Introduction: Dehorning calves is necessary to minimize injury because intensive raising circumstances make horned cows more aggressive. However, acute pain is commonly perceived by farm animals when undergoing painful practices such as dehorning, affecting their health status and quality of life. By quantifying the magnitude of pain and discomfort associated with dehorning, we aim to contribute to a more humane and sustainable cattle farming industry. Methods: The objective of this study was to evaluate the behavioral, physiological, and emotional effects of acute dehorning pain in calves using two methods: dehorning cream and dehorning hot-iron.30 Holstein calves aged 4 days were selected for the study. These calves were randomly assigned to two experimental groups based on the method of disbudding: dehorning cream (n = 15) and hot-iron dehorning (n = 15). Before and after dehorning, we evaluated their physiological indicators of infrared eye temperature, concentrations of substance P, IL-6, cortisol, haptoglobin, as well as emotional state, and pain-related behavioral reactions. Results: Post-dehorning, the duration of lying down decreased significantly in both groups (DI and DC: 0-4 h) after dehorning (p < 0.05). Both groups exhibited increased frequencies of pain-related behaviors such as head shaking (DI: 1-7 h, DC: 1-6 h), ear flicking (DI: 2-7 h, DC: 2-7 h), head scratching (DI: 2-3 h, DC: 1-7 h), and top scuffing (DI: 2 h, DC: 2-7 h) compared to pre-dehorning (p < 0.05). The DC group demonstrated a higher frequency of head-shaking, ear-flicking, head-scratching, and top-rubbing behaviors, along with a longer duration of lying down (0-4 h), compared to the DI group (p < 0.05). Post-dehorning, play behavior reduced significantly in both groups (6-8 h) (p < 0.05), whereas judgment bias and fear levels showed no significant change (p > 0.05). Physiological measures including eye temperature, and blood levels of substance P and IL-6, did not differ significantly between the groups before and after dehorning (p > 0.05). However, 48 h after dehorning, calves in the DC group had significantly higher haptoglobin levels compared to the DI group (p = 0.015). Additionally, salivary cortisol levels in the DC group increased significantly at 3.5 h and 7 h post-dehorning (p = 0.018, p = 0.043). Discussion: Both hot-iron and cream dehorning induced pain in calves, as evidenced by increased pain-related behaviors, elevated salivary cortisol, and higher haptoglobin levels, alongside reduced positive behaviors. Notably, these effects were more pronounced in the DC group than in the DI group, suggesting that dehorning hot-iron may be a comparatively less stressful dehorning method for young calves. Moreover, the brief duration of pain response and weaker response to dehorning observed in 13-day-age calves in this study suggests that dehorning at younger ages may be more advisable and warrants further research.

Adverse stress, such as the long-term restriction of food intake and activity in intensive production, leads to a depression-like mental state in sows. Mood disorder, such as depression, is a widely concerned animal welfare issue. However, little is known about the biological mechanisms that underlie mood disorders in pigs. This study is the first attempt to establish a pig depression model by acute stress. A total of 16 adult Bama pigs were divided into the control and model groups, with 8 pigs (half male and half female) per group. The pigs in the model group were restrained for 24 h in a dark and ventilated environment, with food and water deprivation. After the restraint, behavioral tests (feed intake, sucrose preference test, open field test, and novel object test) were used to evaluate apparent indicators. The levels of COR and ACTH in the serum and the levels of 5-HT, NE, and BDNF in the hippocampus and medial prefrontal cortex were detected using ELISA to identify the physiological state. After acute stress, pigs exhibited decreased feed intake and sucrose preference, increased serum COR levels, decreased hippocampal 5-HT levels, and exhibited more fear. Finally, the model was evaluated according to the weight of the test indicators. The overall score of the model was 0.57, indicating that modeling was feasible. Although the reliability and stability require further verification, this novel model revealed typical depression-like changes in behavior and provided a potential method to establish a model of depression in pigs.

