OBJECTIVES: To investigate whether negative remodeling (NR) detected by intravascular ultrasound (IVUS) of the side branch ostium (SBO) would affect in-stent neointimal hyperplasia (NIH) at the one-year follow-up and the clinical outcome of target lesion failure (TLF) at the long-term follow-up for patients with left main bifurcation (LMb) lesions treated with a two-stent strategy. METHODS: A total of 328 patients with de novo true complex LMb lesions who underwent a 2-stent strategy of percutaneous coronary intervention (PCI) treatment guided by IVUS were enrolled in this study. We divided the study into two phases. Of all the patients, 48 patients who had complete IVUS detection pre- and post-PCI and at the 1-year follow-up were enrolled in phase I analysis, which aimed to analyze the correlation between NR and in-stent NIH at SBO at the 1-year follow-up. If the correlation was confirmed, the cutoff value of the remodeling index (RI) for predicting NIH ≥ 50% was analyzed next. The phase II analysis focused on the incidence of TLF as the primary endpoint at the 1- to 5-year follow-up for all 328 patients by grouping based on the cutoff value of RI. RESULTS: In phase I: according to the results of a binary logistic regression analysis and receiver operating characteristic (ROC) analysis, the RI cutoff value predicting percent NIH ≥ 50% was 0.85 based on the ROC curve analysis, with a sensitivity of 85.7%, a specificity of 88.3%, and an AUC of 0.893 (0.778, 1.000), P = 0.002. In phase II: the TLR rate (35.8% vs. 5.3%, P < 0.0001) was significantly higher in the several NR (sNR, defined as RI ≤ 0.85) group than in the non-sNR group. CONCLUSION: The NR of LCxO is associated with more in-stent NIH post-PCI for distal LMb lesions with a 2-stent strategy, and NR with RI ≤ 0.85 is linked to percent NIH area ≥ 50% at the 1-year follow-up and more TLF at the 5-year follow-up.

RESUMO

As a highly invasive carcinoma, esophageal cancer (EC) was the eighth most prevalent malignancy and the sixth leading cause of cancer-related death worldwide in 2020. Esophageal squamous cell carcinoma (ESCC) is the major histological subtype of EC, and its incidence and mortality rates are decreasing globally. Due to the lack of specific early symptoms, ESCC patients are usually diagnosed with advanced-stage disease with a poor prognosis, and the incidence and mortality rates are still high in many countries, especially in China. Therefore, enormous challenges still exist in the management of ESCC, and novel strategies are urgently needed to further decrease the incidence and mortality rates of ESCC. Although the key molecular mechanisms underlying ESCC pathogenesis have not been fully elucidated, certain promising biomarkers are being investigated to facilitate clinical decision-making. With the advent and advancement of high-throughput technologies, such as genomics, proteomics and metabolomics, valuable biomarkers with high sensitivity, specificity and stability could be identified for ESCC. Herein, we aimed to determine the epidemiological features of ESCC in different regions of the world, especially in China, and focused on novel molecular biomarkers associated with ESCC screening, early diagnosis and prognosis prediction.

RESUMO

The synthesis, conformational structure, electronic property, and anion complexation of novel coronarenes were systematically studied. A number of corona[4]arene[2]tetrazines that contain different combinations of nitrogen atoms with O, S, SO2, and CH2 as bridging units were synthesized conveniently by means of a fragment coupling strategy based on efficient nucleophilic aromatic substitution reaction of easily available aromatic dinucleophiles and 3,6-dichlorotetrazine. The resulting macrocycles adopt crownlike conformational structures with the nitrogen bridge(s) forming conjugation with carbonyl and the other heteroatom linkages with tetrazine. CV and DPV measurements showed that the tetrazine-bearing coronarenes were electron deficient with reduction potentials ranging from -896 to -960 mV. Owing mainly to noncovalent anion-π attractive interactions, N2,O4-corona[4]arene[2]tetrazine formed complexes with anions of varied geometries and shapes yielding diverse assembled structures in the solid state.

Photodynamic therapy (PDT) has been widely applied in cancer treatment due to minimal invasion, negligible side effects and specific tumor ablation. However, the treatment efficiency has been hindered by hypoxia in solid tumors, hydrophobic photosensitizers and their real time tracking. In this paper, we constructed an intelligent and biocompatible bovine serum albumin (BSA)-Ce6-Si QDs-MnO2 (BCSM NPs) nanocomplex as a pH/H2O2 responsive photosensitizer nanocarrier to modulate tumor hypoxia for fluorescence imaging-guided efficient PDT. This versatile nanosystem not only enhanced the loading capacity of the photosensitizer and the formation of cytotoxic singlet oxygen (1O2) owing to the intelligent production of oxygen catalyzed by MnO2 from the endogenous H2O2, but also performed as a dual functional fluorescence and Magnetic Resonance imaging (MRI) probe. In vivo experiments in nude mice further confirmed that BCSM NPs significantly inhibited the growth of HeLa bearing-tumors compared to free Ce6. The results highlight the great potentail of multifunctional BCSM NPs for in vivo imaging as well as enhancing the photodynamic therapy efficiency.

Enzyme-responsive assembly represents one of the increasingly significant topics in biomaterials research and finds feasible applications to the controlled release of therapeutic agents at specific sites at which the target enzymes are located. In this work, based on the concept of host-guest chemistry, a trypsin-responsive supramolecular vesicle using p-sulfonatocalix[4]arene as the macrocyclic host and natural serine protease trypsin-cleavable cationic protein protamine as the guest molecule, is reported. The complexation of p-sulfonatocalix[4]arene with protamine directs the formation of a supramolecular binary vesicle, which is dissipated by trypsin with high selectivity. Therefore, the present system represents a principle-of-concept to build a controlled-release carrier at trypsin-overexpressed sites.

RESUMO

We report that QATPE, an aggregation-induced emission-active tetraphenylethene dye, can be used as a non-sequence-specific ssDNA probe for real-time monitoring of all rolling circle amplification (RCA) reactions, thus making RCA more suitable for biosensing applications.

RESUMO

Two new acylgluconic acids, trans-feruloylgluconic acid (1) and trans-caffeoylgluconic acid (2), together with three known compounds, myo-inositol (3), phthalic acid dibutyl ester (4), and wuzhuyuamide-I (5), were isolated from the water soluble part of the dried nearly ripe fruits of Evodia rutaecarpa (Juss.) Benth. Their structures were determined by spectroscopic methods, including IR, UV, ESITOFMS, HRSIMS, 1D and 2D NMR spectral analyses.
