Climate change is considered to be the primary cause of heat stress (HS) in broiler chickens. Owing to the unique properties of extracted polyphenols, resveratrol-loaded liposomal nanoparticles (Resv-Lipo NPs) were first explored to mitigate the harmful effects of HS. The dietary role of Resv-Lipo NPs in heat-stressed birds was investigated based on their growth performance, antioxidative potential, and the expression of heat shock proteins, sirtuins, antioxidant, immune, and muscle-building related genes. A total of 250 1-day-old Ross 308 broiler chickens were divided into five experimental groups (5 replicates/group, 10 birds/replicate) for 42 days as follows: the control group was fed a basal diet and reared in thermoneutral conditions, and the other four HS groups were fed a basal diet supplemented with Resv-Lipo NPsI, II, and III at the levels of 0, 50, 100, and 150 mg/kg diet, respectively. The results indicated that supplementation with Resv-Lipo NP improved the growth rate of the HS group. The Resv-Lipo NP group showed the most significant improvement in body weight gain (p < 0.05) and FCR. Additionally, post-HS exposure, the groups that received Resv-Lipo NPs showed restored functions of the kidney and the liver as well as improvements in the lipid profile. The restoration occurred especially at higher levels in the Resv-Lipo NP group compared to the HS group. The elevated corticosterone and T3 and T4 hormone levels in the HS group returned to the normal range in the Resv-Lipo NPsIII group. Additionally, the HS groups supplemented with Resv-Lipo NPs showed an improvement in serum and muscle antioxidant biomarkers. The upregulation of the muscle and intestinal antioxidant-related genes (SOD, CAT, GSH-PX, NR-f2, and HO-1) and the muscle-building genes (myostatin, MyoD, and mTOR) was observed with increasing the level of Resv-Lipo NPs. Heat stress upregulated heat shock proteins (HSP) 70 and 90 gene expression, which was restored to normal levels in HS+Resv-Lipo NPsIII. Moreover, the expression of sirtuin 1, 3, and 7 (SIRT1, SIRT3, and SIRT7) genes was increased (p < 0.05) in the liver of the HS groups that received Resv-Lipo NPs in a dose-dependent manner. Notably, the upregulation of proinflammatory cytokines in the HS group was restored in the HS groups that received Resv-Lipo NPs. Supplementation with Resv-Lipo NPs can mitigate the harmful impact of HS and consequently improve the performance of broiler chickens.

The current study was performed to investigate the toxic effects of aflatoxin B1 (AFB1) through the evaluation of kidney function tests and histopathological examination of renal tissues, targeting the therapeutic role of Marjoram (Origanum vulgare essential oil-OEO) in improving health status. Forty-eight New Zealand Whites growing rabbits (four weeks old) weighing on average 660.5 ± 2.33 g were randomly and equally distributed into four groups, each of which had four replicas of three animals as the following: Control group (only basal diet), AFB1 group (0.3 mg AFB1/kg diet), OEO group (1 g OEO/kg diet) and co-exposed group (1 g OEO/kg + 0.3 mg AF/kg diet). Our study lasted eight weeks and was completed at 12 weeks of age. The results revealed that OEO decreased the toxic effects of AFB1 in rabbit kidneys by substantially reducing the cystatin C levels in the AFB1 group. Additionally, OEO decreased oxidative stress and lipid peroxidation levels in the co-exposed group. Moreover, OEO reduced DNA damage and inflammatory response in addition to the down-regulation of stress and inflammatory cytokines-encoding genes. Besides, OEO preserved the cytoarchitecture of rabbits' kidneys treated with AFB1. In conclusion, O. vulgare essential oil supplementation ameliorated the deleterious effects of AFB1 on the rabbits' kidneys by raising antioxidant levels, decreasing inflammation, and reversing oxidative DNA damage.

