RESUMO

A simple method was developed for end-point fluorescence detection of the 735G --> A mutation of the 5'-splice donor site of intron 14 of the dihidropyrimidine dehydrogenase gene (DPYD). The method was based on allele-specific PCR with duplex Scorpion primers. The genotyping results obtained by the fluorescent endpoint PCR technique completely coincided with the results obtained by allele-specific PCR with amplicon detection in agarose gel. Genotyping was performed in 291 DNA samples from residents of Novosibirsk region (Russia), and two heterozygotes (0.69%) were detected.

RESUMO

The addition of the vitamin B complex into the culture medium enhances the productivity of the process ensuring the yield of biomass. The presence of the vitamins of this complex in the culture media intensifies the synthesis of some macromolecular compounds and prolongs the period of the physiological activity of the population.

RESUMO

During the continuous cultivation of typhoid bacteria in Lederberg's synthetic medium the specific rate of glucose and oxygen consumption was shown to differ, depending on the dilution rate, the initial concentration of glucose in the medium and on the oxygen level in the medium. At the same time the conditions ensuring the maximum assimilating properties of the culture did not coincide with the conditions of the maximum biomass production.

RESUMO

For the first time the synthesis of proteolytic enzymes by Corynebacterium diphtheriae has been shown to precede the process of toxin formation. The process of controlling the proteolytic activity of enzymes seems to be a suitable basis for the evaluation of toxin formation in mathematically planned experiments with strictly controlled tasks.
