RESUMO

DNA barcoding has greatly facilitated studies of taxonomy, biodiversity, biological conservation, and ecology. Here, we establish a reliable DNA barcoding library for Chinese snakes, unveiling hidden diversity with implications for taxonomy, and provide a standardized tool for conservation management. Our comprehensive study includes 1638 cytochrome c oxidase subunit I (COI) sequences from Chinese snakes that correspond to 17 families, 65 genera, 228 named species (80.6% of named species) and 36 candidate species. A barcode gap analysis reveals gaps, where all nearest neighbour distances exceed maximum intraspecific distances, in 217 named species and all candidate species. Three species-delimitation methods (ABGD, sGMYC, and sPTP) recover 320 operational taxonomic units (OTUs), of which 192 OTUs correspond to named and candidate species. Twenty-eight other named species share OTUs, such as Azemiops feae and A. kharini, Gloydius halys, G. shedaoensis, and G. intermedius, and Bungarus multicinctus and B. candidus, representing inconsistencies most probably caused by imperfect taxonomy, recent and rapid speciation, weak taxonomic signal, introgressive hybridization, and/or inadequate phylogenetic signal. In contrast, 43 species and candidate species assign to two or more OTUs due to having large intraspecific distances. If most OTUs detected in this study reflect valid species, including the 36 candidate species, then 30% more species would exist than are currently recognized. Several OTU divergences associate with known biogeographic barriers, such as the Taiwan Strait. In addition to facilitating future studies, this reliable and relatively comprehensive reference database will play an important role in the future monitoring, conservation, and management of Chinese snakes.

In this work, a simple one-step hydrothermal method was employed to prepare the Ce-doped Fe2O3 ordered nanorod arrays (CFT). The Ce doping successfully narrowed the band gap of Fe2O3, which improved the visible light absorption performance. In addition, with the help of Ce doping, the recombination of electron/hole pairs was significantly inhibited. The external voltage will make the performance of the Ce-doped sample better. Therefore, the Ce-doped Fe2O3 has reached superior photoelectrochemical (PEC) performance with a high photocurrent density of 1.47 mA/cm2 at 1.6 V vs. RHE (Reversible Hydrogen Electrode), which is 7.3 times higher than that of pristine Fe2O3 nanorod arrays (FT). The Hydrogen (H2) production from PEC water splitting of Fe2O3 was highly improved by Ce doping to achieve an evolution rate of 21 µmol/cm2/h.

The aphid parasitoid Aphelinus asychis Walker is an important biological control agent against many aphid species. In this study, we examined whether the rearing host aphid species (the pea aphid, Acyrthosiphon pisum and the grain aphid, Sitobion avenae) affect the performance of A. asychis. We found that A. pisum-reared A. asychis showed a significantly larger body size (body length and hind tibia length) and shorter developmental time than S. avenae-reared A. asychis. There was no difference in the sex ratio between them. The longevity of A. pisum-reared A. asychis was also significantly longer than that of S. aveane-reared A. asychis. Furthermore, A. pisum-reared A. asychis presented stronger parasitic capacity and starvation resistance than S. aveane-reared A. asychi. In addition, host aphid alteration experiments showed that A. asychis only takes two generations to adapt to its new host. Taken together, these results revealed that A. pisum is a better alternative host aphid for mass-rearing and releasing of A. asychis. The body size plasticity of A. asychis is also discussed.

Effective conservation of threatened biota relies on accurate assessments and scientific guidance. As an unfortunate example, Chinese giant salamanders ( Andrias, CGS) remain critically endangered in nature. Misguided conservation efforts, e.g., commercial propagation and releasing of millions of likely non-indigenous or interspecific hybrids, have further compromised conservation initiatives. Limited information on wild populations of CGS poses a significant conservation challenge. Following 18-month long field monitoring, we now report the discovery of a wild population of CGS in a closed nature reserve in Jiangxi Province, China. Genomic assessments reveal its genetic distinctiveness and do not detect genetic admixture with other species. Based on morphological and molecular evidences, we describe this CGS as a new species Andrias jiangxiensis sp. nov. This is the only known species of CGS today with a genetically pure, reproducing, in situ population. This discovery emphasizes the important role that closed nature reserves play in protecting species, and the necessity of integrating long-term field monitoring and genetic assessments. It sets a new pathway for discovering and conserving endangered species, especially for those biotas that are similarly being extirpated by anthropogenic translocations and overexploitation.

