RESUMO

The onion aphid, Neotoxoptera formosana, poses a significant threat to Allium crops worldwide, causing considerable economic losses and quality degradation. To develop effective pest management strategies, it is crucial to understand the feeding behavior and life history of this pest on different Allium crops. In this study, the electrical penetration graph (EPG) technique was used to monitor the thorn-feeding behavior of the onion aphid on 4 Allium crops: leek, chive, garlic, and shallot. The EPG data revealed distinct feeding patterns, with garlic and shallots being more preferred hosts than chives. Additionally, the aphids primarily fed on the phloem in garlic and shallots. Analysis of life history trait showed that chives provided the most favorable conditions for aphid development and reproduction, while leek exhibited relatively unfavorable conditions. Examination of leaf histology also revealed differences among the crops, which may influence aphid feeding behavior. This study provides valuable insights into the interaction between the onion aphid and different Allium crops, aiding in the development of comprehensive pest control strategies to minimize crop damage and economic losses. The use of advanced techniques like EPG contributes to a more detailed understanding of aphid behavior and shows promise for improving pest management in other plant-pest interactions.

Kin recognition has been widely observed in various taxa. Cannibalism avoidance may be a strong driver for the evolution of kin recognition, as it may avoid a reduction in inclusive fitness. Kin recognition has recently been observed in a generalist phytoseiid, Amblyseius herbicolus (Acari: Phytoseiidae). This study experimentally examined the degree of relatedness needed between prey larvae and cannibal adults of A. herbicolus for the occurrence of kin discrimination. The adults were individually placed in enclosed arenas with two prey, a daughter and a more distant related larva, to observe their cannibalizing choice. The adults of A. herbicolus did not discriminate between close relatives (daughter versus niece) but preferably cannibalized more distant kin (i.e., first and second cousins once removed). Phenotype matching and familiarization seem prominent as recognition mechanisms used by A. herbicolus adults. The effect of learning on kin recognition through prior contact in A. herbicolus requires further investigation. Studies on other adaptive functions of kin recognition of A. herbicolus, such as cooperation and parental care, may provide meaningful insights.

Hydrogen peroxide, as the second messenger in cells, plays an essential regulatory role in cell proliferation, differentiation and migration. How to effectively identify H2O2 signals, and detect H2O2 in circulating tumor cells, are essential for the early diagnosis of tumors. Herein a fluorescence and T1-weighted MR dual-mode imaging material, named MnBQ, has been successfully prepared and characterized. Based on the chemical redox reaction between MnBQ and H2O2, a fluorescence/T1-weighted MRI dual-mode sensor has been developed for quantitative analysis of H2O2, of which the lowest limit detection obtained from fluorescence and MRI could reach 2.841 µmol/L and 43.85 µmol/L, respectively. This imaging sensor has dual advantages of high sensitivity and high spatial resolution, which could be a good candidate for monitoring of H2O2 in vivo.

Cannibalism is commonly found among phytoseiids at almost all mobile life stages. The avoidance of kin cannibalism in the presence of non-kin has been observed in a limited number of phytoseiid species. However, studies examining kin recognition by individuals at different life stages are limited. Early experience with conspecifics can alter individuals' social behaviour at later stages. In this study, we examined the influence of ontogenetic isolation on cannibalism and kin recognition by the predatory mite Amblyseius herbicolus (Chant) (Acari: Phytoseiidae) at different life stages. In a choice test, A. herbicolus protonymphs, deutonymphs, pre-ovipositional adults, and ovipositional adults were used as predators, and larvae (siblings vs. non-siblings) were given as prey. Two rearing treatments were applied to the predators to generate different social experience during ontogeny: reared in isolation or in pairs with ad libitum mixed stages of Carpoglyphus lactis (Acari: Carpoglyphidae). Social experience with siblings during ontogeny significantly reduced the cannibalism rate. Despite the social experience during ontogeny, protonymphs, deutonymphs, and pre-ovipositional adults did not discriminate siblings from non-siblings during cannibalism. The discrimination behaviour between siblings and non-siblings of A. herbicolus occurred after adults became ovipositional. We hypothesize that A. herbicolus adults could learn the characteristics of their larvae through the contact with and assessment of their eggs during oviposition. The result of this study shows that kin discrimination could be phase-specific. Future studies can investigate if contact with non-kin (or non-sibling) eggs and subsequently hatching larvae can influence the discrimination ability of ovipositional adults.

