RESUMO

Axonal motor driven cargo utilizes the microtubule cytoskeleton in order to direct cargo, such as synaptic vesicle precursors (SVP), to where they are needed. This transport requires vesicles to travel up to microns in distance. It has recently been observed that finite microtubule lengths can act as roadblocks inhibiting SVP and increasing the time required for transport. SVPs reach the end of a microtubule and pause until they can navigate to a neighboring microtubule in order to continue transport. The mechanism(s) by which axonal SVPs navigate the end of a microtubule in order to continue mobility is unknown. In this manuscript we model experimentally observed vesicle pausing at microtubule ends in C. elegans. We show that a single rate-constant model reproduces the time SVPs pause at MT-ends. This model is based on the time an SVP must detach from its current microtubule and re-attach to a neighboring microtubule. We show that vesicle pause times are different for anterograde and retrograde motion, suggesting that vesicles utilize different proteins at plus and minus end sites. Last, we show that vesicles do not likely utilize a tug-of-war like mechanism and reverse direction in order to navigate microtubule ends.

We measure the gap density of states and the Fermi level position in thin-film transistors based on pentacene and dinaphtho[2,3-b:2',3'-f]thieno[3,2-b]thiophene (DNTT) films grown on various surfaces using Kelvin probe force microscopy. It is found that the density of states in the gap of pentacene is extremely sensitive to the underlying interface and governs the Fermi level energy in the gap. The density of gap states in pentacene films grown on bare silicon dioxide (SiO(2)) was found to be larger by 1 order of magnitude compared to that in pentacene grown on SiO(2) treated with hexamethyldisilazane and larger by 2 orders of magnitude compared to that of pentacene grown on aluminum oxide (AlO(x)) treated with a self-assembled monolayer (SAM) of n-tetradecylphosphonic acid (HC(14)-PA). When DNTT was grown on HC(14)-PA-SAM-treated AlO(x), the gap density of states was even smaller, so that the Fermi level pinning was significantly reduced. The correlation between the measured gap density of states and the transistor performance is demonstrated and discussed.

Multiple myeloma represents a malignant proliferation of plasma cells derived from a single clone. It is well known that alkylating agents are capable of inducing myelodysplastic syndromes (MDS) and acute myelocytic leukemias (AML). This risk of both diseases in patients with multiple myeloma has been estimated to be 10-20% after 10 years. We aimed to evaluate the time course and the type of genetic abnormalities in melphalan-treated patients in the chronic stage of the disease. We applied fluorescence in situ hybridization methods with probes to 5q31 and 7q31 to mononuclear peripheral blood leukocytes of 18 melphalan-treated patients and compared the results to those of 8 untreated myeloma patients. We found three patients (17%) with a 5q31 deletion in their peripheral white blood cells, but no 7q31 deletion. These findings suggest that 5q- occurs before the overt development of MDS/AML and raise important concerns regarding long-term treatment of myeloma patients with alkylating agents. Also, the performance of cytogenetic evaluation should be considered before autologous transplantation. The clinical and biological implications of these findings should be evaluated in larger clinical and laboratory studies.

Fifteen women pediatric residents, 15 non-medical women academicians similar in age and marital status and 15 women pediatricians at least 10 years past residency were surveyed. Subjects were asked to estimate the amount of time they devote to their professional work, domestic work and the degree to which they feel overworked. Subjects were also questioned regarding their perceptions of the advantages and disadvantages of their life-style and their self-concept as a women and as a professional. The results of this study show that residents, who work significantly more hours per week, have a significantly lower level of work satisfaction than do women in the other two groups. It also suggests the fact that the stress of the residency years affects not only the residents' work satisfaction but their entire self-concept as well. The paper offers possible suggestions the medical profession can take in order to help physicians better cope with their professional stress.