Harmonia axyridis is an important natural enemy that consumes many agricultural and forestry pests. It relies on a sensitive olfactory system to find prey and mates. Odorant-binding proteins (OBPs) as the first-step of recognizing volatiles, transport odors through sensillum lymph to odorant receptors (ORs). However, little is known about the molecular mechanisms of H. axyridis olfaction. In this study, four H. axyridis antenna specific OBP genes, HaxyOBP3, 5, 12, and 15, were bacterially expressed and the binding features of the four recombinant proteins to 40 substances were investigated using fluorescence competitive binding assays. Three-dimensional structure modeling and molecular docking analysis predicted the binding sites between HaxyOBPs and candidate volatiles. Developmental expression analyses showed that the four HaxyOBP genes displayed a variety of expression patterns at different development stages. The expression levels of HaxyOBP3 and HaxyOBP15 were higher in the adult stage than in the other developmental stages, and HaxyOBP15 was significantly transcriptionally enriched in adult stage. Ligand-binding analysis demonstrated that HaxyOBP3 and HaxyOBP12 only combined with two compounds, ß-ionone and p-anisaldehyde. HaxyOBP5 protein displayed binding affinities with methyl salicylate, ß-ionone, and p-anisaldehyde (Ki = 18.15, 11.71, and 13.45 µM). HaxyOBP15 protein had a broad binding profile with (E)-ß-farnesene, ß-ionone, α-ionone, geranyl acetate, nonyl aldehyde, dihydro-ß-ionone, and linalyl acetate (Ki = 4.33-31.01 µM), and hydrophobic interactions played a key role in the binding of HaxyOBP15 to these substances according to molecular docking. Taken together, HaxyOBP15 exhibited a broader ligand-binding spectrum and a higher expression in adult stage than HaxyOBP3, 5, and 12, indicating HaxyOBP15 may play a greater role in binding volatiles than other three HaxyOBPs. The results will increase our understanding of the molecular mechanism of H. axyridis olfaction and may also result in new management strategies (attractants/repellents) that increase the biological control efficacy of H. axyridis.

The insect chemosensory system plays an important role in many aspects of insects' behaviors necessary for their survival. Despite the complexity of this system, an increasing number of studies have begun to understand its structure and function in different insect species. Nonetheless, the chemosensory system in the orange spiny whitefly Aleurocanthus spiniferus, as one of the most destructive insect pests of citrus in tropical Asia, has not been investigated yet. In this study, the sensillum types, morphologies and distributions of the male and female antennae of A. spiniferus were characterized using scanning electron microscopy. In both sexes, six different sensilla types were observed: trichodea sensilla, chaetica sensilla, microtrichia sensilla, coeloconic sensilla, basiconic sensilla, and finger-like sensilla. Moreover, we identified a total of 48 chemosensory genes, including 5 odorant-binding proteins (OBPs), 12 chemosensory proteins (CSPs), 3 sensory neuron membrane proteins (SNMPs), 6 odorant receptors (ORs), 8 gustatory receptors (GRs), and 14 ionotropic receptors (IRs) using transcriptome data analysis. Tissue-specific transcriptome analysis of these genes showed predominantly expression in the head (including antennae), whereas CSPs were broadly expressed in both head (including the antennae) and body tissue of adult A. spiniferus. In addition, the expression profiling of selected chemosensory genes at different developmental stages was examined by quantitative real time-PCR which was mapped to the transcriptome. We found that the majority of these genes were highly expressed in adults, while AspiORco, AspiGR1, AspiGR2, and AspiIR4 genes were only detected in the pupal stage. Together, this study provides a basis for future chemosensory and genomic studies in A. spiniferus and closely related species. Furthermore, this study not only provides insights for further research on the molecular mechanisms of A. spiniferus-plant interactions but also provides extensive potential targets for pest control.

Phylogenetic reconstructions derived from DNA sequence data play a central role in documenting the number of species in a complex. Such analyses are pointing to the existence of many cryptic species, especially in poorly understood groups such as the genus Limnonectes, and the L. kuhlii species complex in particular. To understand the Limnonectes frogs of China, we reconstruct the major matrilineal genealogy of Limnonectes from China and Southeast Asia based on 12S rRNA, tRNAVal and 16S rRNA gene sequences. Based on new data we recognize five species of Limnonectes in China including L. bannaensis, L. fujianensis, L. fragilis, L. taylori (new record), and a new species from southern China and Myanmar. Phylogenetically, the new species is more closely related to the clade comprising L. taylori, L. megastomias, L. isanensis, L. nguyenorum, and L. jarujini from Thailand than to other Chinese species. This study supports previous findings of sympatric members of a species complex that are not each other's closest relatives.