BACKGROUND: The aim of this study was to investigate the role of Vaspin on the chondrogenic differentiation of bone mesenchymal stem cells (BMSCs), and its effect on chondrocyte survival and ECM secretion. We also assessed whether the Akt activation participates in these processes. METHODS: In vivo, immunohistochemistry was used to examine the positive rate of the protein expressions of Akt in Wistar rat articular cartilage and subchondral bone after Vaspin intraperitoneal injection for 14 days. In vitro, we isolated and expanded BMSCs from Wistar rats, and further cultured BMSCs as pellets in a chondrogenic-differentiation medium supplemented with different concentrations of Vaspin. After 21 days, the pellets were processed for cell counting kit assay. The mRNA level of Akt, SOX9 and COL2A1 in the pellets were investigated using quantitative Real-Time polymerase chain reaction, and the protein level of COMP was detected using western blot. RESULTS: During the chondrogenic differentiation of BMSCs, Vaspin promoted the chondrogenic differentiation of BMSCs and chondrocyte survival by activating the Akt pathway. These effects were significantly reduced by treatment with an Akt inhibitor. Moreover, Vaspin promoted chondrogenic differentiation of BMSCs by increasing the expression of markers in cartilage formation and extracellular matrix secretion. Furthermore, our study also found that Vaspin could increase Akt expression in cartilage cavities and subchondral bone in vivo. CONCLUSION: These findings demonstrate that Vaspin can promote the chondrogenic differentiation of BMSCs and chondrocyte survival via Akt activation. Our study provides new insights into the potential ability of Vaspin to ameliorate the chondrogenic differentiation of BMSCs and chondrocyte survival in OA.

OBJECTIVE: To investigate the effect of autophagy expression levels of different weight-bearing states and different stages of osteoarthritis in animal models, as well as the corresponding mechanisms. METHODS: We used the male Sprague-Dawley (SD) rats (12-week-old, SPF) to establish the OA animal models by modified Hulth method, and grouped animal models according to the length of time after surgery and different weight-bearing areas. RT-qPCR was carried out for detection of autophagy-related genes such as Atg7, Atg12, P62, etc. Western blot analysis was used to detect the expression levels of corresponding autophagy-related proteins such as LC3B, P62, etc. T test was performed for statistical analysis to compare different groups, while the differences were deemed statistically significant with P < 0.05. Transmission electron microscopy was used to observe the autophagosome to demonstrate the level of autophagy expression and the status of the chondrocytes. RESULTS: The results of the RT-qPCR testing showed that when the weight-bearing cartilage of the 4-week group (relatively mild) was compared with that of the 10-week group (relatively severe), there were statistically significant differences in all the genes tested, in detail: Atg3 (P < 0.01), Atg7 (P < 0.01), Atg12 (P < 0.01), P62 (P < 0.0001). The expression of autophagy-related mRNA in the 4-week group is increased compared with that of the 10-week group. As for the expression of proteins, Western blotting showed that in the comparison between the 4- and the 10-week groups, statistically significant results include Atg12 (P < 0.01) in the non-weight-bearing area, with decreased autophagy in the 10-week group compared with that of the 4-week group, while expression of LC3B (P < 0.05) protein was significantly higher in the 4-week group than in the control in the non-weight-bearing area. The expression of LC3B (P < 0.0001) and P62 (P < 0.05) in the 10-week group were higher than that of the control. Transmission electron microscope showed that autophagy in the weight-bearing area is stronger than that in the non-weight-bearing area, and autophagy in the 4-week group is stronger than in the 10-week group for the weight-bearing area. CONCLUSIONS: The expression of autophagy varies during different stages of osteoarthritis, in which the autophagy is stronger in the early stage of osteoarthritis, and gradually decreases with the progression of the disease. Autophagy in different weight-bearing areas may also be different.

