Underground coal gasification is of great strategic significance to the effective and clean development of coal resources and scale up production of natural gas worldwide. Selection evaluation is the foundation of the exploration and development of underground coal gasification. In this paper, the differences between mid-deep (500-2200 m) and shallow layer (<500 m) underground coal gasification are analyzed, the key parameters affecting underground coal gasification are clearly identified, and the selection evaluation technology system is established. The results show that there are great differences between mid-deep and shallow layer underground coal gasification in terms of furnace construction site selection, engineering process, gasification efficiency and gasification products, the former is the main development direction in the future considering the resource potential, gasification efficiency, environmental protection and technological progress. The research of mid-deep underground coal gasification selecting should be carried out step by step with "favorable area-favorable zone-favorable zone ranking-favorable target" evaluation system. In the early stage of exploration, the product of thickness and volatiles can be used as a rapid evaluation index. Within the suitable range, the larger the index is, the more favorable it is, and when the index is greater than 3, it is defined as the favorable area. Further, ten key geological parameters of coal seam geological conditions, coal-rock and coal-quality characteristics and structural hydrologic conditions are selected and graded during zone evaluation according to different threshold ranges. At the same time, a favorable zone queuing method based on the two-factor evaluation method is established. Additionally, the final target site selection is established with more detailed evaluation parameters, such as process performance parameters, gasification characteristics parameters, technical recoverable indicators, economic recoverable indicators. The study provides the siting selection technology for the middle deep coal underground gasification, which is of great significant for the development of coal underground gasification industry.

Multicolor two-photon endomicroscopy has become a highly competitive tool for functional imaging in biomedical researches. However, to make the imaging system miniature and applicable for freely behaving animal brain activity, metalenses have received much attention in compact imaging systems. For high resolution multicolor imaging and maximizing fluorescence collection, there is a challenge metalenses faced to achieve large numerical aperture (NA) and focus the NIR excitation and VIS emission lights of multiple fluorophores to the same distance simultaneously because of the limitation of the group delay range of the meta-units. In this paper, we proposed a high NA and polarization-insensitive ultra-broadband achromatic metalens specifically for achromatically focusing the excitation and emission light of multiple fluorophores commonly used in neuroscience studies. TiO2 and Si meta-unit libraries composed of heights, widths and the corresponding phase and group delay were constructed, and the optimal meta-units were selected by particle swarm optimization algorithm to engineer the dispersion of metalens in the VIS band and NIR band, respectively. Combining dispersion engineering with spatial multiplexing, the proposed metalens achieved the maximal effective NA up to 0.8 and large achromatic bandwidth ranging from 500 nm to 1050 nm, which exhibited the coefficient of variation of focal lengths was only 3.41%. The proposed achromatic metalens could successfully achromatically focus different fluorescence with any polarization, which was suitable for most fluorophores. Our results firmly establish that the proposed metalens can open the door to high resolution and minimally invasive multicolor two-photon functional imaging in intravital deep brain.

The yellow-throated marten (Martes flavigula) is a medium-sized carnivore that is widely distributed across much of Asia and occupies an extensive variety of habitats. We reported a high-quality genome assembly of this organism that was generated using Oxford Nanopore and Hi-C technologies. The final genome sequences contained 215 contigs with a total size of 2,449.15 Mb and a contig N50 length of 68.60 Mb. Using Hi-C analysis, 2,419.20 Mb (98.78%) of the assembled sequences were anchored onto 21 linkage groups. Merqury evaluation suggested that the genome was 94.95% complete with a QV value of 43.75. Additionally, the genome was found to comprise approximately 39.74% repeat sequences, of which long interspersed elements (LINE) that accounted for 26.13% of the entire genome, were the most abundant. Of the 20,464 protein-coding genes, prediction and functional annotation was successfully performed for 20,322 (99.31%) genes. The high-quality, chromosome-level genome of the marten reported in this study will serve as a reference for future studies on genetic diversity, evolution, and conservation biology.

