RESUMO

Hydrazine is an alkaline reduction compound which is widely used in synthesis. Based on the structure-activity analysis, to elicit antitumor activity, the presence of the N-methyl group is an absolute requirement. The aim of the research is to synthesize a new hydrazine derivate compound that has potency as a novel anti-breast cancer. 4-hydrazinylphenyl benzenesulfonate was synthesized employing reduction and diazotization methods. Structure characterization was carried out using Fourier transform infrared (FTIR), C13-nuclear magnetic resonance (NMR), H1-NMR, and High Resolution Time-of-Flight Mass Spectrometry (HR-TOF-MS). The anti-cancer activity of this compound against breast cancer Michigan Cancer Foundation-7 (MCF-7) cell line was determined using a PrestoBlue viability assay. The new of hydrazine derivative, 4-hydrazinylphenyl benzenesulfonate, has been successfully synthesized. The reduction and diazotization methods have been successfully used in the synthesis of new compound of hydrazine derivatives. Structure characterization of 4-hydrazinylphenyl benzenesulfonate was established using FTIR, C13-NMR, H1-NMR, and HR-TOF-MS. The anti-cancer activity of this compound against breast cancer MCF-7 cell line was determined using a PrestoBlue viability assay with IC50 0.00246 µg/mL or 9.32 nM. In conclusion, 4-hydrazinylphenyl benzenesulfonate was successfully synthesized as a new candidate for anti-breast cancer compound.

BACKGROUND: The 2',4'-dihydroxy-6-methoxy-3,5-3-dimethylchalcone (ChalcEA) isolated from Eugenia aquea Burm f. leaves has potential anticancer activity against human breast-adenocarcinoma cell lines (MCF-7) with an IC50 value of 250 µM. However, its apoptotic activity on the T47D breast cancer cell lines which is involving caspase-3 has not been investigated. MATERIALS AND METHODS: Therefore, this study aims to evaluate the cytotoxicity of ChalcEA on the T47D cell lines using the 2-(4-iodophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfophenyl)-2H-tetrazolium (WST) method and to predict its possible antagonistic activity on the human estrogen receptor alpha (hERα) using pharmacophore and molecular dynamics (MD) methods. The in vitro test of 10 synthesized ChalcEA derivatives was also performed as an insight into the further development of its structure as an anticancer agent. RESULTS: It is shown that ChalcEA has an IC50 of 142.58 ± 4.6 µM against the hERα-overexpressed T47D breast cancer cell lines, indicating its possible mechanism of anticancer activity as an antagonist of hERα. Pharmacophore study showed that ChalcEA shares similar features with the known hERα antagonist, 4-hydroxytamoxifen (4-OHT), which has hydrogen bond donor (HBD), hydrogen bond acceptor (HBA), ring aromaticity (RA), and hydrophobicity (Hy) features. Molecular docking showed that ChalcEA formed hydrogen bonds with Glu353 and Arg394, and hydrophobic interactions in a similar manner with 4-OHT. Moreover, MD simulations showed that ChalcEA destabilized the conformation of His524, a remarkable behavior of a known hERa antagonist, including 4-OHT. Furthermore, the 10 best chalcone derivatives resulted from pharmacophore- and docking-based screening, were tested against the T47D cell lines. None of the derivatives have better activity than ChalcEA. It is suggested that the functional groups at the B-ring of ChalcEA are interesting to be further optimized in the next studies. CONCLUSION: ChalcEA might act as an antagonist toward hERα, thus warranting further investigation as a potential anticancer agent.

The dataset of curcumin derivatives consists of 45 compounds (Table 1) with their anti cancer biological activity (IC50) against P388 cell line. 45 curcumin derivatives were used in the model development where 30 of these compounds were in the training set and the remaining 15 compounds were in the test set. The development of the QSAR model involved the use of the multiple linear regression analysis (MLRA) method. Based on the method, r2 value, r2(CV) value of 0.81, 0.67 were obtained. The QSAR model was also employed to predict the biological activity of compounds in the test set. Predictive correlation coefficient r2 values of 0.88 were obtained for the test set.

The title compounds, C20H19NO3, (1), and C20H17Cl2NO, (2), are the 3-hy-droxy-benzyl-idene and 2-chloro-benzyl-idene derivatives, respectively, of curcumin [systematic name: (1E,6E)-1,7-bis-(4-hy-droxy-3-meth-oxy-phen-yl)-1,6-hepta-diene-3,5-dione]. The dihedral angles between the benzene rings in each compound are 21.07 (6)° for (1) and 13.4 (3)° for (2). In both compounds, the piperidinone rings adopt a sofa confirmation and the methyl group attached to the N atom is in an equatorial position. In the crystal of (1), two pairs of O-H⋯N and O-H⋯O hydrogen bonds link the mol-ecules, forming chains along [10-1]. The chains are linked via C-H⋯O hydrogen bonds, forming undulating sheets parallel to the ac plane. In the crystal of (2), mol-ecules are linked by weak C-H⋯Cl hydrogen bonds, forming chains along the [204] direction. The chains are linked along the a-axis direction by π-π inter-actions [inter-centroid distance = 3.779 (4) Å]. For compound (2), the crystal studied was a non-merohedral twin with the refined ratio of the twin components being 0.116 (6):0.886 (6).
