One of the most concerning pests that attack strawberries in Brazil is Duponchelia fovealis (Zeller), a non-native moth with no registered control methods to date. Our group recently observed that a fungal consortium formed by two strains of Beauveria bassiana (Balsamo) increased the mortality of D. fovealis more than inoculation with each strain on its own. However, the molecular interaction between the fungal consortium and the caterpillars is unknown. Thus, in this work, we sought to pioneer the evaluation of the molecular interaction between a fungal consortium of B. bassiana and D. fovealis caterpillars. We aimed to understand the biocontrol process involved in this interaction and the defense system of the caterpillar. Seven days after D. fovealis were inoculated with the consortium, the dead and surviving caterpillars were analyzed using GC-MS and LC-MS. Some of the metabolites identified in dead caterpillars have primarily antioxidant action. Other metabolites may have insecticidal potential, such as diltiazem-like and tamsulosin-like compounds, as well as 2,5-dimethoxymandelic acid. In surviving caterpillars, the main mechanisms are pro-inflammatory from 2-Palmitoylglycerol metabolite and the antifungal action of the metabolite Aegle marmelos Alkaloid-C. The metabolites identified in dead caterpillars may explain the increased mortality caused by the consortium due to its antioxidant mechanism, which can suppress the caterpillars' immune system, and insecticide action. In surviving caterpillars, the main resistance mechanisms may involve the stimulus to the immunity and antifungal action.

The spotted wing drosophila (Drosophila suzukii) is one of the main invasive pests of small fruits in the world. Thus, 19 essential oils (EOs) were selected to analyze the effects through toxicity and repellency on oviposition and D. suzukii adults. In addition, their lethal and sublethal effects on the pupal endoparasitoid Trichopria anastrephae were evaluated. The EOs of C. flexuosus and Mentha spp. had the highest toxicity observed in the topical application bioassay for D. suzukii. In contrast, the EOs of C. verum, C. citratus QT citratus, and C. winterianus showed the highest toxicity in the ingestion bioassay for D. suzukii. The dry residues of C. verum and C. citratus QT citratus reduced the oviposition of D. suzukii. In the repellency bioassays, the 19 EOs analyzed repelled ≅ 90% of the D. suzukii females. All EOs evaluated using the LC90 values of the products provided mortality of less than 20% of T. anastrephae adults and did not cause a reduction in the parasitism of surviving T. anastrephae females. We conclude that the EOs evaluated have the potential to be used in the management of D. suzukii. They can also serve as selective active ingredients for the formulation and synthesis of new biopesticides.

The use of two or more microorganisms in a microbial consortium has been increasingly applied in the biological control of diseases and pests. Beauveria bassiana is one of the most widely studied fungal species in biological control, yet little is known about its role in fungal consortiums. In a previous study, our group found that a consortium formed by two strains of B. bassiana had significantly greater biocontrol potential against the polyphagous caterpillars Duponchelia fovealis (Lepidoptera: Crambidae) than either strain on its own. In this study, we use GC-MS and LC-MS/MS to evaluate and discuss the metabolomics of the consortium. A total of 21 consortium biomarkers were identified, corresponding to 14 detected by LC-MS/MS and seven by GC-MS. Antioxidant and anti-inflammatory mechanisms are the main properties of the metabolites produced by the consortium. These metabolites can depress the insect's immune system, increasing its vulnerability and, hence, the fungal virulence of the consortium. In light of these results, we propose an action model of insect mortality due to the metabolites secreted by the consortium. The model includes the inhibition of defense mechanisms such as pro-inflammatory interleukin secretion, cell migration, cell aggregation, Dif, Dorsal and Relish gene transcription, and JAK/STAT and JNK signaling pathways. It also promotes the cleaning of oxidative molecules, like ROS, NOS, and H2O2, and the induction of virulence factors.