Chanodichthys erythropterus is a fierce carnivorous fish widely found in East Asian waters. It is not only a popular food fish in China, it is also a representative victim of overfishing. Genetic breeding programs launched to meet market demands urgently require high-quality genomes to facilitate genomic selection and genetic research. In this study, we constructed a chromosome-level reference genome of C. erythropterus by taking advantage of long-read single-molecule sequencing and de novo assembly by Oxford Nanopore Technology (ONT) and Hi-C. The 1.085 Gb C. erythropterus genome was assembled from 132 Gb of Nanopore sequence. The assembled genome represents 98.5% completeness (BUSCO) with a contig N50 length of 23.29 Mb. The contigs were clustered and ordered onto 24 chromosomes covering roughly 99.49% of the genome assembly with Hi-C data. Additionally, 33,041 (98.0%) genes were functionally annotated from a total of 33,706 predicted protein-coding sequences by combining transcriptome data from seven tissues. This high-quality assembled genome will be a precious resource for future molecular breeding and functional genomics research of C. erythropterus.

In this study, the complete mitochondrial genome of Steppe Whiskered Bat was sequenced for the first time using muscular tissue. The whole mitochondrial genome was 16,771 bp in length, consisting of two ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes, and one control region (D-loop). Phylogenetic analysis using PAUP based on mitochondrial genome (12 PCGs, except ND6) of 16 other Vespertilionidae species revealed the close relationship of M. aurascens with other related Myotis species.

The Taodonggou group of Middle Permian is an important source rock in Taibei sag of Turpan-Hami basin. Due to its deep burial, drilling has only been revealed in recent years. Based on organic petrology and organic geochemistry experiments, this paper studies the organic petrology, organic geochemistry, sedimentary environment, and hydrocarbon generation potential of source rocks in Taibei sag, Turpan-Hami basin, and reveals the influence of the sedimentary environment on the organic matter abundance of source rocks. The results are as follows: (1) The organic matter of the Middle Permian source rocks in Taibei sag of Turpan-Hami basin is mainly sapropelite and exinite. The vitrinite is mainly vitrodetrinite, and the exinite is mainly lamalginiite. (2) The total organic carbon content value is 0.55-6.08 wt %, and the average value is 2.58 wt %. The PG value ranges from 0.78 mg HC/g to 30.86 mg HC/g, and the average value is 4.88 mg HC/g. Chloroform asphalt "A" is 0.046-0.8767 wt %, and the average value is 0.285 wt %. The types of organic matter are mainly III and II-III, and the R o value is 0.628-1.49 wt % (average = 0.988 wt %). The T max distribution is 329-465 °C. The average temperature is 434.7 °C, which is in the mature stage (oil window stage). The Middle Permian source rocks are mainly very good to excellent source rocks with a good hydrocarbon generation potential. (3) The source rocks are deposited in a semihumid and semiarid climate. Organic matter is input as a mixed source. The early and late stages is dominated by terrestrial higher plants. The middle stage is dominated by lower aquatic organisms, and the sedimentary environment consists of weak reduction and weak oxidation environments. (4) In the study area, the abundance of organic matter has a weak negative correlation with CPI and a positive correlation with Pr/Ph and ∑C21-/∑C22+. Under the coaction of paleoclimate, organic matter input, and redox environment, the enrichment model of organic matter with high productivity and weak oxidation environment characteristics can also form excellent source rocks. This study is of great significance and provides theoretical guidance for the exploration of deep oil and gas resources.

