RESUMO

Distant metastasis is the principal cause of mortality for breast cancer patients. Targeting specific mutations that have been acquired during the evolution process of advanced breast cancer is a potential means of enhancing the clinical efficacy of treatment strategies. In metastatic breast cancer, ARID1A is the most prevalent mutation of the SWI/SNF complex, which regulates DNA repair, recombination, and gene transcription. The low expression of ARID1A is associated with poor disease-free survival and overall survival of patients with luminal A or HER2-rich breast cancer. In addition, ARID1A plays a prominent role in maintaining luminal characteristics and has an advantage for identifying responses to treatment, including endocrine therapies, HDAC inhibitors and CDK4/6 inhibitors. The therapeutic vulnerabilities initiated by ARID1A alterations encourage us to explore new approaches to cope with ARID1A mutant-related drug resistance or metastasis. In this review, we describe the mutation profiles of ARID1A in metastatic breast cancer and the structure and function of ARID1A and the SWI/SNF complex as well as discuss the potential mechanisms of ARID1A-mediated endocrine resistance and therapeutic potential.

OBJECTIVE: To find the best synthesis method of 6-benzyl-1-[(benzyloxy)methyl]-3-hydroxy-5-(hydroxymethyl) pyrimidine-2,4(1H,3H)-dione e for observing the change of its biological activity after N-3 hydroxylation. METHODS: After trying some N-hydroxylation methods, the target compound was successfully synthesized via one-pot oxidizing process by sodium hydride (NaH) and 3-chloroperbenzoic acid(m-CPBA); the anti-HIV reverse transcriptase (RT) activity and integrase (IN) activity of the target compound was assayed via enzyme-linked immunesorbent assay (ELISA) and phosphorylation of DNA package method. RESULTS: The target compound could be obtained through the improved m-CPBA oxidative method by only one step, and the yield of the reaction could reach 60%-70%. And the structure of this compound was identified by 1H NMR, 13C NMR and MS; The activity result showed it added the anti-HIV IN activity after N-3 hydroxylation as well as retained the anti-HIV RT activity. CONCLUSION: The improved m-CPBA oxidative method is a convenient and efficient way to prepare the compound 6-benzyl-1-[(benzyloxy)methyl]-3-hydroxy-5-(hydroxymethyl)pyrimidine-2,4(1H,3H)-dione e which has both anti-HIV RT and IN activity.

OBJECTIVE: To investigate whether the administration of the ultra-filtration extract from Danggui Buxue Decoction (EDBD) was able to protect cardiomyocytes from oxidative injury of rats induced by hydrogen peroxide (H(2)O(2)) and its potential mechanism. METHODS: Myocardial cells from 1- to 2-day-old neonatal rats were cultured in Dulbecco's modified Eagle's medium low-glucose and Ham's F12 medium (1:1), and the cellular injury was induced by H(2)O(2). The ultra-filtration extract mixture from Angelica sinensis and Hedysarum polybotrys was given in three doses of 3.75, 7.5, and 15 mg/mL. Morphological changes of cardiomyocytes were observed by microscope. Survival rate of myocardial cells was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The cardiomyocyte damages were estimated by detecting lactate dehydrogenase (LDH) and creatine kinase (CK) releases in the medium, superoxide dismutase (SOD) activities, and intracellular malondialdehyde (MDA) and myeloperoxidase (MPO) contents. The levels of caspase-3 and heat shock protein 70 (hsp70) mRNA expression in cardiomyocytes were measured by reverse transcription polymerase chain reaction. RESULTS: The EDBD could protect the cardiomyocytes from H(2)O(2) injury in a dosedependent manner (3.75, 7.50, and 15.00 mg/mL). The EDBD could significantly decrease LDH and CK leakages and intracellular MDA and MPO contents, increase SOD activity, up-regulate hsp70 expression, and down-regulate caspase-3 expression. CONCLUSION: The EDBD has protection on cardiomyocytes injured by H(2)O(2) through improving cell antioxidant ability, up-regulating hsp70 expression, and inhibiting caspase-3 activity.