INTRODUCTION: Grasshoppers and locusts are widely distributed worldwide, causing significant losses in agriculture. The origin and functions of their haemocytes are not entirely understood. OBJECTIVES: Insect haemocytes arbitrate cellular defence and participate in humoral defences. Due to their importance, the haemocytes of 35 species of grasshoppers and locusts from China were morphologically examined in this study. We aim to highlight a simple method for the morphological examination of insect haemocytes. METHODS: The haemocytes were observed, counted and compared under a light microscope after Wright-Giemsa staining. RESULTS: High complexity in form and shape were observed in the haemocytes. These include prohaemocytes, plasmatocytes, granulocytes, vermicytes, podocytes and megakaryocytes. No clear relationship was seen between the haemocyte type and their phylogenetic relationship among the three families examined. The high abundance of plasmatocytes and granulocytes suggests their importance in the immunity of grasshoppers and locusts. The minor haemocyte populations including prohaemocytes, vermicytes and podocytes may not be always present in individuals. CONCLUSION: All examined species shared similarities in their haemocyte types. Wright-Giemsa staining is a simple and efficient method for evaluating haemocytes.

The plastic flow behaviors of FCC polycrystalline aluminum after pre-cyclic tension-compression deformation are mainly investigated in tension-torsion stress space by the physically based crystal plasticity model introducing a back-stress. A global finite element model (GFEM) constructed of sufficient grains was established to simulate the same-size thin-walled tube specimen constrained and loaded as the experiments of yield surfaces. The computational results showed that the shape of subsequent yield surfaces and the plastic flow directions directly depended on the given offset strain levels and the applied re-loading paths under different pre-cyclic deformations. The angle deviation between the plastic flow direction and the theoretical orthogonal direction further indicated that there was a large difference between them in the inverse pre-straining direction, but the difference was negligible in the pre-straining direction. From the influence of the anisotropic evolution of the subsequent yield surfaces on plastic flow, we found that the plastic normality rule followed the smooth yield locus; conversely, the significant non-associated flow was attributed to the distorted yield locus. Furthermore, it was also demonstrated that the anisotropic evolution and the plastic flow trend of the subsequent yield surfaces obtained by experiments can be better reproduced by the crystal plasticity model.

Compact-tension (CT) specimens made of low alloy 30CrMo steels were hydrogen-charged, and then subjected to the fracture toughness test. The experimental results revealed that the higher crack propagation and the lower crack growth resistance (CTOD-R curve) are significantly noticeable with increasing hydrogen embrittlement (HE) indexes. Moreover, the transition in the microstructural fracture mechanism from ductile (microvoid coalescence (MVC)) without hydrogen to a mixed quasi-cleavage (QC) fracture and QC + intergranular (IG) fracture with hydrogen was observed. The hydrogen-enhanced decohesion (HEDE) mechanism was characterized as the dominant HE mechanism. According to the experimental testing, the coupled problem of stress field and hydrogen diffusion field with cohesive zone stress analysis was employed to simulate hydrogen-assisted brittle fracture behavior by using ABAQUS software. The trapezoidal traction-separation law (TSL) was adopted, and the initial TSL parameters from the best fit to the load-displacement and J-integral experimental curves without hydrogen were calibrated for the critical separation of 0.0393 mm and the cohesive strength of 2100 MPa. The HEDE was implemented through hydrogen influence in the TSL, and to estimate the initial hydrogen concentration based on matching numerical and experimental load-line displacement curves with hydrogen. The simulation results show that the general trend of the computational CTOD-R curves corresponding to initial hydrogen concentration is almost the same as that obtained from the experimental data but in full agreement, the computational CTOD values being slightly higher. Comparative analysis of numerical and experimental results shows that the coupled model can provide design and prediction to calculate hydrogen-assisted fracture behavior prior to extensive laboratory testing, provided that the material properties and properly calibrated TSL parameters are known.

Bone regenerative engineering provides a great platform for bone tissue regeneration covering cells, growth factors and other dynamic forces for fabricating scaffolds. Diversified biomaterials and their fabrication methods have emerged for fabricating patient specific bioactive scaffolds with controlled microstructures for bridging complex bone defects. The goal of this review is to summarize the points of scaffold design as well as applications for bone regeneration based on both electrospinning and 3D bioprinting. It first briefly introduces biological characteristics of bone regeneration and summarizes the applications of different types of material and the considerations for bone regeneration including polymers, ceramics, metals and composites. We then discuss electrospinning nanofibrous scaffold applied for the bone regenerative engineering with various properties, components and structures. Meanwhile, diverse design in the 3D bioprinting scaffolds for osteogenesis especially in the role of drug and bioactive factors delivery are assembled. Finally, we discuss challenges and future prospects in the development of electrospinning and 3D bioprinting for osteogenesis and prominent strategies and directions in future.