With the novel capabilities of engineering the optical wavefront at the nanoscale, the dielectric metalens has been utilized for fluorescence microscopy imaging system. However, the main technical difficulty is how to realize the achromatic focusing and light modulation simultaneously by a single-layer metalens in the two-photon excitation STED (TPE-STED) endomicroscopy imaging system. Herein, by combining the spatial multiplexing technology and vortex phase modulation, a single-layer multitasking vortex-metalens as a miniature microscopy objective on the end of fiber was proposed. The multitasking vortex-metalens with 36-sectors interleaving (diameter of 100 µm) could focus the excitation beam (1050 nm) and depletion beam (599 nm) to the same focal distance, modulate a doughnut-shaped depletion spot with vortex phase and reshape the focal spots to further make improvement in the quality and symmetry. According to the TPE-STED theory, a symmetrical effective fluorescent spot with the lateral resolution of 30 nm was obtained by the proposed metalens. Thus, with the advantage of ultra-compact and lightweight, we prospect that the subminiature multitasking metalens will help guide future developments in high-performance metalenses toward high-resolution and real-time images for deep biological tissue in vivo and enable scientific high-end miniature endomicroscopy imaging system.

The high gas content of deep coal seams is a driving force for the exploration and development of deep coalbed methane (CBM). The nanoscale pores, which are the main spaces for adsorption and storage of CBM, are closely related to the burial depth. Based on integrated approaches of vitrinite reflectance (Ro), maceral composition, scanning electron microscope (SEM), proximate analysis, fluid inclusion test, low-temperature N2 adsorption-desorption, and CH4 isothermal adsorption, the nanoscale pore structure of coals recovered at depths from 650 to 2078 m was determined, and its influence on the CH4 adsorption capacity was discussed. The results show that the coal rank has a good linear relationship with the current burial depth of the coal seams; that is, the influences of the burial depth on the coals can be reflected by the influences of the coal rank on the coals. With the increase in the coal rank, the moisture and volatile content decrease, and the fixed carbon content increases. The variation in the pore volume and specific surface area with the increase in the coal rank can be divided into two stages: the rapid decline stage (when 0.75% 1.0%, the FD of the micropores is close to 2. This indicates that with the increase in the degree of coalification, the surface of the micropores is simpler. The above results show that the gas adsorption capacity of coal first slightly decreases (when 0.75% < Ro < 1.0%) and then increases (when 1.0% < Ro < 1.35%), and the coincident results are shown in the Langmuir volume (VL) test results.

The α-enolase (ENO1) plays pivotal roles in several types of cancer, but its clinical significance, functional role, and possible mechanism in colorectal cancer (CRC) have remained unclear. Expression level of ENO1 in CRC tissues was examined by qRT-PCR, Western blot, and immunohistochemistry. The effects of ENO1 on cell growth were investigated by MTT, colony formation, flow cytometry assays, and in vivo tumorigenic capacity analysis. The impacts of ENO1 on cell migration and invasion were also explored by scratch-healing, Transwell or Matrigel chamber assays, and in vivo metastatic capacity analysis. Our results showed that the expression level of ENO1 was significantly elevated in CRC tissues. High expression level of ENO1 was associated with disease progression in CRC patients. Overexpression of ENO1 in HCT116 cell line promoted cell proliferation, migration, and invasion in vitro as well as tumorigenesis and metastasis in vivo. In other hand, ablation of ENO1 in HCT116 cells led to totally reverse effects. Mechanistically, we revealed ENO1 could regulate AMPK/mTOR signaling pathway. AMPK pathway activation or mTOR pathway suppression blocked these ENO1 induced alterations. Together, our results demonstrated that ENO1 is a potent promoter of CRC genesis and metastasis at least in part though regulating AMPK/mTOR pathway. These findings also suggested that ENO1 may be a promising therapeutic target in CRC patients.

