RESUMO

Chlorpyrifos (CPF) has been shown to have male reproductive toxicity in mice and rat. However, the association of CPF and male reproduction in pigs remains unknown. Therefore, this study attempts to investigate the damage of CPF on male reproduction in pigs and its potential molecular mechanisms. First, ST cells and porcine sperms were treated with CPF and then cell proliferation, apoptosis, motility of sperm, and oxidative stress levels were examined, respectively. Meanwhile, RNA sequencing was performed on ST cells before and after the treatment of CPF. The results of experiments in vitro showed that CPF had broad-spectrum toxic effects on ST cells and porcine sperms. The RNA-sequencing data and WB results indicated that CPF may regulate cell survival through the PI3K-AKT pathway. In conclusion, this study may lay the foundation for improving male fertility in pigs and provide theoretical information for human infertility.

The Period circadian regulator (PER) gene family, including PER1, PER2 and PER3, codes transcriptional repressors which could accurately control biological rhythms. PER1/2 gene was proved to be associated with bone mass and PER1 gene was associated with insulin-like growth factor binding proteins 3 (IGFBP3) levels in serum. However, it was few studies reported genetic effects of PER gene on growth traits at the individual level. In this study, we identified the potential insertion/deletion (indel) loci in PER1/2/3 gene, and then explored the relationship between goat growth traits and the frequency of genotype in Shaanbei white cashmere goats (n = 827). As a result, a 9 bp indel within PER1 gene (g.27528003-27528011 del.TGCTGCTGC; rs642467689) was identified using molecular biology techniques. In addition, there existed significant correlation between the 9 bp indel and body height, height at hip cross, chest depth, body length index and cannon circumference index of goats. These results suggested that the 9 bp indel variation in PER1 gene was associated with goat growth traits, providing the theoretical basic for the role of PER1 gene in goat breeding.

A new reflective display technology, the electrowetting display (EWD), has the advantages of high contrast ratio, high reflectivity, and ultralow power consumption. The response speed of EWDs has an important influence on optical performance, and pixel structure is one of the key factors affecting the response speed of EWDs. In order to improve the response speed, a new multi-electrode pixel structure is proposed in this paper. This structure was realized by dividing the pixel into four square-shaped sub-electrodes, and a three-dimensional EWD simulation model was established. In this model, a driving voltage was first applied to one of these sub-electrodes, and the oil was ruptured. Then, its two adjacent sub-electrodes were also supplied with driving voltages, so as to spur the oil to move to a pixel corner quickly. Simulation results showed that the response speed of EWDs can be effectively improved by using the proposed multi-electrode pixel structure. Compared with a single-electrode pixel structure, the oil rupture response time of the multi-electrode pixel structure was advanced by 0.6 ms. The pixel achieved a 2.7 ms faster response time than the single-electrode pixel for reaching a 50% aperture ratio in an opening process, and the maximum aperture ratio was increased by 6.2%.

Copy number variations (CNVs) have many forms of variation structure, and they play an important role in the research of variety diversity, biological evolution and disease correlation. Since CNVs have a greater impact on gene regulation and expression, more studies are being finalized on CNVs in important livestock and poultry species. The protein phosphatase 3 catalytic subunit alpha (PPP3CA) is a key candidate gene involved in the goat fecundity trait, and has important effects on precocious puberty, estrogen signal transduction pathways and oocyte meiosis. Additionally, PPP3CA also has a dephosphorylation effect in the process of spermatogonial stem cell meiosis and spermatogenesis. So far, there is no research on the relationship between the copy number variations of the PPP3CA gene and reproduction traits. Therefore, the purpose of this study was to determine the association between copy number variations in the goat PPP3CA gene and litter size and semen quality in Shaanbei white cashmere goats (SBWC) (n = 353) and Guizhou Heima goats (n = 64). Based on the association analysis, the results showed that only CNV1 and CNV2 within the PPP3CA gene were distinctly related to the first-birth litter size in female goats (p = 7.6802 × 10-11; p = 5.0895 × 10-9, respectively) and they were also significantly associated with the semen quality of SBWC goats (p < 0.05). In addition, individuals with Loss genotypes demonstrated better phenotypic performance compared to those with other types. Therefore, CNV1 and CNV2 of the PPP3CA gene are potentially useful for breeding, as they are linked to important goat reproduction traits.

