Obesity has emerged as a prominent global health concern, with heat stress posing a significant challenge to both human health and animal well-being. Despite a growing interest in environmental determinants of obesity, very few studies have examined the associations between heat stress-related environmental factors and adiposity. Consequently, there exists a clear need to understand the molecular mechanisms underlying the obesogenic effects of heat stress and to formulate preventive strategies. This study focused on culturing porcine subcutaneous preadipocytes at 41.5 ℃ to induce heat stress, revealing that this stressor triggered apoptosis and fat deposition. Analysis demonstrated an upregulation in the expression of HSP70, BAX, adipogenesis-related genes (PPARγ, AP2, CEBPα and FAS), the p-AMPK/AMPK ratio and SIRT1, PGC-1α in the heat stress group compared to the control group (P < 0.05). Conversely, the expression of lipid lysis-related genes (ATGL, HSL and LPL) and Bcl-2 decreased in the heat stress group compared to the control group (P < 0.05). Furthermore, subsequent activator and/or inhibitor experiments validated that heat stress modulated HSP70 and AMPK signalling pathways to enhance lipogenesis and inhibit lipolysis in porcine subcutaneous preadipocytes. Importantly, this study reveals, for the first time, that EGCG mitigates heat-stress-induced fat deposition by targeting HSP70 through the activation of AMPK-SIRT1-PGC-1α in porcine subcutaneous preadipocytes. These findings elucidate the molecular mechanisms contributing to heat stress-induced obesity and provide a foundation for the potential clinical utilisation of EGCG as a preventive measure against both heat stress and obesity.

As an important precursor of secondary inorganic aerosols (SIAs), ammonia (NH3) plays a key role in fine particulate matter (PM2.5) formation. In order to investigate its impacts on haze formation in the North China Plain (NCP) during winter, NH3 concentrations were observed at a high-temporal resolution of 1 min by using the SP-DOAS in Tai'an from December 2021 to February 2022. During the observation period, the average NH3 concentration was 11.84 ± 5.9 ppbv, and it was determined as an ammonia-rich environment during different air quality conditions. Furthermore, the average concentrations of sulfate (SO42-), nitrate (NO3-) and ammonium (NH4+) were 9.54 ± 5.97 µg/m3, 19.09 ± 14.18 µg/m3 and 10.72 ± 6.53 µg/m3, respectively. Under the nitrate-dominated atmospheric environment, aerosol liquid water content (ALWC) was crucial for NH3 particle transformation during haze aggravation, and the gas-particle partitioning of ammonia played an important role in the SIAs formation. The reconstruction of the molecular composition further indicated that ammonium nitrate (NH4NO3) plays a dominant role in the increase of PM2.5 during haze events. Consequently, future efforts to mitigate fine particulate pollution in this region should focus on controlling NH4NO3 levels. In ammonia-rich environments, NO3- formation is more dependent on the concentration of nitric acid (HNO3). The sensitive analysis of TNO3 (HNO3 + NO3-) and NHX (NH3 + NH4+) reduction using the thermodynamic model suggested that the NO3- concentration decreases linearly with the reduction of TNO3. And the concentration of NO3- decreases rapidly only when NHX is reduced by 50-60 %. Reducing NOX emissions is the most effective way to alleviate nitrate pollution in this region.

Tropospheric reactive bromine is important for atmospheric chemistry, regional air pollution, and global climate. Previous studies have reported measurements of atmospheric reactive bromine species in different environments, and proposed their main sources, e.g. sea-salt aerosol (SSA), oceanic biogenic activity, polar snow/ice, and volcanoes. Typhoons and other strong cyclonic activities (e.g. hurricanes) induce abrupt changes in different earth system processes, causing widespread destructive effects. However, the role of typhoons in regulating reactive bromine abundance and sources remains unexplored. Here, we report field observations of bromine oxide (BrO), a critical indicator of reactive bromine, on the Huaniao Island (HNI) in the East China Sea in July 2018. We observed high levels of BrO below 500 m with a daytime average of 9.7 ± 4.2 pptv and a peak value of ∼26 pptv under the influence of a typhoon. Our field measurements, supported by model simulations, suggest that the typhoon-induced drastic increase in wind speed amplifies the emission of SSA, significantly enhancing the activation of reactive bromine from SSA debromination. We also detected enhanced BrO mixing ratios under high NOx conditions (ppbv level) suggesting a potential pollution-induced mechanism of bromine release from SSA. Such elevated levels of atmospheric bromine noticeably increase ozone destruction by as much as ∼40% across the East China Sea. Considering the high frequency of cyclonic activity in the northern hemisphere, reactive bromine chemistry is expected to play a more important role than previously thought in affecting coastal air quality and atmospheric oxidation capacity. We suggest that models need to consider the hitherto overlooked typhoon- and pollution-mediated increase in reactive bromine levels when assessing the synergic effects of cyclonic activities on the earth system.