Environmental enrichment can improve animal welfare. As a method of environmental enrichment, the effect of different auditory stimulations on the behavior response and welfare of laying hen chicks has yet to be investigated. Therefore, this study was aimed at exploring the impact of various auditory exposures on the behavior, learning ability, and fear response of 4-week-old laying hen chicks. A total of 600 1-day-old chicks were randomly assigned to five different groups: C (control group), LM (Mozart's String Quartets, 65 to 75 dB), LN (recorded ventilation fans and machinery, 65 to 75 dB), HN (recorded ventilation fans and machinery, 85 to 95 dB), and HM (Mozart's String Quartets, 85 to 95 dB). The experiment was conducted from day 1 until the end of the experiment on day 28. Groups LM and LN were exposed to music and noise stimulation ranging from 65 to 75 dB. Groups HN and HM, meanwhile, received noise and music stimulation ranging from 85 to 95 dB. The control group (C) did not receive any additional auditory stimuli. During the experimental period, continuous behavioral recordings were made of each group of chicks from day 22 to day 28. On day 21, the PAL (one-trial passive avoidance learning) task was conducted. On days 23 and 24, OF (open field) and TI (tonic immobility) tests were performed, and the levels of serum CORT (corticosterone) and DA (dopamine) were measured. The results indicated that exposure to music and noise at intensities ranging from 85 to 95 dB could reduce comforting, preening, PAL avoidance rate, the total number of steps and grid crossings of OF, and the concentration of DA in 4 WOA chicks (p < 0.05), increase the freezing times of OF (p < 0.05); 65 to 75 dB of noise stimulation could reduce preening and total number steps of OF in 4 WOA chicks (p < 0.05), increase the freezing times of OF (p < 0.05); and 65 to 75 dB of music exposure could reduce the concentration of CORT in 4 WOA chicks (p < 0.05). Therefore, 65 to 75 dB of music exposure could produce positive effects on chicks and showed relatively low CORT level, whereas 85 to 95 dB of music and noise exposure could reduce comforting and preening behavior, impair learning ability, and increase the fear responses of chicks.

Many factors influence the effects of exogenous organic chromium (EO-Cr) on the growth performance and carcass qualities of weaned and growing-finishing pigs, such as pig growth stages, types of EO-Cr, period of supplementation, and farm management. However, it is challenging to comprehensively consider all factors in one study. To solve this problem, we searched all relative literature published from 1 January 2000 to 1 January 2023, to systematically analyze and review the effects of EO-Cr on pig growth performance and carcass qualities via meta-analysis. Thirty-five papers were filtered and analyzed, which involved 4366 pigs. The results showed that, for weaned piglets, EO-Cr diets significantly increased the average daily gain (ADG, p < 0.001) and average daily feed intake (ADFI, p = 0.022) but reduced the feed-gain ratio (p = 0.004). In addition, for growing-finishing pigs, EO-Cr supplementation significantly increased the ADG (p < 0.001), carcass lean ratio (p = 0.020), and loin muscle area (p < 0.001), but had no significant effect on the ADFI (p = 0.071), feed-gain ratio (p = 0.692), dressing percent (p = 0.989), or back fat thickness (p = 0.142). Moreover, the effect of EO-Cr was greater in weaned piglets than in growing-finishing pigs. In terms of the dose effect of the supplement, chromium nicotinate is the most suitable EO-Cr type for weaned piglets with an optimal dosage range of 0.125-0.150 mg/kg. On the other hand, chromium picolinate is the most suitable EO-Cr type for growing-finishing pigs with an optimal dosage range of 0.250-0.300 mg/kg. In conclusion, EO-Cr supplementation is beneficial for enhancing the growth performance and carcass qualities of both weaned and growing-finishing pigs.