Heliothrips haemorrhoidalis is a polyphagous thrips species that has been reported as a potential pest in strawberry cultivation in Brazil. Since it has only recently emerged as a pest species in production areas, understanding seasonal trends in the species' activity is particularly important for designing management strategies. Therefore, this study aims to investigate the influence of climate factors (temperature and relative humidity) and six strawberry cultivars ('Albion', 'Aromas', 'Camino Real', 'Monterey', 'Portola', and 'San Andreas') on the population dynamics of H. haemorrhoidalis during the 2018-2019 and 2019-2020 growing seasons in southern Brazil. The greatest peak in thrips populations in the cultivars assessed occurred between January and March (summer) in both growing seasons. The relationship between the incidence of H. haemorrhoidalis and temperature was observed to be negative during the autumn, spring, and summer months but positive during winter. Relative humidity levels above 70% had a negative influence on the abundance of insects. Populations of H. haemorrhoidalis varied between the cultivars as follows: 'Aromas' > 'Monterey' > 'San Andreas' > 'Albion' > 'Camino Real' > 'Portola'. Cross-sections of strawberry leaves showed that 'Monterey' had the highest density of nonglandular trichomes/mm2 (7.26), while 'Portola' cultivar, the greatest predominance of glandular trichomes/mm2 (12.67), which may have contributed to the incidence of H. haemorrhoidalis. The demographic information presented on H. haemorrhoidalis in strawberry cultivars provides a starting point to better understand the insect's population dynamics. These results also indicate that climatic conditions are a limiting factor for the development of H. haemorrhoidalis.

Lema bilineata Germar (Coleoptera: Chrysomelidae) was recently reported to damage Physalis peruviana crops in Brazil. Given the potential for inflicting damage on other Solanaceae species and the lack of alternatives for controlling this pest, we assessed the pathogenicity of 15 Beauveria isolates against L. bilineata adults in vitro. In addition, three of these isolates were tested for their ovicidal effect against L. bilineata eggs. Fungal strains were isolated from mummified corpses of L. bilineata collected in a non-commercial field in Paraná, Brazil. The isolates were identified as Beauveria bassiana using molecular markers. Lema bilineata adults were susceptible to conidial suspensions of all these isolates at a concentration of 108 conidia mL-1. Deaths caused by fungal extrusion were confirmed. Three strains were found to be more virulent against L. bilineata adults and showed ovicidal effects. This is the first study on entomopathogenic fungi isolated from dead insects collected from P. peruviana crops and tested against L. bilineata carried out in Brazil. The results obtained in the laboratory indicate the high potential of the use of three B. bassiana strains against L. bilineata as a biocontrol agent.

Actinobacteria have been drawing attention due to their potential for the development of new pest control products. We hereby assess the effects of Streptomyces isolated from marine and caatinga biomes against Duponchelia fovealis Zeller (Lepidoptera: Crambidae), a pest associated with the strawberry culture at a global scale. To this end, eggs deposited by adults were immersed for 5 s in a bacterial suspension, and the larvae were fed on leaflets placed in glass tubes containing bacterial suspensions. In both treatments, the control was a saline solution. The bioassays demonstrated that the Streptomyces strains were able to cause the death of D. fovealis eggs (≈ 40%) and larvae (≈ 65%) compared to untreated eggs (1.4%) and larvae (2.0%). The crude extract of strain T49 and the chitinase extract of strain T26 affected larval growth when applied directly to the thorax of first-instar larvae (larval-adult lifespan of 65.3 ± 0.5 days and 67.5 ± 0.7 days, respectively; survival of 61.2 ± 1.2%) in relation to the control treatment (larval-adult lifespan of 41.75 ± 0.2 days and survival of 83.7 ± 2.6%). The Streptomyces spp. strains T41, T49, and T50 caused antifeeding activity. Apart from larval mortality, the adults that emerged from the larvae exposed to the extracts presented morphological abnormalities, and the moths' chitin spectra showed clear alterations to the pupa and wings. Our studies show, for the very first time, that Streptomyces isolated from the marine environment and the Caatinga biome are effective at provoking the mortality of D. fovealis and are promising agents for developing new products with biological control properties.