Ozone (O3) profiles are crucial for comprehending the intricate interplay among O3 sources, sinks, and transport. However, conventional O3 monitoring approaches often suffer from limitations such as low spatiotemporal resolution, high cost, and cumbersome procedures. Here, we propose a novel approach that combines multiaxis differential optical absorption spectroscopy (MAX-DOAS) and machine learning (ML) technology. This approach allows the retrieval of O3 profiles with exceptionally high temporal resolution at the minute level and vertical resolution reaching the hundred-meter scale. The ML models are trained using parameters obtained from radiative transfer modeling, MAX-DOAS observations, and a reanalysis data set. To enhance the accuracy of retrieving the aqueous phosphorus from O3, we employ a stacking approach in constructing ML models. The retrieved MAX-DOAS O3 profiles are compared to data from an in situ instrument, lidar, and satellite observation, demonstrating a high level of consistency. The total error of this approach is estimated to be within 25%. On balance, this study is the first ground-based passive remote sensing of high time-height-resolved O3 distribution from ground to the stratopause (0-60 km). It opens up new avenues for enhancing our understanding of the dynamics of O3 in atmospheric environments. Moreover, the cost-effective and portable MAX-DOAS combined with this versatile profiling approach enables the potential for stereoscopic observations of various trace gases across multiple platforms.

Formaldehyde (HCHO) is considered one of the most abundant gas-phase carbonyl compounds in the atmosphere, which can be directly emitted through transportation sources. Long-Path Differential Optical Absorption Spectroscopy (LP-DOAS) was used to observe HCHO in the river channel of Wusong Wharf in Shanghai, China for the whole year of 2019. Due to the impact of ship activity, the annual average HCHO level in the channel is about 2.5 times higher than that in the nearby campus environment. To explain the sources of HCHO under different meteorological conditions, the tracer-pair of CO and Ox (NO2+O3) was used on the clustered air masses. The results of the source appointment show that primary, secondary and background account for 24.14% (3.34 ± 1.19 ppbv), 44.78% (6.20 ± 2.04 ppbv) and 31.09% (4.31 ± 2.33 ppbv) of the HCHO in the channel when the air masses were from the mixed direction of the city and channel, respectively. By performing background station subtraction at times of high primary HCHO values and resolving the plume peaks, directly emitted HCHO/NO2 in the channel environment and plume were determined to be mainly distributed between 0.2 and 0.3. General cargo ships with higher sailing speeds or main engine powers tend to have higher HCHO/NO2 levels. With the knowledge of NO2 (or NOx) emission levels from ships, this study may provide data support for the establishment of HCHO emission factors.

Increases in litter size, which are influenced by ovulation, are responsible for between 74% and 96% of the economic value of genetic progress, which influences selection. For the selection and breeding of highly prolific goats, genetic mechanisms underlying variations in litter size should be elucidated. Here, we used single-nucleus RNA sequencing to analyze 44,605 single nuclei from the ovaries of polytocous and monotocous goats during the follicular phase. Utilizing known reference marker genes, we identified 10 ovarian cell types characterized by distinct gene expression profiles, transcription factor networks, and reciprocal interaction signatures. An in-depth analysis of the granulosa cells revealed three subtypes exhibiting distinct gene expression patterns and dynamic regulatory mechanisms. Further investigation of cell-type-specific prolificacy-associated transcriptional changes elucidated that "downregulation of apoptosis", "increased anabolism", and "upstream responsiveness to hormonal stimulation" are associated with prolificacy. This study provides a comprehensive understanding of the cell-type-specific mechanisms and regulatory networks in the goat ovary, providing insights into the molecular mechanisms underlying goat prolificacy. These findings establish a vital foundation for furthering understanding of the molecular mechanisms governing folliculogenesis and for improving the litter size in goats via molecular design breeding.

