RESUMO

The microbial diversity on the carposphere (berry) surface of the grape cultivar Cabernet Sauvignon grown in eight different locations/vineyards of Henan Province was determined by high-throughput sequencing of the bacterial 16S rRNA gene and fungal 18S rRNA gene. The structure of bacterial and fungal communities varied according to the sampling sites, but with some common phyla. Proteobacteria and Ascomycota were dominant/common phyla for bacteria and fungi, respectively. A total of 27 and 20 bacterial and fungal families, respectively, and 39 and 20 bacterial and fungal genera, respectively, with statistically significant differences, were found among different sampling sites. The difference for metabolic pathways of bacteria among the sampling sites existed. In addition, various abundances of enzymes from different sites might indicate that different function patterns exist in microbiota from different sites. The results revealed that locations of grape vineyards might play a significant role in shaping the microbiome, as well as the fact that vineyards can be distinguished based on the abundance of several key bacterial and fungal taxa. Overall, these findings extend our understanding of the similarities and differences in microbial community and their metabolic function on Cabernet Sauvignon grape surfaces from different geographic locations.

Tobacco-specific alkaloids and nitrosamines are important biomarkers for the estimation of tobacco use and human exposure to tobacco-specific nitrosamines that can be monitored by wastewater analysis. Thus far their analysis has used solid phase extraction, which is costly and time-consuming. In this study, we developed a direct injection liquid chromatography-tandem mass spectrometry method for the quantification of two tobacco-specific alkaloids and five nitrosamines in wastewater. The method achieved excellent linearity (R2 > 0.99) for all analytes, with calibration ranging from 0.10 to 800 ng/L. Method limits of detection and quantification were 0.17 ng/L (N-nitrosonornicotine, NNN) and 1.0 ng/L (N-nitrosoanatabine (NAT) and NNN), with acceptable accuracy (100 % ± 20 %) and precision (± 15 %). Analyte loss during filtration was < 15 %, and the relative matrix effect was < 10 %. The method was applied to 43 pooled wastewater samples collected from three wastewater treatment plants in Australia between 2017 and 2021. Anabasine and anatabine were detected in all samples at concentrations of 5.0 - 33 ng/L and 12 - 41 ng/L, respectively. Three of the five tobacco-specific nitrosamines (NAT, NNN, and (4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol) (NNAL)) were detected, in < 50 % of the wastewater samples, with concentrations nearly ten times lower than the tobacco alkaloids (< 1.0 - 6.2 ng/L). In-sewer stability of the nitrosamines was also assessed in this study, with four (NAT, NNAL, NNN, and N-nitrosoanabasine (NAB)) being stable (i.e. < 20 % transformation over 12 h in both control reactor (CR) and rising main reactor (RM) and 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) being moderately stable (< 40 % loss over 12 h in RM). This direct injection method provides a high-throughput approach in simultaneous investigation of tobacco use and assessment of public exposure to tobacco-specific nitrosamines.

Wastewater-based epidemiology (WBE) is proposed as a cost-effective approach to objectively monitor the antidepressant use but it requires more accurate correction factors (CF) than what had been used in previous studies. Amitriptyline is a popular prescription medicine for treating depression and nerve pain, which could be prone to misuse and need monitoring. The CF of amitriptyline employed in previous WBE studies varied from 10 to 100, leading to substantial disparities between WBE estimates and expected mass of antidepressants in wastewater. Hence, this study aimed to take amitriptyline as a case study and refine the CF by correlating mass loads measured in wastewater from 12.2 million inhabitants collected during the 2016 Census with corresponding annual sales data. The triangulation of WBE data and sales data resulted in a newly-derived CF of 7, which is significantly different from the CF values used in previous studies. The newly derived CF was applied to a secondary, multi-year (2017 to 2020) WBE dataset for validation against sales data in the same period, demonstrating the estimated amitriptyline use (380 ± 320 mg/day/1000 inhabitants) is consistent with sales data (450 ± 190 mg/day/1000 inhabitants). When we applied the new CF to previous studies, the wastewater consumption loads matched better to prescription data than previous WBE estimations. The refined CF of amitriptyline can be used in future WBE studies to improve the accuracy of the consumption estimates.

