RESUMO

Structural characteristics of discharged and undischarged nematocysts from the hydrozoans Millepora alcicornis and Millepora complanata, two fire corals collected in the Mexican Caribbean, were examined using transmission electron, scanning and light microscopy. In this study, we report for the first time images of the nematocysts found in these Mexican Caribbean venomous species. Two types of nematocysts were observed in both species, the more abundant identified as macrobasic mastigophore and the other a stenotele type. Macrobasic mastigophores were present in medium and large size classes while stenoteles appeared in only one size.

UNLABELLED: The characterization of lung damage in an experimental model of collapsing glomerulopathy (CG) in rats is described. METHODS: 12 rats were divided into two groups and injected intravenously (iv) with 1 mg/saline in a final volume of 1 ml/ day in the tail vein for 5 days, with fractionated serum from control and CG subjects. Proteinuria was quantified, and the Glomerular filtration rate was calculated based on creatinine clearance (CC). Rats were sacrificed by perfusion fixation at day 5. RESULTS: Rats injected with serum from CG patients developed proteinuria (p<0.001). A decrease in CC (0.68+/-0.19) in these rats was also observed. Glomerular tuft retraction and mesangial proliferation was observed in all rats receiving serum from the CG patients. Peribronchiolar infiltrate integrated mainly by lymphocytes, was identified in all CG rats. In some areas this infiltration disrupted the basement membrane and damaged the epithelium. No histopathological abnormalities in the kidney or lungs were found in rats receiving control serum. CONCLUSION: Patchy pulmonary lymphoid infiltrates were found in the CG model. Up to now there was no information about pulmonary lymphoid infiltration in CG patients. Besides fluid overload due to renal insufficiency or a nephrotic syndrome, other causes of pulmonary involvement in CG patients should be explored.

Nerve growth factor (NGF) induces morphological and physiological changes in cultured pancreatic beta-cells, including the extension of neurite-like processes. This latter effect is potentiated by dibutyryl cAMP (dbcAMP). beta-cells cultured under these conditions maintain their immunoreactivity to insulin and gamma-amino-butyric acid (GABA). NGF, dbcAMP, and high glucose concentrations also increase the expression of the GABA-synthesizing enzyme glutamic acid decarboxylase-65 in cultured beta-cells. The aim of this work was to study the effect of NGF alone or in combination with dbcAMP on pancreatic beta-cell ultrastructural morphology, after 10 days in culture. We used light microscopy, scanning electron microscopy, and transmission electron microscopy to analyze the modifications in cell surface and neurite-like projections. Morphometric analysis showed that NGF and/or dbcAMP treatment substantially increased the insulin and GABA content in granules and rough endoplasmic reticulum. Given that pancreatic beta-cells express NGF receptors and that NGF is synthesized and secreted by beta-cells, these results further suggest that NGF could have trophic actions on pancreatic hormone synthesis and/or storage.

The volcano mouse Neotomodon alstoni alstoni is a genus endemic to the higher elevations of the Mexican transvolcanic belt. In the present study we examined for the first time the morphological features of the spermatozoa taken from the caudal epididymis of this species by transmission and scanning electron microscopy. Spermatozoan motility was studied in sucrose and bicarbonate solutions; vitality and morphology were observed by light microscopy. Transmission electron microscopy shows that the head of spermatozoon is asymmetric and possesses a large and curved hook. The axoneme of the spermatozoan tail is highly developed at fibers 1, 5, and 6. Absolute and relative measurements of the length of the head, the midpiece, and the rest of the tail were also obtained. N. alstoni alstoni spermatozoa were hyperactive in the presence of 290 mM sucrose and 10 and 20 nM bicarbonate solutions exhibited high motility (180-190 microm/sec), and high flagellum beating frequency (10-12 Hz). In contrast, the spermatozoa in 310 mM sucrose solution showed scarce motility (13.5 +/- 3.8 microm/sec) and low beating frequency (1.5 +/- 0.4 Hz). It is proposed that the volcano mouse spermatozoa possess some features very similar to other mammalian spermatozoa and that bicarbonate triggers caudal epididymal sperm motility of this species. J. In scanning electron microscopy (SEM) most biological specimens lack conductance of electrons, requiring of coating with conducting element, which prevent damage from the electron beam. The present study combines the use of osmium-thiocarbohydrazide-osmium (OTO) with Peldri II and hexamethyldisilazane (HMDS), as an alternative procedure to improve the image of biological specimens for SEM. We examined freeze-fractured rat's myocardium. The tissue was fixed with cacodylate buffer, followed by 2.5% glutaraldehyde in buffer. Specimens were remove and fixed for two hours at 4 degrees C. The first group of tissues were processed with standard technique, the second with OTO and Peldri II and the third was treated with OTO and HMDS. All specimens were examined in a SEM. The routine processed specimens allowed an appropriate visualization of myocytes and their subcellular elements including nucleus, myofibrils, mitochondria. Likewise tissue processed with the combination of OTO in addition HMDS and OTO plus Peldri II, allowed a high quality study. The procedure described herein may offer an alternative for the study of biological specimens by SEM.