Essential oils (EOs) produced by plants in the Lamiaceae family may provide new insecticidal molecules. Novel control compounds are needed to control Drosophila suzukii (Matsumura), a severe economic invasive pest of thin-skinned fruit crops. Thus, we characterized the main compounds of EOs from three rosemary Rosmarinus officinalis ecotypes (ECOs) and evaluated their toxicity to D. suzukii adults, deterrence of oviposition behavior, and histological alterations in larvae. Additionally, we analyzed the lethal and sublethal effect on the pupal parasitoid Trichopria anastrephae. The main compounds identified in the R. officinalis ECOs were α-pinene, camphor and 1,8-cineole. In bioassays via topical application or ingestion, ECOs and their major compounds showed high toxicity on D. suzukii adults and a lower concentration could kill 50% and 90% of flies compared to spinetoram. The dry residues of a-pinene, 1,8-cineole, and camphor provided a repellent effect by reducing D. suzukii oviposition by ~47% compared to untreated fruit. Histological sections of 3rd instar larval D. suzukii posttreatment revealed damage to the fat body, Malpighian tubules, brain, salivary gland, and midgut, which contributed to high larval and pupal mortality. Survival and parasitism by adult T. anastrephae were not affected. Thus, R. officinalis EO and their compounds have potential for developing novel insecticides to manage D. suzukii.

The presence of weeds in the margins of strawberry crops can enhance populations of predatory mites as a measure to support conservation biological control. The aims of this study were (i) to assess the composition of the acarofauna associated with strawberries and the accompanying herbaceous plants in an organic farming system, and (ii) to evaluate the possible relationships between phytophagous and predatory mites occurring in this system. Strawberry leaves and whole plants of weeds were sampled biweekly from August 2014 to February 2015 in Lapa (Paraná, Brazil). In total, 23 weed species belonging to 10 families were identified; 3768 mite individuals (from 15 families and 4 suborders) were recovered, 77% on strawberries and 23% on weeds. Abundance of predatory mites on weeds was greater than on strawberry cultivars. On strawberries, the most abundant family was Tetranychidae (84%) followed by Phytoseiidae (11.6%). In total, 16 predatory mite species from the Phytoseiidae family were identified, 13 of them occurring on strawberry leaflets. Typholodromalus aripo, Neoseiulus californicus and Typhlodromips mangleae were the most abundant mite species on strawberry leaves. On weeds, most individuals were predatory mites (59%), whereas phytophagous mites represented 17.2%. The most abundant family was Phytoseiidae (36.4%). On weeds, the phytoseiid mite T. aripo was the most abundant species, representing 34.7%. Besides being found on strawberry leaflets, T. aripo was associated with 15 weed species. Among the weeds, Bidens pilosa showed the highest values of the Shannon index (1.97), Margalef index (3.04), and Pielou's evenness index (0.95). This study emphasizes the importance of surrounding weeds as a shelter for beneficial mitefauna in strawberry fields, likely contributing to enhance conservation biological control.

The diverse flora of the Atlantic Forest is fertile ground for discovering new chemical structures with insecticidal activity. The presence of species belonging to the genus Baccharis is of particular interest, as these species have shown promise in pest management applications. The objective of this study is to chemically identify the constituents expressed in the leaves of seven species of Baccharis (B. anomala DC., B. calvescens DC., B. mesoneura DC., B. milleflora DC., B. oblongifolia Pers., B. trimera (Less) DC. and B. uncinella DC.) and to evaluate the toxicological and morphological effects caused by essential oils (EOs) on the larvae and adults of Drosophila suzukii (Diptera: Drosophilidae). Chemical analysis using gas chromatography-mass spectrometry (GC-MS) indicated that limonene was the main common constituent in all Baccharis species. This constituent in isolation, as well as the EOs of B. calvescens, B. mesoneura, and B. oblongifolia, caused mortality in over 80% of adults of D. suzukii at a discriminatory concentration of 80 mg L-1 in bioassays of ingestion and topical application. These results are similar to the effect of spinosyn-based synthetic insecticides (spinetoram 75 mg L-1) 120 h after exposure. Limonene and EOs from all species had the lowest LC50 and LC90 values relative to spinosyn and azadirachtin (12 g L-1) in both bioassays. However, they showed the same time toxicity over time as spinetoram when applied to adults of D. suzukii (LT50 ranging from 4.6 to 8.7 h) in a topical application bioassay. In olfactometry tests, 92% of D. suzukii females showed repellent behavior when exposed to the EOs and limonene. Likewise, the EOs of B. calvescens, B. mesoneura, and B. oblongifolia significantly reduced the number of eggs in artificial fruits (≅ 7.6 eggs fruit-1), differing from the control treatment with water (17.2 eggs fruit-1) and acetone (17.6 eggs fruit-1). According to histological analyses, the L3 larvae of D. suzukii had morphological and physiological alterations and deformations after exposure to treatments containing EOs and limonene, which resulted in high larval, pupal, and adult mortality. In view of the results, Baccharis EOs and their isolated constituent, limonene, proved to be promising alternatives for developing bioinsecticides to manage of D. suzukii.