BACKGROUND: Blunt injuries/contusion on eyes might cause retina blunt trauma. This study is to evaluate the protective function of BN52021 against retinal trauma. METHODS: A total of 70 cats, 6 months old, were divided into six groups: Group A to E (n = 12) and normal control (N) group (n = 10). The right eyes in Group A to E were contused. All experiments were performed under general anesthetization. Retrobulbar injections of medication in right eyes were performed. Cats were administrated with 0.5 mL of normal saline (NS), dimethyl sulphoxide, 0.2 g/L BN52021, 1 g/L BN52021 and 5 g/L BN52021, respectively. Cats in Group N were administrated with 0.5 mL of NS. Intraocular pressure (IOP), flash electroretinogram (ERG), and retinal nerve fiber layer (RNFL) thickness were measured. Hematoxylin and eosin (HE) staining and transmission electron microscope (TEM) were detected. RESULTS: No significant difference was observed in IOP levels among groups. Comparing with cats in Group N, those in Group A to E showed significant lower amplitudes of rod a- and b-waves (P < 0.05). Amplitudes of rod a- and b-waves were increased by administration of high concentration of BN52021 (≥ 1 g/L). Moreover, high concentration of BN52021 decreased the RNFL thickness increased by contusion. Axons in RNFL in Group E arranged neatly at 7 days after modeling. CONCLUSIONS: The degenerated axons caused by contusion were repaired by BN52021. The administration of high concentration of (≥ 1 g/L) BN52021 could partially repair retinal function in contused cat eyes.

The objective of this study is to define the population dynamics of Semiaphis heraclei in the main-producing district of Lonicera japonica in Shandong, and screen for highly efficient, safety control technique. Through fixed field investigation, we tested the toxicity of eight kinds of insecticides by using dipping methods, and carried out the field experiment. The results showed that the aphids' emergence peak appeared in May. The aphids on the Sijihua variety of L. japonica was more susceptible and the peak was also seven days earlier than Damao variety of L. japonica. The aphid populations on Sijihua were 1 fold than those on the Daomao in happened peak. Comparing the eight kinds of insecticides, the LC50 of lambda-cyhaothrin, abamectin, imidacloprid and pyrethrin to wingless aphids were 1.494, 1.690, 2.840, 2.861 mg x L(-1), respectively, whose toxicity were higher, the toxicity of matrine, pymetrozine and azadirachtin were also high. The field efficacy trials indicated that during the period of aphids occurred, 25% imidacloprid wettable powder, 1.8% abamectin missible oil, 2.5% lambda-cyhaothrin missible oil, 25% pymetrozine wettable powder, 5% pyrethrin missible oil, 1% matrine water aqua were sprayed at concentrations of 20,000, 2,000, 2,500, 5,000, 500 and 50 times, respectively,the control effect achieved 91.69%, 98.90%, 96.18%, 95.06%, 99.24%, 90.10%, respectively, after 5 days. During the growing period of L. japonica in spring, application of thiamethoxam, thiacloprid, pymetrozine and imidacloprid, all of the control effect against aphids achieved above 98.88% after 50 days. The result indicated that May was the S. heraclei Takahashi's emergence peak in Pingyi, Shandong. The efficient safety and environmentally friendly insecticides by spraying and systemic insecticide of pymetrozine and imidacloprid by root application were all efficient controlled aphids. These insecticides were long for controlling S. heraclei Takahashi and worthy of being widely applied.

BACKGROUND: Optimal management of patients with acute type aortic dissection is not clear. The clinical experience with total arch replacement combined with stented elephant trunk technique in the treatment of acute type B aortic dissection involving the aortic arch was analyzed. METHODS: From January 2006 to April 2011, 24 consecutive patients with acute type B aortic dissection involving the aortic arch underwent total aortic arch replacement with stented elephant trunk implantation. Concomitant procedures included the Bentall procedure in 3 patients, David procedure in 1 patient, and ascending aortic replacement in 7 patients. RESULTS: The times of cardiopulmonary bypass and selective cerebral perfusion averaged 168±41 and 21±4 minutes, respectively. One patient died of multiorgan failure 3 days after surgery. Complications occurred in 3 patients (3 of 24; 12.5%), including hoarseness and mediastinal bleeding requiring reoperation, each in 1 patient. One patient experienced transient consciousness dysfunction. The in-hospital mortality was 4.1% (1 of 24). No paraplegia occurred postoperatively. Follow-up was available in 22 patients (22 of 23; 95.7%). During the follow-up, 1 patient died of massive gastrointestinal bleeding 2 months after surgery. Type II endoleak occurred in 1 patient. One patient underwent thoracoabdominal aortic replacement 14 months after surgery. The complete thrombosis ratio of the false lumen was 86.4% (19 of 22) 6 months after surgery. The 5-year survival was 91.7%. CONCLUSIONS: Total arch replacement combined with stented elephant trunk technique was safe and feasible for acute type B aortic dissection involving the aortic arch. A low rate of morbidity and mortality and a satisfactory 5-year survival rate were achieved, as well as improved thrombosis of the false lumen.

