RESUMO

MMP-21 is a newly identified member of the matrix metalloproteinase family and has been reported to regulate both embryonic development and tumor progression. However, the roles of MMP-21 in hemofiltrate C-C chemokine (HCC) remain largely unclear. In this study, we used western blot, qPCR and immunohistochemistry (IHC) to determine the upregulation of MMP-21 in HCC tissues, and showed that the increase in MMP-21 was associated with vascular invasion and poor prognosis. Although changing levels of MMP-21 in HCC cell lines had no significant effect on cell migration or invasion abilities in in vitro transwell tests, both IHC analysis and in vivo mouse models proved that upregulated MMP-21 promoted metastasis. Functional enrichments of MMP-21 using The Cancer Genome Atlas (TCGA) data suggested that MMP-21 might regulate metastasis via macrophages. Further experiments proved that MMP-21 enhanced macrophage recruitment by increasing CCL-14 levels and promoted M2-type polarization of macrophage by elevating the expression of CSF-1 and FGF-1. Taken together, this study revealed that MMP-21 controlled the tumor microenvironment remodeling and functional regulation of macrophages to regulate HCC metastasis.

Various metabolic diseases are closely related to lipid metabolism disorders, but the regulatory effect of long noncoding RNAs (lncRNAs) on the function of lipids has been poorly elucidated. Previous our work has found that lncNONMMUG027912 (abbreviated as lnc027912) involved in cholesterol metabolism. Here, we further explored the novel function of lipid metabolism-associated lnc027912. We found that upregulated lnc027912 in AML12 cells treated with oleic acid (OA) and palmitic acid (PA) showed a significant decrease in lipid accumulation, triglyceride (TG) levels, and lipid biosynthesis genes. In terms of regulatory mechanisms, lnc027912 increased the expression of p-AMPKα, inhibited p-mTOR levels, decreased the expression of SREBP1C in nuclei, decreased the promoter activity of SREBP1C, and inhibited the expression of lipid synthesis genes. Most importantly, lnc027912 could reduce lipid accumulation and liver inflammation through AMPKα/mTOR signal axis in nonalcoholic fatty liver disease (NAFLD) mice model. Altogether, our study revealed a novel molecular mechanism of lnc027912 in lipid metabolism through the AMPKα/mTOR/SREBP1C signaling axis and highlights the potential of lnc027912 as a new treatment target for lipid disorder diseases (such as NAFLD).

Due to loss and degradation of natural wetlands, waterbirds increasingly rely on surrounding human-dominated habitats to obtain food. Quantifying vigilance patterns, investigating the trade-off among various activities, and examining the underlying mechanisms will help us understand how waterbirds adapt to human-caused disturbances. During two successive winters (November-February of 2012-13 and 2013-14), we studied the hooded crane, Grus monacha, in the Shengjin Lake National Nature Reserve (NNR), China, to investigate how the species responds to human disturbances through vigilance and activity time-budget adjustments. Our results showed striking differences in the behavior of the cranes when foraging in the highly disturbed rice paddy fields found in the buffer zone compared with the degraded natural wetlands in the core area of the NNR. Time spent vigilant decreased with flock size and cranes spent more time vigilant in the human-dominated buffer zone. In the rice paddy fields, the birds were more vigilant but also fed more at the expense of locomotion and maintenance activities. Adult cranes spent more time vigilant and foraged less than juveniles. We recommend habitat recovery in natural wetlands and community co-management in the surrounding human-dominated landscape for conservation of the hooded crane and, generally, for the vast numbers of migratory waterbirds wintering in the middle and lower reaches of the Yangtze River floodplain.