ABSTRACT: Increased production of the Cape gooseberry (Physalis peruviana L.) in Brazil has given rise to interest in identifying the phytophagous species that might damage this crop to inform preventive control and integrated pest management strategies. In this study, we report the occurrence and describe the damage that larvae and adults of Lema bilineata Germar (Coleoptera: Chrysomelidae) cause in P. peruviana. The number of L. bilineata individuals, both larvae and adults, significantly affected the total consumption of P. peruviana leaves. We also report, for the first time, three natural enemies, including a fungus, a fly, and an ant, which are associated with this pest in Brazil and may play a role in biological control strategies.


RESUMO: Fisális, Physalis peruviana L., é uma cultura em expansão no Brasil, dessa forma a identificação de espécies fitófagas que causam danos nesta cultura é importante para desenvolver controle preventivo e estratégias para o Manejo Integrado de Pragas. Neste estudo, relatamos a ocorrência de Lema bilineata Germar (Coleoptera: Chrysomelidae) no cultivo de P. peruviana e descrevemos os danos causados por suas larvas e adultos. Observou-se que o número de indivíduos de L. bilineata, tanto larvas como adultos, influenciaram significativamente o consumo total de folhas de P. peruviana. Além disso, relatamos pela primeira vez a presença de três inimigos naturais, incluindo um fungo, uma mosca e uma formiga, associados a esta praga no Brasil que poderiam ser utilizados como agentes de controle biológico.

The European pepper moth, Duponchelia fovealis (Lepidoptera: Crambidae), is a key pest in strawberry production. Entomopathogenic fungi (EF) and parasitoids of the Trichogrammatidae family are effective biological control agents of this pest with the potential to be used jointly for improved efficacy. This study aims to evaluate the susceptibility of Trichogramma atopovirilia and Trichogramma pretiosum to two Beauveria bassiana strains (B2 and B3) and two commercial bioinsecticides (Bovemax® and Methamax®) by applying them to D. fovealis eggs in pre- and post-parasitism periods. Pre-parasitism application of B2 and B3 did not affect the percentage of D. fovealis eggs parasitized by either Trichogramma species, except in the case of T. atopovirilia when eggs were sprayed with B3 at 1.5 × 105 conidia mL-1 (16.7% less than the control). In contrast, eggs sprayed with 1.5 × 108 conidia mL-1 of the commercial bioinsecticides were not parasitized by any Trichogramma species. Overall, the EF tested reduced the parasitism rate, adult emergence, and longevity of Trichogramma adults by less than 30% in all cases. The adverse effects of the B. bassiana strains and commercial products on the biological traits of both Trichogramma species were minimal, meaning that these agents can be used jointly in D. fovealis control strategies.