This research aims at uncovering the effects and investigating the molecular mechanisms of dietary resveratrol (RES) supplementation on antioxidant capacity and meat quality of pigs. In this study, 20 µM RES could activate the KEAP1-NRF2 antioxidant defense pathway in response to oxidative stress in porcine skeletal muscle satellite cells was firstly found. Then, twenty-four healthy crossbred castrated boars were allocated to 4 treatments that were fed with a basal diet (control) and a basal diet supplemented with 200 mg, 400 mg or 600 mg RES per Kilogram (kg) of feed for 41 days, respectively. 400 and 600 mg/kg RES-supplemented diet can effectively improve the meat quality traits and activities of antioxidizing enzymes via the KEAP1-NRF2 signaling pathway of pigs. The molecular dynamic simulation further revealed that RES could directly binding to KEAP1 to reduce the tightness of KEAP1-NRF2 protein-protein interaction. More importantly, dietary supplementation of RES also improves antioxidant capacity through a series of KEAP1-NRF2 pathway-related lncRNAs were found by RNA sequencing (RNA-seq). Altogether, this study demonstrated that RES improves meat quality traits by effectively increasing antioxidant levels via the lncRNA-KEAP1-NRF2 axis in vivo and/or in vitro. These results provide new insights into the molecular mechanisms by which RES, as a nutritional agent, regulates antioxidant capacity and improves meat quality in pigs.

Resveratrol (RES) is one of the best-known bioactive polyphenols that has received much attention in recent years because of its importance to anti-obesity. However, the exact mechanism underlying this effect and whether it can improve lipid metabolism by regulating the long-chain noncoding RNA (lncRNA) remains unclear. In this study, 24 healthy crossbred castrated boars were fed a basal diet (control) and a basal diet supplemented with 200 mg, 400 mg or 600 mg RES per Kilogram (kg) of feed for 41 d, respectively. We found that 400 mg/kg and 600 mg/kg RES-supplemented diet did not affect growth rate, but reduced significantly subcutaneous adipose thickness, carcass fat rate, greater dramatically the serum concentration of adiponectin and high-density lipoprotein in pigs. Further, we verified that RES could inhibit the formation and accumulation of lipid droplets by AdipoQ-AdipoR1-AMPKα and AdipoQ-AdipoR2-PPARα signal pathway in vivo and vitro (3T3-L1 preadipocytes). Transcriptome analyses found that 5 differently expressed (DE) lncRNAs and 77 mRNAs in subcutaneous adipose between control group and 400 mg/kg RES group, which mainly involved in "adipocytokine signaling pathway," "Wnt signaling pathway," "PI3K-Akt signaling pathway" and "MAPK signaling pathway." In conclusion, RES can inhibit the formation and accumulation of lipid droplets through AdipoQ signal pathway and lipid metabolism-related lncRNAs. Our results provide a new insight on the molecular mechanism of RES as a nutritional agents to the prevention and treatment for obesity.

MicroRNAs (miRNAs) are small non-coding RNAs that post-transcriptionally regulate several pathway intermediates and affect the skeletal muscle development in mice, pigs, sheep, and cattle. However, to date, only a small number of miRNAs have been reported in the muscle development of goats. In this report, the longissimus dorsi transcripts of one- and ten-month-old goats were analyzed by sequencing RNAs and miRNAs. The results showed that the ten-month-old Longlin goats had 327 up- and 419 down-regulated differentially expressed genes (DEGs) compared with the one-month-old. In addition, 20 co-up-regulated and 55 co-down-regulated miRNAs involved in the muscle fiber hypertrophy of goats were identified in ten-month-old Longlin and Nubian goats compared with one-month-old. Five miRNA-mRNA pairs (chi-let-7b-3p-MIRLET7A, chi-miR193b-3p-MMP14, chi-miR-355-5p-DGAT2, novel_128-LOC102178119, novel_140-SOD3) involved in the goat skeletal muscle development were identified by miRNA-mRNA negative correlation network analysis. Our results provided new insight into the functional roles of goat muscle-associated miRNAs, allowing a deeper understanding of the transformation of miRNA roles during mammalian muscle development.

