RESUMO

We investigated the production of oxytocin (OT) and oxytocin-neurophysin (bNpI) by bovine granulosa cells cultured in presence of 10% foetal calf serum, a condition known to induce spontaneous luteinization of these cells. The production of immunoreactive OT was significantly higher in the cultures of granulosa cells harvested from large follicles than in those derived from small follicles. Chromatography on Sephadex G-25 showed similar elution sites of ovarian and synthetic OT, while high performance liquid chromatography revealed two peaks of OT-immunoreactivity, one of which (+/- 65% of the total immunoreactivity) coincided with synthetic OT. In another experiment, we could observe a gradual increase of OT, bNp I and progesterone production by granulosa cells derived from large follicles, in relation with the incubation time. The mean molar ratio OT: bNp I was 2.2 +/- 0.5 (SEM), and we found a significant positive correlation between the production of OT and bNp I (r = 0.77; P less than 0.01) and between the production of OT and progesterone (r = 0.80; P less than 0.01). Furthermore, the cellular OT and bNp I content of large follicles-derived granulosa cells before culture was 4-5 times lower than the total amount of OT and bNp I produced during a 72-h incubation, suggesting an active synthesis of these peptides. These data show that bovine granulosa cells are able to produce OT and bNp I, probably by an active biosynthesis as observed in the hypothalamo-neurohypophysial system and that the granulosa productions of OT, bNp I and progesterone are closely related.

Four Romney ewes were actively immunized with a partially purified preparation of inhibin derived from bovine follicular fluid and their ovulation rates in four successive oestrous cycles were compared with those of four ewes receiving adjuvant alone. The ovulation rates of the ewes immunized with the inhibin preparation were significantly higher than those of the control ewes (2.06 +/- 0.16 (S.E.M.) vs 1.31 +/- 0.06 ovulations/ewe, n = 4). Plasma concentrations of FSH and LH, measured in blood samples taken three times a week for 11 weeks, during which time each ewe was immunized three times, were not significantly different between the two treatment groups. These results suggest that active immunization with inhibin-enriched follicular fluid may be a potential means of increasing fecundity in sheep.

Matrex gel red A purified follicular fluid has been used to study whether or not this material contains sialic acid residues and their importance in maintaining the biological activity of inhibin both in vitro and in vivo. It appears that sialic acid is present in these preparations and can be released either by neuraminidase treatment of acid hydrolysis. The addition of intact and desialylated inhibin-containing material to isolated rat pituitary cells in culture gives similar inhibition of LHRH-induced FSH release of these cells indicating that sialic acid is not required for inhibin activity in vitro. The injection of intact inhibin preparations leads to a reduction of the uterine weight increase seen in immature female mice primed with human chorionic gonadotropin. By contrast, the inhibition of this uterine weight increment by 80% desialylated inhibin-containing material is significantly reduced, suggesting that sialic acid residues play an important role in maintaining the biological activity of inhibin in vivo.

Radioactive sulphur (35SO4) is incorporated in the proteoglycans of chicken embryonic cartilage. In fact, the molecular weight of the macromolecules which bind the sulphur corresponds to monomeric proteoglycans and proteoglycan-hyaluronic acid complexes. This incorporation does not take place when the cartilage has been previously killed. The capture of 35S is stimulated by human serum, in a dose-dependent way. At doses less then, equal to and greater than the maximal plasma concentrations observed following a single oral dose (600 mg), acetyl salicylic acid and sodium salicylate inhibit the incorporation of radioactive sulphur in the cartilage. Under the same conditions, tiaprofenic acid and oxametacine have no effect on this metabolic parameter.

RESUMO

Prostatic acid phosphatase has been isolated from human seminal plasma. The purification method utilizes gel filtration on Sephadex G100, ammonium sulfate precipitation and a series of chromatographical steps including concanavalin A Sepharose 4B, anion exchange and gel filtration chromatography. The final product appears homogenous when analyzed by gel filtration on Sephadex G100. It gives one major band on SDS polyacrylamide gels. The specific activity is similar to that obtained by other purification schemes. The yield of the method described above has allowed to set up a sensitive radioimmunoassay of prostatic acid phosphatase.

The FSH secretion-inhibiting action of inhibin in vitro under basal conditions and also in the presence of LH-RH is suppressed by the addition of MIX, a phosphodiesterase inhibitor. In the presence of LH-RH, inhibin reduces significantly the intracellular level of cAMP in isolated pituitary cells. In contrast, the simultaneous addition of MIX and inhibin raises the cAMP level, and this stimulation is comparable to the increase observed when MIX is added alone. These observations suggest that one mode of action of inhibin could be mediated by a reduction in cAMP within the pituitary gonadotropic cell.