In this paper, torsional fatigue failure of 30CrMnSiNi2A steel which exhibited non-Masing behavior was studied under different constant shear strain amplitudes, using thin-walled tubular specimens. The relationship between shear fatigue and the evolution of meso-deformation inhomogeneity and the prediction method of the torsional fatigue life curve were investigated. Shear fatigue of the material under constant amplitude was researched by numerical simulation with reference to tests, by using crystal plasticity of polycrystalline representative volume element (RVE) as the material model. Considering the non-Masing behavior of material, when determining the parameter values of the crystal plasticity model the correlation between these parameters and strain amplitude was taken into account. The meso-deformation inhomogeneity with increments in the number of cycles was characterized by using the statistical shear strain standard deviation of RVE as the basic parameter. Considering the effect of strain amplitude on fatigue damage, ratio cycle peak stress/yield stress was taken as the weight to measure the torsional fatigue damage and an improved fatigue indicator parameter (FIP) to measure the inhomogeneous deformation of the material was proposed. The torsional fatigue life curve of 30CrMnSiNi2A steel was predicted by the critical value of the FIP and then the result was confirmed.

This study compares the longitudinal histological characteristics of proximal humeral implants with different spatial structures in rabbits. Thirty skeletally-mature male rabbits were divided into a trabecular structure group and regular hexahedron structure group according to the different spatial structures of a biological titanium alloy screw inserted into the greater tuberosity of the proximal humerus. Samples were collected 3, 6, and 12 weeks after the implantation surgery. Histological results showed that the amount of bone in-growth in the porous cavity of the screw implant increased over time. Quantitative analysis showed there was significantly more bone in-growth in the trabecular structure group than the classic structure group 3 weeks (25.4% ± 6.9% vs 19.6% ± 3.7%, P < 0.05) and 6 weeks (31.2% ± 1.7% vs 26.9% ± 5.3, P < 0.05) after the implantation surgery. No significant difference was detected between the two groups 12 weeks after the surgery (41.7% ± 2.5% vs 39% ± 4.1%, P > 0.05). Our data found that bone in-growth significantly differed among the three time points (P < 0.05) in both groups, but not between the implants with different spatial structures 12 weeks after the surgery.

In this study, we investigated whether the measurement of patellar tracking can be used as a diagnostic parameter of patellofemoral joint disease. Patellar tracking is defined as the movement of the patella in relation to the femorotibial joint within the full range of flexion and extension of the knee joint. The PubMed, EMBASE, Medline, PsychINFO, and AMED databases were used to find relevant articles. Analyzed were the patellar tracking coordinate system and the measurement objects, precision, methods used in those studies, as well as the results obtained. Origin points for coordinate systems varied across the studies. The research object and methods of patellar tracking varied in the studies. Most studies focused on a static description of the internal and external displacement and the internal and external inclination. The in vivo, noninvasive, and six degrees of freedom evaluation of patellar tracking reflect patellar motion more comprehensively, though each of these methods does so in different ways. Dynamic and quantitative evaluation of patellar tracking is still lacking in clinical work. Accurate and quantitative patellar tracking measurement could provide clinicians with a comprehensive evaluation of the stability of the knee joint.

This study aims at introducing the back stress of anisotropic strain-hardening into the crystal plasticity theory and demonstrating the rationality of this crystal plasticity model to describe the evolution of the subsequent yield surface of polycrystalline aluminum at the mesoscopic scale under complex pre-cyclic loading paths. By using two different scale finite element models, namely a global finite element model (GFEM) as the same size of the thin-walled tube specimen used in the experiments and a 3D cubic polycrystalline aggregate representative volume element (RVE) model, the evolution of the subsequent yield surface for different unloading cases after 30 pre-cycles is further performed by experiments and numerical simulations within a crystal plasticity finite element (CPFE) frame. Results show that the size and shape of the subsequent yield surfaces are extremely sensitive to the chosen offset strain and the pre-cyclic loading direction, which present pronounced anisotropic hardening through a translation and a distortion of the yield surface characterized by the obvious "sharp corner" in the pre-deformation direction and "flat" in the reverse direction by the definition of small offset strain, while the subsequent yield surface exhibits isotropic hardening reflected by the von Mises circle to be distorted into an ellipse by the definition of large offset strain. In addition, the heterogeneous properties of equivalent plastic strain increment are further discussed under different offset strain conditions. Modeling results from this study show that the heterogeneity of plastic deformation decreases as a law of fraction exponential function with the increasing offset strain. The above analysis indicates that anisotropic hardening of the yield surface is correlated with heterogeneous deformation caused by crystal microstructure and crystal slip. The crystal plasticity model based on the above microscopic mechanism can accurately capture the directional hardening features of the yield surface.

