RESUMO

Epigenetic methylation has been shown to play an important role in transcriptional regulation and disease pathogenesis. Recent advancements in detection techniques have identified DNA N6-methyldeoxyadenosine (6mA) and RNA N6-methyladenosine (m6A) as methylation modifications at the sixth position of adenine in DNA and RNA, respectively. While the distributions and functions of 6mA and m6A have been extensively studied in prokaryotes, their roles in the mammalian brain, where they are enriched, are still not fully understood. In this review, we provide a comprehensive summary of the current research progress on 6mA and m6A, as well as their associated writers, erasers, and readers at both DNA and RNA levels. Specifically, we focus on the potential roles of 6mA and m6A in the fundamental biological pathways of the mammalian genome and highlight the significant regulatory functions of 6mA in neurodegenerative diseases.

Because of the difficulty in collecting fresh brains of humans at different ages, it remains unknown how epigenetic regulation occurs in the primate brains during aging. In the present study, we examined the genomic distribution of 5hmC, an indicator of DNA methylation, in the brain regions of non-human primates (rhesus monkey) at the ages of 2 (juvenile), 8 (young adult), and 17 (old) years. We found that genomic 5hmC distribution was accumulated in the monkey brain as age increased and displayed unique patterns in the cerebellum and striatum in an age-dependent manner. We also observed a correlation between differentially hydroxymethylated regions (DhMRs) and genes that contribute to brain region-related functions and diseases. Our studies revealed, for the first time, the brain-region and age-dependent 5hmC modifications in the non-human primate and the association of these 5hmC modifications with brain region-specific function and potentially aging-related brain diseases.

RESUMO

Nanocellulose is a type of nanomaterial with high strength, high specific surface area and high surface energy. Additionally, it is nontoxic, harmless, biocompatible and environmentally friendly and can be extracted from biomass resources. The surface groups of cellulose show high surface energy and binding activity on the nanoscale and can be modified by using various methods. Because nanocellulose has a high elastic modulus, rigidity and a low thermal expansion coefficient, it is an excellent material for polymer reinforcement. This paper summarizes the reinforcement mechanisms of nanocellulose polymer composites with a focus on the role of theoretical models in elucidating these mechanisms. Furthermore, the influence of various factors on the properties of nanocellulose reinforced polymer composites are discussed in combination with analyses and comparisons of specific research results in related fields. Finally, research focus and development directions for the design of high-performance nanocellulose reinforced polymer composites are proposed.

Carcinoembryonic antigen-related cell adhesion molecule 5 (CEACAM-5) assays are employed in routine clinical settings to diagnose tumor. We selected two nanobodies with high-affinity to CEACAM-5, termed Nb11C12 and Nb2D5, using phage-display technology. The Nb2D5 fused with calf intestinal alkaline phosphatase (CAP), human placental alkaline phosphatase (HAP), or Pyrococcus abyssi alkaline phosphatase (PAP) were expressed in human embryonic kidney (HEK293) cells. The enzymatic activity of Nb2D5-HAP fusion protein was the best and remained stable at 60 °C for 7 days. The affinity of Nb2D5-HAP fusion protein to CEACAM-5 reached 42 pM. A chemiluminescent enzyme immunoassay (CLEIA) based on Nb2D5-HAP fusion protein was established for quantitative CEACAM-5 assay in clinical settings. The CLEIA exhibited a wide linear range of 0.31-640 ng/mL toward CEACAM-5, with a limit of detection (LOD) of 0.85 ng/mL. No cross-reactivity occurred with CEACAM-1, CEACAM-3, CEACAM-6, or CEACAM-8, and no interference was observed with rheumatoid factors. The CLEIA based on Nb2D5-HAP fusion protein was stable for 8 weeks at 37 °C and 50% relative humidity. The CLEIA developed from Nb2D5-HAP fusion protein had much better stability and linearity with similar reproducibility compared with the enzyme-linked immunosorbent assay developed from conventional monoclonal antibodies, which have been widely used in clinics over the past several decades. A high monosaccharide conversion rate of hemicellulose in a green solvent and under moderate reaction conditions for industrialization is one of the most important keys in a lignocellulosic biorefinery. The behavior of Populus tomentosa hemicellulose polysaccharides, crystallinity and the furfural formation in the autohydrolysis process under moderate subcritical water conditions (160-180 °C, 0.618-1.002 MPa) were studied. The results have shown that the hemicellulose was converted to corresponding monosaccharides at an ultra-high hydrolysis rate. Factor analysis indicates that the temperature is the most important factor affecting hemicellulose autohydrolysis. When the autohydrolysis temperature increased from 160 to 180 °C for 2 h, the hydrolysis rate of xylose, rhamnose, galactose, mannose, and glucose from hemicellulose increased from 70% to 91%, 71% to 100%, 82% to 95%, 42% to 58%, and 34% to 37%, respectively. Arabinose was completely dissolved in 30 min. The xylose, rhamnose, galactose, and arabinose from hemicellulose could be almost completely removed under the conditions. The hemicellulose removal rate obtained herein exceeded the values reported for most acid, alkali, ionic liquid, or deep eutectic solvent treatments. It is notable that almost all glucose in hemicellulose was dissolved and the glucose in cellulose was partially hydrolyzed. An analysis of the sugar composition and the crystallinity change in the process at 180 °C demonstrate that hydrolysis reaction started to shift from amorphous regions to crystalline regions, due to the partial hydrolysis of crystalline cellulose after 90 min at 180 °C. Overall, these results show that the moderate subcritical water autohydrolysis of hemicellulose in Populus tomentosa may be a potential bio-refinery process.

The plasmon resonances of nanostructures enable wide applications from highly sensitive sensing to high-resolution imaging, through the improvement of photogeneration rate stimulated by the local field enhancement. However, quantitative experimental studies on the localized heating and the thermal transport process in the vicinity of plasmonics are still lacking because of the diffraction limit in conventional optothermal methodologies. In this work, we demonstrate an approach based on Raman thermometry to probe the near-field heating caused by plasmonics. An array of Au nanoparticles (AuNPs) fabricated by the template-assisted method is used to generate the near field effect. Multi-walled carbon nanotubes (MWCNTs) dispersed on the AuNPs are employed to quantify the near-field heating from their Raman peak shifts. Results show that the temperature rise in MWCNTs on AuNPs is much higher than that in a control group under the same laser irradiation. Further analysis indicates that the enhanced photon absorption of MWCNTs attributed to plasmon resonances is partially responsible for the different heating effect. The nonuniform thermal hot spots at the nanoscale can result in the quasi-ballistic thermal transport of phonons in MWCNTs, which is another reason for the temperature rise. Our results can be used to understand plasmonic heating effects as well as to explore quasi-ballistic thermal transport in carbon-based low-dimensional materials by tailoring the geometry or size of plasmonic nanostructures.

The synthesis process of the phenolic resin adhesive was developed in the 19th century, and its excellent environmental resistance and high bonding strength make it one of the main wood adhesives. With the development of industry, phenolic resin adhesive is not only used in plywood, wood processing and laminate, but also in automobile, aerospace, composite materials and other fields. Herein we review the main synthetic processes and latest research progress for phenolic resin adhesives, the capacity distribution of major domestic phenolic resin enterprises, analysis of domestic phenolic resin consumption, import and export volume and price, Chinese plywood production, and the main production and export markets. Furthermore, research and discussion on the commercial application of domestic phenolic resin adhesives in plywood were carried out, which provides a reference for development of better phenolic resin adhesives that meet the development needs of the country, along with quality and economic competitiveness.