OBJECTIVE: To evaluate the efficacy, predictability, stability, and safety of the surgical correction myopia using implantable Collamer lenses (ICL) in phakic eyes. METHODS: A prospective study was performed to analyze 91 eyes of 48 patients who had the implantation of ICL for the treatment of myopia from July 2008 to February 2010. Patients were examined preoperatively and followed at 1 week, 1, 3, 6, and 12 months postoperatively. The examinations included the uncorrected visual acuity (UCVA), best corrected visual acuity (BCVA), refraction, contrast sensitivity, wavefront aberration, intraocular pressure, space between crystal lens and intraocular lens (vault), endothelial cell density (ECD), anterior chamber depth (ACD), surgical complication, etc. RESULTS: Successful implantation was achieved in all patients. The mean follow-up time was (9.54 ± 4.12) months (SD)(range 1 to 12 months). The mean preoperative SE was -12.38 diopters (D) (range -5.0D to -23.0D). Postoperatively, UCVA was maintained or improved in all eyes. UCVA achieved 1.0 in 58 eyes (64%) and BCVA gained more than 1 line in 69 eyes (75.9%). The glare and no glare contrast sensitivity were improved at 3cd, 12cd and 18cd, with the exception of 6cd. The aberration decreased in RMS, spherical and coma. Post operative ACD (1 week after operation) diminished 8.92% as compared with the preoperative ACD. The mean vaulting was (452 ± 216.38) µm (6 months) and ranged 130 - 1080 µm at different follow-up periods. There was no statistically significant difference in vaulting between postoperative data at different periods (t = 0.200, P > 0.05). The mean postoperative ECD decreased but had no statistically difference compared with the preoperative ECD. ACD decreased 31% after surgery in 2 eyes (2.1%). Transient high IOP was observed in 13 eyes (2.1%) one week after the operation. CONCLUSION: These results indicate that ICL implantation in the treatment of myopia is efficient, predictable, safe, and reliable.

OBJECTIVE: To summarize the surgical experience of aortic arch operation under deep hypothermic circulatory arrest. METHODS: 22 patients suffering from aortic dissection or descending aorta aneurysm with the involvement of aortic arch received operation under deep hypothermic circulatory arrest. Eight patients underwent ascending aorta and partial aortic arch replacement, one patient received aortic root, ascending aorta, and partial aortic arch replacement, 2 patients received ascending aorta and total arch replacement, 2 patients received aortic valve replacement plus ascending aorta and partial aortic arch replacement, 8 patients underwent ascending aorta and total arch replacement plus elephant trunk technique (stunted elephant trunk used in 6 cases), and 1 patient received left partial aortic arch and descending aorta replacement. Coronary artery bypass grafting was performed concomitantly in 4 cases. RESULTS: Three patients died peri-operatively with a mortality rate of 13.6%. One patient had aortic dissection rupture before operation leading to cardiac tamponade, acute inferior myocardial infarction, and cardiac arrest. This patient received operation while resuscitation. After operation, the patient had severe right heart failure and died 16 hours later. One patient had bleeding and multi-organ failure, and died 3 days later. The third patient, with acute aortic dissection did not awake after operation, had pulmonary infection and multi-organ failure, and died 39 days later. Re-thoracotomy for bleeding was required in 3 cases; delayed awareness occurred in 3 cases; and 2 cases had renal failure after operation. CONCLUSIONS: Aortic arch operation includes partial aortic arch replacement, total arch replacement, and total arch replacement with elephant trunk technique. The operation procedure is selected according to the primary lesion and how aortic arch has been affected. Deep hypothermic circulatory arrest with selective cerebral perfusion facilitates complicated aortic arch operation, resulting in a reduction of mortality and morbidity for arch aneurysms or dissection.

OBJECTIVE: To investigate the visual evoked potentials in adults with migrainous vertigo (MV). METHODS: Totally 113 patients with MV were enrolled from vertigo clinic. Patients received necessary laboratory examinations as well as pattern visual evoked potential (PVEP) testing. RESULTS: Definite MV accounted for 46.9% (53/113) and probable MV accounted for 53.1% (60/113). Among 74 patients who received PVEP, the results were normal in 35 patients (47.3%) and abnormal in 39 patients (52.7%). The abnormal manifestations included lowered N75-P100 amplitude, elongated latency of P100, and lowered N75-P100 amplitude combined with delayed latency of P100. Seven patients with MV had unilateral lowered N75-P100 amplitude and 4 had bilateral abnormal amplitude. Nine patients had unilateral delayed latency of P100 and 11 had bilateral abnormal latency. Four patients had unilateral and 4 had bilateral abnormal N75-P100 amplitude and latency of P100. CONCLUSIONS: MV patients usually have abnormal PVEP. PVEP may become a useful electrophysiological test in the diagnosis of MV.
