RESUMO

AIM: Analysis of epidemic manifestations of natural-foci infections (NFI), clarification of spectrum of their causative agents, determination of epizootic activity of natural foci in the Crimea Federal District (KFD). MATERIALS AND METHODS: Epizootologic examination of 10 administrative districts of KDF was carried out. 291 pools (2705 specimens) of ixodes ticks and 283 samples of organs of small mammals were studied by PCRmethod for the presence of DNA/RNA of causative agents of a number of NFI. RESULTS: Morbidity by NFI in KFD was registered by 6 nosologies: Lyme borreliosis, Marseilles fever, leptospirosis; tularemia, intestine yersiniosis and tick-borne viral encephalitis, wherein, transmissive infections made up 91.6%. Circulation of causative agents of Crimea hemorrhagic fever, Q fever, group of tick-borne spotted fever, Lyme borreliosis, human granulocytic anaplasmosis, human monocytic ehrlichiosis, hemorrhagic fever with renal syndrome, West Nile fever, tularemia and leptospirosis was established. CONCLUSION: Due to activity of natural foci of NFI further monitoring of epidemiologic and epizootologic manifestations of these infections in the Crimea, including using genetic methods of analysis, is necessary for ensuring sanitary-epidemiologic welfare of KFD population.

AIM: Analysis ofresults of epidemiologic monitoring especially dangerous, natural-foci and other infectious diseases, asill as epizootologic activity of natural foci of infection on,the terri- tory, of city-resort Sochi. MATERIALS AND METHODS: Laboratory studies of 820. samples by PCR; im- mune- and bacteriologic methods were carried out, among those 344 - clinical material, 12 - water from open bodies and 321 - field material. Molecular-genetic identification of-143 strains of Vibrio-cholerae, isolated from open water bodies of city-resort Sochi, was-carried out,. RESULTS: Circulation of causative agents of Q fever, tularemia and hemorrhagic fever with renal syndrome of Dobrava-Adler genotypes was established, -as well as rickettsia of tick spotted fever group Predomination of rotaviruses (70.9%) was detected during study of samples of clinical material in etiologic structure of sporadically emerging acute intestine infections. Relatively high temperature values of river water during summer months (from 18, to 30 C), exit of sulfide mineral waters into the riverbed, that create-alkaline medium, seasonal reduction of river debit and speed of water current facilitated sustenance of contamination of water of Agura river by V.cholerae. CONCLUSION: Activity of natural foci of the infectious disease and contamination of Agura iver by V.cholerae Ο1, (atoxigenic) gives evidence on the necessity of constant control of epidemiologic situation by dangerous and natural-foci infectious diseases on the territory of city-resort Sochi, as well as study regional features, of their causative agents, including using genetic methods.

The possibility of detecting antigens of plague, tularemia, and brucellosis microbes with magnetic latex (ML)-based test systems has been demonstrated. MLs were prepared from latexes (polyacroleine microspheres, 1.2-1.8 +/- 0.1 microm) by exposing the particles to a 25-35%-solution of ferrous sulfate for 0.5 h and then to a 15-25%-aqueous solution of ammonia for 0.5 h in a 100 degrees C water bath and dehydrating after each operation. The possibility of preparing magnetic latex immunosorbents (MLIS) by ligand immobilization on ML and using them in magnetic latex ELISA (ML-ELISA) for the detection of microbial antigens was demonstrated. The detection limit in ML-ELISA equaled 10(2)-10(3) microbial cells in 1 ml (cells/ml). Relative experimental error was not higher than 8%.

RESUMO

The polymerase chain reaction (PCR) with and without pre-treatment of the samples of the fleas Citellophilus tesquorum with magnoimmunosorbents was used to detect the plague microbe in the samples containing 142, 168, 193 or more bacterial cells. PCR analysis reduced the time of tests of ectoparasites for plague to 6 hours. The use of a combination of PCR and magnoimmunosorbent simplified the most time-consuming and longest stage of isolation of plague microbial DNA from the suspensions of the fleas, which allows the time of a test to be reduced to 3 hours. The administration of an affine sorbent having magnetic properties during treatment of samples with plague-infected exoparasites for PCR analysis makes it possible to have a specific concentration of Yersinia pestis and to prevent the inhibitory effect of flea tissues on the polymerase chain reaction, to use the boiling method for isolating microbial DNA and for disinfecting the material to be tested.

RESUMO

The matrix for obtaining silica-based diagnosticum was selected, its activation with surfactant was optimized and its immobilization with tularemia immunoglobulins was carried out. In the glass suspension agglutination test (SAT) the sensitivity of the diagnosticum was 3.125 x 10(6) to 6.25 x 10(6) microbial cells per ml; the duration of SAT, including the evaluation of its results, was 1-5 minutes.

RESUMO

The improvement of the laboratory diagnostics of quarantine infections (QI) is aimed at obtaining the result at a shorter time, the simplification of making serological reactions, enhancing the specificity and sensitivity of the results due to the introduction of new = generation diagnostic preparations into practical use. The wide practical use of erythrocytic and immunofluorescent diagnostic preparations has shown that they cannot completely meet all requirements simultaneously. The optimized scheme of the indication of the causative agents of QI must ensure the combined use, at different stages of the analysis, of magneto-immunosorbents, enzyme immunoassay, as well as immuno-suspension preparations for the simplification and acceleration of obtaining results with sufficient sensitivity and specificity provided.

RESUMO

A combined method for the indication of the causative agent of anthrax (Bacillus anthracis), including the preparation of the material to be tested, the exposure of the magneto-imunosorbent in the sample, cultivation in selective medium, DNA extraction with subsequent testing in the polymerase chain reaction with primers to genes cap, pag and chromosomal sequence Ba813, the registration and interpretation of results, has been developed. All determinations, including the preparation of samples, last not more than 6 hours. The indication of B. anthracis by the proposed method makes it possible not only to confirm its presence in the sample Under test, but also to evaluate its epidemic potential.

RESUMO

The use of new diagnostic preparations (magnetic immunosorbents, polyvalent fluorescent immunoglobulins and immunoperoxidase conjugates), as well as the combination of magnetic immunosorbents with the polymerase chain reaction, for the rapid diagnostics of dangerous infectious diseases has been shown to have good prospects. Information on their successful use for laboratory diagnostics in emergency situations is presented.