The present study aimed to analyze the chemical composition of five species of the genus Piper (P. aduncum L.; P. crassinervium Kunth.; P. malacophyllum Prels.; P. gaudichaudianum Kunth.; P. marginatum L.), and assess their toxicity to the adults of Drosophila suzukii (Diptera: Drosophilidae) and the pupal parasitoid Trichopria anastrephae Lima (Hymenoptera: Diapriidae). The major compounds were monoterpene hydrocarbons (5.3-60.9%); oxygenated monoterpenes (13.3%); sesquiterpenes hydrocarbons (8.3-45.3%), oxygenated sesquiterpenes (5.2-58.8%); and arylpropanoids (15.2-29.6%). In bioassays of ingestion and topical application, essential oils (EOs) from P. aduncum, P. gaudichaudianum, and P. marginatum killed approximately 100% of adults of D. suzukii, similarly to the insecticide based on spinetoram (75 mg L-1) (96.2% of mortality). Besides, the dry residues from P. aduncum, P. gaudichaudianum, and P. marginatum provided a repellent effect on oviposition (≅ 7 eggs/fruits) and negative effects on egg viability (≅ 2 larvae/fruits) of D. suzukii on artificial fruits. Based on the estimate of the lethal concentration required to kill 90% of exposed flies, EOs from P. aduncum, P. crassinervium, P. gaudichaudianum, P. malacophyllum, and P. marginatum provided low toxicity to the parasitoid T. anastrephae in a bioassay of ingestion and topical application (mortality < 20%), similarly to the water treatment (≅ 5% of mortality). EOs of Piper species tested in this work showed to be promising plant insecticides for the management of D. suzukii.

ABSTRACT: Considering the damage caused by terrestrial gastropods in strawberry producing areas in southern Brazil, this study aimed to i) identify the species associated with Fragaria x ananassa in rural properties in municipalities of Paraná State, Brazil; ii) describe and illustrate the diagnostic morphological and conchology features of the mollusk species; iii) construct an illustrated key based on these morphological and conchology features; iv) analyze two species collected in March 2017 to investigate the possible occurrence of parasitic nematodes of medical importance. Taxonomic determination of the mollusks was performed through morphological analysis of samples, and parasitological analysis was carried out using artificial digestion. The following gastropod species were identified: Bradybaena similaris (Bradybaenidae), Rumina decollata (Subulinidae), Deroceras leave (Agriolimacidae), Limacus flavus (Limacidae), and Meghimatium pictum (Philomycidae). Rumina decollata, L. flavus,and M. pictum were recorded for the first time as damaging strawberries in Brazil. No larvae of medical or veterinary importance, associated with samples, were reported. The main diagnostic features are described and illustrated. Besides, the potential of the gastropods as crop pests and vectors of nematodes associated with zoonotic diseases are discussed. Results can facilitate identification of gastropod species in strawberry crops and provide background information for future studies on bioecology and pest control.


RESUMO: Considerando os danos causados por moluscos terrestres em regiões produtoras de morangueiro no Sul do Brasil, neste estudo tivemos como objetivos i) identificar as espécies de moluscos associadas à Fragaria x ananassa em propriedades rurais localizadas em municípios do estado do Paraná; ii) descrever e ilustrar as características morfológicas e conquiliológicas diagnósticas das espécies de moluscos encontradas; iii) construir uma chave pictórica com base em características morfológicas e conquiliológicas; iv) analisar parasitologicamente amostras de duas espécies coletadas em março de 2017, visando a identificação de possíveis nematódeos de interesse médico. A determinação taxonômica foi realizada através da análise morfológica das amostras e a parasitológica através da técnica de digestão artificial. Foram identificados os caracóis Bradybaena similaris (Bradybaenidae), Rumina decollata (Subulinidae), Deroceras leave (Agriolimacidae), Limacus flavus (Limacidae) e Meghimatium pictum (Philomycidae). Rumina decollata, L. flavus e M. pictum foram pela primeira vez registradas causando danos ao morangueiro no Brasil. Larvas de interesse médico e/ou veterinário não foram encontradas associadas às amostras analisadas. Os principais caracteres diagnósticos para cada espécie são descritos e ilustrados, assim como discutida a potencialidade das mesmas como pragas agrícolas e hospedeiras de nematódeos de importância médico-veterinária. Os resultados auxiliam a identificação destas espécies na cultura do morangueiro, servindo de base para estudos de bioecologia e controle nestas áreas.

