RESUMO

Methyl Orange, an azo dye, is a widely used colouring agent in the textile industry. The study aimed to investigate the efficiency of bioremediating bacteria in degrading methyl orange. Escherichia coli (E. coli), a Methyl Orange-degrading bacterium, was isolated from cow dung and its biochemical properties were analysed using 16S rRNA sequencing, and MALDI-TOF MS. A pre-cultured strain of Pseudomonas aeruginosa was co-cultured with E. coli in 1:1 ration in a microbial fuel cell (MFC) for simultaneous electricity production and methyl orange degradation. The degradation was combined with biological wastewater treatment at varying Methyl Orange concentrations, and the electrochemical characteristics were analysed through polarisation study, cyclic voltammetry, and electrochemical impedance spectroscopy. The impact of parameters such as anolyte pH, dye concentration, incubation time, and substrate concentrations were also studied. This study confirmed E. coli as an effective methyl orange degrading bacteria with a maximum % degradation efficiency of 98% after 48 h incubation at pH 7.0. The co-culture of isolated microorganisms at 250 mg/L of methyl orange concentration showed a maximum power density 6.5 W/m3. Further, anode modification with Fe2O3 nanoparticles on the anode surface enhanced power production to 11.2 W/m3, an increase of 4.7 W/m3.

Philadelphia-like (Ph-like) acute lymphoblastic leukemia (ALL) is a subgroup of B-cell precursor ALL (BCP-ALL) with a gene expression profile analogous to Philadelphia-positive ALL and recurrent IKAROS Family Zinc Finger 1 (IKZF1) gene deletion despite lacking BCR-ABL1 (Breakpoint cluster region-ABL protooncogene) translocation. Although recognized to occur at all ages, the proportion of cases among BCP-ALL varies (< 10% in children and up to 30% in adolescents). In all age groups, males are more commonly affected. Generally, Ph-like ALL is associated with adverse clinical features and an increased risk of treatment failure with conventional approaches. Genetic alterations such as aberrant expression, point mutations, or fusion translocations lead to activation of cytokine receptors and signaling kinases, which affect the ABL1 (ABL class fusion) or Janus Kinase (JAK) signaling pathways. Several clinical trials are being conducted to understand whether specific tyrosine kinase inhibitor therapy can improve cure rates. This review summarizes the current literature available about this entity.

OBJECTIVE: To explore the role of VEGF in attenuating endoplasmic reticulum stress in placental trophoblast cells. STUDY DESIGN: Study was divided into following parts: 1. Serum Analysis of GRP78 and VEGF using sandwich ELISA. 2. Expression of VEGF and GRP78 in placentae by immunohistochemistry (IHC). 3. In Vitro experiments. Status of ER stress markers (GRP78, eIF2α, XBP1, ATF6 and CHOP) was assessed at various time points (8 h, 14 h, 24 h) when trophoblast cells were treated with varying concentration(s) of VEGF and also by adding recombinant VEGF at protein (Immunofluorescence, Western blot) and transcript levels (qRT-PCR). RESULTS: Increased GRP78 and decreased VEGF protein levels in sera and placentae of preeclamptic pregnant women and reduced expression of various ER stress markers at both transcript and protein levels was observed in trophoblast cells when they were exposed to recombinant VEGF thereby indicating positive role of VEGF in alleviating ER stress. CONCLUSIONS: Reduced expression of ER stress markers in trophoblast cells against increased VEGF highlighted a new window to explore prospective drugs that can be designed to modulate the activities of various ER stress sensors in order to alleviate ER stress in pregnant women with preeclampsia.

The complete genome sequence of a garlic common latent virus (GarCLV) isolate collected from NHRDF, Karnal, India was determined. The whole genome of this GarCLV isolate consists of 8573 nucleotides, excluding the 3'-end poly-A tail. The sequence is 91-93% similar to rest of the five whole genome sequences available till date. It contains six open reading frames (ORFs) which encode polypeptide of 219 kDa, 25 kDa, 11 kDa, 7 kDa, 32.74 kDa and 15 kDa from ORF1, ORF2, ORF3, ORF4, ORF5 and ORF6 respectively. The phylogenetic study based on coat protein sequence demonstrated high diversity present among Indian population of GarCLV.

AIM: To compare efficacy and safety of 90,000 IU versus 300,000 IU oral single dose vitamin D for treatment of nutritional rickets. STUDY DESIGN: Randomized controlled trial. SETTING: Tertiary care hospital. PARTICIPANTS: One hundred ten children (6 months to 5 years, median age 10.5 months) with rickets. Exclusion criteria were disease affecting absorption, intake of calcium/vitamin D preparation in last 6 months, abnormal renal function, and rickets other than nutritional. INTERVENTION: Vitamin D3 as a single oral dose 90,000 IU (group A, n = 55) or 300,000 IU (group B, n = 55). METHODOLOGY: Severity of rickets was scored on knee and wrist X-ray as per Thacher's radiographic score. Baseline serum levels of calcium, SAP, 25(OH)D, iPTH were measured. Follow up was done at 1 week, 4 weeks, and 12 weeks. OUTCOME VARIABLE: Primary - Radiographic score at 3 months. Secondary - Serum levels of 25(OH)D, SAP, and iPTH at 3 months, clinical and biochemical adverse effects. RESULTS: Eighty-six subjects (43 in each group) completed the study. The radiographic score reduced from 6.90 to 0.16 in group A and from 6.93 to 0.23 in group B. The levels of 25(OH)D, ALP, and PTH were similar between the groups at baseline and follow up. Hypercalciuria and hypercalcemia were seen more often in group B as was hypervitaminosis D. There were no clinical adverse events. CONCLUSIONS: Single oral dose vitamin D3 90,000 IU is safe and effective in achieving healing of rickets.

In this study, peste des petits ruminants virus (PPRV) was detected in frozen pooled tissue samples from a dead Asiatic lion (Panthera leo persica). The samples were negative for canine distemper virus and positive for PPRV nucleic acids when tested with one-step RT-PCR using the appropriate virus-specific primers. Subsequent amplification, cloning, and sequencing of the partial nucleocapsid, matrix, and fusion genes confirmed the presence of PPRV nucleic acid. Comparative sequence and phylogenetic analyses of the structural genes of the isolated virus confirmed that the virus belonged to Asian lineage IV and was closely related to PPRV circulating in India.
