RESUMO

Aeromonas dhakensis is reported as an emerging pathogenic species within the genus Aeromonas and is widely distributed in tropical coastal areas. This study provided a detailed description and characterization of a strain of A. dhakensis (202108B1) isolated from diseased Ancherythroculter nigrocauda in an inland region of China. Biochemical tests identified the isolate at the genus level, and the further molecular analysis of concatenated housekeeping gene sequences revealed that the strain belonged to the species A. dhakensis. The isolated A. dhakensis strain was resistant to five antibiotics, namely, penicillin, ampicillin, clindamycin, cephalexin and imipenem, while it was susceptible or showed intermediate resistance to most of the other fifteen tested antibiotics. The isolated strain of A. dhakensis caused acute haemorrhagic septicaemia and tissue damage in artificially infected A. nigrocauda, with a median lethal dose of 7.76×104 CFU/fish. The genome size of strain 202108B1 was 5043286 bp, including one chromosome and four plasmids. This is the first detailed report of the occurrence of infection caused by an A. dhakensis strain causing infection in an aquaculture system in inland China, providing important epidemiological data on this potential pathogenic species.

The area around Poyang Lake is the main aquaculture area in Jiangxi Province, China, and an important base for the supply of freshwater aquatic products. Aquaculture in the Poyang Lake area is severely threatened by diseases caused by bacterial pathogens, and Aeromonas veronii has been the main pathogen in recent years. In this paper, ERIC-PCR genotyping, virulence gene and antimicrobial resistance gene detection, and drug susceptibility tests were carried out on 46 A. veronii isolates obtained from aquaculture systems in the Poyang Lake area from 2016 to 2020. The results showed that the A. veronii strains in the Poyang Lake area had high genetic diversity, and 46 strains produced 36 ERIC genotypes. There were no geographical and temporal differences in the cluster analysis results and no dominant clones. All 13 virulence genes tested were detected, and all isolates had harbored 2 or more virulence genes, with a maximum of 12 virulence genes detected. Among the 22 antimicrobial resistance genes selected, 15 were detected; 97.8% of the isolates contained 2 or more antimicrobial resistance genes, with a maximum of 9 antimicrobial resistance genes. Drug susceptibility tests showed that some strains were resistant to a variety of traditionally effective drugs for Aeromomas, such as enrofloxacin and florfenicol. This study provides a reference for exploring the impact of aquaculture in the Poyang Lake area on public health.

Materials with excellent high-temperature strength are now sought for applications in hypersonics, fusion reactors, and aerospace technologies. Conventional alloys and eutectic multiprincipal-element alloys (MPEAs) exhibit insufficient strengths at high temperatures due to low melting points and microstructural instabilities. Here, we report a strategy to achieve exceptional high-temperature microstructural stability and strength by introducing eutectic carbide in a refractory MPEA. The synergistic strengthening effects from the multiprincipal-element mixing and strong dislocation blocking at the interwoven metal-carbide interface make the eutectic MPEA not only have outstanding high-temperature strength (>2 GPa at 1473 K) but also alleviate the room-temperature brittleness through microcrack tip blunting by layered metallic phase. This strategy offers a paradigm for the design of the next-generation high-temperature materials to bypass the low-melting point limitation of eutectic alloys and diffusion-dominated softening in conventional superalloys.

Multi-principal element alloys (MPEAs) exhibit outstanding mechanical properties because the core effect of severe atomic lattice distortion is distinctly different from that of traditional alloys. However, at the mesoscopic scale the underlying physics for the abundant dislocation activities responsible for strength-ductility synergy has not been uncovered. While the Eshelby mean-field approaches become insufficient to tackle yielding and plasticity in severely distorted crystalline solids, here we develop a three-dimensional discrete dislocation dynamics simulation approach by taking into account the experimentally measured lattice strain field from a model FeCoCrNiMn MPEA to explore the heterogeneous strain-induced strengthening mechanisms. Our results reveal that the heterogeneous lattice strain causes unusual dislocation behaviors (i.e., multiple kinks/jogs and bidirectional cross slips), resulting in the strengthening mechanisms that underpin the strength-ductility synergy. The outcome of our research sheds important insights into the design of strong yet ductile distorted crystalline solids, such as high-entropy alloys and high-entropy ceramics.

