RESUMO

Gestational diabetes mellitus (GDM) has become a worldwide problem in recent years. Macrosomia, a primary consequence of GDM, has short-term and life-long consequences in the offspring of mothers with GDM. Our previous study showed that miR-517a was dysregulated in placenta and plasma of fetal growth restriction through inhibiting invasion of trophoblast and might be closely related with the regulation of birth weight by the placenta. To further investigate the mechanism of miR-517a, we conducted genome-wide microarray profile of lncRNAs. lncRNA-SNX17 was found to be significantly upregulated in the placenta of diabetic macrosomia by qRT-PCR, and the expression of miR-517a and IGF-1 were measured by qRT-PCR and Western blot. Interestingly, significant inverse correlations of the miR-517a with both lncRNA-SNX17 and IGF-1 expression were revealed in the placenta of diabetic macrosomia. Bioinformatic prediction also revealed that both lncRNA-SNX17 and IGF-1 possessed binding sites for miR-517a, which were then confirmed by luciferase report assay. LncRNA-SNX17 overexpression reduced the expression of miR-517a and increased the IGF-1 expression in HTR-8/SVneo human trophoblast cell line and thus enhanced the proliferation of HTR-8/SVneo. The enhancement of HTR-8/SVneo proliferation by lncRNA-SXN17 could be nullified by co-transfection of miR-517a mimics. The data suggested that lncRNA-SNX17 might promote the trophoblast proliferation through miR-517a/IGF-1 pathway and might play a role in the placentation of diabetic macrosomia.

OBJECTIVE@#To investigate a reliable clinical indication for predicting the therapeutic response of decitabine therapy in the patients with myelodysplastic syndromes (MDS).@*METHODS@#The clinical efficacy of decitabine for 55 cases of MDS was analyzed retrospectively. According to the lymphocyte level at d28 after the first time treatment with decitabine, the patients were divided into high lymphocyte level group (H-Lym≥1.2×10/L) and low lymphocyte level group (L-Lym<1.2×10/L), and the overall response rate (ORR) and the progression-free survival (PFS) time in 2 groups were compared.@*RESULTS@#As compared with L-Lym group, the ORR and PFS time in H-Lym group were significantly enhanced [(76.0% vs 50.0%) (P<0.05) and median time (15.7 months vs 8.5 months)(P<0.05), respectively];the ratio of platelet level ≥100×10/L in H-Lym group was very significantly higher than that in L-Lym group (72.0% vs 20.0%)(P<0.01). Multivariat analysis showed that the risk of disease progression in L-Lym group was 4.45-fold of H-Lym group (95% CI:1.58-12.59)(P<0.05).@*CONCLUSION@#The patients with lymphocyte level ≥1.2×10/L at day 28 after the first time treatment with decitabine show the higher ORR and longer PFS time, therefore. the lymphocyte level at day 28 after first time treatment with decitabine can be used as an early clinical indicator for predecting the response to decitabine treatment.

Objective To explore the main characteristics and influencing factors of military pharmacy postgraduates' creative ability,providing the basis for the creative ability training and education management of military pharmacy postgraduate students.Methods Full-time graduate students of three military medical colleges and universities were participated in this study,and questionnaire survey was carried out on 428 military pharmacy postgraduates by cluster random sampling.SPSS 16.0 was used to process descriptive analysis,t-test,F-test,correlation analysis and multiple regression analysis for data analysis.Results The creative ability of military pharmacy postgraduates is better than average (the median is 3),and the ability to construct knowledge was significantly higher than that of finding problems,solving problems and the ability of improvement and transformation (F=39.024,P<0.001).The creative ability of doctoral students in grade three was higher than that of the master students in all grades (F=2.602,P<0.05).The extrinsic motivation was significantly higher than intrinsic motivation (t=4.327,P<0.001),and the interaction between teachers and students was significantly higher than that among students them-selves (t=6.755,P<0.001).On the institutional category,it showed statistically significant in the training concept,the training subject,the training condition,the intrinsic motivation,the interaction between teachers and students,and the interaction among students themselves (t=-1.962,P<0.05;t=2.697,P<0.01;t=-2.593,P<0.01;t=2.938,P<0.01;t=4.595,P<0.001;t=2.984,P<0.01).The significant predictive effects on improving the creative ability of military pharmacy postgraduates were shown by training concept,training methods,academic atmosphere,intrinsic motivation,interaction between teachers and students,and the interaction among students themselves (F=48.711,P<0.001).Conclusion The creative ability of military pharmacy postgraduates is in the middle level,and the important influencing factors lie in themselves and schools.The education departments should construct the innovation ability training model from the concept of innovation training,teaching methods,scientific research team,atmosphere of innovation,incentive mechanism and other aspects.

China contributes 23 % of global carbon emissions, of which 26 % originate from the household sector. Due to vast variations in both climatic conditions and the affordability and accessibility of fuels, household carbon emissions (HCEs) differ significantly across China. This study compares HCEs (per person) from urban and rural regions in northern China with their counterparts in southern China. Annual macroeconomic data for the study period 2005 to 2012 were obtained from Chinese government sources, whereas the direct HCEs for different types of fossil fuels were obtained using the IPCC reference approach, and indirect HCEs were calculated by input-output analysis. Results suggest that HCEs from urban areas are higher than those from rural areas. Regardless of the regions, there is a similarity in per person HCEs in urban areas, but the rural areas of northern China had significantly higher HCEs than those from southern China. The reasons for the similarity between urban areas and differences between rural areas and the percentage share of direct and indirect HCEs from different sources are discussed. Similarly, the reasons and solutions to why decarbonising policies are working in urban areas but not in rural areas are discussed.
