This study aimed to assess the genetic differentiation and relationship among five sea cucumber species from the Red Sea in Egypt, namely Holothuria atra, H. impatiens, H. leucospilota, Actinopyga crassa and A. mauritiana, using Inter Simple Sequence Repeated (ISSR) and Start Codon Targeted (SCoT) markers. A collection of 100 specimens, with 20 individuals per species, was gathered for the analysis. With ten ISSR primers, 135 amplified bands were detected, including 11 distinct species-specific bands, indicating high-level polymorphism among species. Using ten SCoT primers, 151 amplicons were generated, including 30 species-specific bands, with 52% polymorphic bands indicating high-level polymorphism among species. The degree of genetic similarity (GS) among the different genotypes of species was calculated based on ISSR bands analysis, which ranged from 93% between H. atra and H. impatiens to 86% between H. atra and A. crassa. The highest genetic similarity was observed between H. atra and H. impatiens (90%), while the lowest was identified between A. crassa and A. mauritiana (75%) using SCoT bands. Notably, the ISSR and SCoT-based DNA analysis revealed similar genetic relationships between H. atra and H. impatiens compared to other sea cucumber species studied. This study provides new insights into the genetic diversity and relationship among sea cucumber species in the Red Sea, which could have implications for their conservation and management.

Catfish (Clarias gariepinus) meat isn't preferred by consumers in compared to other fish species meat, thus to enhance the use of catfish meat, ready to eat catfish fingers were prepared with some plants additives (amla and ginger powder). The mean objective of this study was to assess the impact of amla and ginger powder on catfish finger qualities in terms of physical, chemical, microbiological, and sensorial parameters under stored at a low temperature (5 ± 1 °C). The obtained results were compared with those of a control sample (basic formula) and a sample containing the synthetic antioxidant. During the storage period, the levels of pH, thiobarbituric acid, total volatile basic nitrogen, trimethylamine, total bacteria count, psychrophilic bacteria, molds and yeasts counts increased dramatically, although the values remained within acceptable ranges. The findings also revealed, that amla and ginger powder considerably (p < 0.05) reduced the changes in quality parameters, as well as there was a considerable increase in the quality parameter in all treated samples than in the control. Finally, amla and ginger powder can be a substitute for synthetic antioxidants and antimicrobials. These findings suggest that the powder of amla and ginger are suitable for use as a natural antioxidants and antimicrobials to extend the shelf-life of animal products.

Abstract Study aimed to evaluate the effect of replacing fish meal with different levels of full fat soybean meal (FFSBM) on growth and digestive enzyme activities in the stomach, Liver and intestine for Clarias gariepinus. Four diets (D1, D2, D3 and D4) were formulated with 0, 15, 20 and 20 g 100-1 protein + DL-methionine by alternating FFSBM with fish meal. The growth of C. gariepinus was found to be significantly decreased when FFSBM replacement increased. Final body weight was 89.69, 79.70, 70.82 and 68.29 g for fish fed on D1, D2, D3 and D4, respectively, with significant differences between treatments. Specific growth rate (SGR) ranged between 3.11 to 2.78%. Proteolytic activity was higher only with alkaline pHs, whereas only very low activity was shown with acidic. Results of liver showed approximately similar results at acid and alkaline. In contrast, higher proteolytic activity in the stomach was observed at acid pHs 3.0 and 4.0 μg tyrosine-1 minute-1 mg-1 protein) whereas lower values were observed at neutral pH 7.0 g tyrosine-1 minute-1 mg-1 protein for catfish fed on the experimental diets. Moreover, trypsin activity was higher for the stomach, followed by the intestine and liver. However, higher amount of amylase observed in the liver than intestine and stomach.


Resumo Objetivou-se avaliar o efeito da substituição da farinha de peixe por diferentes níveis de farelo de soja integral (FSI) sobre o crescimento e atividades de enzimas digestivas no estômago, fígado e intestino de Clarias gariepinus. Quatro dietas (D1, D2, D3 e D4) foram formuladas com 0, 15, 20 e 20 g 100-1 proteína + DL-metionina, alternando FSI com farinha de peixe. O crescimento de C. gariepinus foi significativamente reduzido quando aumentou a substituição de farinha de peixe por FSI . O peso corporal final foi de 89,69, 79,70, 70,82 e 68,29 g para peixes alimentados com D1, D2, D3 e D4, respectivamente, com diferenças significativas entre os tratamentos. Taxa de crescimento específico (TCE) variou entre 3,11 a 2,78%. A atividade proteolítica foi maior apenas com pHs alcalinos, enquanto que atividades muito baixas foram somente mostradas com ácido. Os resultados no fígado mostraram valores aproximadamente semelhantes tanto em meio ácido quanto alcalino. Em contraste, observou-se maior atividade proteolítica no estômago em pH ácido (3,0 e 4,0 g de tirosina-1 minuto-1 mg-1 de proteína), ao passo que valores mais baixos foram observados em pH neutro (7,0 g de tirosina-1 minuto-1 mg-1 de proteína) para os bagres que se alimentaram com as dietas experimentais. Além disso, a atividade da tripsina foi maior para o estômago, seguida pelo intestino e pelo fígado. No entanto, observou-se maior quantidade de amilase no fígado,intestino e estômago.