Oxidative stress is considered the main etiologic factor involved in inflammatory bowel disease (IBD). Integration of nanocarriers for natural therapeutic agents with antioxidant and anti-inflammatory potential is a novel promising candidate for curing IBD. Herein, the colonic antioxidant and anti-inflammatory effects of different concentrations of quercetin nanoparticles (QT-NPs) were evaluated using a dextran sulfate sodium (DSS)-induced colitis model. Following colitis induction, the efficacy and mechanistic actions of QT-NPs were evaluated by assessing lesion severity, molecular aids controlling oxidative stress and inflammatory response, and histopathological and immunohistochemistry examination of colonic tissues. Administration of QT-NPs, especially at higher concentrations, significantly reduced the disease activity index and values of fecal calprotectin marker compared to the colitic group. Colonic oxidant/antioxidant status (ROS, H2O2, MDA, SOD, CAT, GPX and TAC) was restored after treatment with higher concentrations of QT-NPs. Moreover, QT-NPs at levels of 20 mg/kg and, to a lesser extent, 15 mg/kg reduced Nrf2 and HO-1 gene expression, which was in line with decreasing the expression of iNOS and COX2 in colonic tissues. Higher concentrations of QT-NPs greatly downregulated pro-inflammatory cytokines; upregulated genes encoding occludin, MUC-2 and JAM; and restored the healthy architectures of colonic tissues. Taken together, these data suggest that QT-NPs could be a promising alternative to current IBD treatments.

Optimal combinations of essential oils (EOs) can enhance performance and maintain poultry productivity. The effects of EOs with black pepper oil (BPO) or radish seed oil (RSO) on performance and the expression of digestive enzymes, lipogenesis, immunity, and autophagy-related genes in broiler chickens were explored. Six dietary treatments for 300 one-day-old chicks were allocated as follows: controls were fed a basal diet, one group was fed an EO-supplemented diet (1.5 g/kg diet of parsley, mint, and carrot seed oils (1:1:1)), and other groups received Eos + BPO0.25, Eos + BPO0.5, Eos + RSO0.25, and Eos + RSO0.5 treatments, with a basal diet containing EOs plus BPO or RSO at the level of 0.25 or 0.5 g/kg, respectively. Supplementation with 0.5 g/kg of EOs plus BPO or RSO resulted in the most improved maximum BWG and FCR in broiler chickens. The lactobacilli population was increased in Eos + BPO0.5, followed by EOs + RSO0.5, unlike in the control. The highest expression of the CCK and PNLIP genes was identified in the Eos + BPO group. The FAS and ACC genes were upregulated, while the IgA and IL-10 genes were downregulated, with EOs plus RSO or BPO. The group that received Eos + BPO0.5, followed by Eos + RSO0.5, displayed patterns of higher expression for atg5, atg7, and atg12, with lower expression of mTOR. In summary, a new combination of EOs with 0.5 g/kg BPO had potential growth-promoting and immune-boosting effects in broiler chickens.

Insecticides are widely employed in agriculture to control pests and as major factors for enhancing crop productivity. Thiacloprid (TH) is one of the most-used insecticides worldwide. In this study, the negative impact of TH on the brain tissue of developing chicken embryo models and the modulatory effect of chicoric (CA) and rosmarinic (RA) acids were investigated. The eggs were injected in ovo with different doses of TH (0.1, 1, 10, and 100 µg/egg). TH significantly increased the oxidative damage in the brain of exposed embryos in a dose-dependent manner (p < 0.05). TH significantly elevated the oxidative stress markers; protein carbonyl, malondialdehyde content, and DNA damage (p < 0.05). Myeloperoxidase activity and nitric oxide significantly increased with overexpression of the pro-inflammatory cytokines (interferon gamma, tumor necrosis factor alpha, and interleukin-1 beta) and stress-related and apoptotic genes (NF-KB, Caspase-3) in the brain tissue on both biochemical and molecular levels (p < 0.05), while downregulating the expression of antiapoptotic Bcl-2. Co-treatment of CA and RA with TH markedly decreased the insecticide-induced toxicity with a prominent synergistic effect (p < 0.05). In conclusion, TH is suggested to be a possible neurotoxic to embryos of vertebrates including human. The study also revealed the antioxidant, anti-inflammatory, genoprotective, and antiapoptotic property of CA and RA against TH toxicity.

