This study aimed to compare the efficacy and effect on lipid profiles of Ainuovirine (ANV)- and efavirenz (EFV) -based regimens in treatment-naïve people living with HIV-1 (PLWH) at week 24. The proportion of PLWH achieving HIV-1 RNA < the limit of quantification in the ANV group was significantly higher than that in the EFV group (89.18% vs. 76.04%, P = 0.002). The mean change of log10 HIV-1 RNA from baseline was greater (-4.34 vs. -4.18, P < 0.001), the median change from baseline in CD4+ T cell count increased more (106.00 cells/µL vs. 92.00 cells/µL, P = 0.007) in the ANV group, while the CD4+/CD8+ ratio was similar (0.15 vs. 0.20, P = 0.167) between the two groups. The mean changes from baseline in total cholesterol (-0.02 for ANV vs. 0.25 mmol/L for EFV, P < 0.001), triglyceride (-0.14 for ANV vs. 0.11 mmol/L for EFV, P = 0.024), and low-density lipoprotein cholesterol (-0.07 for ANV vs. 0.15 mmol/L for EFV, P < 0.001) was significantly different between the two groups. The percentage of patients with improved lipid profiles was significantly higher in the ANV group (37.44 %) than in the EFV group (29.55%, P = 0.0495). The incidence of any adverse events in the ANV group was significantly lower than that in the EFV group at week 12 (6.2% vs. 30.7%, P < 0.001) and was comparable at week 24 (3.6% vs. 5.5%, P = 0.28). The ANV-based regimen was well tolerated and lipid-friendly in treatment-naïve PLWH.

Ainuovirine (ANV), a novel non-nucleoside reverse-transcriptase inhibitor (NNRTI), was approved in China in 2021. In a previous randomized phase 3 trial, ANV demonstrated non-inferior efficacy relative to efavirenz (EFV) and was associated with lower rates of dyslipidemia. In this study, we aimed to explore lipid changes in treatment-experienced people with human immunodeficiency virus (HIV)-1 (PWH) switching to ANV from EFV in real world. At week 24, 96.65% of patients in the ANV group and 93.25% in the EFV group had HIV-1 RNA levels below the limit of quantification (LOQ). Median changes from baseline in CD4 +T cell counts (37.0 vs 36.0 cells/µL, P = 0.886) and CD4+/CD8 +ratio (0.03 vs 0.10, P = 0.360) were similar between the two groups. The ANV group was superior to the EFV group in mean changes in total cholesterol (TC, -0.06 vs 0.26 mmol/L, P = 0.006), triglyceride (TG, -0.6 vs 0.14 mmol/L, P < 0.001), high-density lipoprotein cholesterol (HDL-C, 0.09 vs 0.08 mmol/L, P = 0.006), and low-density lipoprotein cholesterol (LDL-C, -0.18 vs 0.29 mmol/L, P < 0.001) at week 24. We also observed that a higher proportion of patients demonstrated improved TC (13.55% vs 4.45%, P = 0.015) or LDL-C (12.93% vs 6.89%, P = 0.017), and a lower proportion of patients showed worsened LDL-C (5.57% vs 13.52%, P = 0.017) with ANV than with EFV at week 24. In conclusion, we observed good efficacy and favorable changes in lipids in switching to ANV from EFV in treatment-experienced PWH in real world, indicating a promising switching option for PWH who may be more prone to metabolic or cardiovascular diseases.

BACKGROUND: Consistent and complete adherence is considered an essential requirement for patients on antiretroviral therapy (ART). This study aimed to evaluate the impact of ART duration on ART adherence, identify the trend of complete adherence, and compare the factors associated with ART adherence between short-term and long-term ART group among men who have sex with men (MSM) living with HIV in Jinan of China. METHODS: MSM living with HIV aged 18 or above and currently on ART were recruited from October to December 2020 using convenience sampling. Univariate and multivariable logistic regressions were used to evaluate the impact of ART duration on adherence and compare factors associated with ART adherence between subgroups. The Mann-Kendall test was used to identify the trend of complete adherence. RESULTS: A total of 585 participants were included in analysis, consisting of 352 on short-term ART (ART initiation ≤ 3 years) and 233 on long-term ART (ART initiation > 3 years). Significant difference of complete ART adherence between short-term and long-term ART group was detected (79.8% vs. 69.1%, P = 0.003). Multivariable analysis showed that men with longer ART duration were less likely to report complete ART adherence (AOR = 0.88, 95% CI 0.81-0.95). A descending trend of complete adherence was identified (Z = 1.787, P = 0.037). Alcohol use and lack of medication reminders were barriers to complete adherence for both of the subgroups. CONCLUSIONS: Sustained efforts to encourage maintaining adherence for a lifetime are necessary, especially for those on long-term ART. Future interventions should be tailored to subgroups with different ART duration and individuals with specific characteristics.