Currently, the three-dimensional (3D) distribution and characteristics of air pollution cannot be understood based on the application of any single atmospheric monitoring technology. Long-term, high-precision and large-scale 3D atmospheric monitoring might become practical by combining heterogeneous modern technologies; for this purpose, the Space-Air-Ground integrated system is a promising concept. In this system, optical remote sensing technologies employing fixed or mobile platforms are used as the main means for ground-based observations. Tethered balloons, unmanned aerial vehicles (UAV) and airborne platforms serve as the air-based observation segment. The final part, satellite remote sensing, corresponds to space-based observations. Aside from obtaining the 3D distribution of air pollution, research on emission estimation and pollution mechanisms has been extensively implemented based on the strengths of this system or some portion of it. Moreover, further research on the fusion of multi-source data, optimization of inversion algorithms, and coupling with atmospheric models is of great importance to the realization of this system.

Nitrous acid (HONO) is considered as one of the main sources of the hydroxyl radical (OH), the most relevant oxidant in the atmosphere. Multi-AXis-Differential Optical Absorption Spectroscopy (MAX-DOAS) measurements were conducted to obtain the vertical profiles of aerosol and HONO from November 1, 2020 to January 31, 2021 at a suburb site of Shanghai, China. HONO was mainly distributed near the surface, but high values HONO occasionally occurred around 0.7 km, indicating an unaccounted source of daytime HONO at high altitudes. The positive correlation between HONO and aerosols suggested that the photo-enhanced heterogeneous reactions on the aerosol surface were an important source of daytime HONO at high altitudes. To obtain the vertical distribution of OH production by HONO photolysis (P(OH)HONO), the vertical profiles of photolysis rate of HONO (JHONO) were calculated by establishing a method of combining observations with empirical relationship based on heterogeneous atmospheric and radiative transfer models. The JHONO increased approximately linearly with increasing altitudes and the noontime averages value of JHONO near the ground were 6.68 × 10-4 s-1, which was strongly negatively affected by aerosols in the morning and afternoon. The P(OH)HONO profile varied in different months (November, December, January) that the changes were mainly affected by HONO and JHONO. P(OH)HONO was more positively affected by JHONO at high altitude and noon but greatly influenced by HONO concentrations in the morning and afternoon.

Responses to the COVID-19 pandemic led to major reductions on air pollutant emissions in modern history. To date, there has been no comprehensive assessment for the impact of lockdowns on the vertical distributions of nitrogen dioxide (NO2) and formaldehyde (HCHO). Based on profiles from 0 to 2 km retrieved by Multi-AXis-Differential Optical Absorption Spectroscopy observation and a large volume of real-time data at a suburb site in Shanghai, China, four types of machine learning models were developed and compared, including multiple linear regression, support vector machine, bagged trees (BT), and artificial neural network. Ultimately BT model was employed to reproduce NO2 and HCHO profiles with the best performance. Predictions with different meteorological and surface pollution scenarios were conducted from 2017 to 2019, for assessing the corresponding impacts on the changes of NO2 and HCHO profiles during COVID-19 lockdown. The simulations illustrate that the NO2 decreased in 2020 by 43.8%, 45.5%, and 44.6%, relative to 2017, 2018, and 2019, respectively. For HCHO, the lockdown-induced situation presented the declines of 28.6%, 32.1%, and 10.9%, respectively. In the comparisons of vertical distributions, NO2 maintained decreasing at all altitudes, while HCHO decreased at low altitudes and increased at high altitudes. During COVID-19 lockdown, the reduction of NO2 and HCHO from the variation of surface pollutants was dominated below 0.5 km, while the relevant meteorological factors played a more significant role above 0.5 km.

