To understand the relationship between kindergarten school-based research (SBR) and preschool teachers' teaching ability and identify the mechanism by which SBR affects teachers' teaching ability, a survey of randomly sampled preschool teachers in Sichuan Province (N = 625, Mage = 26.61) was conducted online using three scales assessing community learning, teaching ability and teaching philosophy. The survey results showed that the level of SBR reached the upper-middle level. The level of SBR positively predicted the teaching beliefs and teaching ability of preschool teachers. SBR influenced teaching ability through direct and indirect effects, with indirect effects accounting for 23.5% of the total effect; however, only constructivist teaching beliefs played a mediating role in the relationship between SBR and teaching ability via an indirect effect. SBR focused on improvement in teaching ability should not only examine the connection between SBR and practical problems to promote improvement in teachers' teaching ability but also should examine the role of teachers' beliefs and influence teachers' teaching abilities by changing teachers' teaching beliefs.

BACKGROUND Glioma is a common brain malignancy, but the effects of the γδ T cells and their subsets in peripheral blood in patients with glioma have not been reported. MATERIAL AND METHODS Flow cytometry was used to analyze the functions and expressions of δ T cells and their subsets in peripheral blood in healthy controls and patients with glioma. The Vδ2 T cells and the activation of killing function-related signaling pathway were analyzed by Western blot assay; the immunosuppressive functions of Vδ1 T cells were detected by CFSE proliferation assay; and the Vδ2 T cell killing functions were detected by killing assay. RESULTS Compared with the healthy controls, the ratio of Vδ1 T cells was significantly increased and the ratio of Vδ2 T cells was significantly decreased. After in vitro culture and anti-TCR gd antibody stimulation and in the presence of IL-2, in the patients with glioma, the Vδ1 T cells dominated and Vδ2 T cells were scarce. Flow cytometry staining showed that expression of immunosuppression-related molecules on the Vδ1 T cell surface was significantly increased, while the expression of killing function-related molecules and the activation of killing function-related signaling pathway in the Vδ2 T cells were significantly decreased. Functional test results showed that the immunosuppressive function of Vδ1T cells was enhanced and the killing function of Vδ1T cells was reduced. CONCLUSIONS The ratio and function changes of Vδ1 T cells and Vδ2 T cells are possibly associated with the pathogenesis of glioma.

This study investigated the effects of dietary fresh fermented soybean meal (FSM) on the intestinal microbiota and metabolites, bacterial enzyme activity and intestinal morphology of weaning piglets. A total of 64 weaned piglets were randomly allocated into two treatments. A corn-soybean-based diet was used as the control and other treatment was fed the same basal diet containing 15% fresh FSM. The feeding trial lasted for 21 days. Bacterial community structure and diversity in the cecum and colon were assessed using pyrosequencing-based analysis. The results showed that the phylum level, Firmicutes, Bacteroidetes, Proteobacteria and Tenericutes were dominant in the cecum or colon. Gut Firmicutes increased, while Bacteroidetes and Proteobacteria decreased in the fresh FSM-fed piglets. At the genus level, the relative abundances of butyrate-producing bacteria, Lactobacillus and Prevotella were higher in both cecum and colon of fresh FSM fed piglets. Meanwhile, fresh FSM could promote the development of intestinal morphological and reduce the incidence of diarrhea. The results indicated that fresh FSM might change intestinal function by influencing intestinal microenvironment.

The present study aims to investigate the effects of miR-146a on the proliferation and apoptosis of human osteoarthritis (OA) chondrocytes by targeting tumour necrosis factor receptor-associated factor 6 (TRAF6) through nuclear factor-κB (NF-κB) signalling pathway. Human normal and OA chondrocytes were selected and divided into the normal group, blank group, negative control (NC) group, miR-146a mimics group, miR-146a inhibitors, miR-146a inhibitor + si-TRAF6 group and si-TRAF6 group. Quantitative real-time PCR (qRT-PCR) was applied to detect the expressions of miR-146a, TRAF6 mRNA and NF-κB mRNA. Western blotting was used to detect the protein expressions of TRAF6 and NF-κB. CCK-8 assay and flow cytometry were used to detect cell proliferation and apoptosis. Compared with normal chondrocytes, the expression of miR-146a decreased, while the mRNA and protein expressions of TRAF6 and NF-κB increased in OA chondrocytes. OA chondrocytes had a lower proliferation rate and a higher apoptosis rate than the normal chondrocytes. Compared with the blank, NC and si-TRAF6 groups, the expression of miR-146a increased in the miR-146a mimics group, but decreased in the miR-146a inhibitors and miR-146a inhibitor + si-TRAF6 groups. Compared with the blank, NC and miR-146a inhibitor + si-TRAF6 groups, the mRNA and protein expressions of TRAF6 and NF-κB decreased, cell proliferation rate increased and cell apoptosis rate decreased in the miR-146a mimics and si-TRAF6 groups, while opposite trends were observed in the miR-146a inhibitors group. Our study suggests that miR-146a could promote proliferation and inhibit apoptosis of OA chondrocytes by inhibiting TRAF6 expression and suppressing the activation of NF-κB signalling pathway.