Natural killer (NK) cells are an important part of the body's innate immune system. As the first line of defense against pathogens, they need to be transformed into a mature state under the control of various cell signaling molecules and transcription factors to play cytotoxic and immune regulatory roles. Under the interaction of activated receptors and inhibitory receptors, NK cells are activated to perform a direct cell killing effect by secreting perforin and granzyme, or indirectly eliminate pathogenic microorganisms in the body by secreting various cytokines, such as type I and type II interferons. These functions of NK cells play a very important role in antiviral and anti-autoimmune diseases, especially in anti-tumor.

Circular dichroism spectroscopy is frequently used to characterize the chiral biomolecules by measuring the absorption spectra contrast between the left-handed circularly polarized light and the right-handed circularly polarized light. Compared with biomolecules, chiral metal plasmonic nanostructures also produce a strong circular dichroism response in the range of near-infrared. However, due to the large damping rate, the non-adjustable resonant frequency of the conventional metals, the applications of chiral metal plasmonic nanostructures in the fields of photoelectric detection and chemical and biochemical sensing are restricted. Here, we present a chiral graphene plasmonic Archimedes' spiral nanostructure that displays a significant circular dichroism response under the excitation of two polarizations of circularly polarized light. By manipulating the material and geometric parameters of the Archimedes' spiral, the stronger circular dichroism responses and modulation of the resonant wavelength are achieved. The optimized plasmonic nanostructure has outstanding refractive index sensing performance, where the sensitivity and figure of merit reach 7000nm/RIU and 68.75, respectively. Our proposed chiral graphene plasmonic Archimedes' spiral nanostructure might find potential applications in the fields of optical detection and high performance of index sensing.

We propose the monolayer graphene plasmonic waveguide (MGPW), which is composed of graphene core sandwiched by two graphene metamaterial (GMM) claddings and investigate the properties of plasmonic modes propagating in the waveguide. The effective refraction index of the GMMs claddings takes negative (or positive) at the vicinity of the Dirac-like point in the band structure. We show that when the effective refraction index of the GMMs is positive, the plasmons travel forward in the MGPW with a positive group velocity (vg > 0, vp > 0). In contrast-for the negative refraction index GMM claddings-a negative group velocity of the fundamental mode (vg < 0, vp > 0) appears in the proposed waveguide structure when the core is sufficiently narrow. A forbidden band appears between the negative and positive group velocity regions, which is enhanced gradually as the width of the core increases. On the other hand, one can overcome this limitation and even make the forbidden band disappear by increasing the chemical potential difference between the nanodisks and the ambient graphene of the GMM claddings. The proposed structure offers a novel scheme of on-chip electromagnetic field and may find significant applications in the future high density plasmonic integrated circuit technique.

Multiple Fano resonances (FRs) can be produced by destroying the symmetry of structure or adding additional nanoparticles without changing the spatial symmetry, which has been proved in noble metal structures. However, due to the disadvantages of low modulation depth, large damping rate, and broadband spectral responses, many resonance applications are limited. In this research paper, we propose a graphene plasmonic metamolecule (PMM) by adding an additional 12 nanodiscs around a graphene heptamer, where two Fano resonance modes with different wavelengths are observed in the extinction spectrum. The competition between the two FRs as well as the modulation depth of each FR is investigated by varying the materials and the geometrical parameters of the nanostructure. A simple trimer model, which emulates the radical distribution of the PMM, is employed to understand the electromagnetic field behaviors during the variation of the parameters. Our proposed graphene nanostructures might find significant applications in the fields of single molecule detection, chemical or biochemical sensing, and nanoantenna.