We have studied the charge to spin conversion in Bi1-x Sb x /CoFeB heterostructures. The spin Hall conductivity (SHC) of the sputter-deposited heterostructures exhibits a high plateau at Bi-rich compositions, corresponding to the topological insulator phase, followed by a decrease of SHC for Sb-richer alloys, in agreement with the calculated intrinsic spin Hall effect of Bi1-x Sb x . The SHC increases with increasing Bi1-x Sb x thickness before it saturates, indicating that it is the bulk of the alloy that predominantly contributes to the generation of spin current; the topological surface states, if present, play little role. Unexpectedly, the SHC is found to increase with increasing temperature, following the trend of carrier density. These results suggest that the large SHC at room temperature, with a spin Hall efficiency exceeding 1 and an extremely large spin current mobility, is due to increased number of thermally excited Dirac-like electrons in the L valley of the narrow gap Bi1-x Sb x alloy.

The iron and steel industry discharges large quantities of wastewater. The environmental impact of the wastewater is traditionally assessed from the quantitative aspect. However, the water quality of discharged wastewater plays a more significant role in damaging the natural environment. Moreover, comprehensive assessment of multi-pollutants in wastewater from both quality and quantity is still a gap. In this work, a total environmental impact score (TEIS) is defined to assess the environmental impact of wastewater discharge, by considering the volume of wastewater and the quality of main processes. To implement the comprehensively qualitative and quantitative assessment, a field monitoring and measurement of wastewater discharge volume and the quality is conducted to acquire pH, suspend solids (SS), chemical oxygen demand (COD), total nitrogen (TN), total iron (TFe), and hexavalent chromium (Cr(VI)). The sequence of TEIS values is obtained as steelmaking > ironmaking > sintering > hot rolling > coking > cold rolling and TN > COD > SS > pH > Cr(VI) > TFe. The TEIS of the investigated steel plant is 26.27. The leading process lies in steelmaking with a TEIS of 19.98. The dominant pollutant is TN with a TEIS of 15.00. Finally, a sensitivity analysis is performed to validate the feasibility and generalisability of the TEIS.

Voltage-controlled magnetic anisotropy (VCMA) in an epitaxially grown Fe/Fe1-xCox/Pd/MgO system was investigated using spin-wave spectroscopy. The spin-wave resonant frequency linearly depended on the bias-voltage. The resonant-frequency shift increased with the Co fraction in Fe1-xCox/Pd. We achieved a VCMA of approximately 250 fJ/Vm at the Co/Pd/MgO region.

RESUMO

Various disturbances do exist in the image taking process of scanning transmission electron microscopes (STEM), which seriously reduces the resolution and accuracy of STEM images. In this paper, a deep understanding of the scanning distortion influence on the real and reciprocal spaces of STEM images is achieved via theoretical modeling and simulation. A scanning distortion correction algorithm is further developed based on two images scanned along perpendicular directions, which is able to effectively correct scanning distortion induced deviations and significantly increase the signal to noise ratio of STEM images.