AIMS AND OBJECTIVES: A high-performance liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for determining hypaphorine, a potential agent for treating osteoclast- based bone loss, was developed and validated in rat plasma. MATERIALS AND METHODS: Plasma samples were pretreated by the protein precipitation. Chromatographic separation was performed using an Inertsil ODS-3 column (50 mm × 4.6 mm, 5 µm). The mobile phase consisted of water (containing 0.1% formic acid) and acetonitrile in a gradient mode at a flow rate of 0.5 mL/min. The transitions from protonated precursor ion [M + H]+ to the particular daughter ion were acquired using selected reaction monitoring (SRM). The mass transitions of hypaphorine and IS were found to be 247 → 188 and m/z 219 → 188, respectively. The method was validated in terms of selectivity, linearity, accuracy and precision, extraction recovery and matrix effect, stability, and carryover. RESULTS: It showed good linearity over the range of 1-2000 ng/mL (R2 = 0.9978). The intra-batch accuracy was within 93.95-105.81%, and the precision was within 4.92-11.53%. The inter-batch accuracy was within 96.18-100.39% with a precision of 6.22-11.23%. The extraction recovery and matrix factors were acceptable. CONCLUSION: The simple and rapid method was successfully applied to the pharmacokinetic study in rats following oral administration of hypaphorine at the doses of 0.5, 1.5, and 4.5 mg/kg.

Electrophoretic displays (EPDs) have excellent paper-like display features, but their response speed is as long as hundreds of milliseconds. This is particularly important when optimizing the driving waveform for improving the response speed. Hence, a driving waveform design based on the optimization of particle activation was proposed by analyzing the electrophoresis performance of particles in EPD pixels. The particle activation in the driving waveform was divided into two phases: the improving particle activity phase and the uniform reference grayscale phase. First, according to the motion characteristics of particles in improving the particle activity phase, the real-time EPD brightness value can be obtained by an optical testing device. Secondly, the derivative of the EPD brightness curve was used to obtain the inflection point, and the inflection point was used as the duration of improving particle activity phase. Thirdly, the brightness curve of the uniform reference grayscale phase was studied to set the driving duration for obtaining a white reference grayscale. Finally, a set of four-level grayscale driving waveform was designed and validated in a commercial E-ink EPD. The experimental results showed that the proposed driving waveform can cause a reduction by 180 ms in improving particle activity phase and 120 ms in uniform reference grayscale phase effectively, and a unified reference grayscale can be achieved in uniform reference grayscale phase at the same time.

As a kind of paper-like display technology, power consumption is a very important index for electrowetting displays (EWDs). In this paper, the influence of driving waveforms on power consumption of the EWDs is analyzed, and a driving waveform with rising gradient and sawtooth wave is designed to reduce the power consumption. There are three stages in the proposed driving waveform. In the initial stage, the driving voltage is raised linearly from the threshold to the maximum value to reduce the invalid power consumption. At the same time, the oil breakup can be prohibited. And then, a section of sawtooth wave is added for suppressing oil backflow. Finally, there is a section of resetting wave to eliminate the influence of charge leakage. Experimental results show that the power consumption of the ultra-low power driving waveform is 1.85 mW, which is about 38.13% lower than that of the conventional used square wave (2.99 mW), when the aperture ratio is 65%.

The aim of our prospective study was to investigate the clinical results and advantages of a minimally invasive sinus tarsi approach with cannulated screw fixation combined with vacuum-assisted closure for the treatment of severe open calcaneal fractures with medial wounds. A total of 31 patients (32 feet) with open calcaneal fractures who were admitted to our hospital from January 2008 to May 2013 were selected for the study and randomly divided into 2 groups: the cannulated screw group (n = 16 patients, 16 feet) and the plate group (n = 15 patients, 16 feet). The Böhler and Gissane angles were compared before and after surgery. The clinical results were evaluated using according to the American Orthopaedic Foot and Ankle Society ankle-hindfoot scale and the rate of infection. The follow-up duration for all patients ranged from 10 to 36 (mean 24) months. No statistically significant differences were found in the radiologic indicators, incidence of early postoperative complications, or American Orthopaedic Foot and Ankle Society ankle-hindfoot scores (p > .05) between the 2 groups. However, a statistically significant difference was seen in the duration of hospitalization (p < .05) between the 2 groups. A minimally invasive sinus tarsi approach with cannulated screw fixation combined with vacuum-assisted closure is an effective method for the treatment of severe open calcaneal fractures with medial wounds. It provides good reduction and requires fewer days of hospitalization.
