Renal transplantation is the preferred treatment option for patients with end-stage renal disease (ESRD) in a clinical setting. Antibody mediated rejection (AMR) is one of the leading causes of graft dysfunction. To address the current shortcomings in the early diagnosis and treatment of AMR in clinical practice, this article analyzes the distribution of different circulating T follicular helper (cTfh) cell subtypes and B cell subpopulations in peripheral blood and detects the cytokine levels of chemokine ligand 13 (CXCL13), interleukin-21 (IL-21), and interleukin-4 (IL-4) related to cTfh cells in peripheral blood of kidney transplant recipients. Moreover, we also explore the correlation between cTfh cells, peripheral blood memory B cells, and AMR, their value as early predictive indicators of AMR, and explore potential therapeutic targets for AMR patients. Our results indicate that the proportion of cTfh cells increased at the onset of AMR, which plays an important role in antigen-specific B-cell immune regulation. Activation of cTfh cells in AMR patients correlates with phenotypes of memory B cells and plasma blasts. cTfh cells and memory B cells have promising diagnostic efficacies and predictive values for AMR. The proportion of cTfh cells to CD4+ T cells and the proportion of memory B cells to CD19+ B cells are correlated with serum creatinine levels, indicating that cTfh cells and memory B cells may be involved in the progression of AMR. In addition, the CXCL13, IL-21, and IL-4, which were associated with cTfh cells, may be involved in the onset of AMR.

As a water-soluble polymer, the widely used polyvinyl alcohol (PVA) is produced from hydrolysis of polyvinyl acetate. Microbial PVA carbon backbone cleavage via a two-step reaction of dehydrogenation and hydrolysis has been well studied. Content of acetyl group is a pivotal factor affecting performance of PVA derivatives in industrial application, and deacetylation is a non-negligible part in PVA degradation. However, the genetic and biochemical studies of its deacetylation remain largely elusive. Here, Comamonas sp. strain NyZ500 was isolated for its capability of growing on acetylated PVA from activated sludge. A spontaneous PVA-utilization deficient mutant strain NyZ501 was obtained when strain NyZ500 was cultured in rich media. Comparative analysis between the genomes of these two strains revealed a fragment (containing a putative hydrolase gene dacApva ) deletion in NyZ501 and dacApva-complemented strain NyZ501 restored the ability to grow on PVA. DacApva, which shares 21% identity with xylan esterase AxeA1 from Prevotella ruminicola 23, is a unique deacetylase catalyzing the conversion of acetylated PVA and its derivatives to deacetylated counterparts. This indicates that strain NyZ500 utilizes acetylated PVA via acetate as a carbon source to grow. DacApva also possessed the deacetylation ability for acetylated xylan and the antibiotic intermediate 7-aminocephalosporanic acid (7ACA) but the enzymes for the above two compounds had no activities against PVA derivatives. This study enhanced our understanding of the diversity of microbial degradation of PVA and DacApva characterized here is also a potential biocatalyst for the eco-friendly biotransformation of PVA derivatives and other acetylated compounds.IMPORTANCE: Water-soluble PVA, which possesses a very robust ability to accumulate in the environment, has a very grave environmental impact due to its widespread use in industrial and household applications. On the other hand, chemical transformation of PVA derivatives is currently being carried out at high energy consumption and high pollution conditions using hazardous chemicals (such as NaOH, methanol) under high temperatures. The DacApva reported here performs PVA deacetylation under mild conditions, then it has a great potential to be developed into an eco-friendly biocatalyst for biotransformation of PVA derivatives. DacApva also has deacetylation activity for compounds other than PVA derivatives, which facilitates its development into a broad-spectrum deacetylation biocatalyst for production of certain desired compounds.

Mapping knowledge domain (MKD) is an important application of visualization technology in Bibliometrics, which has been extensively applied in psychology, medicine, and information science. In this paper we conduct a systematic analysis of the development trend on road safety studies based on the Science Citation Index Expanded (SCIE) and Social Sciences Citation Index (SSCI) articles published between 2000 and 2018 using the MKD software tools VOSviewer and Sci2 Tool. Based on our analysis, we first present the annual numbers of articles, origin countries, main research organizations and groups as well as the source journals on road safety studies. We then report the collaborations among the main research organizations and groups using co-authorship analysis. Furthermore, we adopt the document co-citation analysis, keywords co-occurrence analysis, and burst detection analysis to visually explore the knowledge bases, topic distribution, research fronts and research trends on road safety studies. The proposed approach based on the visualized analysis of MKD can be used to establish a reference information and research basis for the application and development of methods in the domain of road safety studies. In particular, our results show that the knowledge bases (classical documents) of road safety studies in the last two decades have focused on five major areas of "Crash Frequency Data Analysis", "Driver Behavior Questionnaire", "Safety in Numbers for Walkers and Bicyclists", "Road Traffic Injury and Prevention", and "Driving Speed and Road Crashes". Among the research topics, the five dominant clusters are "Causation and Injury Severity Analysis of Road Accidents", "Epidemiologic Study and Prevention of Road Traffic Injury", "Intelligent Transportation System and Active Safety", "Young drivers' driving behavior and psychology", and "Older drivers' psychological and physiological characteristics". Finally, the burst keywords in research trends include Cycling, Intelligent Transportation Systems, and Distraction.