Nos sistemas de produção do morangueiro, a adubação com boro é uma das práticas culturais mais empregadas pelos produtores, contudo o seu uso de forma incorreta pode comprometer a produção e a qualidade das frutas. O objetivo da presente pesquisa foi avaliar os efeitos da aplicação de boro no solo (0 e 4kg ha-1) combinada com pulverizações na parte aérea das plantas (0, 240, 480, 720 e 960g B ha-1) sobre a planta, número, produção, acidez titulável, teor de sólidos solúveis, pH da polpa e firmeza das frutas do morangueiro. A aplicação de 4kg B ha-1 no solo proporcionou redução da produção de massa seca radicular e aérea das plantas de morangueiro. A aplicação foliar de boro aumentou o número e a produção de frutas com maior diâmetro, e a máxima produção total de frutas foi obtida com a aplicação foliar de 568g B ha-1 durante o período reprodutivo. Tanto a aplicação de boro na parte aérea quanto no solo aumentaram o teor foliar deste nutriente, sendo que o teor foliar adequado para a segunda florada do morangueiro oscilou entre 69 a 88mg kg-1. A aplicação foliar de boro aumentou a firmeza das frutas, porém independentemente da forma de aplicação de boro houve redução no teor de sólidos solúveis das frutas do morangueiro.


In strawberry production systems, fertilization with boron is one of the most cultural practices employed by producers, however its use incorrectly can compromise the production and quality of fruit. The aim of this study was to evaluate the effects of applying boron in the soil (0 and 4kg ha-1) combined with spraying in the shoots (0, 240, 480, 720 B and 960g ha-1) on plant, number, production, titratable acidity, soluble solids, pH and pulp firmness of strawberry fruit. The application of 4kg B ha-1 soil resulted in reduced production of root biomass and shoot of strawberry plants. Foliar application of boron increased the number and fruit production with bigger diameter and maximum total production of fruits was obtained with foliar application of 568g B ha-1 during the reproductive period. Both the application of boron in the air and in the soil increased the level of this nutrient leaf, and the leaf content suitable for the second blooming strawberry ranged from 69 to 88mg kg-1. Foliar application of boron increased the firmness of the fruit, but regardless of the form of application of boron decreased the soluble solids content of fruit in strawberry.

RESUMO

A avaliação da qualidade microbiológica de produtos armazenados, utilizados como plantas medicinais, é fundamental para garantir a segurança alimentar, em função do potencial micotoxigênico apresentado por algumas espécies de fungos. No presente trabalho foram realizados o isolamento, a quantificação e a identificação de fungos potencialmente produtores de micotoxinas em amostras de chás de camomila (Chamomilla recutita L.), erva-doce (Pimpinella anisum L.) e erva-mate (Ilex paraguariensis St.-Hil.), adquiridas em estabelecimentos comerciais da cidade de Curitiba, PR, Brasil. As amostras foram analisadas nas formas de infusão fria, de infusão tradicional e de cocção. Não houve diferenças significativas nos valores de UFC/g nas amostras de camomila, erva-mate e erva-doce analisadas na forma de infusão. Foram observadas diferenças significativas em amostras de camomila e erva-doce em forma de infusão fria em relação às demais formas de processamento de amostras, contudo essas não foram evidentes nas amostras de erva-mate. Os principais gêneros de fungos isolados das amostras analisadas foram Aspergillus sp (35,9%); Penicillium sp (9,4%); Fusarium sp (0,21%); Rhizopus sp (11,5%), Ulocladium sp (18,4%) e Mycelia sterilia (6,84%). Entre os fungos potencialmente toxigênicos, o Aspergillus sp foi o mais frequente nos três tipos de chás, seguido de Penicillium sp e Fusarium sp. Considerando que os fungos com potencial micotoxigênico persistem mesmo após a infusão ou cocção, recomenda-se que estratégias sejam desenvolvidas para garantir a qualidade e a segurança alimentar dos produtos consumidos pela população.