To reveal the relationship between grain size and twinning deformation of magnesium alloys under cyclic strain, this study carried out a group of strain-controlled low-cycle fatigue experiments and statistical analysis of microstructures. Experimental results show that the shape of the hysteresis loop exhibits significant asymmetry at different strain amplitudes, and the accumulation of residual twins plays an important role in subsequent cyclic deformation. For the different strain amplitudes, the statistical distribution of the grain size of magnesium alloy approximately follows the Weibull probability function distribution, while the statistical distribution of twin thickness is closer to that of Gaussian probability function. The twin nucleation number (TNN) increases with the increase of grain size, but there is no obvious function relationship between twin thickness and grain size. Twin volume fraction (TVF) increases with the increase of grain size, which is mainly due to the increase of TNN. This work can provide experimental evidence for a more accurate description of the twinning deformation mechanism.

The relationship between strain fatigue life and evolution of meso-deformation inhomogeneity was studied, through the cyclic process of numerical simulation of crystal plasticity compared with the fatigue test of steel hot-rolled ribbed-steel bar 400 (HRB400). The statistical characterization parameters at grain level, including the standard deviation of the dot product of longitudinal stress and strain, the product of the macro stress and the standard deviation of the longitudinal strain, and the product of the macro stress ratio and the standard deviation of the longitudinal strain, were proposed and respectively applied to measure the meso-deformation inhomogeneity of materials. These parameters take the effect of peak stress into account, distinct from the pure strain statistical parameters. The numerical results demonstrate that the low-cycle fatigue life curves of materials are predictable using the new parameters as FIPs (fatigue indicator parameters), and the predictions are more rational than by utilizing the FIPs without considering the peak stress effect.

An improved model based on the Chaboche constitutive model is proposed for cyclic plastic behavior of metal and low cycle fatigue of notched specimens under cyclic loading, considering the effect of strain gradient on nonlinear kinematic hardening and hysteresis behavior. The new model is imported into the user material subroutine (UMAT) of the finite element computing software ABAQUS, and the strain gradient parameters required for model calculation are obtained by calling the user element subroutine (UEL). The effectiveness of the new model is tested by the torsion test of thin copper wire. Furthermore, the calibration method of strain gradient influence parameters of constitutive model is discussed by taking the notch specimen of Q235 steel as an example. The hysteresis behavior, strain distribution and fatigue failure of notched specimens under cyclic loading were simulated and analyzed with the new model. The results prove the rationality of the new model.

Oxya chinensis is one of the most widespread grasshopper species found in China and one of the most common pests against rice. In view of the importance of haemocytes in insect immunity in general, and the lack of information on the haemocytes of O. chinensis, we examined the haemocytes of this species in detail. We challenged the cellular response of this grasshopper with the bacteria Escherichia coli, Staphylococcus aureus, and Bacillus subtilis Haemocyte morphology was observed using light, scanning electron and transmission electron microscopy, which revealed distinct morphological varieties of haemocytes. Granulocytes and plasmatocytes responded to the bacterial challenge by phagocytosis. Histochemical staining indicated the presence of acid phosphatase in plasmatocytes and granulocytes. We also observed non-phagocytic prohemocytes and vermicytes, but their functions in the circulation are unclear. Insect haemocytes play a crucial role in cellular immunity, and further research is needed for a comprehensive understanding.