Ubiquitin specific protease 35 (USP35) is a member of deubiquitylases (DUBs). It remains largely unknown about the biological role and the regulation mechanism of USP35. Here, we first identified miR let-7a as a positive regulator of USP35 expression and showed that USP35 expression positively correlates with miR let-7a expression in different cancer cell lines and tissues. Then, we showed that USP35 expression was decreased dramatically in the tumor tissues compared with the adjacent non-cancerous tissues. USP35 overexpression inhibited cell proliferation in vitro and inhibited xenograft tumor growth in vivo. Furthermore, we revealed that USP35 acts as a functional DUB and stabilizes TNFAIP3 interacting protein 2 (ABIN-2) by promoting its deubiquitination. Functionally, both ABIN-2 and USP35 could inhibit TNFα-induced NF-κB activation and overexpression of ABIN-2 alleviated USP35-loss induced activation of NF-κB. Collectively, our data indicated that miR let-7a-regulated USP35 can inhibit NF-κB activation by deubiquitination and stabilization of ABIN-2 protein and eventually inhibit cell proliferation. Overall, our study provides a novel rationale of targeting miR let-7a-USP35-ABIN-2 pathway for the therapy of cancer patients.

OBJECTIVE: To investigate the role of miR-221/222 in cell proliferation and apoptosis in human prostate cancer cells, and examine the effects of miR-221/222 on caspase-10 expression. METHODS: Prostate cancer cells were transfected with miR-221/222 mimics or inhibitors. Cell proliferation was assessed by MTT assay. The expression levels of miR-221/222 were detected with quantitative real-time PCR. Apoptosis was induced with TNF-α/CHX treatment, and evaluated by Hoechst 33342 staining, propidium iodide (PI) flow cytometric analysis, caspase-3 activity measurement, and Western blot analysis. Luciferase activity assay, quantitative real-time PCR, and Western blot were performed to evaluate the effects of miR-221/222 on caspase-10 expression. RESULTS: Our results showed that miR-221/222 could promote the proliferation of prostate cancer cells, including LNCaP and PC3 cells. After transfection and apoptosis induction, Hoechst 33342 staining and PI flow cytometric assay showed that apoptosis was dramatically decreased in prostate cancer cells treated with miR-221/222 mimics. Moreover, caspase-3 activity was dramatically decreased, and the cleaved forms of caspase-3 were reduced, in the miR-221/222 mimic-treated group. On the contrary, miR-221/222 knockdown sensitized the prostate cancer cells to TNF-α/CHX-induced apoptosis. In addition, a negative correlation was observed between the expressions of miR-221/222 and caspase-10 in prostate cancer cells. miR-221/222 could repress the expression of caspase-10, which was confirmed by the luciferase reporter assay. CONCLUSION: miR-221/222 promote cell proliferation and repress apoptosis, through suppressing caspase-10, in prostate cancer cells. Our results provide promising evidence for the miRNA-based therapeutic strategy of prostate cancers.

FBXW7 (F-box and WD40 domain protein 7) is a tumor suppressor frequently inactivated in human cancers. The precise molecular mechanisms by which FBXW7 exerts antitumor activity remain under intensive investigation and are thought to relate in part to FBXW7-mediated destruction of key cancer-relevant proteins. Enolase 1 (ENO1) possesses oncogenic activity and is often overexpressed in various human cancers, besides its critical role in glycolysis. However, the detailed regulatory mechanisms of ENO1 expression remain unclear. Here we show that the elevated expression of ENO1 was identified in FBXW7-depletion HCT116 cells through two-dimensional protein electrophoresis and mass spectrometry assays (2DE-MS). Subsequent western blotting and immunohistochemical assays confirmed that ENO1 expression reversely correlates with FBXW7 expression in several cells and colon cancer tissues. Furthermore, we show that FBXW7 physically binds to ENO1 and targets ENO1 for ubiquitin-mediated degradation. Functionally, we found that FBXW7 suppresses the ENO1-induced gene expression, lactate production, cell proliferation and migration. These findings suggest that ENO1 is a novel substrate of FBXW7, and its activity can be negatively regulated by FBXW7 at the posttranslational level. Our work provides a novel molecular insight into FBXW7-directed tumor suppression through regulation of ENO1.
