BACKGROUND: Myocardial ischemia-reperfusion injury (I/RI) is a major cause of perioperative cardiac-related adverse events and death. Studies have shown that sevoflurane postconditioning (SpostC), which attenuates I/R injury and exerts cardioprotective effects, regulates mitochondrial dynamic balance via HIF-1α, but the exact mechanism is unknown. This study investigates whether the PI3K/AKT pathway in SpostC regulates mitochondrial dynamic balance by mediating HIF-1α, thereby exerting myocardial protective effects. METHODS: The H9C2 cardiomyocytes were cultured to establish the hypoxia-reoxygenation (H/R) model and randomly divided into 4 groups: Control group, H/R group, sevoflurane postconditioning (H/R + SpostC) group and PI3K/AKT blocker (H/R + SpostC + LY) group. Cell survival rate was determined by CCK-8; Apoptosis rate was determined by flow cytometry; mitochondrial membrane potential was evaluated by Mito Tracker™ Red; mRNA expression levels of AKT, HIF-1α, Opa1and Drp1 were detected by quantitative real-time polymerase chain reaction (qRT-PCR); Western Blot assay was used to detect the protein expression levels of AKT, phosphorylated AKT (p-AKT), HIF-1α, Opa1 and Drp1. RESULTS: Compared with the H/R group, the survival rate of cardiomyocytes in the H/R + SpostC group increased, the apoptosis rate decreased and the mitochondrial membrane potential increased. qRT-PCR showed that the mRNA expression of HIF-1α and Opa1 were higher in the H/R + SpostC group compared with the H/R group, whereas the transcription level of Drp1 was lower in the H/R + SpostC group. In the H/R + SpostC + LY group, the mRNA expression of HIF-1α was lower than the H/R + SpostC group. There was no difference in the expression of Opa1 mRNA between the H/R group and the H/R + SpostC + LY group. WB assay results showed that compared with the H/R group, the protein expression levels of HIF-1α, Opa1, P-AKT were increased and Drp1 protein expression levels were decreased in the H/R + SpostC group. HIF-1α, P-AKT protein expression levels were decreased in the H/R + SpostC + LY group compared to the H/R + SpostC group. CONCLUSION: SpostC mediates HIF-1α-regulated mitochondrial fission and fusion-related protein expression to maintain mitochondrial dynamic balance by activating the PI3K/AKT pathway and increasing AKT phosphorylation, thereby attenuating myocardial I/R injury.

China is prone to broad land degradation and thus has been implementing ecological restoration projects (ERPs) since the reform and opening up. The extent of ERPs, as well as the varied planting efforts including tree gain projects (TGPs), grass gain projects (GGPs), and shrub gain projects (SGPs), have remained largely unknown. In addition, the mixed success of ERPs on preventing soil erosion and improving biodiversity is not well known. Based on a land use and land cover (LULC) product and a trajectory-based change detection approach, we successfully generated the first national map of ERPs associated with land use and land cover change (LUCC) and its three associated subcategories. Then, we applied the Integrated Valuation of Ecosystem Services and Tradeoffs (InVEST) model to evaluate the dynamics of sediment retention and habitat quality. In addition, we explored the heterogeneous patterns for the ecological impacts of ERPs. Our results suggested that during the past 40 years, a total ERP area of 9.54 × 106 hm2 was observed nationwide, mainly in the northwestern provinces of China. Of the three ERP subcategories, TGPs accounted for the largest area (48.55%), followed by GGPs (47.50%) and SGPs (3.96%). The national average sediment retention experienced a significant increase, whereas the national average habitat quality experienced a significant decline. ERP-driven increases in habitat quality were offset partly by the LUCCs induced by economic development policies in some regions, especially in northeast China. The simultaneous effect of construction land expansion and ERP implementation on sediment retention made the synchronization between ERP implementation and sediment retention improvement insignificant. We also suggested the optimal direction for ERP implementation.

Global land-cover datasets are key sources of information for understanding the complex inter-actions between human activities and global change. They are also among the most critical variables for climate change studies. Over time, the spatial resolution of land cover maps has increased from the kilometer scale to 10-m scale. Single-type historical land cover datasets, including for forests, water, and impervious surfaces, have also been developed in recent years. In this study, we present an open and synergy framework to produce a global land cover dataset that combines supervised land cover classification and aggregation of existing multiple thematic land cover maps with the Google Earth Engine (GEE) cloud computing platform. On the basis of this method of classification and mosaicking, we derived a global land cover dataset for 6 years over a time span of 25 years. The overall accuracies of the six maps were around 75% and the accuracy for change area detection was over 70%. Our product also showed good similarity with the FAO and existing land cover maps.