Osteoarthritis (OA) is a prevalent degenerative joint disease whose pathogenesis remains unclear. The research aims to investigate the roles of Circ_0136474/miR-127-5p/MMP-13 axis in OA. Differentially expressed circRNAs and miRNAs in OA cartilage tissue were screened out and visualized by R project based on RNA-seq data and microarray data respectively. qRT-PCR was carried out for detection of relative expression levels of Circ_0136474, miR-127-5p, MMP-13 and other inflammatory factors and Western blot analysis was conducted to detect the protein expression level of MMP-13. CCK-8 assay and flow cytometry were conducted to determine cell proliferation and cell apoptotic ability respectively. RNA-fluorescence in situ hybridization (RNA-FISH) experiments were conducted to confirm the immune-localization of the Circ_0136474 and MMP-13 in human tissues. Targeted relationships were predicted by bioinformatic analysis and verified by dual-luciferase reporter assay. Our findings revealed that the expression levels of both Circ_0136474 and MMP-13 in OA cartilage tissue were significantly higher than that in normal cartilage tissue. Circ_0136474 could suppress cell proliferation by facilitating MMP-13 expression and suppressing miR-127-5p expression in OA. Overexpression of miR-127-5p negatively regulated MMP-13 expression to enhance cell proliferation. Our study demonstrated that Circ_0136474 and MMP-13 suppressed cell proliferation, while enhanced cell apoptosis by competitive binding to miR-127-5p in OA, which may well provide us with a new therapeutic strategy for osteoarthritis.

OBJECTIVE: To investigate the effect of tumor necrosis factor alpha (TNF-α) on the proliferation of fibroblast-like synoviocytes (FLS) and the expression of programmed cell death factor 5 (PDCD5) in an inflammatory microenvironment, for the further understanding of the mechanism of action of TNF-α in promoting the proliferation of synovial cells and the apoptosis of the chondrocytes. METHODS: Articular carriage specimens were obtained from 21 cases with osteoarthritis and 12 cases with femoral neck fractures as healthy controls during arthroplasties. The expression of PDCD5 was evaluated by immunofluorescence analyzed by mean option density (MOD) detected using the software ImagePro Plus. Real-time PCR was performed to evaluate the transcriptions of PDCD5 and TNF-α in synovium. FLS cells derived from rheumatoid arthritis patients were cultured in vitro and incubated with different concentrations of TNF-α. The effects of TNF-α at different concentrations on the proliferation of FLS cells were detected by Cell Counting Kit-8 (CCK-8) assay to evaluate the cell proliferation rate. After incubation with the absence or presence of recombinant human TNF-α at different concentrations, the FLS cells were isolated for detection of PDCD5 protein and PDCD5 gene. The expression of PDCD5 protein was detected by western-blot and the transcription of PDCD5 gene from the cells was detected by real-time quantitative PCR. RESULTS: The MOD of PDCD5 as well as TNF-α of osteoarthritis cartilage sections were significantly increased compared with those of the controls, and in synovium there was a positive correlation between transcriptions of their mRNA. When the concentration of TNF-α was 1 ng/mL, the cell proliferation rate was not significantly different from that of the control group (P = 0.592), while the proliferation of FLS cells was significantly promoted when the concentration of TNF-α was 5, 10, 15, or 20 ng/mL, and the proliferation-promoting rates were 35.64% ± 6.96%, 48.72% ± 7.69%, 45.60% ± 8.85%, and 39.32% ± 6.18%, respectively (P < 0.01). The transcription of PDCD5 gene was significantly downregulated, which was 80.44% ± 4.07% and 84.30% ± 5.48%, respectively (P < 0.05), in the FLS cells incubated with TNF-α at the concentration of 10 and 15 ng/mL for 24 h. When the concentration of TNF-α was 1, 5, or 20 ng/mL, the transcription of PDCD5 mRNA in FLS cells was not significantly different from that in the control group (P > 0.05). The expression of PDCD5 protein was only significantly downregulated when the concentration of TNF-α was 10 ng/mL (P < 0.01), while the expression of PDCD5 protein in FLS cells was not significantly different from that in the control group (P > 0.05). CONCLUSION: The expression of PDCD5 as well as TNF-α in osteoarthritis cartilage and synovium was significantly higher than in healthy tissues, and TNF-α can promote the proliferation of FLS cells in patients with rheumatoid arthritis, and inhibit the expression of PDCD5. PDCD5 may be involved in the abnormal proliferation of synoviocytes and the degeneration of chondrocytes stimulated by TNF-α.