After standard glutaraldehyde-osmium tetroxide fixation procedures, the majority of microfilament bundles in BHK-21 cells exhibit relatively uniform electron density along their long axes. The inclusion of tannic acid in the glutaraldehyde fixation solution results in obvious electron density shifts along the majority of microfilament bundles. Striated patterens are frequently observed which consist of regularly spaced electron dense (D) and electron lucid (L) bands. A striated pattern is also observed along many BHK-21 stress fibers after processing for indirect immunofluorescence utilizing BHK-21 myosin antiserum. A direct correlation of these periodicities seen by light and electron microscope techniques is impossible at the present time. However, comparative measurements indicate that the overall patterns seen in the immunofluorescence and electron microscope preparations are similar. The ultrastructural results provide an initial clue for the ultimate determination of the supramolecular organization of contracile proteins other than actin within the microfilament bundles of non-muscle cells.

A Ca2+-dependent modulator protein has been isolated from BHK-21 cells. The purification requires heat treatment, ion-exchange chromatography, and gel filtration. The protein appears homogenous on sodium dodecyl sulfate--polyacrylamide and isoelectric focusing gels. The protein comigrates with purified smooth muscle and brain modulators. BHK-21 modulator is characterized by a high content of aspartic and glutamic acids and by a high phenylalanine/tyrosine ratio. It lacks both cysteine and tryptophan. The protein is effective in activating brain-modulator-deficient phosphodiesterase. It can also be used in assay systems to generate Ca2+-sensitive actin activation of both BHK-21 and smooth muscle myosins. Therefore, it is proposed that the BHK-21 modulator protein is a component of the Ca2+-dependent mechanism involved in the regulation of actin--myosin interactions in BHK-21 cells.

A fraction has been obtained from baby hamster kidney (BHK-21) cells that will stimulate the actin-moderated ATPase (ATP phosphohydrolase, EC 3.6.1.3) activity of both BHK-21 myosin and gizzard smooth muscle myosin. This activation is associated with the specific phosphorylation of the myosin 20,000-dalton light chain. The BHK-21 myosin light chain kinase preparation contains a major protein of approximately 105,000 molecular weight as determined by sodium dodecyl sulfate gel electrophoresis. Both the actin activation and phosphorylation events require the presence of Ca2+ and the so-called modulator or calcium-dependent regulator protein that has been isolated from smooth muscle, brain, and other tissues. On the basis of these results we propose that this kinase system constitutes a Ca2+-dependent regulatory mechanism for myosin-actin interactions in nonmuscle mammalian cells.

Myosin has been isolated from baby hamster kidney cells (BHK21/C13) in high yield and characterized biochemically and immunologically. The subunit composition consists of 2 heavy chains, approximately 200,000 Daltons each, and 2 classes of light chains of approximately 16,000 and 20,000 Daltons. The myosin exhibits ATPase activity in the presence of K+-EDTA or Ca2+ but very little activity with Mg2+-ATP. The Mg2+-ATPase activity is stimulated only about 2-fold by skeletal actin, but a much larger activation is obtained in the presence of a protein kinase isolated from chicken gizzard. The increase in actin activation is accompained by the phosphorylation of the 20,000-Dalton light chain. BHK21 myosin is insoluble at low ionic strength and forms typical biopolar thick filaments. A specific antiserum generated against this protein forms a single precipitin line with the antigen but does not crossreact with either skeletal or smooth muscle myosin. The antiserum also specifically stains stress fibres in BHK21 cells as shown by indirect immunofluorescence.

The localization and organization of actin-like microfilaments in normal, SV-40 and adenovirus transformed cells are determined by the coordinated use of light optical, electron optical and biochemical techniques. In adenovirus-type 5 transformed hamster embryo cells, microfilament meshworks appear to be the predominant organizational form of cellular action, while in normal hamster cells, microfilament bundles are prevalent. Differences between 3T3 and SV-40 transformed 3T3 cells are less apparent and may be related to the packing and intracellular distribution of microfilament bundles. Attempts at relating these ultrastructural changes in transformed cells to the images obtained following reaction with fluorescein-labelled myosin fragments and indirect immunofluorescence with smooth muscle myosin antibody are discussed. In several instances the fluorescence microscope images to not correspond to the ultrastructural observations. The results are discussed in terms of the possible relationships between alterations in cytoplasmic contractile elements and the abnormal behavior of transformed cells.
