RESUMO

Functional near-infrared spectroscopy (fNIRS) can dynamically respond to the relevant state of brain activity based on the hemodynamic information of brain tissue. The cerebral cortex and gray matter are the main regions reflecting brain activity. As they are far from the scalp surface, the accuracy of brain activity detection will be significantly affected by a series of physiological activities. In this paper, an effective algorithm for extracting brain activity information is designed based on the measurement method of dual detectors so as to obtain real brain activity information. The principle of this algorithm is to take the measurement results of short-distance channels as reference signals to eliminate the physiological interference information in the measurement results of long-distance channels. In this paper, the performance of the proposed method is tested using both simulated and measured signals and compared with the extraction results of EEMD-RLS, RLS and fast-ICA, and their extraction effects are quantified by correlation coefficient (R), root-mean-square error (RMSE), and mean absolute error (MAE). The test results show that even under low SNR conditions, the proposed method can still effectively suppress physiological interference and improve the detection accuracy of brain activity signals.

Human DNA topoisomerase 1 (Top1) is a crucial enzyme responsible for alleviating torsional stress on DNA during transcription and replication, thereby maintaining genome stability. Previous researches had found that non-working Top1 interacted extensively with chromosomal DNA in human cells. However, the reason for its retention on chromosomal DNA remained unclear. In this study, we discovered a close association between Top1 and chromosomal DNA, specifically linked to the presence of G-quadruplex (G4) structures. G4 structures, formed during transcription, trap Top1 and hinder its ability to relax neighboring DNAs. Disruption of the Top1-G4 interaction using G4 ligand relieved the inhibitory effect of G4 on Top1 activity, resulting in a further reduction of R-loop levels in cells. Additionally, the activation of Top1 through the use of a G4 ligand enhanced the toxicity of Top1 inhibitors towards cancer cells. Our study uncovers a negative regulation mechanism of human Top1 and highlights a novel pathway for activating Top1.

Lethal lipid peroxidation caused by reactive oxygen species occurs in different types of programmed cell death, especially in ferroptosis. Ferroptosis inducers, which serve as small-molecule probes, can provide insight into the mechanism of ferroptosis and facilitate drug discovery. The classical ferroptosis inducers indirectly lead to lipid peroxidation; thus, it is difficult to explore lipid regulation during the ferroptotic process. In this study, we designed two quinazolinone-based lipophilic probes BODIQPy-TPA and QPy-TPA, which proved to directly induce lipid peroxidation by light irradiation in vitro. The probe BODIQPy-TPA, which was mainly distributed in the endoplasmic reticulum (ER), specifically triggered ferroptosis in B16 and HepG2 cells upon light irradiation. As a comparison, the probe QPy-TPA, which was mainly distributed in lipid droplets (LDs), induced cell death by a nonferroptotic pathway. Further lipidomic analysis revealed that these two probes caused different patterns of lipid regulation and lipid peroxidation, suggesting that ferroptosis might activate distinct lipid regulation.

In recent years, research on emotion recognition has become more and more popular, but there are few studies on emotion recognition based on cerebral blood oxygen signals. Since the electroencephalogram (EEG) is easily disturbed by eye movement and the portability is not high, this study uses a more comfortable and convenient functional near-infrared spectroscopy (fNIRS) system to record brain signals from participants while watching three different types of video clips. During the experiment, the changes in cerebral blood oxygen concentration in the 8 channels of the prefrontal cortex of the brain were collected and analyzed. We processed and divided the collected cerebral blood oxygen data, and used multiple classifiers to realize the identification of the three emotional states of joy, neutrality, and sadness. Since the classification accuracy of the convolutional neural network (CNN) in this research is not significantly superior to that of the XGBoost algorithm, this paper proposes a CNN-Transformer network based on the characteristics of time series data to improve the classification accuracy of ternary emotions. The network first uses convolution operations to extract channel features from multi-channel time series, then the features and the output information of the fully connected layer are input to the Transformer netork structure, and its multi-head attention mechanism is used to focus on different channel domain information, which has better spatiality. The experimental results show that the CNN-Transformer network can achieve 86.7% classification accuracy for ternary emotions, which is about 5% higher than the accuracy of CNN, and this provides some help for other research in the field of emotion recognition based on time series data such as fNIRS.

Small-molecule-based fluorescent chemosensors provide powerful tools for analytical chemistry. However, their organic essence often "cursed" them for aggregation-caused quenching (ACQ) in an aqueous context. Albeit the praxis of the disaggregation-induced emission (DIE) strategy as a potential solution, it still awaited improvement due to the uncontrollability of the aggregation/segregation process. To address this issue, herein, we supposed that sulfur substitution on a molecule could serve as a promising strategy to achieve an evolved ACQ-DIE probe. To prove this concept, a precursor G-quadruplex (G4) ligand CQ was modified to get its thionation version CTQ. Strikingly, CTQ exhibited more arranged aqueous segregation behavior, as compared with CQ, and therefore enhanced fluorescence performance. Our research, in the meantime, manifested that CTQ remained to possess favorable G4 selectivity, whereby it could function as an evolved probe for more accurate in vitro G4-related assays, specifically, a classification assay for distinguishing virus variants.

RESUMO

An extramedullary plasmacytoma (EMP) is a rare neoplasm characterized by monoclonal proliferation of plasma cells without features of multiple myeloma. Most EMPs occur in the head and neck region, especially in the aerodigestive tract. We herein report a case of an EMP arising from the nasal inferior turbinate. The mass was surgically removed, and a short cycle of radiotherapy was performed after the surgery. There was no recurrence of the tumor after 1 year of follow-up. These result may be useful for physicians who encounter similar situations in clinical practice.

Pleurotus tuoliensis is a popular edible and medical mushroom, but it is highly perishable during postharvest storage. The quality parameters, chemical composition, malondialdehyde (MDA) concentration, and activity of metabolic enzymes were studied during 12 days of storage at 4 °C and 6 days of storage at 25 °C. Degradation was well described by changes in quality parameters, losses in nutritional value, increased metabolic enzyme activity, the accumulation of MDA concentrations, and the increase of total phenolic (TP) content. The phenylalanine ammonia lyase (PAL) significantly positively correlated with TP, which suggested an underlying mechanism of browning that the increased PAL activity stimulates the biosynthesis of phenols through the phenylalanine pathway. These results suggest that increased activity of laccase, lipoxygenase, PAL, TP and MDA accumulation, together with polysaccharide degradation, are the main factors involved in the deterioration of P. tuoliensis during storage.