OBJECTIVE: To explore the relationships between traditional Chinese medicine (TCM) constitutional types and overweight or obesity so as to provide evidence for adjusting constitutional bias and preventing and treating obesity. METHODS: The data comes from a cross-sectional survey on TCM constitution of 18 805 samples aged above 18 in Beijing and 8 provinces (Jiangsu, Anhui, Gansu, Qinghai, Fujian, Jilin, Jiangxi and Henan) in China. The survey of TCM constitution was performed by standardized constitution in Chinese medicine questionnaire (CCMQ). Discriminatory analysis method was used to judge the individual's constitutional type (gentleness type, qi-deficiency type, yang-deficiency type, yin-deficiency type, phlegm-dampness type, dampness-heat type, blood-stasis type, qi-depression type and special diathesis type). The relationships between TCM constitution types and overweight or obesity was investigated by logistic regression analysis. RESULTS: Compared with gentleness type, the risk of overweight (OR, 2.05; 95% CI, 1.79-2.35) and obesity (OR, 4.34; 95% CI, 3.52-5.36) in phlegm-dampness type is significantly increased; the risk of obesity (OR, 1.60; 95% CI, 1.30-1.98) in qi-deficiency type is significantly higher; the risk of overweight and obesity in yang-deficiency type, blood-stasis type, and qi-depression type is significantly lower. CONCLUSION: Phlegm-dampness type and qi-deficiency type are the main constitutional risk factors of overweight or obesity.

OBJECTIVE: To investigate the changes of endocrine, cyclic nucleotide and immune systems in subjects of yang deficiency constitution, and to explore the relationship among characteristics and causes of yang deficiency constitution, the physiological and biochemical parameters. METHODS: Based on the diagnostic criteria for the clinical epidemiological investigation, sixty subjects of yang deficiency constitution and fifty of normal constitution were selected. Eight milliliters venous blood were taken from overnight fasted subjects at 8:00-9:00. The sera were obtained by centrifugation of the blood at the speed of 3000 r/min for 5 minutes, and they were stored at -70 degrees C until use. The serum levels of corticosterone, cortisol, adrenocorticotrophic hormone (ACTH), cyclic adenosine monophosphate (cAMP), cyclic guanosine monophosphate (cGMP), free triiodothyronine (FT3), free thyroxine (FT4), thyrotropic-stimulating hormone (TSH), interleukin-1beta and interleukin-2 were measured by enzyme-linked immunosorbent assay; the cAMP/cGMP ratio was also computed; and the differences of the above indexes were compared between the two types of subjects. RESULTS: The serum levels of corticosterone, interleukin-1beta, TSH and cAMP/cGMP ratio of yang deficiency constitution significantly increased as compared with those of normal constitution, and the serum levels of cortisol, ACTH, cGMP and FT4 of yang deficiency constitution significantly decreased in comparison with those of normal constitution. CONCLUSION: Subjects of yang deficiency constitution may be not only related to hypothalamic-pituitary-adrenal axis and hypothalamic-pituitary-thyroid axis, but also related to the functional disorders of cyclic nucleotide and immune systems.

OBJECTIVE: To treat pulmonary fibrosis and study its mechanisms. METHODS: Choosing randomly 24 Wistar rats with normal sodium intratracheal injection as normal control group, the rests with heomycin A5 induced fibrosis were divided randomly into model group 24 as positive control, prednisone group 24, total hedysarum polybotyssaccharide (THPS) group 24, THPS and small dose prednisone group 24, and treated with different drugs. 6 rats of every group were put to death and observed pathological section, using imaging processing computer to quantitative analysis histomorphology, collagen, and transforming growth beta1, (TGF-beta1) on 7, 14, 30, 60 days. RESULTS: The group treated by THPS combination small dose prednisone showed up the most effects in the 3 treatment groups. CONCLUSION: THPS combination small dose prednisone to treat pulmonary fibrosis of rats is better than classic ways and its efficacy of inhibition TGF-beta1 may be a mechanism.