Previous studies have shown that intensive macropinocytosis occurs in cancer cells and neutral red (NR) is noted for its capability to enter into the cell massively through a process mimetic to macropinocytosis. In addition, trans-cinnamic acid (tCA) has been found to be an inhibitor of histone deacetylase (HDAC). In the present study, cinnamoylphenazine (CA-PZ) that consists of NR and tCA moieties was synthesized and evaluated. As shown, CA-PZ massively entered into colon carcinoma HT-29 cells and pancreatic carcinoma MIA PaCa-2 cells and this entry was blocked by 5-(N-ethyl-N-isopropyl) amiloride (EIPA, an inhibitor of macropinocytosis), indicating a macropinocytosis-mediated uptake. Furthermore, CA-PZ markedly increased the protein expression levels of acetyl-H3, acetyl-H4 and p21 in HT-29 cells and MIA PaCa-2 cells. CA-PZ significantly inhibited the growth of colon carcinoma HT-29 and pancreatic carcinoma MIA PaCa-2 xenografts. By in vivo imaging, CA-PZ displayed prominent accumulation in the tumor xenografts. The study indicates that the newly synthesized CA-PZ acts as an HDAC inhibitor in association with intensive macropinocytosis-mediated intracellular delivery in cancer cells. The use of neutral red for preparation of chimeric molecules with the attribute of macropinocytosis-mediated intracellular delivery might open an alternative way for development of HDAC inhibitors.

RESUMO

Nanoporous graphene- based electric-double-layer transistors (EDLTs) are successfully fabricated. Transport measurements of the EDLTs demonstrate that the ambipolar electronic states of massless Dirac fermions with a high carrier mobility are well preserved in 3D nanoporous graphene along with anomalous nonlinear Hall resistance and exceptional transistor on/off ratio. This study may open a new avenue for device applications of graphene.

Multidrug resistance is a major limitation for microtubule-binding agents in cancer treatment. Here we report a novel microtubule inhibitor (2-morpholin-4-yl-5-nitro-benzoic acid 4-methylsulfanyl-benzyl ester, IMB5046), its cytotoxicity against multidrug-resistant cell lines and its antitumor efficacy in animal models. IMB5046 disrupted microtubule structures in cells and inhibited purified tubulin polymerization in vitro. It bound to the colchicine pocket of tubulin. IMB5046 displayed potent cytotoxicity against multiple tumor cell lines with an IC50 range of 0.037-0.426 µM. Notably, several multidrug-resistant cell lines which were resistant to colchicine, vincristine and paclitaxel remained sensitive to IMB5046. IMB5046 was not a P-glycoprotein substrate. IMB5046 blocked cell cycle at G2/M phase and induced cell apoptosis. Microarray assay indicated that the differentially expressed genes after IMB5046 treatment were highly related to immune system, cell death and cancer. In a mouse xenograft model IMB5046 inhibited the growth of human lung tumor xenograft by 83% at a well-tolerated dose. It is concluded that IMB5046 is a tubulin polymerization inhibitor with novel chemical structure and can overcome multidrug resistance. It is a promising lead compound for cancer chemotherapy, especially for treatment of multidrug-resistant tumors.

There have been numerous efforts to develop creep-resistant materials strengthened by incoherent particles at high temperatures and stresses in response to future energy needs for steam turbines in thermal-power plants. However, the microstructural instability of the incoherent-particle-strengthened ferritic steels limits their application to temperatures below 900 K. Here, we report a novel ferritic alloy with the excellent creep resistance enhanced by coherent hierarchical precipitates, using the integrated experimental (transmission-electron microscopy/scanning-transmission-electron microscopy, in-situ neutron diffraction, and atom-probe tomography) and theoretical (crystal-plasticity finite-element modeling) approaches. This alloy is strengthened by nano-scaled L21-Ni2TiAl (Heusler phase)-based precipitates, which themselves contain coherent nano-scaled B2 zones. These coherent hierarchical precipitates are uniformly distributed within the Fe matrix. Our hierarchical structure material exhibits the superior creep resistance at 973 K in terms of the minimal creep rate, which is four orders of magnitude lower than that of conventional ferritic steels. These results provide a new alloy-design strategy using the novel concept of hierarchical precipitates and the fundamental science for developing creep-resistant ferritic alloys. The present research will broaden the applications of ferritic alloys to higher temperatures.