The stall-housing system is commonly used in the modern swine industry in many countries; however, long-term space restrictions can cause affective and physiological abnormalities in sows. The pupil light reflex (PLR) can reflect the psychological and neurological changes in animals, and confined sows show higher pupillary rigidity. However, the PLR differs between same-parity sows, suggesting differences in behaviors and affective states between parity groups. We subjected confined Yorkshire × Landrace sows of parity 0, 2, and 5 to a PLR test and accordingly assigned them to the weak PLR (WR) group (n = 20) or the strong PLR (SR) group (n = 22). We then observed the sows' behaviors and performed a sucrose/quinine response test and novel object test (NOT) to assess the differences in their affective states. The standing and lateral lying behaviors of the sows were less frequent in WR than in SR (p < 0.05), whereas ventral lying and sitting behaviors was more frequent in WR than in SR (p < 0.05). No changes in chewing behaviors and sucrose/quinine responses were observed (p > 0.05); however, the numbers and duration of novel object contact were lower and the novel object response latency time was longer in WR than in SR (p < 0.05). Regarding parity, standing and lateral lying behaviors were less frequent and ventral lying and sitting behaviors were more frequent at parity 5 than at parity 0 (p < 0.05). Bar-biting, rooting, trough-biting, and sucrose response score were lower at parity 5 than at parity 0 (p < 0.05), and vacuum chewing behavior and quinine response score were higher in sows of parity 5 than in those of parity 0 (p < 0.05). NOT showed that the number of contacts and contact duration in sows decreased with increasing parity (p < 0.05), and the response latency time was longer in sows of parity 5 than in those of lower parity (p < 0.05). In conclusion, the behavioral expression and responses of confined sows to novel objects differed between PLRs. The evaluation of the affective state of sows also revealed marked differences with increasing parity. Thus, confined sows with WR and high parity apparently suffer from more severe psychological problems, and PLR may be a potent indicator for evaluating the affective state of confined sows.

This study investigated the proteomic characteristics of colostrum for sows housed under different conditions. Among 12 gilts, four were housed in a gestation-crate and farrowing-crate combined housing system (CC) as controls, four were housed in a gestation-pen and farrowing-pen combined housing system (PP), and four were housed in a gestation-pen and farrowing-crate combined housing system (PC). Differentially expressed proteins in the colostrum (PP versus CC, and PC versus CC) were screened by proteomics technology, and bioinformatics analysis was then performed. Results showed that 93 proteins were differentially expressed in PP versus CC, and that 126 proteins were differentially expressed in PC versus CC. The differentially expressed proteins in the PP versus CC comparison were mainly enriched in interleukin (IL)-17, transforming growth factor-ß, and nuclear factor-κ B signaling pathways, and in metabolic pathways, including glutathione metabolism, peroxisome, and carbon metabolism. In contrast, differentially expressed proteins in the PC versus CC comparison were enriched in the IL-17 signaling pathway, cholesterol metabolism, and peroxisome proliferator-activated receptor signaling pathway. In conclusion, the housing environment appeared to affect the colostrum composition of sows by acting on their immune system and metabolic processes, particularly fat metabolism.

It is essential to monitor the ground temperature over large areas to understand and predict the effects of climate change on permafrost due to its rapid warming on the Qinghai-Tibet Plateau (QTP). Land surface temperature (LST) is an important parameter for the energy budget of permafrost environments. Moderate Resolution Imaging Spectroradiometer (MODIS) LST products are especially valuable for detecting permafrost thermal dynamics across the QTP. This study presents a comparison of MODIS-LST values with in situ near-surface air temperature (Ta), and ground surface temperature (GST) obtained from 2014 to 2016 at five sites in Beiluhe basin, a representative permafrost region on the QTP. Furthermore, the performance of the thermal permafrost model forced by MODIS-LSTs was studied. Averaged LSTs are found to strongly correlated with Ta and GST with R2 values being around 0.9. There is a significant warm bias (4.43-4.67 °C) between averaged LST and Ta, and a slight warm bias (0.67-2.66 °C) between averaged LST and GST. This study indicates that averaged MODIS-LST is supposed to be a useful data source for permafrost monitoring. The modeled ground temperatures and active-layer thickness have a good agreement with the measurements, with a difference of less than 1.0 °C and 0.4 m, respectively.

