We investigated whether the expression of progesterone-binding sites on human sperm acrozomal membrane is correlated with in vitro fertilization outcome. In motile sperm we evaluated percentage of progesterone-binding sperm (%PB) and of acrozome-reacted sperm (%AR), collected from 39 male partners undergoing an in vitro fertilization program (IVF), by labeling the sperm with progesterone-conjugated BSA-labeled FITC (P-BSA-FITC) and pisum sativum agglutinin labeled-FITC (PSA-FITC). Mean %PB in 30 fertilized cases was significantly higher than that in 9 fertilization failures (9.0 +/- 5.1 vs. 22.3 +/- 14.8%, p < 0.01). Furthermore, %PB shows a strong positive correlation with fertilization rate in IVF (r = 0.44) and %AR (r = 0.62), but not with sperm concentration and sperm motility. These results suggest that the expression of progesterone receptor on the sperm was related to the sperm fertilizability, and the evaluation of %PB was useful for predicting the fertilization outcome of IVF.

A large number of CD56(bright) natural killer (NK) cells, which comprise a very small fraction of peripheral blood lymphocytes, appear in the endometrium during the late secretory phase and early pregnancy. These cells are thought to immunologically maintain or inhibit pregnancy. However, the details regarding their contribution to the immuno-elimination systems of embryonic cells or decidual stromal cells remain unclear. Recently, leukocyte function-associated antigen-1 (LFA-1) was shown to play a critical role in the regulation of NK cytolysis in peripheral blood lymphocytes. We speculated that LFA-1 on the decidual CD56(bright) NK may be involved in the regulation of pregnancy. The expression of LFA-1 on the decidual CD56(bright) NK cells was analysed using flow cytometry with fluorescent monoclonal antibodies; CD56 (NKH1), CD16 (Fcg-R3) and CD11a (LFA-1 alpha-chain). In comparison with non-pregnant endometrium during the late secretory phase, the subpopulation of CD56(bright)CD16(-) cells in decidual lymphocytes was significantly increased during normal pregnancy, but was less than that in early pregnancy loss (P < 0.05). Furthermore, the number of CD56(bright) NK cells expressing LFA-1 was significantly higher in early pregnancy loss, and the late secretory phase, than during normal pregnancy (P < 0.05). Our results indicate that up-regulation of LFA-1 on CD56(bright) NK cells is related to spontaneous abortion or the onset of menstruation.

RESUMO

A micro-assay method for evaluating the allergenicity of soybean allergen was developed by using the mouse antiserum against Gly m Bd 30K, a major soybean allergen, and RBL-2H3, a rat mucosal mast cell line. The antiserum against Gly m Bd 30K was prepared by subcutaneously immunizing BALB/c mice with the allergen. The behavior by affinity-chromatography and the properties against heat treatment show that the reaginic activity of the antiserum resided in the IgE antibody specific for Gly m Bd 30K. The developed assay method is shown to be useful for simulating IgE mediated type-I allergy and to be highly sensitive for detecting the allergen.

RESUMO

We describe a case of gastric cancer with multiple liver metastasis that clearly decreased in size after administration of 5'-DFUR. The patient (a 63-year-old male) was inoperable because of multiple liver metastasis. 5'-DFUR 600 mg/day reduced the size of the multiple liver tumors (reduction rate 90%, partial response for four months), but the gastric tumor was unaltered. A rapid decrease in alkali-phosphatase indicated reduction of the liver tumors only one week after the initiation of 5'-DFUR.

1. The sulfite oxidases from various vertebrates, including rat, rabbit, chicken, frog, and eel, were partially purified and their physiochemical, kinetic, and immunochemical properties compared. The physicochemical and kinetic properties of these five enzymes were similar. 2. Antibody against the molybdenum-containing peptide prepared from purified rat liver sulfite oxidase was raised in rabbits. In Ouchterlony double diffusion and quantitative immunoprecipitation tests, the antibody could form precipitates with the enzymes from rats and rabbits, but no reaction was observed with enzymes from other sources. 3. The sulfite-cytochrome c reductase activity of the enzymes from these five animals were inhibited by the antibody, though the enzymes from chicken, frog, and eel were less sensitive to the antibody than those from rat and rabbit. 4. The inhibition of binding between 3H-labeled rat sulfite oxidase and its antibody by unlabeled enzymes from other animals demonstrated that 10 to 20% of the antibody could cross-react with these enzymes.

The surface charge of Bacillus megaterium QM B1551 spores was estimated to be negative, -0.2 ad -0.4 mueq/mg by colloidal titration using glycol chitosan (GCh) and methylglycol chitosan (MGCh), respectively, as positive colloids. MGCh, which reacts with all of the negatively charged groups including carboxylate, inhibited the second stage of the germination to result in semirefractile spores, but GCh, which reacts only with strong acidic groups such as phosphate, did not. The spores produced in a medium with limited phosphate had coats with low phosphate content and carried less negative charge, and they were induced to germinate with 0.4 mM KNO3, which is one-tenth of the minimum concentration required for the germination of the control spores. A similar increase in germinability was observed in spores incubated with calcium acetate. The results suggest that the role of the surface charge in germination is as follows. Strong acidic groups (such as phosphate) in the coat may block the action of ionic germinants and act as a barrier against the initiation of ionic germination. Positively charged compounds (such as calcium) may compensate for this blocking effect. Weak acidic groups (such as carboxylate) may be involved in the later stage of germination.

RESUMO

A case of agenesis of the left internal carotid artery with an aneurysm of the anterior communicating artery was reported in detail. The patient was a 48-year-old man who had an episode of subarachnoid hemorrhage about 10 years ago. This time left heminumbness and motor weakness of the left lower extremity occured and he was brought our hospital. Right common carotid angiography, left retrograde brachial serial angiography and aortography demonstrated an agenesis of the left internal carotid artery, an aneurysm of the anterior communicating artery, tortuous megadolic hobasilar artery and anomalies on the circle of Willis such as absence of supra optic portion of the right anterior cerebral artery, and large left posterior communicating artery. We confirmed latter findings by operation. After successful neck clipping of the aneurysm, his clinical findings were remarkably improved.