Elizabethkingia meningoseptica has been recognised as an occasional but serious opportunistic bacterial pathogen to human beings. Recently, it was frequently isolated from tiger frog, Rana tigerina rugulosa, with cataract disease, which is the most common disease of unknown aetiology of frogs in Hainan, China. The purpose of this study was to identify and characterise the bacterial strains isolated from the recent outbreaks of cataract disease in farmed tiger frog in Hainan, China, and to evaluate their pathogenicity to the frog and their sensitivity to 20 chemotherapeutic agents. The 16S rRNA gene sequences of strains W0701 (1478bp), W0702 (1477bp) and W0703 (1478bp) showed 98.6-98.7% similarity with the sequence of E. meningoseptica type strain (ATCC 13253) and 99.9-100% similarity with that of E. meningoseptica NTU 870424-IL. Six strains (W0701-W0706) were selected to represent 24 isolates retrieved from six moribund frogs. The morphological, physiological and biochemical characteristics of the six representative isolates were consistent with those of E. meningoseptica strains. The organisms were only susceptible to vancomycin and moderately susceptible to cefoperazone among the 20 investigated chemotherapeutic agents. Virulence test with strain W0702 was conducted and pathogenicity (by intramuscular injection) was demonstrated in the tiger frog. In conclusion, 24 isolates obtained from frogs with cataract disease were the E. meningoseptica strains highly pathogenic to tiger frog, and this is the first report of E. meningoseptica as a pathogen for tiger frog.

The control arithmetic is proposed for axial blood pump driven by extracorporeal alternating magnet field based on ventricular work. According to health physiological parameters, the control model is verified by calculation and experiment. The control model used for driving blood pump is derived from the relationship of natural heart and artificial heart. By comparison with others, the control arithmetic based on ventricular work is more helpful in appraising the property of left ventricular aided device (LVAD), in meeting human body's natural need, and in effective use of motor for avoiding motor running at high speed and hence preventing the blood from the destruction caused by a blood pump at high temperature.

BACKGROUND & OBJECTIVE: Asiaticoside (ATS), a triterpene extracted from Centella asiatica (L.) Urban, a traditional Chinese herb, possesses good wound healing activities because of its stimulative effect on collagen synthesis. Recently, the anti-tumor effect of asiaticiside has been reported. This study was to examine the induction of apoptosis in cancer cells, and the enhancement of vincristine (VCR) cytotoxicity by asiaticoside. METHODS: MTT assay was used to evaluate inhibitory effect of asiaticoside combined with vincristine on proliferation of several cancer cell lines, including KB, KBv200, MCF-7, and MCF-7/ADM. Cell cycle, and apoptosis of KB cells were analyzed by flow cytometry; apoptosis induction was also proved by electrophoresis,and morphologic assessment; the expression of apoptosis-, and cell cycle-related proteins were determined by Western blot. RESULTS: The IC(50) values of asiaticoside for KB, KBv200, MCF-7, and MCF-7/ADM cells detected by MTT assay were (1.11+/-0.13) mg/ml, (1.82+/-0.08) mg/ml, (1.58+/-0.15) mg/ml, and (3.25+/-0.46) mg/ml, respectively. Multidrug resistant KBv200, and MCF-7/ADM cancer cells displayed similar sensitivity to asiaticoside as their parental counterparts (KB, and MCF-7 cells). Moreover, asiaticoside induced apoptosis in KB cells. At sub-cytotoxicity concentration, asiaticoside showed synergistic effect with vincristine in these 4 cell lines. The apoptosis rates were much higher in asiaticoside plus vincristine groups than in vincristine or asiaticoside groups. Bcl-2 phosphorylation levels were higher in the combination groups than in vincristine or asiaticoside alone groups. The activated caspase-3 protein was only presented in the combination groups. Asiaticoside plus vincristine enhanced S-G(2)/M arrest, up-regulated Cyclin B1 protein expression, and down-regulated P34(cdc2) protein expression in KB cells. CONCLUSION: Asiaticoside, as a biochemical modulator, may induce apoptosis,and enhance anti-tumor activity of vincristine in cancer cells, might be useful in cancer chemotherapy.
