Introduction: The use of controlled-release nitrogen (N) fertilizers has been shown to improve yield and N-use efficiency (NUE) in mechanical transplanted rice. However, the fertilizer requirements for mechanical direct-seeding rice differ from those for mechanical transplanted rice. The effects of controlled-release fertilizers on yield, NUE, and quality in mechanical direct-seeding rice are still unknown. Methods: Hybrid indica rice varieties Yixiangyou 2115 and Fyou 498 were used as test materials, and slow-mixed N fertilizer (120 kg hm-2) as a base (N1), N1+urea-N (30 kg hm-2) once as a base (N2), N1+urea-N (30 kg hm-2) topdressing at the tillering stage (N3), N1+urea-N (30 kg hm-2) topdressing at the booting stage (N4) four N fertilizer management to study their impact on the yield, NUE and quality of mechanical direct-seeding rice. Results and discussion: Compared with Yixiangyou 2115, Fyou 498 significantly increased photosynthetic potential, population growth rate, root vigor, and N transport rate by 3.34-23.88%. This increase further resulted in a significant improvement in the yield and NUE of urea-N topdressing by 1.73-5.95 kg kg-1. However, Fyou 498 showed a significant decrease in the head rice rate and taste value by 3.34-7.67%. All varieties were treated with N4 that significantly increase photosynthetic potential and population growth rate by 15.41-62.72%, reduce the decay rate of root vigor by 5.01-21.39%, promote the N transport amount in stem-sheaths (leaves) by 13.54-59.96%, and then significantly increase the yields by 4.45-20.98% and NUE of urea-N topdressing by 5.20-45.56 kg kg-1. Moreover, the rice processing and taste values were optimized using this model. Correlation analysis revealed to achieve synergistic enhancement of high-yield, high-quality, and high-NUE in rice, it is crucial to focus on increasing photosynthetic potential, population growth rate, and promoting leaf N transport. Specifically, increasing the contribution rate of N transport in stem-sheaths is the most important. These findings offer an effective N management strategy for 4R nutrient stewardship (right source, right method, right rate and right timing) of mechanical direct-seeding hybrid indica rice.

With the continuous promotion of "smart cities" worldwide, the approach to be used in combining smart cities with modern advanced technologies (Internet of Things, cloud computing, artificial intelligence) has become a hot topic. However, due to the non-stationary nature of environmental sound and the interference of urban noise, it is challenging to fully extract features from the model with a single input and achieve ideal classification results, even with deep learning methods. To improve the recognition accuracy of ESC (environmental sound classification), we propose a dual-branch residual network (dual-resnet) based on feature fusion. Furthermore, in terms of data pre-processing, a loop-padding method is proposed to patch shorter data, enabling it to obtain more useful information. At the same time, in order to prevent the occurrence of overfitting, we use the time-frequency data enhancement method to expand the dataset. After uniform pre-processing of all the original audio, the dual-branch residual network automatically extracts the frequency domain features of the log-Mel spectrogram and log-spectrogram. Then, the two different audio features are fused to make the representation of the audio features more comprehensive. The experimental results show that compared with other models, the classification accuracy of the UrbanSound8k dataset has been improved to different degrees.

Anemarrhena asphodeloides Bunge (Liliaceae) is an important Traditional Chinese Medicine herb, which contains up to 6 % total steroidal saponins (timosaponins) and has multiple pharmacological properties. However, the timosaponin biosynthetic pathway has not been extensively investigated. Here we conducted de novo transcriptome sequencing and analysis of A. asphodeloides Bunge and screened for candidate genes involved in the timosaponin biosynthetic pathway. Targeted metabolite analysis showed that timosaponins primarily accumulated in rhizomes, while phytosterols (including cholesterol) were distributed throughout various organs. Most of the identified candidate genes of the timosaponin biosynthetic pathway were also highly expressed in the rhizome, consistent with the results of metabolic analysis. Based on the transcriptome results, two candidate 7-dehydrocholesterol reductase genes were cloned and heterologously expressed in the yeast Saccharomyces cerevisiae. The purified and identified products supported that Aa7DR1 possessed Δ7-reduction activity in yeast and therefore may be involved in the timosaponins biosynthetic pathway in A. asphodeloides Bunge. Phylogenetic analysis showed Aa7DR1 belongs to monocotyledonous Δ7 reductase of phytosterol biosynthesis. These data expand our understanding of timosaponin biosynthesis.

Ordered polycyclization catalyzed by oxidosqualene synthases (OSCs) morph a common linear precursor into structurally complex and diverse triterpene scaffolds with varied bioactivities. We identified three OSCs from Iris tectorum. ItOSC2 is a rare multifunctional α-amyrin synthase. Sequence comparisons, site-directed mutagenesis and multiscale simulations revealed that three spatially clustered residues, Y531/L256/L258 form an unusual Y-LL triad at the active site, replacing the highly conserved W-xY triad occurring in other amyrin synthases. The discovery of this unprecedented active site architecture in ItOSC2 underscores the plasticity of terpene cyclase catalytic mechanisms and opens new avenues for protein engineering towards custom designed OSCs.
