OBJECTIVES: The aim of this study was to investigate the protective effects of a sustained release form of dexamethasone (dex) loaded chitosan-based genipin-cross-linked hydrogel (CBGCH) in a guinea pig model of cisplatin (CP) induced hearing loss. METHODS: Implantation of CBGCH was made by intratympanic (IT) injection. Ototoxicity was produced by intraperitoneal (IP) single dose of 14 mg/kg CP. Animals were randomly divided into four groups with 6 guinea pigs in each. Group 1 received only IP CP; group 2 received only IT dex-loaded CBGCH injections. Group 3 and group 4 received IP CP, plus IT nondrug CBGCH and IT dex-loaded CBGCH respectively 24 h prior to IP CP injections. Distortion product otoacoustic emissions (DPOAEs) and auditory brainstem response (ABR) measurements were obtained before the treatments and solely ABR measurements were done after 3 and 10 days. The ultrastructural effects were investigated by scanning electron microscopy (SEM) analysis. RESULTS: The postCP ABR thresholds at 4, 8, 12, 16, 32 kHz frequencies were significantly better in group 4 than groups 1 and 3 (p < 0.05). The comparison of time effective ABR thresholds between groups 1 and 4 and between groups 3 and 4 showed significantly lower ABR thresholds in group 4 (p < 0.05). The SEM analysis showed that stereocilia of inner and outer hair cells were preserved in group 4, almost like group 2, whereas cytotoxic degenerations were noted in groups 1 and 3. CONCLUSIONS: Intratympanic administration of dex-loaded CBGCH has been shown to provide functional and structural protection against CP-induced ototoxicity.

The capacities of montmorillonite K10 (K10), aluminum pillared K10 (Al-K10), and iron pillared K10 (Fe-K10) to eliminate zearalenone (ZEN) from synthetic media and the aqueous part of canned corn were studied. Original clay and pillared clays were characterized in terms of X-ray powder diffraction analysis and N(2) adsorption-desorption isotherms. The maximum amounts of adsorption of ZEN by K10, Al-K10, and Fe-K10 at 25°C and pH 7 were 0.202, 1.305, and 1.028 mg/g and 0.264, 0.096, and 0.255 mg/g, calculated from Langmuir and Freundlich isotherms, respectively. The adsorption of ZEN was also studied as a function of adsorbent amount (1 to 30 mg), ZEN concentration (2 to 20 mg/liter), pH of solution (pH 4 to 10), and contact time. Pillared clays could be an excellent alternative for removing ZEN in contaminated food samples and are potentially low-cost adsorbents with a promising future as an alternative to more costly materials.

Many algae contain secondary metabolites with the potential to gain importance as pharmaceutically active secretions. Caulerpa racemosa var. cylindracea and Caulerpa prolifera are very abundant on the Mediterranean coastlines. The methanolic extracts of C. racemosa and C. prolifera were tested for inhibitory effects on soybean lipoxygenase. The extract of C. prolifera showed potent inhibitory effect in a lipoxygenase enzyme activity assay. HPLC comparison revealed that C. racemosa extract contained less caulerpenyne, the major secondary metabolite of both algae. In accordance with these findings, purified caulerpenyne inhibited lipoxygenase with an IC(50) of 5.1 µM. The enzyme kinetic studies indicated that both K (M) and V (max) decreased from 0.041 to 0.019 mM and 312.5 to 151.5 U mL(-1) in the presence of 5 µM caulerpenyne, revealing an un-competitive type of inhibition kinetics. The major secondary metabolite of Caulerpa species, caulerpenyne, is thus a novel lipoxygenase inhibitor that can be easily obtained in high quantities from the abundant algae.