Zinc (Zn) is an important trace element in fish diets that is required for growth, immunity and antioxidant defense mechanisms. The current study assessed the effects of both organic and nanoparticle zinc oxide (ZnO and ZnO-NPs, respectively) on growth performance, immune response and the antimicrobial effect against Pseudomonas aeruginosa in African catfish Clarias gariepinus. Fish were fed either a control diet or diets supplemented with organic ZnO at concentrations of 20 and 30 mg kg-1 or ZnO-NPs at concentrations of 20 and 30 mg kg-1. After 60 d, a subset of the fish was injected intraperitoneally with 3 × 107 CFU ml-1 of P. aeruginosa. Results showed that body weight gain, feed conversion ratio and specific growth rates were significantly increased in ZnO-NPs20 compared to all other groups. The dietary supplementation with 20 mg kg-1 of ZnO-NPs improved the antioxidant status of fish. Moreover, IgM, lysozyme and nitric oxide showed a significant increase in the fish which received the ZnO-NPs20-supplemented diet. A significant upregulation of growth and stress-related genes was seen in the ZnO-NPs20-supplemented group compared to other groups. However, there was no significant difference in the expression of immune-related genes among ZnO-NPs20, ZnO-NPs30 and ZnO30 groups. These findings highlight the potential use of nano-ZnO for improving growth performance, antioxidant status, immunological status and antibacterial activity against P. aeruginosa in African catfish.

The current study was conducted to investigate the impact of thermal stress on growth performance, blood biomarkers, metabolic hormones, immunological response, antioxidant activity, and expression of stress-related genes in broilers. One hundred and fifty one-day-old chicks (Ross 308) were utilized in this work. On the 21st day of age, birds were subjected to three environmentally controlled treatments with five replicate pens of 10 birds per each, representing an initial density of 10 birds/m2-control: reared in a thermoneutral condition; THS1 and THS2: exposed to 4 and 6 h of daily thermal stress at 40 ± 1 °C, respectively, until the 42 days of age. The results demonstrated that thermal stress for 4 and 6 h significantly reduced (P < 0.001) daily weight gain, daily feed intake, blood leukocyte and lymphocyte counts, serum immunoglobulins (IgM, IgA, IgG), and insulin-like growth factor-1 (IGF-1), while serum levels of aspartate aminotransferase, alanine aminotransferase, glucose, cholesterol, low-density lipoprotein, and lactate dehydrogenase were elevated relative to the thermoneutral group. Additionally, the corticosterone level and the ratio of heterophil:lymphocyte increased significantly (P < 0.001) in thermal-stressed groups. The antioxidant enzymes were affected by thermal stress as represented by a significant decrease in the activity of serum catalase (CAT) and glutathione peroxidase (GSH-Px) along with an increase in malonaldehyde concentration. Thermal stress affected gene expression by upregulating heat shock protein 70, heat shock factors 1 and 3, nuclear factor kappa B, interleukin-4, and uncoupling protein, and downregulating GSH-Px, CAT, and IGF-1 transcript levels. However, no changes were observed in interleukin-2 expression levels. It can be concluded that thermal stress destructively influences productivity, physiological status, and gene expression by upregulating heat shock protein 70, heat shock factors 1 and 3, nuclear factor kappa B, interleukin-4, and uncoupling protein, and downregulating GSH-Px, CAT, and IGF-1 transcript levels of broiler chickens.

