RESUMO

Objective: To study the inhibitory effect of iridoid glycosides from Boschniakia rossica (IGBR) combined with 5-Fu on epithelial-mesenchymal transition induced by TGF-β1 in human hepatoma SK-Hep1 and HepG2 cells, and compare the efficacy of drugs. Methods: The survival ability of HepG2 and SK-Hep1 cells was detected by MTT and the combination index (Q value) was calculated to judge the interaction of combined drugs. The EMT model of HepG2 and SK-Hep1 cells was established. The cell adhesion rate was detected by MTT. The expression of matrix metalloproteinase (MMP) MMP2, MMP7, MMP9, Snail, and Slug was detected by Western blotting. The localization and expression intensity of E-cadherin and Vimentin was detected by immunofluorescence. Results: MTT showed that compared with the control group, the 5-FU group, IGBR group and combination group cell survival ability were decreased (P < 0.05) at 48 h after administration; IGBR and 5-Fu had an additive or synergistic effect. Compared with the model group, the adhesion rate of 5-FU group, IGBR group and combination group was reduced (P < 0.05). Western blotting results showed that compared with the control group, the expression of MMP2, MMP7, MMP9, Snail, Slug were up-regulated (P < 0.05) in the model group. Compared with the model group, the expression of MMP2, MMP7, MMP9, Snail and Slug were down-regulated (P < 0.05) in 5-FU group, IGBR group and combination group. Compared with the control group, immunofluorescence showed that the E-cadherin fluorescence intensity was decreased in the model group, but the Vimentin fluorescence intensity was increased. Compared with the model group, the E-cadherin fluorescence intensity was increased in 5-FU group, IGBR group and combined group, but the Vimentin fluorescence intensity was decreased. Conclusion: IGBR and 5-Fu can inhibit human hepatoma EMT. The combined drugs have the combined effect on HepG2 cells and synergistic effect on SK-Hep1 cells. The therapeutic effect on SK-Hep1 cells is better than HepG2 cells.

Dichromate is a widespread contaminant in wastewater, threatening the health of humans and other organisms. Therefore, effective detection and removal of dichromate from water is of great significance. Herein, a tetraphenylethylene functionalized cationic organic network (CON-LDU2) was constructed via a facile quaternization reaction. CON-LDU2 was successfully integrated with both detection and removal functionalities toward dichromate. On the one hand, benefiting from the strong fluorescence, CON-LDU2 was employed as a chemosensor, it could efficiently and selectively probe Cr2O72- in water with "turn-off" fluorescent response. On the other hand, the cationic skeleton and free anions inside framework make CON-LDU2 an excellent adsorbent for Cr2O72-, it could capture Cr2O72- from water with rapid kinetics and high capacity. The kinetic constant for adsorption of Cr2O72- can reach up to 1.784 g mg-1 min-1, while the capacity is determined as 325 mg g-1. Furthermore, CON-LDU2 displayed good recyclability and can be reused for at least 5 cycles. Therefore, CON-LDU2 can serve as an ideal candidate not only in detection but also in removal of Cr2O72- in water medium.

RESUMO

We demonstrate herein a facile approach for constructing -C[double bond, length as m-dash]N- linked COFs from acetals. Three new COFs (imine-linked LZU-20, hydrazone-linked LZU-21, and azine-linked LZU-22) were synthesized by the direct condensation of dimethyl acetals and amines. All the synthesized COFs are highly crystalline and exhibit good thermal stability.

Objective To discuss the method for defining personalized materials properties of the fresh human long bone with alcohol treatment and the effect from the number of bone materials on finite element results. Methods Based on images from CT scans, a three dimensional solid model of the long bone was established in Mimics, which was then classified into the cortical bone, cancellous bone and marrow in Hypermesh. Based on relevant empirical formulas, material parameters of the cortical bone and cancellous bone were given, respectively, and 5 finite element analysis (FEA) models with different numbers of materials were set up. The simulation for linear elasticity of the compression was carried out in Abaqus and the results were validated by in vitro verification test. Results Under the end displacement ranging from 0 to 1 mm, the average relative error between the simulation results and the experimental data for holistic force-displacement was about 10%, when the materials number was defined as 1 kind of the cancellous bone and over 10 kinds of the cortical bone. And the average relative error between the simulation results and the experimental data for the deformation of the measurement points was 14.6%. The error of holistic force displacement for 1 kind of the cortical bone was 2.83% when the end displacement ranged from 0 to 0.5 mm. Conclusions (1) Using the gray value of CT scans, materials properties of the main component of the bone could be defined accurately. (2) The simulation result was greatly affected by the material number of the cortical bone, and defining 10 kinds of the cortical bone could satisfy the FEA need. (3) The FEA model with 1 kind of the cortical bone also could satisfy the need of analysis under small deformation.