OBJECTIVE: To study the erythrocyte immuno-regulatory effect of Patrinia scabra Bunge extracts extracted by macroporous adsorptive resins in tumor bearing mice. METHODS: Patrinia scabra Bunge was extracted by macroporous adsorptive resins, and the amount of polysaccharides and saponins in the extract were determined. Mice bearing S180 tumor were treated with the extract and their survival prolongation rate, erythrocyte rosette formation rates of C3b receptor (ERR-CR), immune complex (ERR-IC) and tumor cell (ERR-TC), as well as the CD35 and CD44s were observed. RESULTS: Polysaccharide content was 21.4%, saponin 41.8% in the extract. As compared with the model group, the survival rate was increased, the erythrocyte immune function was improved (showed increase of ERR-CR and ERR-TC, decrease of ERR-IC), and the amount of CD35 and CD44s in red blood cell membrane increased in mice after being treated with the extract (P < 0.05 or P < 0.01). CONCLUSION: Extract of Patrinia scabra Bunge extracted by macroporous adsorptive resins can regulate the erythrocyte immune function to a certain extent.

OBJECTIVE: To optimize the conditions for the extraction of Patrinia scabra Bunge saponins. METHODS: Orthogonal experimental design and ultrasonic method were employed to examine the conditions for the extraction by determination of saponins. RESULTS: The optimun condition for the extraction of Patrinia scabra Bunge saponins was as follows: 65% ethanol for 40 minutes, 55 degrees C and 210 watt of ultrasonic efficinecy. CONCLUSION: The extraction method of Patrinia scabra Bunge sponins is simple and efficient.

AIM: To investigate the effects of Fuzheng Yiliu granules (body-resistance strengthening and tumor-suppressing granules) in patients with esophageal carcinoma. METHODS: We compared the immune adherent properties of red blood cells (RBCs), the expression of metastasis protein CD44, and the metastasis inhibition factor nm23, in esophageal carcinoma tumor cells of patients before and after radiotherapy in the presence and absence of orally administered Fuzheng Yiliu granules. Sixty-three hospitalized patients with esophageal carcinoma were treated with standard radiotherapy and randomly divided into treatment group (n = 30) treated with both radiotherapy and Fuzheng Yiliu granules and control group (n = 33) given radiotherapy only. Blood samples and tumor tissue were obtained before and after 21 d of treatment. The rosette rates for complement receptor type 3b (C3bRR) and immune complex receptor (ICRR) on RBCs were measured by erythrocyte immunological methods. Expression of CD44 and nm23 in tumor tissue sections was determined by immunohistochemical staining with monoclonal antibodies CD44v6 ad nm23H-1, respectively. RESULTS: The positivity of RBC-C3bRR before and after 21 d of treatment increased from 7.78% +/- 1.59% to 10.03% +/- 2.01% in the double treatment group, while it changed only slightly from 7.18% +/- 1.29% to 7.46% +/- 1.12% in the radiotherapy group. The positive rate for RBC-ICRR decreased from 37.68% +/- 2.51% to 22.55% +/- 1.65% after the double treatment, and from 37.28% +/- 2.41% to 24.69% +/- 1.91% in radiotherapy group at the same time points. The difference in erythrocyte immune adherent function between the two groups was significant (P < 0.01, t-test). The CD44(+)-cases were reduced from 21 (70.00%) to 12 (40.00%) after treatment with Fuzheng Yiliu granules, whereas the CD44(+)-cases (69.70%) in the radiotherapy group remained unchanged. The difference between the treatment (40.00%) and control (69.70%) groups was significant (P < 0.05). Although the nm23(+)-cases were increased from 4 (13.33%) to 6 (20.00%) in the double treatment group and from 6 (18.18%) to 7 (21.21%) in the radiotherapy group, the difference was not significant (P > 0.05). CONCLUSION: Fuzheng Yiliu granules enhance the immune adhesion function of RBCs and reduce the number of CD44(+)-cells in esophageal carcinoma patients, suggesting a potential role of these Chinese herbals in suppression of invasion and metastasis of malignant cells. However, this anti-metastatic effect has yet to be validated in vivo.