Objective: Physical activity restriction in sows may lead to behavioral abnormalities and affective disorders. However, the psychophysiological state of these sows is still unclear. As miRNAs can be used as effective markers of psychopathy, the present study aimed to assess the difference in microRNA expression between the long-term activity restricted sows and activity free sows, thus contributing to the understanding of abnormal sow behaviors.. Methods: Four dry sows (sixth parity, Large × White genetic line) were selected from activity restricted crates (RC) or activity free pens (FP) separately. microRNAs in the ventromedial prefrontal cortex (vMPFC) and serum were examined using RT-PCR, and the correlation between the miRNAs expressed in the vMPFC and serum was evaluated. Results: miR-134 (1.11 vs 0.84) and miR-1202 (1.09 vs 0.85) levels were higher in the vMPFC of the RC sows than in the FP sows (p < 0.01). Furthermore, miR-132 (1.27 vs 1.08) and miR-335 (1.03 vs 0.84) levels were also higher in the RC sows than in FP sows (p < 0.05); however, miR-135a, miR-135b, miR-16 and miR-124 levels were not different (p > 0.05). The relative expression of serum miR-1202 was higher in the RC sows than in the FP sows (1.04 vs 0.54) (p < 0.05). , and there was a strong correlation (R=0.757, p < 0.05) between vMPFC and Serum levels of miR-1202. However, no significant difference was observed in miR-16 levels in the serum of the RC sows and FP sows (p > 0.05). Conclusion: This pilot study demonstrates that long-term activity restriction in sows likely results in autism or other complex psychopathies with depression-like behaviors. These observations may provide new insights for future studies on abnormal behavior in sows and contribute to research on human psychopathy.

Thermokarst lakes are a ubiquitous landscape feature, which widely distributed in the pan-arctic and some low latitude regions, and are associated with regional hydrological processes. The studies were taken to obtain a better understanding of the water balance of thermokarst lakes in the Qinghai-Tibet Plateau (QTP) in order to gain insight of the regional hydrological cycle. The characteristics of the stable isotopes δ 18O and δ D were investigated in precipitation, permafrost meltwater, and thermokarst lake water in the continuous permafrost region of the QTP and analyzed the lake water balance using the isotope mass model. The results showed that the δ D-δ 18O relationship in the thermokarst lakes (δ D = 5.45 δ 18O - 18.95) differed from that of the local precipitation (δ D = 8.30 δ 18O + 18.49) and permafrost meltwater (δ D = 5.78 δ 18O - 23.41), and the mean isotope compositions in the thermokarst lakes were -7.2‰ in δ 18O and -58.0‰ in δ D. The more positive isotope signals in thermokarst lakes than in the precipitation and permafrost meltwater revealed that the lakes had experienced stronger isotope enrichment. Additionally, the evaporation-to-inflow ratio (E/I) values were < 1 in most of the thermokarst lakes (84%), which might be explained by the recent expansion of the lake surfaces. However, 16% of the thermokarst lakes had shrunk, owing to thermokarst erosion, lateral expansion as the temperature increases, and lower recharge volume. Moreover, precipitation on the lake surface was only 14-18% of the inflow volume in the thermokarst lakes, and the surface-subsurface inflow and permafrost meltwater are very important for recharging the lakes and maintaining the water balance. The results of this study provide a comprehensive understanding of the influence of climate warming on hydrological processes in the permafrost regions in the QTP.