Resveratrol (RES) has a wide range of biological and pharmacological activities with various health benefits for humans as a food additive. In animal production, RES has been considered a potential functional feed additive for producing high-quality pork. Long noncoding RNAs (lncRNAs) have emerged as essential regulators of fat metabolism, and phytochemicals can regulate fat metabolism through lncRNA. However, it is unclear whether RES can improve back-fat thickness by regulating lncRNA. In this study, we identified a novel lncRNA, which was named a long intergenic non-protein coding RNA, a regulator of fat metabolism (LincRNA-ROFM), from our previous lncRNA sequencing data. LincRNA-ROFM can inhibit adipocyte proliferation and differentiation. In-depth analyses showed that LincRNA-ROFM acts as a molecular sponge for miR-133b, and adiponectin (AdipoQ) is a direct target of miR-133b in porcine preadipocytes. In addition, the expression of LincRNA-ROFM was positively correlated with AdipoQ. RES can promote the expression of LincRNA-ROFM by PPARα and C/EBPα. Altogether, our research showed that LincRNA-ROFM acts as a ceRNA to sequester miR-133b and is upregulated by RES, leading to heightened AdipoQ expression, and thus decreased adipocyte proliferation and differentiation, which reduces back-fat thickness of pigs. Taken together, the RES/LincRNA-ROFM/miR-133b/AdipoQ regulatory network preliminarily explains the mechanism of action of RES in inhibiting fat deposition, which provides new insight into the downstream mechanism of RES inhibition of fat deposits by regulating the lncRNA.

As a passive remote sensing technique, MAX-DOAS method was widely used to investigate the vertical profiles of aerosol and trace gases in the lower troposphere. However, the measurements for midlatitude marine boundary layer are rarely reported, especially during the storm weather system. In this study, the MAX-DOAS was used to retrieve the aerosol, HCHO and NO2 vertical distribution at Huaniao Island of East China Sea in summer 2018, during which a strong tropical cyclone developed and passed through the measurement site. The observed aerosol optical depth (AOD), HCHO- and NO2-VCDs (Vertical Column Density) were in the range of 0.19-0.97, (2.57-12.27) × 1015 molec/cm2, (1.24-4.71) × 1015 molec/cm2, which is much higher than remote ocean area due to the short distance to continent. The vertically resolved aerosol extinction coefficient (AEC), HCHO and NO2 presented the decline trend with the increase of height. After the typhoon passing through, the distribution of high levels of aerosol and HCHO stretched to about 1 km and the abundances of the bottom layer were found as double higher than before, reaching 0.51 km-1 and 2.44 ppbv, while NO2 was still constrained within about 300 m with 2.59 ppbv in the bottom layer. The impacts of typhoon process forced air mass were also observed at the suburban site in Shanghai in view of both the aerosol extinction and chemical components. The different changes on air quality associated with typhoon and its mechanism in two different environments: coastal island and coastal city are worthy of further investigation as it frequent occurred in East Asia during summer and fall.

This study investigated the effects of resveratrol (RES) supplementation on the growth performance, carcass and meat quality, blood lipid levels and ruminal bacterial microbiota of fattening goats. A total of forty castrated Nubian goats (28.25 ± 0.26 kg body weight) were randomly divided into four groups and provided with diets containing different levels of RES (0, 150, 300 and 600 mg/kg) for 120 d. The results showed that RES increased redness and intramuscular fat content, whilst reducing shear force in the longissimus dorsi muscle of goats (p < 0.05). In addition, the final weight, average daily gain, hot carcass weight, net meat weight, carcass lean percentage and eye muscle area of goats were significantly increased in the 150 mg/kg RES group compared with the other three groups, while those in the 600 mg/kg RES group significantly decreased (p < 0.05). RES significantly decreased serum triacylglycerol and LDL-C contents (p < 0.05), and increased HDL-C content and the HDL-C/TC ratio (p < 0.05). Supplementation with 150 mg/kg RES also increased the proportion of Acetitomaculum and Moryella, genera comprising short-chain fatty acid-producing bacteria. The present study indicated that an appropriate supplemental level of RES could improve the growth performance, neat percentage, meat quality, ruminal microbiota and serum lipid levels of fattening goats.