Vitamin K3, also known as menadione, is a synthetic lipid-soluble 2-methyl-1,4- naphthoquinone analogs of vitamin K. The vitamin K derivatives exhibit potent cytotoxicity against several cancer cell lines through ROS induction and mitochondrial dysfunction. We investigated vitamin K3-inspired derivatives as potential apoptotic inducers and analyzed their mechanisms beyond apoptosis. The cytotoxicity of a panel of vitamin K3 analogs was screened against 10 doxorubicin-sensitive and -resistant cancer cell lines overexpressing ATP-binding cassette transporters (P-glycoprotein, ABCB5, BCRP) or oncogenes (ΔEGFR) or with knockout of tumor suppressors (p53), Cell cycle arrest, apoptosis, cell migration, and microtubule formation were further investigated. The online tool SwissTargetPrediction was utilized for target prediction. Among the screened compounds, one vitamin K3 thio-derivative (No. 45, VKT-1) exhibited the most potent cytotoxicity specifically against both drug-sensitive and -resistant cancer cell lines. In addition, VKT-1 arrested the cells at the G2/M phase and induced apoptosis as detected by flow cytometry. As predicted by SwissTargetPrediction, VKT-1 targeted microtubule-associated tau protein. Indeed, VKT-1 dramatically inhibited cell migration and microtubule formation in vitro. In conclusion, the synthetic vitamin K3 thio-derivative (VKT-1) inhibited doxorubicin-sensitive and -resistant tumor cells by cell arrest, apoptosis induction, as well as, migration inhibition, and microtubule deterioration of U2OS-GFP-α-tubulin cells.

The present investigation was performed to evaluate the efficiency of Zinc oxide (ZnO) as a fish feed additive in immunomodulation of Oreochromis niloticus. Fish were fed on ZnO nano-particles (nZnO) and conventional (ZnO) in two concentrations (30 and 60 mg/kg diet), in addition to the control fish which was fed on Zn free diet. After 6° days, the highest survival rate was recorded in the nZnO30 -supplemented group. The total antioxidant capacity (TAC) and antioxidant enzymes were improved in different dietary Zn supplementation, obviously in the nZnO30 -supplemented group, while the lowest antioxidant status was noticed nZnO60 supplemented fish. The lipid peroxides (MDA) level was diminished upon Zn supplementation, particularly in nZnO30-supplemented group but showed a significant elevation in the nZnO60-supplemented group. Furthermore, the immune parameters examined, lysozyme activity, bactericidal activity, and IgM were significantly higher in ZnO60, and nZnO30 supplemented groups. The C-reactive protein (CRP) level showed no significant increase in response to Zn supplementation in the both forms at level of 30 mg/kg diet, but showed marked elevation in nZnO60- supplemented group. The mRNA expression profile of both interleukin 8 (IL-8), interleukin 1, beta (IL-1ß) encoding genes showed an up-regulation that was found in all Zn- supplemented groups, but more pronounced in nZnO60-supplemented group. On the other hand, the expression pattern of myxovirus resistance (Mx)-encoding gene showed no remarkable difference between the Zn- supplemented and control fish. The expression level of CXC-chemokine, toll-like receptor 7 (TLR-7), immunoglobulin M heavy chain (IgM heavy chain) and interferon gamma (IFN-γ) gene was upregulated in Zn-supplemented groups particularly in the nZnO30- supplemented group. While, the lowest expression was found in nZnO60- and ZnO30-supplemented groups. Here, Zn supplementation promoted the immune and antioxidant strength in fish mainly in nano form at the level of 30 mg/kg diet but not at 60 mg/kg diet that disrupt the immune and antioxidant status and promote inflammatory response.

The phenylpyrazole insecticide, fipronil, isused for the eradication of insects in agriculture, which also exposes various non-target groups such as birds and animals. Our aim was to assess the cardiac and pulmonary consequences of sub-acute administration of fipronil (1∕5 LD50; 2.26 mg/kg) in the Japanese quail for fifteen days and to determine the tissue recovery over a period of 60 days. Fipronil exposure led to a significant decrease in the body weight of the treated birds. Its exposure also induced cardiac and pulmonary damage of varying degrees. Fipronil increased the lipid peroxide (LPO) and nitric oxide (NO) contents as well as indices of tissue injury in the serum of exposed birds. Furthermore, it decreased the antioxidant indices in both the organs. Most of these changes gradually reversed and the histological changes, particularly of the heart, reversed completely by day-60 of recovery. Furthermore, alterations in the mRNA gene expressions of Nuclear factor kappa B (NF-κB), Interleukin 6 (IL-6), and Tumor necrosis factor-alpha (TNF-α) were monitored by quantitative polymerase chain reaction (RT-PCR). In both the tissues, a significant up-regulation of the transcripts was recorded after fipronil administration, which was reversed during the recovery period in the heart tissue except for TNF-α, while the transcripts in the lung tissue declined non-significantly. This study showed that the exposure of Japanese quail to fipronil has a profound negative impact on heart and lung including oxidative injury and tissue inflammation. Fipronil can induce the activity of NF-κB inflammatory -signaling pathway that play a role in the associated tissue inflammation. Although most of the cardiac changes could be reversed after a recovery period of sixty days, the pulmonary changes did not reverse much.