With a long history of fermentation technology and rich flavors, bread is widely consumed by people all around the world. The consumer market is huge and the demand is wide. However, the formation mechanism of bread baking flavor has not been completely defined. In order to improve the breadmaking process and the quality of bread, the main flavor substances produced in bread baking, the formation mechanism, and the detection technology of bread baking flavor are carefully summarized in this paper. The generation conditions and formation mechanism of flavor substances during the bread baking process are expounded, and the limitations of some current bread flavor detection technologies are proposed, which will provide theoretical basis for effectively regulating the generation of flavor substances in the bread baking process and making bread with good flavor and rich nutrition in the future.

BACKGROUND: Intra-articular injection is a classic strategy for the treatment of early osteoarthritis (OA). However, the local delivery of traditional therapeutic agents has limited benefits for alleviating OA. Exosomes, an important type of extracellular nanovesicle, show great potential for suppressing cartilage destruction in OA to replace drugs and stem cell-based administration. METHODS: In this study, we developed a thermosensitive, injectable hydrogel by in situ crosslinking of Pluronic F-127 and hyaluronic acid, which can be used as a slow-release carrier to durably retain primary chondrocyte-derived exosomes at damaged cartilage sites to effectively magnify their reparative effect. RESULTS: It was found that the hydrogel can sustainedly release exosomes, positively regulate chondrocytes on the proliferation, migration and differentiation, as well as efficiently induce polarization of M1 to M2 macrophages. Intra-articular injection of this exosomes-incorporated hydrogel significantly prevented cartilage destruction by promoting cartilage matrix formation. This strategy also displayed a regenerative immune phenotype characterized by a higher infiltration of CD163+ regenerative M2 macrophages over CD86+ M1 macrophages in synovial and chondral tissue, with a concomitant reduction in pro-inflammatory cytokines (TNF-α, IL-1ß, and IL-6) and increase in anti-inflammatory cytokine (IL-10) in synovial fluid. CONCLUSION: Our results demonstrated that local sustained-release primary chondrocyte-derived exosomes may relieve OA by promoting the phenotypic transformation of macrophages from M1 to M2, which suggesting a great potential for the application in OA.

The postharvest senescence of Chinese winter jujube fruit can be effectively delayed by refrigerated storage. However, chilling injury often occurs in jujube fruit during cold storage. In this study, Chinese winter jujubes were sprayed with CaCl2 (4%) 3 times at intervals of 2 hr on the day of refrigeration. The results presented that maximum difference of 2.7 N firmness, 3.42% TAC, and 0.8 OD280 /g polyphenol content were detected in calcium-treated fruit during cold storage, but the levels of O2- , MDA, hydrogen peroxide, browning rate, electrolyte leakage, and weight loss rate were significantly inhibited (p < .05). The maximum difference of enzymes activity of CAT, POD, SOD was 2.1, 10.8, and 40.6 mol h-1 kg-1 respectively, but 21.1 mol h-1 kg-1 PPO was restrained in the treated group. In conclusion, the results provided a reliable method for inhibiting cold injury and explained the internal molecular mechanism of the fruit regulated by calcium. PRACTICAL APPLICATIONS: Refrigerated storage is an important method for extending the storage time of Chinese winter jujube fruit. However, cold damage may occur when the jujubes are stored at low temperature for long-term. It is, therefore, of great significance to find a new method and reveal the molecular mechanism. We believe that our study makes a significant contribution to the literature because it provides an effective method of maintaining higher quality and mechanistic insights into the resistance against the chilling stress of jujubes.

