RESUMO

Changes in foaming properties and foam structure of raw whole, raw skim, pasteurised whole, and pasteurised skim milk during storage at 4 °C, evaluated by mechanical mixing, air and steam injection, were investigated. The results showed that storage of milk until the end of their shelf-life (day 3 for raw milk and day 21 for pasteurised milk) did not induce any significant change in pH, particle size, viscosity and their foaming properties (foaming capacity, foam stability and size of air bubbles) although there was a slight increase in free fatty acid content. Regarding foaming methods, the air injection method produced much less foam volume than mechanical mixing and steam injection, except for raw milk where mechanical mixing showed the least foamability. However, an opposite trend was observed for foam stability. Although air injection induced the largest size of air bubbles, it produced the most stable foam, which was much more stable than foam produced by mechanical mixing and steam injection.

RESUMO

BACKGROUND/AIM: Early and accurate diagnosis of functional visual loss (FVL) allows optimum management. Visual evoked potentials (VEPs) offer a means of objectively estimating acuity and therefore could assist with early and accurate diagnosis. The aim of this study was to assess the sensitivity and specificity of the step VEP in diagnosing FVL. METHODS: A retrospective audit was conducted in 36 school-aged children presenting with reduced visual acuity and clinical suspicion of FVL. All had undergone step VEP testing as part of their investigation. Medical notes were reviewed, and where necessary, referring centres, general practitioners or electronic clinical portals were consulted to obtain longer-term outcome data. RESULTS: Twenty-seven of the 36 patients (75%) were classified as having had FVL: all had a normal step VEP spatial threshold. Nine patients (25 %) had an organic cause for their acuity loss, of whom seven had abnormal step VEP spatial thresholds: the other two patients had some functional overlay to their organic disease. The step VEP sensitivity was 78% (95% confidence interval 40-96%), and specificity was 100% (95% confidence interval 84-100%). CONCLUSION: The high specificity of the step VEP for FVL warrants increased suspicion of an organic cause should the step VEP spatial threshold be abnormal.

RESUMO

Tissue engineering scaffolds with complex geometries can provide an architecture that directs tissue formation. Drug delivery from these scaffolds to promote regeneration is often challenging due to the complex fabrication processes. Surface-mediated DNA delivery from multiple channel bridges was applied to deliver lipoplexes in vivo to the injured spinal cord. The surface properties of the polymer, DNA deposition with or without drying, and the presence of ECM components were investigated. In vitro studies revealed that fibronectin produced greater expression levels and immobilization efficiencies compared with collagen, laminin, and no coating. In addition, lipoplex incubation on ECM-coated PLG increased expression relative to either of the drying methods. Additionally, the incubation method had more homogeneously distributed lipoplexes and a higher number of transfected cells relative to the dried conditions. Translation to three-dimensional bridges led to high levels of transgene expression in vitro. In vivo, lipoplexes immobilized to the bridge produced transgene expression levels in a rat spinal cord hemisection model that were 2-fold greater than naked plasmid. Additionally, expression with lipoplexes persisted for at least three weeks. Surface-mediated delivery can be applied to scaffolds with complex geometries to promote transgene expression in vivo.