Adsorption method may play an important role to remove ochratoxin A (OTA) from wine by bentonite (B), nonylammonium bentonite (NB), dodecylammonium bentonite (DB), KSF-montmorillonite (KSF), and chitosan bead (CB). The optimum conditions of OTA adsorption from synthetic solutions were revealed at room temperature and pH 3.5. The adsorption equilibria of B and NB were almost established within 120 and 240 min, respectively. DB, KSF, and CB had about 90 min of equilibration time. The adsorption efficiency carried out in the synthetic OTA solution did not change remarkably when the amounts of adsorbents were 25 mg for bentonite, DB, and KSF and 100 mg for NB and CB. Furthermore, 25 mg of adsorbents was used at all adsorption studies in synthetic solution. The adsorption isotherm was fitted with mostly a Freundlich equation with respect to the correlation coefficients. The adsorption data were evaluated using Langmuir and Freundlich equations having Kf values ranging from 0.011 to 9.5 with respect to correlation coefficients (R2 = 0.900-0.977). DB and KSF have the highest adsorption capacity for OTA in synthetic solutions. In wine, the removal of OTA was succeeded at a percentage of 60-100 by KSF and CB. Furthermore, the highest adsorption capacity of OTA for red wine was obtained by using 250 mg of KSF, which caused less damage to the nature of wine and also low adsorption of polyphenols and anthcyans.

Chitosan bead was synthesized for the removal of a cationic dye malachite green (MG) from aqueous solution. The effects of temperature (303, 313 and 323 K), pH of the solution (2-11) on MG removal was investigated. Preliminary kinetic experiment was carried out up to 480 min. The sorption equilibrium was reached within 5 h (300 min). In order to determine the adsorption capacity, the sorption data were analyzed using linear form of Langmuir and Freundlich equation. Langmuir equation showed higher conformity than Freundlich equation. Ninety-nine percent removal of MG was reached at the optimum pH value of 8. From kinetic experiments, it was obtained that sorption process followed the pseudo-second-order kinetic model. This study showed that chitosan beads can be excellent adsorbents at high pH values. Activation energy value for sorption process was found to be 85.6 kJ mol(-1). This indicates that sorption process can be assumed as chemical process. Due to negative values of Gibbs free energy, sorption process can be considered as a spontaneous. In order to determine the interactions between MG and chitosan bead, FTIR analysis was also conducted.

This study presents the adsorption of cationic drug, promethazine hydrochloride from aqueous solution onto K10 montmorillonite. The effects of pH and temperature on adsorption process were investigated. Maximum adsorption pH was obtained to be about 7.5. Thermodynamic parameters found in this study depict the exothermic nature of adsorption. The process was favorable and spontaneous. From kinetic studies, it was found that adsorption process obeyed the pseudo-second-order kinetic model. The Langmuir, Freundlich, Dubinin-Radushkevich (DR) models were applied to describe the equilibrium isotherms and the isotherm constants were determined. The fit of the Langmuir and DR models appeared to be good. Physisorption mainly controls the whole adsorption process but chemisorption also shows a particular contribution.

In order to increase the adsorption capacities of bentonite, sepiolite, and illite for the removal of boron form aqueous solution, the clay samples were modified by nonylammonium chloride. Specific surface areas of the samples were determined as a result of N2 adsorption-desorption at 77 K using the BET method. X-ray powder diffraction analysis of the clays and modified clays was used to determine the effects of modifying agents on the layer structure of the clays. The surface characterization of clays and modified clay samples was conducted using the FTIR technique before and after the boron adsorption. For the optimization of the adsorption of boron on clays and modified clays, the effect of pH and ionic strength was examined. The results indicate that adsorption of boron can be achieved by regulating pH values in the range of 8-10 and high ionic strength. In order to find the adsorption characteristics, Langmuir, Freundlich, and Dubinin-Radushkevich adsorption isotherms were applied to the adsorption data. The data were well described by Freundlich and Dubinin-Radushkevich adsorption isotherms while the fit of Langmuir equation to adsorption data was poor. It was reached that modification of bentonite and illite with nonylammonium chloride increased the adsorption capacity for boron sorption from aqueous solution.