The aim was to elucidate the effects of strain and the duration of thermal stress (36 °C) on the carcass yield, metabolic hormones, immunological parameters and the relative expression of liver heat shock protein 90 (HSP90) and Myogenin genes. Two hundred and seventy day-old chicks (135 of each breed; Ross 308 and Cobb 500) were used in this trial. The birds were divided into 3 equal groups (CON: thermoneutral condition; S1 and S2 groups were subjected to 4 and 6 h of daily thermal stress, respectively. The relative bursa weight in the thermonuteral Ross broilers, but not Cobb broilers was significantly greater than both heat-stressed groups (P = 0.001). The serum Na and phosphorus levels were significantly decreased when both broiler strains subjected to 6 h of thermals stress (P = 0.020 and 0.022, respectively). A linear decrease in the serum T3 level was recorded in both broiler strains as the duration of thermal stress increases (P = 0.001). In both broiler strains, heat stress up-regulate the expression of liver HSP90 gene compared with the thermoneutral group (P = 0.012). Meanwhile, the S2 group of Ross broilers showed a significantly lower IgG level (P = 0.027). Moreover, thermal stress for 6 h down-regulate the relative expression of Myogenin gene compared with the S1 and thermoneutral groups (P = 0.005). In conclusion, thermal stress for 6 h down-regulate the mRNA expression of liver Myogenin, concomitantly with an increase in the expression of HSP90 gene in both broiler strains. These results could be helpful in the markers assisted selection to develop more heat-tolerant broiler strains.

The effects of rutin on growth performance, hematological and biochemical profiles, antioxidant capacity, economics and the relative expression of selected antioxidants and lipid-related genes were studied in broiler chickens over 42 days. A total of 200 one-day-old female Ross-308 broiler chickens were distributed into four groups, with five replicates of 10 individuals per replicate. They were fed with 0 (control), 0.25, 0.5 or 1 g rutin/kg supplementation in their basal diet. Dietary rutin supplementation, especially the 1 g/kg diet, increased body weight gain, the protein efficiency ratio (p < 0.001) and both white blood cell and lymphocyte counts (p < 0.001). However, it had no effect on total protein, albumin, globulin, or alanine transaminase. A high concentration of rutin (0.5 and 1 g/kg) also significantly reduced serum total cholesterol, triacylglycerol and low-density lipoprotein cholesterol concentrations (p < 0.001), as well as malondialdehyde concentrations (p = 0.001). A high concentration diet also increased the activity of superoxide dismutase, catalase and glutathione peroxidase. Of the lipid-related genes examined, acetyl CoA carboxylase and fatty acid synthase were significantly down-regulated in the livers of rutin-fed individuals, whereas carnitine palmitoyl transferase 1 and peroxisome proliferator-activated receptor alpha were significantly up-regulated. Therefore, rutin supplementation at 1 g/kg has the potential to improve the productive performance and health status of broiler chickens.