OBJECTIVE: To study the effect of a new human sperm freezing method on the sperm recovery rate and search for an optimal method for cryopreservation of human epididymal sperm. METHODS: We collected semen samples from 76 men with obstructive azoospermia by percutaneous epididymal sperm aspiration and divided each sample into two parts to be cryopreserved with a self-made metal freezing plate (the experimental group) or by slow freezing (the control group), respectively. We measured the percentage of progressively motile sperm (PMS) with the computer-assisted semen analysis system and compared the membrane function, DNA fragmentation index (DFI), acrosin activity and morphological abnormality of the sperm between the two groups before and after cryopreservation. RESULTS: After thawing, both the percentages of PMS and hypotonically swollen sperm were significantly higher in the experimental than in the control group (ï¼»12.0 ± 7.5ï¼½% vs ï¼»8.0 ± 5.1ï¼½%, P < 0.05; ï¼»22.0 ± 17.5ï¼½% vs ï¼»18.0 ± 20.5ï¼½%, P < 0.05), though both decreased in comparison with the pre-freezing parameters (ï¼»20.7 ± 8.8ï¼½% and ï¼»30.0 ± 13.5ï¼½%) (P < 0.05). The sperm acrosin activity was remarkably higher in the experimental than in the control group after thawing (ï¼»75.2 ± 9.5ï¼½ vs ï¼»55.7 ± 8.3ï¼½ µIU/106sperm, P < 0.05), though decreased as compared with the baseline (ï¼»120.0 ± 10.5ï¼½ µIU/106 sperm, P < 0.05). No statistically significant differences were observed between the experimental and the control groups after thawing in the percentage of morphologically abnormal sperm (ï¼»98.7 ± 8.8ï¼½% vs ï¼»98.5±9.2ï¼½%, P > 0.05) or sperm DFI ï¼»38.2 ± 8.5ï¼½% vs ï¼»39.5 ± 10.2ï¼½%, P > 0.05), though both markedly elevated in comparison with the pre-freezing parameters (ï¼»97.2 ± 9.5ï¼½% and ï¼»30.8 ± 9.7ï¼½%) (P < 0.05). The post-thaw recovery rate of sperm was significantly higher in the experimental than in the control group (ï¼»65.2 ± 12.0ï¼½% vs ï¼»52.3 ± 18.0ï¼½%, P < 0.05). CONCLUSIONS: The self-made metal freezing plate, with its advantages of low cost, high efficiency, and easy operation, can be used as an effective method for cryopreservation of human sperm to achieve a high post-thaw sperm recovery rate, progressive sperm motility, and sperm acrosin activity.

Although research has demonstrated that Hg is methylated in the reducing conditions of the dental clinic wastewater collection system, studies are inconclusive as to whether further methylation occurs in the aeration basin of activated sludge wastewater treatment plant (WWTP) which typically treats this waste. Given the high levels of methyl Hg reported in dental wastewater (DWW), it is important to determine whether additional methylation occurs once it enters the WWTP. To achieve this objective, we incubated DWW under conditions designed to mimic the oxidized conditions of the activated sludge aeration basin in a WWTP. Duplicate bioreactors were charged with raw DWW collected from a 12-chair dental clinic and incubated both with and without aeration. Aeration was continued for 15 days, consistent with the typical mean cell residence time (MCRT) necessary for both heterotrophic carbon oxidation (typically 5-6 days) and nitrification (typically 12-15 days), thus ensuring that incubation time exceeded those for most conceivable MCRTs used in the activated sludge process. Results demonstrate a rapid increase in pH concomitant with an increase in dissolved oxygen (DO) to near saturation in the aerated reactor. The non-aerated reactor remained low or at zero DO due to low surface reaeration coupled with the high levels of organic matter. The rate of mercury methylation increased in the unaearated reactors rapidly upon incubation, reaching highest levels when DO was at the lowest levels during the experiment. In great contrast, methyl mercury levels were much lower and net mercury methylation does not appear to occur at any significant rate under aeration. These results imply that although some mercury methylation may occur in the sewer collection system (or anaerobic digesters), net methylation is unlikely to occur in the aeration basin in activated sludge WWTPs, and thus methyl Hg influent levels from DWW represent an upper bound on effluent levels.

