination of eye-acupuncture and rTMS is more effective in improving cognitive function after traumatic brain injury than either alone.

Objective To observe any effect when eye acupuncture is combined with computer-assisted cognition training to ameliorate cognitive dysfunction after stroke.Methods Stroke patients with cognitive impairment were randomly divided into an eye acupuncture group (n=30),a computer group (n=30) and a combination group (n=30).The eye acupuncture group was given eye acupuncture,the computer group was given rehabilitative cognition training with specialized equipment and the combination group was given both eye acupuncture and the computeraided training.The treatment lasted 2 months.The Loewenstein occupational therapy cognitive asessment (LOTCA)and the modified Barthel index were used to evaluate the patients' cognitive function and daily life ability before and after treatment.Results Before training there was no significant difference in average LOTCA and MBI results among the three groups.After 2 months of training,all three groups had significantly higher scores in both evaluations.Moreover,the combination group's average score on organization ability,perceptual ability,thinking operation ability and concentration,and also their average total score and MBI score were significantly better than those of the other two groups.Conclusions Eye acupuncture combines synergistically with cognitive rehabilitation training to ameliorate cognitive dysfunction after stroke.Together they promote cognition,ADL ability and an early return to normal family and social life better than either alone.

ObjectiveTo explore the relationship between serum markers and liver fat content predicted by hepatorenal ratio in type 2 diabetic mellitus (T2DM) patients.Methods A total of 120 T2DM patients were recruited from January 2011 to September 2012 in the Affi liated Hospital of Qingdao University. The sagittal liver-right kidney diagrams of the patients were obtained by two-dimensional ultrasonic examination, and the hepatorenal ratio were analyzed by NIHimage. Then, the related serum markers were tested 24 hours later. The relationship between hepatorenal ratio and serum markers was analyzed by Spearman rank correlation on the patients of T2DM. Then, the affective factors on the hepatorenal ratio were analysed by multivariate linear regression analysis, with hepatorenal ratio as dependent variable, and the concentration of triglyceride (TG,X1) , total cholesterol (TC,X2), high density lipoprotein cholesterol (HDL-C,X3), low density lipoprotein cholesterol (LDL-C,X4), alanine aminotransferase (ALT,X5), aspartateaminotransferase (AST,X6), glutamyltransferase (γ-GT,X7) , and alkaline phosphatase (ALP,X8) as independent variables. The receiver operating characteristic curve (ROC) was drawn to diagnose the point of the hepatorenal ratio when the TG began to increase (>1.92 mmol/L).Results There was positive correlation between hepatorenal ratio and TG, AST, ALT,γ-GT (r=0.420,P=0.000;r=0.383,P=0.000;r=0.309,P=0.001;r=0.253,P=0.005), while no correlation between hepatorenal ratio and glycosylated hemoglobin, TC, HDL-C, LDL-C, ALP or blood uric acid (BUA) (r=0.0.067,P=0.368;r=0.145,P=0.115;r=?-0.148,P=0.106;r=0.002,P=0.986;r=0.160,P=0.081;r=0.064,P=0.489) were found; the linear regression analysis showed that TG level (X1) was the only markers which had correlation with hepatorenal ratio in the T2MD patients with the regression equation ofY=1.245+0.062X. The ROC curve analysis showed that the optimal cutoff value for hepatorenal ratio to in diagnosinge increased TG (>1.92 mmol/L) was 1.236 in T2DM patients, and the area under the curve was 0.677. The sensitivity and specifi city were 86.7% and 45.3% respectively. Conclusions Hepatorenal ratio can be a reliable indicator to predict liver fat content, which has correlation with TG, ALT, AST andγ-GT. The increased TG level can refl ect the increasing fat content in the liver, and TG begin to increase when the hepatorenal ratio reach 1.236.

Objective To observe the effect of core stability training combined with eye acupuncture on motor function in patients with hemiplegia after stroke.Methods Eighty patients with hemiplegia after stroke were randomly divided into a treatment group and a control group (40 cases in each).Both groups were given regular rehabilitation training, but the patients in the treatment group also were taught core stability training of their waist and abdominal muscle groups, their shoulder muscles, trunk and pelvis.They were also given eye acupuncture.A trunk control test (TCT) , Berg's balance scale (BBS), the modified Barthel index (MBI), functional ambulation categories (FACs) and the Fugl Meyer assessment scale (FMA) were used to assess motor function before and after treatment.Results At the end of 6 weeks of treatment, all of the measurements in both groups had significantly improved.The average TCT, BBS, MBI, FMA, and FAC scores of the treatment group significantly exceeded those of the control group.Conclusions Core stability training and eye acupuncture together can significantly improve the motor function of hemiplegic stroke survivors.The combination therapy is worthy of clinical popularization and application.

<p><b>OBJECTIVE</b>To examine the temporal and spatial expression of vascular endothelial growth factor (VEGF) and angiopoietins (Ang) in rat brain after cerebral ischemia, and to elucidate the roles they played in angiogenesis and vascular permeability.</p><p><b>METHODS</b>Rats were subjected to either middle cerebral artery occlusion (MCAO) or sham operation. Reverse transcriptase-polymerase chain reaction, Western blotting, and immunohistochemistry were used to detect the expression of VEGF, Ang-1 and Ang-2 at different time points after ischemia. CD31 was used to label endothelial cells after MCAO. Vascular permeability was determined by Evans blue.</p><p><b>RESULTS</b>VEGF was markedly increased at 2 h, had an initial peak at 12 h (0.7249 ± 0.1933, P < 0.01), and a second peak at 7 days (0.5264 ± 0.1519, P < 0.01). Ang-2 mRNA and protein significantly increased after MCAO, both of them peaked at 12 h (0.6747 ± 0.2416, P < 0.01; 1.1197 ± 0.1780, P < 0.01). In contrast, Ang-1 mRNA and protein gradually decreased after MCAO, respectively reaching a minimum at 3 d (0.3220 ± 0.1427, P < 0.01) and 1 d (0.1298 ± 0.0293, P < 0.01). Changes in the expression of these factors correlated with the progress of angiogenesis and vascular permeability. Evans blue test revealed that the vascular permeability gradually increased, and peaked at day 1 after ischemia [(6.219 ± 0.887) µg/g, P < 0.01].</p><p><b>CONCLUSION</b>Dynamic temporal changes in VEGF, Ang-1 and Ang-2 expression stimulate the cerebral angiogenesis after focal cerebral ischemia.</p>