Beiluhe basin is underlain by warm and ice-rich permafrost, and covered by vegetation and soils characteristic of the Qinghai-Tibet Plateau. A field monitoring network was established to investigate permafrost conditions and to assess potential impacts of local factors and climate change. This paper describes the spatial variations in permafrost conditions from instrumented boreholes, controlling environmental factors, and recent thermal evolution of permafrost in the basin. The study area was divided into 10 ecotypes using satellite imagery based classification. The field investigations and cluster analysis of ground temperatures indicated that permafrost underlies most of the ground in swamp meadow, undisturbed alpine meadow, degrading alpine meadow, and desert alpine grassland, but is absent in other cover types. Permafrost-ecotope relations examined over a 2-year (2014-2016) period indicated that: (i) ground surface temperatures varied largely among ecotopes; (ii) annual mean ground temperatures ranged from -1.5 to 0°C in permafrost, indicating sensitive permafrost conditions; (iii) active-layer thicknesses ranged from 1.4m to 3.4m; (iv) ground ice content at the top of permafrost is high, but the active-layer soil is relatively dry. Long-term climate warming has driven thermal changes to permafrost, but ground surface characteristics and soil moisture content strongly influence the ground thermal state. These factors control local-scale spatial variations in permafrost conditions. The warm permafrost in the basin is commonly in thermal disequilibrium, and is sensitive to future climate change. Active-layer thicknesses have increased by at least 42cm and the mean annual ground temperatures have increased by up to 0.2°C in the past 10years over the basin. A permafrost distribution map was produced based on ecotypes, suggesting that permafrost underlies 64% of the study region.

The aim of the present study was to study the effects of straw enriched environment on behaviors of nursery piglets reared in the farrowing pens. Fourteen litters (Large White x Landrace) weaned at 35 days of age were reared in the modified farrowing pens, flatdecks (F) or straw enriched pens (P), until 70 days of age. The behavior was observed from 7 to 10 weeks of age. Results showed that straw enriched pens significantly increased walking, total exploring and active behavior, reduced lying and exploring behavior direct to pen, but not that direct to penmates. Meanwhile, in wk8-wk10, the number of fighting piglets in P was significantly more than that in F. With increasing age, piglets exploring in total or direct to pen, and active piglets decreased gradually in F. In P, piglets exploring in total or that direct to straw decreased, and reached a trough in wk9, then rose up. Lying piglets in F increased with age while that in P increased only at 9 or 10 weeks of age. Walking piglets decreased significantly with age in both environments. The number of fighting piglets in F was a maximum in wk7 while it in P was fewer in wk7 or wk8. Furthermore, the activity of piglets in F was at peak during 08:00-10:00 hr and reached a trough during 11:00-13:00 hr. In P, refreshed straw kept piglets at a more active state during morning, shortened the activities trough at noon, and showed high activity in the afternoon. In conclusion, present straw enriched pen can prevent fighting, increase total exploring, reduce exploring direct to pen, and even affect the rhythm of behavior. It is applicable for improving welfare of nursery piglets.

This study estimated the fermentation characteristics and nutrient value of corn-lablab bean mixture silages relative to corn silages. The effects of feeding corn-lablab bean mixture silages on nutrient apparent digestibility and milk production of dairy cows in northern China were also investigated. Three ruminally cannulated Holstein cows were used to determine the ruminal digestion kinetics and ruminal nutrient degradability of corn silage and corn-lablab bean mixture silages. Sixty lactating Holstein cows were randomly divided into two groups of 30 cows each. Two diets were formulated with a 59:41 forage: concentrate ratio. Corn silage and corn-lablab bean mixture silages constituted 39.3% of the forage in each diet, with Chinese wildrye hay constituting the remaining 60.7%. Corn-lablab bean mixture silages had higher lactic acid, acetic acid, dry matter (DM), crude protein (CP), ash, Ca, ether extract concentrations and ruminal nutrient degradability than monoculture corn silage (p<0.05). Neutral detergent fiber (NDF) and acid detergent fiber (ADF) concentrations of corn-lablab bean mixture silages were lower than those of corn silage (p<0.05). The digestibility of DM, CP, NDF, and ADF for cows fed corn-lablab bean mixture silages was higher than for those fed corn silage (p<0.05). Feeding corn-lablab bean mixture silages increased milk yield and milk protein of dairy cows when compared with feeding corn silage (p<0.05). The economic benefit for cow fed corn-lablab bean mixture silages was 8.43 yuan/day/cow higher than that for that fed corn silage. In conclusion, corn-lablab bean mixture improved the fermentation characteristics and nutrient value of silage compared with monoculture corn. In this study, feeding corn-lablab bean mixture silages increased milk yield, milk protein and nutrient apparent digestibility of dairy cows compared with corn silage in northern China.