Ground-based Multi-Axis Differential Optical Absorption Spectroscopy (MAX-DOAS) instruments were used to carry out observation of aerosol in the urban and suburban areas of Shanghai from October 17 to November 21, 2019. Fudan University (FDU) site is a typical urban environment, surrounded by residential areas, commercial areas and arterial roads, while Dianshan Lake (DSL) site is a suburban environment with high vegetation coverage and no pollutant emission sources. The aerosol retrieved by MAX-DOAS was in good correlation with the observation of sun photometer and the PM2.5 concentration of the corresponding site, which demonstrates that the aerosol retrieved by MAX-DOAS is reliable and feasible. Comparing the mean aerosol extinction coefficient (AEC) profiles during the observation period between urban and suburban areas, it was found that the occurrence of high aerosol concentration at FDU was nearly 3 h later than that of DSL at suburban site. And the aerosol at DSL was concentrated at an altitude of 0.3- 0.5 km, with a mean peak value of 0.486 km-1, which was slightly higher than the peak AEC of 0.453 km-1 at FDU of 0.2- 0.4 km. The difference in aerosol characteristics between the two sites may be due to the fact that the influences of aerosol transport and boundary layer dynamics are different between the two sites. The backward trajectories analysis also presents that there were mutual transports of aerosol between urban and suburban areas, which affect the optical properties of the aerosol in these two sites. In a case of aerosol pollution, we visualized the transport pathway of aerosol from the western part of the North China Plain to Shanghai using AEC profiles and backward trajectories, providing the evidence that the local aerosol pollution in Shanghai was affected by long-distance transport.

Litter size is an important component trait of doe reproduction. By improving it, production efficiency and economic benefits can be significantly provided. Genetic marker-assisted selection (MAS) based on proven molecular indicators could enhance the efficacy of goat selection, as well as litter size trait. Many molecular markers have been identified that they can be used to improve litter size in different goat breeds. However, the presence and value of these markers vary among goat breeds. In the present study, we used the reported loci on other breeds of goat as candidate loci to detect whether these loci appear in this Nubian goat population; then we proceed to genotype and detect surrounding loci (50 bp) by multiplex PCR and sequencing technology. As a result, 69 mutations (59 SNPs and 10 indels) were screened out from 23 candidate genes in Nubian goat population, 12 loci were significantly associated with the litter size of first-parity individuals; 5 loci were significantly associated with the litter size of second-parity individuals; 3 loci were significantly associated with the litter size of third-parity individuals. In addition, five loci were significantly associated with the average litter size. The additive effect value of KITLG: g.18047318 G > A in first parity, KITLG: g.18152042G > A in third parity, KISS-1: g.1341674 C > G in first parity, and GHR: g.32134187G > A in second parity exceed more than 0.40, and the preponderant alleles are G, C, A and G, respectively. Further, linkage disequilibrium analysis of 21 mutation loci shows that 3 haplotype blocks are formed, and the litter size of combination type AACC in KISS-1 gene and AAGG in KITLG gene are significantly lower than that of other combinations genotype in first parity ( P < 0.05 ). These findings can provide effective candidate DNA markers for selecting superior individuals in Nubian goat breeding.

Heat stress (HS) results in health problems in animals. This study was conducted to investigate the effect and the underlying mechanism of HS on the proliferation and differentiation process of 3T3-L1 preadipocytes. 3T3-L1 preadipocytes were treated at 37 °C or 41.5 °C. HS up-regulated the mRNA and protein expression level of heat shock protein 70 (HSP70). Furthermore, the proliferation of 3T3-L1 preadipocytes were significantly inhibited after HS treatment for 2 days. A large number of accumulated lipid droplets were observed under the microscope after HS treatment for 8 days. Notably, the result of oil red O staining showed that the number of lipid droplets increased significantly and the differentiation ability of the cells was enhanced after HS. Moreover, after 2 and 8 d of differentiation, HS increased the transcription levels of fat synthesis genes including peroxisome proliferators activated receptor γ (PPARγ), fatty acid binding protein 2 (AP2), fatty acid synthase (FAS) and CCAAT enhancer binding protein α (CEBPα) genes, while decreasing the transcription levels of lipid decomposition genes including ATGL and HSL genes. In addition, HS reduced the expression of AMPK and PGC-1α, as well as the dephosphorylation of AMPK. 5-Aminoimidazole-4-carboxamide ribonucleotide (AICAR) can eliminate HS induced lipogenesis by activating AMPK. These results indicated that HS inhibited the proliferation of 3T3-L1 preadipocytes and promoted lipid accumulation by inhibiting the AMPK-PGC-1α signaling pathway in 3T3-L1 preadipocytes. This work lays a theoretical foundation for improving the effect of HS on meat quality of livestock and provides a new direction for the prevention of obesity caused by HS.
