RESUMO

Ectopic bone formation in the left atrioventricular valves and cardiac fibroskeleton, with systemic circulatory disturbance, is reported in a 4-year-old crossbred heavy horse. Microscopically, there was fibrosis, chondral metaplasia and mature bone, with bone marrow within the left atrioventricular cusps and in the annuli of the aortic and right atrioventricular valves.

The comonomer compositional distribution and thermal and morphological characteristics were investigated for five bacterially synthesized poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] samples with 3HV content of 45, 49, 70, 80, and 96 mol %. All these samples were fractionated into many fractions with widely different 3HV content by changing solvent/nonsolvent volume ratio of chloroform/n-heptane mixtures. Bacterial P(3HB-co-3HV) samples investigated in this study were found to have broad comonomer compositional distribution. The tendencies of the fractional precipitation of the P((3HB-co-3HV)s with 3HV content lower than 60 mol % and those with 3HV higher than 80 mol % were found to be contrary. The 3HV content dependences of the thermal properties and crystalline structures were investigated for bacterial poly(3-hydroxybutyrate) [P(3HB)] and a series of compositionally well-fractionated P(3HB-co-3HV) samples with 3HV content ranged from 14 to 98 mol % by DSC, WAXD, and solid-state (13)C NMR. It was found that P(3HB-co-3HV) samples with 3HV content lower than about 47 mol % form the crystalline lattice having the P(3HB) homopolymer type lattice including the 3HV unit as the crystal constituent, and those with a 3HV content higher than about 52 mol % form the crystalline lattice having the P(3HV) homopolymer type lattice including the 3HB units. Thus, P(3HB-co-3HV)s show the crystalline structural change in a very narrow range of 3HV content.

RESUMO

Poly(3-hydroxybutyrate) [P(3HB)] and its copolymers with hydroxyalkanoates are naturally occurring thermoplastic materials produced by bacteria. There are many potential uses for these copolyesters owing to their biodegradability and biocompatibility. The physical properties of the copolyesters vary depending on the chemical structure as well as the composition of the comonomers. Usually, we expect, copolymers to have a narrow chemical composition distribution (CCD). Several reports, however, have pointed out that some bacterial copolyesters have broad and/or multimodal CCD. Fractionation based on the chemical composition has also been reported for several bacterial copolyester samples. In this review, the literature concerning CCD and fractionation based on chemical composition is summarized. The width of CCD is approximated based on the data of diad sequence distribution. Generality of the complex CCD in bacterial copolyesters is also discussed.

RESUMO

The possibilities of the biosynthesis of polyesters from 20 kinds of naturally occurring L-amino acids by Alcaligenes eutrophus were studied. It was found that several amino acids were used efficiently to synthesize copolyesters of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3 HV) by A. eutrophus under the 'loose nitrogen-limiting' condition, but the other amino acids were scarcely utilized as a carbon source for the synthesis of polyesters. When L-threonine or L-isoleucine were used as the sole carbon source, copolyesters with higher 3HV content were produced.

RESUMO

It was found that an optically active copolyester, poly(3-hydroxybutyrate-co-3-hydroxyvalerate), denoted as P(3HB-co-3HV), is synthesized by Alcaligenes eutrophus H16 from several amino acids under various fermentation conditions. The optimum condition for the biosynthesis from one amino acid, threonine, was investigated and its biosynthetic pathway was discussed on the basis of the relation between the fermentation condition and the co-monomer composition of the produced polyesters.

RESUMO

A series of copolyesters of 3-hydroxybutyrate (3HB) and 3-hydroxyvalerate (3HV) was biosynthesized by Alcaligenes eutrophus from an amino acid, threonine. The 3HV content of these polyesters ranged from less than 0.1% to 30%.

Alcaligenes eutrophus H16 was grown on mixtures of 1H- and 2H-acetate as carbon sources. The accumulation of deuterated poly(3-hydroxybutyrate) (P(3HB)) was observed. The deuterium distributions in the isolated P(3HB)s were determined from 1H and 2H-n.m.r. spectra and confirmed by 13C-n.m.r. spectra. Although one would expect to synthesize P([2,2,4,4,4-2H5]3HB) when the cells were grown on 2H-acetate as the sole carbon source, the methyl, methylene and methine groups of the P(3HB) contained both deuterium and proton. This observation indicates some substitution from 2H to 1H during the P(3HB) synthesis. The 2H content in the methyl groups was larger than that in the methylene groups, which suggests a kinetic isotope effect in the P(3HB) synthesizing process. The deuterium distributions in the two magnetically non-equivalent methylene protons were determined to be different, which indicates stereoselectivity at the C2 site.

Changes in action potential (AP) and summating potential (SP) were investigated in guinea pigs immunized with type II collagen through the stylomastoid foramen. Endolymphatic hydrops could be induced in four of 11 guinea pigs. The striking feature of the electrocochleographic waveform in guinea pigs with endolymphatic hydrops was the negative SP recording in response to high frequency tone bursts. Furthermore, abnormal changes in AP were observed in three of four hydropic guinea pigs. Morphologic study of the cochleas in these three guinea pigs with light microscopy revealed spiral ganglion cell degeneration in addition to endolymphatic hydrops and almost normal sensory hair cells. These results suggest that guinea pigs with hydrops as produced by our procedure can serve as a useful model of Meniere's disease, that autoimmune response may play an important role in the etiopathogenesis of Meniere's disease, and that spiral ganglion cell degeneration together with endolymphatic hydrops seems to contribute to abnormal changes in AP.

RESUMO

A case of a new-born infant, aged 35 days old, with laryngocele was successfully treated by laryngomicroscopic operation under general anaesthesia. He suffered from congenital inspiratory dyspnea due to cystic swelling of the left arytenoid region. Although therapeutic efforts had been made by repeated punctions for the laryngeal cystic swelling, inspiratory dyspnea did not improve. Total removal of laryngocele by laryngomicro-surgery seems to be appropriate for this lesions.