Collateral Channel is a heavily polluted former navigation slip to the Chicago Sanitary and Ship Canal (Illinois, USA). Characterization of sediment cores taken in the channel show high levels of heavy metals, polycyclic aromatic hydrocarbons (PAHs) and other contaminants in deposited sediment dating back to the 1800's. Of these, PAHs were the contaminants of greatest concern based upon exceedance of sediment contamination criteria (Sigma(16) PAHs up to 1500mg/kg). Benthic animal counts revealed a lack of biodiversity, with relatively low levels of small tubificid oligochaetes (generally <3000/m(2)) in surficial sediments. Comparison of surficial sediment contaminant levels between 1995 and 2005 showed few decreases in contaminant levels, indicating a lack of "natural recovery" processes occurring in the channel. These results led to an analysis of sediment amendments for an active capping demonstration project in the channel using transport models developed in our previous work (Viana et al., 2008). Based on the sediment characterization and modeling results, the active capping design will be focused on organic contaminant sequestration through the use of organoclay. A site-specific difficulty is the substantial rates of gas ebullition from anaerobic organic matter biodegradation in the sediments, particularly in the summer months. These gases can open advective channels that may result in substantial pollution release and compromise cap effectiveness, and thus the capping scenario must control for such releases. The active capping layer will underlay a sloped sand layer and a high permeability gas venting system to allow biogenically-produced gas migration to shoreline collectors through an innovative support grid. The cap will include an overlaying wetland to remove nutrients from the adjoining Chicago River and provide a public recreational space.

Dental wastewater (DWW) was collected over two months from a 12-chair clinic and a single-chair office to identify conditions that may affect Hg methylation. DWW was settled for 24 h and samples were collected from the top and bottom of the supernatant to simulate a range of particles that may escape in-line traps. Total Hg spanned 5 orders of magnitude (0.02-5000 microM), following a log-normal distribution with p10, p50, and p90 concentration values of 0.24, 31 and 4000 microM, respectively; typically well in excess of free aqueous Hg solubility. Methyl Hg was present in high levels (2-270 nM), also following a log-normal distribution with p10, p50, and p90 concentration values of 2.8, 17, and 100 nM, respectively. There were no statistically significant differences (90% CI) in p50 methyl Hg or total Hg between the clinic and office. Methyl Hg was predicted from total Hg data by (+/- 95% CI): Log (Me-Hg) = 0.33 (+/- 0.06) x Log (T-Hg) - 2.27 (+/- 0.13). Total methyl Hg from DWW to U.S. wastewater collection systems is estimated to be 2-5 kg yr(-1). Equilibrium speciation modeling predicted that DWW Hg was primarily in sulfide-Hg complexes, except at high total Hg levels where organo-Hg complexes become significant. DNA extracts amplified by quantitative polymerase chain reaction with primers for total eubacteria and sulfate-reducing bacteria (SRB) indicated that the total eubacterial DNA was composed primarily of SRB, and highly significant correlations were found between methyl Hg and both amplified Desulfobacteraceae (p < 0.0001) and Desulfovibrionacaea DNA (p < 0.00001). Both are known Hg methylators. In marked contrast, there was no significant correlation between methyl Hg and amplified Desulfobulbus DNA, a genus generally not known to methylate Hg at high rates. These results strongly suggest that SRB are implicated in DWW Hg methylation.

OBJECTIVE: To purify and identify recombinant Varicella-Zoster Virus Glycoprotein E. METHODS: The recombinant plasmid pGEX-VZVgE was induced by isopropyl-beta-D-thiogalactoside (IPTG), the fusion protein was purified with affinity chromatography column; then the purified fusion protein was cleaved by thrombin, and the product's antigenicity was examined by Western blot. RESULTS: The product of pGEX-VZVgE induced by IPTG was separated from the mixture proteins by the affinity chromatography column, the expressed fusion protein's relative molecular mass was about 98 x 10(3). After cleavage, the obtained VZV Glycoprotein E's relative molecular mass was about 72 x 10(3); the purified fusion protein and VZV Glycoprotein E were single band by SDS-PAGE. The available antigenicity of Glycoprotein E was confirmed by Western blot. CONCLUSION: Purification of VZV Glycoprotein E with affinity chromatography is an effective method. It provides a foreground for studies on the application of VZV gE.