OBJECTIVE: To study the clinical efficacy of Fuzheng Jiedu Granule (FJG) in coronary heart disease (CHD) patients in long term contact with nickel (Ni) and its effect on electrocardiogram (ECG) and cardial enzymes. METHODS: Sixty cases were randomly assigned to two groups with 30 cases in each group, the treatment group treated with FJG plus Western medicine and the control group given Western medicine alone. The therapeutic course in both groups was 4 weeks. Another 15 healthy subjects from the same region were taken as the healthy control. Changes of clinical symptoms and ECG were observed, and the serum levels of creatine kinase (CK), lactic acid (LD), lactic dehydrogenase (LDH), Na+-K+-ATPase, Ca2+-ATPase and Ni were detected before and after treatment. RESULTS: The ameliorative effects on symptoms, signs and ECG in the treatment group were superior to those in the control group (P<0.05). Compared with healthy subjects, the serum levels of CK, LD, LDH and Ni were higher and the serum levels of Na+-K+-ATPase and Ca2+-ATPase were lower in patients before treatment; after treatment, the decrease of serum CK, LD, LDH and Ni levels and increase of serum Na+-K+-ATPase and Ca2+-ATPase were more significant in the treatment group than those in the control group (P<0.05 or P<0.01). CONCLUSION: FJG can decrease the serum Ni level, recover the activity of myocardial enzymes, thus to improve symptoms and abnormal ECG figures in CHD patients.

OBJECTIVE: To observe the effect of traditional Chinese herbs Fuzheng Yiliu Granule (FYG)-contained serum from tumor bearing mice on apoptotic rate, free radicals content and mitochondrial membrane potential of hepatoma cell lines H22 in vitro. METHODS: The effect of FYG drug-serum on apoptosis of hepatoma cell line H22 was determined using flow cytometry. The changes of DNA RNA, free radicals and mitochondrial membrane potential (delta psi m) in H22 cell were detected through laser scanning confocal microscope. RESULTS: FYG-contained serum can induce the apoptosis of H22 cell, enhance the free radicals content, and reduce the content of DNA RNA and delta psi m of H22 cell in vitro. CONCLUSION: The apoptosis of hepatoma cell line H22 induced by FYG is probably correlated to the change of free radicals content and delta psi m.

OBJECTIVE: to study depressant effect of total hedysarum polybotys saccharids (THPS) on S180 tumor-bearing-mice and its mechanisms. METHODS: THPS was extracted from Radix Xedysari with water and precipitated with ethanol, determining its molecular weight, purity, saccharide and aldonic acid content. 90 Kunming mice were divided into 9 groups randomly. One group was the normal group, the others were divided into 8 groups randomly after inculating S180 tumors and were treated with THPS and THPS combination cyclophosphamide (CY) in low, moderate and high dose, to put them to death after 14 days. To determine every tumor weight, the rate of depressant tumor, the contents of IL-2 and TNF-alpha with ELISA, and NF-kappaB with immunochemistry. RESULTS: The moderate dose THPS conspicuously possessed a depressant effect on S180 tumor and joint action with combination CY and increased the contents of IL-2, TNF-alpha and NF-kappaB of mice. CONCLUSION: THPS of moderate dose possesses a depressant effect on S180 tumor through increasing the contents of IL-2, TNF-alpha and NF-kappaB of mice.

RESUMO

OBJECTIVE: To research the erythrocyte immunoregulation effects of Patrinia scabra extracts by macroporous adsorptive resins on mice burdened transplanted tumor. METHODS: Extracts of Patrinia scabra Bunge were separated by macroporous adsorptive resins, ingredients were analysised. Mice burdened transplanted tumor were given extracted drugs. Life prolongation rate was observed, erythrocyte immunologic function and the CD35, CD44s contents of red blood cell were evaluated. RESULTS: Polysaccharide and saponin accounted for 8.4% and 48.4%. Extracts could porolong life expectancy of mice, improve erythrocyte immunolgic function and increase the CD35 and CD44s contents of red blood cell. CONCLUSION: Extracts of Patrinia scabra Bunge by macroporous adsorptive resins have erythrocyte immunoregulation effects on mice burdened transplanted tumor.