The objective was to investigate the effects of genetic type and the duration of chronic thermal stress (36 °C) on the growing efficiency, carcass traits, antioxidant status, and the expression of liver heat shock protein 70 (HSP70), growth hormone (GH) and superoxide dismutase (SOD) genes. Two hundred and seventy one-day-old chicks (135 male chicks of each breed; Ross 308 and Cobb 500) were used in this work. On the 21st day of age, birds were allocated randomly into 3 equal groups till the 42 days of age (CON:raised in a thermoneutral condition; HS1 and HS2 groups were subjected to 4 and 6 h of daily thermal stress, respectively). Regardless of genetic type, thermal stress decreased the dressing percentage in broilers when compared with the thermoneutral conditions (p = 0.039). In both broiler strains, thermal stress for 6 h (HS2) increased the heterophil to lymphocyte ratio (p = 0.036) and the serum albumin, cholesterol and triglyceride levels (p = 0.023, 0.012 and 0.005, respectively) compared with the thermoneutral group. Under the thermonuteral and heat stress conditions, the Ross broiler chickens showed a significant lower serum triiodothyronine level compared with the Cobb boilers (p = 0.042). It is interesting to note that the expression of HSP70 in the liver of heat-stressed Ross broilers, either 4 or 6 h, was significantly (p = 0.002) higher than that reported in the heat-stressed Cobb broilers. In both broiler strains, the thermal stress for 6 h up-regulate the expression of SOD gene (p = 0.001), but down-regulate the expression of GH gene (p = 0.021) when compared with the CON group. In conclusion, chronic thermal stress down-regulate the mRNA expression of liver GH, concomitantly with an increase in the expression of HSP70 and SOD genes in both broiler strains. This could be useful in the identification of molecular genetic markers to assist in selecting broilers that are more tolerant to heat stress.

The aim of this study was to explore the potential effects of quercetin (QUR) on doxorubicin (DOX)-induced nephrotoxicity. Fifty male rats were assigned to five groups (10 rats each): a control group, a DOX-treated group (total dose, 15 mg/kg bw, intraperitoneally), a QUR-treated group (50 mg/kg bw/day, orally), a prophylaxis co-treated group, and a therapeutic co-treated group. Biochemical parameters and renal function were measured. Moreover, kidney tissues were homogenized for inflammatory marker evaluation and real-time qPCR analysis to determine the changes in intermediate filament protein mRNA levels (desmin, vimentin, connexin 43 and nestin). QUR exhibited a significant nephroprotective effect, particularly when it was administered prior to and simultaneously with DOX treatment (prophylaxis co-treated group). This role was biochemically demonstrated by the significant modulation of DOX-induced body weight loss, hypoproteinemia, and elevated serum creatinine and urea. Moreover, QUR attenuated the inflammatory response as shown by decreased renal nitric oxide, tumor necrosis factor-α production and myeloperoxidase activity elicited by DOX injection. These biochemical improvements were accompanied by a significant histopathological restoration of rat kidney tissue and successful down-regulation of the intermediate filament protein mRNA levels, indicating amelioration of DOX-induced podocyte injury. Taken together, these results conclusively demonstrated that QUR administration has a prophylactic effect on DOX-induced injury in the rat kidney.

Understanding the genetic information of related genes is helpful for the selection and breeding course through marker assisted selection. The aim of the current study was to detect polymorphisms of the KiSS1 gene in 137 animals, including Baladi, Zaraibi and Damascus goat breeds by PCR-RFLP, and DNA sequencing and to investigate the association between these variants and reproductive traits. Comparison of the nucleotide sequence indicated the substitution of T with A at position 121 (T121A) in the intron 1 of the KiSS1 gene in all goat breeds. This substitution distorts the restriction site of the XmnI restriction enzyme and consequently two genotypes were detected (TA and TT). The T121A SNP is associated significantly with litter size in Damascus and Zaribi breeds (p=0.025 and 0.001, respectively). The animals with the TT genotype in Damascus and Zaribi breeds had a significantly higher estradiol17ß level than that recorded in TA genotype at estrus phase (p=0.013 and 0.028, respectively) and late-luteal phase (p=0.067 and 0.041, respectively) of the estrus cycle. Furthermore, animals with the TT genotype in Damascus and Zaribi breeds had significant higher progesterone level at mid-luteal (p=0.037 and 0.045, respectively) phase. Meanwhile, there were no significant differences in progesterone level in late-luteal phase between both genotypes in Zaribi breed (p=0.267). The current trial indicated that the prolific TT genotype in both Damascus and Zaribi breeds had superior estradiol17ß level at estrus phase and an eminent progesterone level at both early and mid-luteal phases of the estrous cycle.