The montmorillonite has been subjected to modification through ion-exchange reaction by N,N'-didodecyl-N,N'-tetramethylethanediammoniumdiiodide (DEDMAI). The modified sample was studied by X-ray diffraction (XRD), Fourier transform infrared (FTIR) spectroscopy, and thermogravimetric analysis (TGA). The basal spacing of modified montmorillonite was determined as 28.72 A. The IR spectra of organomontmorillonite showed changes in C-H vibrations. The characterization of N,N'-didodecyl-N,N'-tetramethylethanediammonium montmorillonite (DEDMAM) and the adsorption of 2,4-dichlorophenoxyacetic acid (2,4-D), 2,4-dichlorophenoxybutyric acid (2,4-DB), 4-chloro-2-methylphenoxyacetic acid (MCPA), and 2,4,5-trichlorophenoxyacetic acid (2,4,5-T) on organomontmorillonite was studied as a function of the solution concentration. The adsorption energy (E) and adsorption capacity (qm) for phenoxyalkanoic acid herbicides adsorbing on organomontmorillonite (DEDMAM) were estimated using the Dubinin-Radushkevic (D-R) equation. These isotherms were modeled according to Freundlich and Dubinin-Radushkevic adsorption isotherms. The sorption of the herbicides on DEDMAM increased in order of MCPA<2,4-D<2,4-DB<2,4,5-T.

In this study, adsorption of boron onto Siral 5, Siral 40, and Siral 80 samples was studied in a batch system with a function of temperature and contact time. The results of adsorption studies indicate that adsorption process was well described with the Freundlich equation and Dubinin-Radushkevich (DR) equation. The Langmuir model does not appear to fit the adsorption better than Freundlich or DR. It has also been found that the magnitudes of free energies are less than 8 kJ mol(-1) and within the energy range of physical adsorption. The intraparticle diffusion model and pseudo-first-order and pseudo-second-order kinetic model were tested to find out rate constants of adsorption. From kinetic experiments, it is realized that boron adsorption onto Siral samples obeyed the pseudo-second-order kinetic model. The thermodynamic studies showed that adsorption process was not spontaneous nature and endothermic. It could also be added that the randomness increased during the adsorption.

The adsorption of 2,4-dichlorophenoxyacetic acid (2,4-D) on organophilic sepiolite (dodecylammonium sepiolite, DAS) was studied as a function of solution concentration and temperature. The observed adsorption rates were found to be equal to the first-order kinetics. The rate constants were calculated for temperatures ranging between 25 and 40 degrees C at constant concentration. The adsorption energies, E, and adsorption capacity, q(m), for 2,4-D adsorption on organophilic sepiolite was estimated using the Dubinin-Radushkevic equation. Thermodynamic parameters (Deltag(a), Deltah(a), Deltas(a)) were determined by a new approximation from the isotherm of 2,4-D adsorption on DAS. Also, DeltaS(0) and DeltaH(0) values were calculated from the van't Hoff equation. These isotherms were modeled according to the Freundlich and Dubinin-Radushkevic adsorption equations. The amount of adsorption of this herbicide on organophilic sepiolite was found to be dependent on the relative energies of adsorbent-adsorbate, adsorbate-solvent, and adsorbate-adsorbate interaction.

Phospholipid and triacylglycerol fatty acid compositions of whole animals from all life stages of Eurygaster integriceps, including eggs, nymphs, pre-diapausing adults and diapausing adults, were determined. The fatty acid composition of total lipids of their food, wheat, was also determined. The major components of the insects and their food were the expected C16 and C18 saturated and unsaturated fatty acids. Since fatty acid compositions of third-stadium nymphs were not similar to the profiles of their food, most likely, dietary fatty acids are modified by the insect. The fact is that the food does not provide C20 polyunsaturated fatty acids, but the insect tissue lipids include these components. We suggest biosynthesis of the C20 components by elongation/desaturation of C18:2n-6, an abundant component of the diets. We also show differences in fatty acid profiles from each of the life stages.