OBJECTIVE: To evaluate the effects of Fuzheng Yiliu Granules (FZYLG) on apoptotic rate and mitochondrial membrane potential (Delta psi m) of hepatocellular carcinoma cell line H22 from mice. METHODS: Forty-eight mice inoculated with H22 cells were randomly divided into four groups: untreated group, cyclophosphamide-treated group, high-dose FZYLG-treated group and low-dose FZYLG-treated group. After 14 days of corresponding treatment, H22 cells in each group were stained with propidium iodide, and the apoptotic rates were detected by flow cytometry (FCM). The rhodamine 123 was used as a fluorescence probe to label the H22 cells, and the fluorescence intensities were observed with laser scanning confocal microscope. The fluorescence intensity of H22 cells indicated the Delta psi m of H22 cells. RESULTS: FZYLG could significantly increase the apoptotic rate while reduce the Delta psi m of H22 cells from mice as compared with those in the untreated group. CONCLUSION: The antitumor effects of FZYLR on H22 cells from mice are related to decreasing the Delta psi m and then inducing the apoptosis of the H22 cells.

A HPLC method with photodiode array detection (PDA) was developed for the determination and a pharmacokinetic study of eleutheroside E (ELU E) and eleutheroside B (ELU B) in rat plasma and tissue following an eleutherococcus injection. The analysis was performed on a Kromasil C18 column, using water-acetonitrile as the gradient mobile phase and 0.8 mL/min flow rate. Detection wavelengths of ELU E and ELU B were 220 and 206 nm, respectively. Protein from the biological sample was deposited using acetonitrile. ELU E and ELU B were extracted from the biological samples using acetonitrile, separated by solid-phase extraction, and eluted from the cartridge using 60% methanol. The extraction recovery of ELU E and ELU B was 91.2 and 88.8%, respectively. The limit of detection was 37.6 ng/mL for ELU E and 37.0 ng/mL for ELU B (S/N = 3) in plasma. Blood drug level-time cuvers of ELU E and ELU B in Wister rats following administration of an eleutherococcus injection into femoral vein were shown to fit a three-compartment model. The half-life (t1/2) was 4.662 h for ELU E and 2.494 h for ELU B. Following administration of a single eleutherococcus injection, the concentration of ELU E and ELU B in the tissue was Cliver > Ckidney > Cspleen > Cheart and Ckidney > Cliver > Cheart. We believe the method described in the present paper is accurate and reliable and can be used for pharmacokinetic studies of ELU E and ELU B in rats. In addition, the method for sample preparation, using solid phase extraction, is precise, simple and rapid.

OBJECTIVE: To study the effects of Fuzheng Yiliu Granule (FZYLG)-medicated serum on apoptosis of liver cancer cells H22 from mice and its mechanism. METHODS: Liver cancer cells H22 from mice were incubated in culture media containing sera from rabbits medicated with different doses of FZYLG. Flow cytometry was used to examine the cell cycle and analyze the apoptotic rate of the H22 cells. The morphological changes of the H22 cells were observed by transmission electron microscope and the apoptosis related proteins Bcl-2 and Bax were examined by streptavidin-biotin peroxidase complex (SABC) method. RESULTS: FZYLG-medicated serum could influence the cell cycle and stop the proliferation of H22 cells at the G(1)/G(0) phase with apoptotic peak being detected. In culture media with FZYLG-medicated sera, the expression of Bcl-2 decreased while that of Bax increased as compared with that in culture medium with non-medicated serum (P<0.05). CONCLUSION: FZYLG-medicated serum can induce apoptosis of the liver cancer cells H22 by influencing the cell cycle, down-regulating the expression of Bcl-2 and up-regulating the expression of Bax.