The current research aims to demonstrate the therapeutic effect of sour orange albedo extract (SOAE) and two flavanones loaded-tetraethylorthosilicate (TEOS) using sol-gel technique, in adose100 mg/kg body weight taken orally or45 days against acrylamide (ACR)toxicity in rats. This was achieved through measuring the activities of specific biochemical parameters related to liver functions in tissue of ACR intoxicated rats as compared to normal one. Liver functions included alanine and aspartate aminotransferases, antioxidants and oxidative stress biomarkers; superoxide dismutase, catalase, glutathione and lipid peroxide (malondialdehyde, MDA). Moreover, histological examination of liver was performed to confirm the biochemical findings. The present results clearly indicated disturbances in all biochemical parameters, such as increase in the liver function enzyme activities and MDA level. Results of ATPase enzyme activities revealed significant decrease in ACR intoxicated rats and liver biomarker enzymes declared significant decrease. On the other hand, treatment of intoxicated rats with the previous different nano-particles natural product demonstrated improvement in all biochemical parameters under investigation.

OBJECTIVE: To describe a case of adult-onset hypophosphatemic osteomalacia treated with orally administered phosphate and complicated by tertiary hyperparathyroidism. METHODS: We present pertinent clinical, radiologic, and laboratory details of the study patient for a period of more than 20 years and discuss the few reported cases of tertiary hyperparathyroidism attributable to prolonged phosphate therapy. RESULTS: A 49-year-old Jordanian man, who had been diagnosed at age 26 years as having sporadic adult-onset hypophosphatemic vitamin D-resistant osteomalacia, presented with severe right hip pain, severe osteopenia with lytic bone lesions, and hypercalcemia after prolonged oral treatment with phosphate and vitamin D. These clinical, radiologic, and biochemical findings, in conjunction with a very high serum parathyroid hormone level, indicated the diagnosis of tertiary hyperparathyroidism, which was substantiated histopathologically. CONCLUSION: Physicians should be aware of the potential for development of tertiary hyperparathyroidism in patients receiving prolonged oral phosphate therapy.

Wolfram syndrome (WS) is a recessively inherited disorder associated with recognised clinical features. Bleeding tendency was noticed in some of our patients, although this has not been reported before. We therefore studied this problem in all our WS patients and tried to postulate a possible pathogenesis. At the same time, a genetic linkage study provided evidence of locus heterogeneity of this syndrome and showed that the majority of our patients belong to the second WS locus identified in that study. Our study group consisted of 13 WS patients, belonging to WSF2 locus (group I). Controls consisted of 4 healthy siblings of WS patients (group II) and 7 diabetics who do not have WS (group III). Relevant clinical data were obtained, and a coagulation screen was carried out for all groups. All individuals in the three study groups have normal platelet count, thrombin time (TT), prothrombin time (PT), activated partial thromboplastin time (aPTT), clot retraction, Factor VIII activity (FVIIIc) and von Willebrand factor antigen (vWAg). Eleven of the WS patients have prolonged template bleeding time (BT) compared with both control groups. Patients with WS have a longer BT (mean 9.6 min, 95% CL 8.61-10.53 min) than the siblings group (mean 6.75 min, 95% CL 5.52-7.98 min) and the diabetic group (mean 5.49 min, 95% CL 4.56-6.42 min). The differences between the study group and controls are statistically significant, p = 0.02 and 0.0002, respectively. In the three groups, platelet aggregation studies were normal using adenosine diphosphate (ADP), ristocetin and epinephrine. Aggregation with collagen was either absent or impaired, with failure of secondary wave being noticed in 11 of the WS patients (85%) and normal in the control groups. The pathogenesis of this problem is not known, but could be due to an inhibitory effect of vWAgII, deficiency of thrombospondin or a defect in the platelet membrane GPIa/IIa. Bleeding diathesis is a new additional feature to the clinical spectrum of WS, which is probably a feature of the disorder WFS2 and not WFS1, as bleeding has never been reported in the latter. This provides further evidence for the phenotypic and genotypic heterogeneity of this complex disorder and may provide clues to the search for the second gene responsible for this phenotype.

To study the prevalence of recognized and unrecognized hyperglycaemia (defined as fasting blood glucose > or = 140 mg/dL and/or random blood glucose > or = 200 mg/dL on two or more occasions), data were collected from Jordan University Hospital on all hospital deaths in those > or = 20 years of age, for the years 1995-97. The prevalence of diagnosed diabetes was 35.3% and 19.2% were missed cases of hyperglycaemia; of the missed cases, 65.7% had a blood glucose level > 250 mg/dL. Thus, the prevalence of missed hyperglycaemia among hospitalized patients is high, and greater vigilance needs to be exercised by physicians and other health professionals in reviewing laboratory results, especially those related to diabetes.
