RESUMO

This study aimed to assess the dose-response relationship (DRR) between measures of training load (TL) and anaerobic performance in female short-track speed skaters. TL data from 22 female short-track speed skaters (mean ± SD; calendar age 20.7 ± 2.01 years, training length 4.18 ± 1.23 years, height 164.20 ± 5.53 cm, weight 52.00 ± 5.58 kg) was collected over a six week period. Participants completed two anaerobic capacity assessments (cycling parameters: total work, maximal anaerobic power [MAnP], power duration indices, maximal blood lactate, fatigue indices, and peak power output [PPO]) pre- and post-training, which included three tests (short-term, intermediate-term, and 120 s maximal anaerobic test). The mean weekly TL measures calculated were total distance, red zone (time spent > 85% of peak heart rate), Edwards training impulse (eTRIMP), Stagno training impulse (gTRIMP), Lucia training impulse, and session ratings of perceived exertion. The gTRIMP identified a curvilinear relationship and explained 79% and 63% of the variance in changes of MAnPi and PPO (R2 = 0.79, 95%CI = 0.64 to 1.00, turn point 621AU; R2 = 0.63, 95%CI = 0.27 to 1.00, turn point 633AU, respectively). Likewise, The eTRIMP identified a curvilinear relationship and explained 62% and 54% of the variance in changes of MAnPi and PPO (R2 = 0.62, 95%CI = 0.11 to 1.00, turn point 485AU; R2 = 0.54, 95%CI = 0.07 to 1.00, turn point 515AU, respectively). All other variables exhibited a DRR below moderate. This study investigated a TL strategy to improve the speed endurance of female short-track speed skaters and suggested the superiority of gTRIMP and eTRIMP methods of internal TL.

Purpose: This study is aimed at exploring how soleus H-reflex change in poststroke patients with spasticity influenced by body position. Materials and Methods: Twenty-four stroke patients with spastic hemiplegia and twelve age-matched healthy controls were investigated. Maximal Hoffmann-reflex (Hmax) and motor potential (Mmax) were elicited at the popliteal fossa in both prone and standing positions, respectively, and the Hmax/Mmax ratio at each body position was determined. Compare changes in reflex behavior in both spastic and contralateral muscles of stroke survivors in prone and standing positions, and match healthy subjects in the same position. Results: In healthy subjects, Hmax and Hmax/Mmax ratios were significantly decreased in the standing position compared to the prone position (Hmax: p = 0.000, Hmax/Mmax: p = 0.016). However, Hmax/Mmax ratios were increased in standing position on both sides in poststroke patients with spasticity (unaffected side: p = 0.006, affected side: p = 0.095). The Hmax and Hmax/Mmax ratios were significantly more increased on the affected side than unaffected side irrespective of the position. Conclusions: The motor neuron excitability of both sides was not suppressed but instead upregulated in the standing position in subjects with spasticity, which may suggest that there was abnormal regulation of the Ia pathway on both sides.

6-Gingerol, an active herbal ingredient of ginger has various bioactivities such as anti-neurodegenerative disease, anti-inflammatory and so on. The aim of the present study was to enhance the oral bioavailability and brain distribution of 6-Gingerol via polymeric micelles. A polymeric micelles drug delivery system of 6-Gingerol consisting of D-α-Tocopheryl polyethylene glycol 1000 succinate (TPGS) and Poly (ethylene glycol)-poly (ε-caprolactone) (PEG-PCL) was prepared via solvent injection method. The developed 6-Gingerol-loaded TPGS/PEG-PCL micelles (6-GTPMs) were characterized based on particle size, polydispersity index (PDI), zeta potential, encapsulation efficiency (EE), drug loading (DL) and in vitro release profile. The pharmacokinetics and tissue distribution studies were also evaluated. The nanoformulation produced a particle size of 73.24 ± 2.84 nm with acceptable PDI (0.129 ± 0.03), zeta potential (-2.74 ± 0.92 mV), DL (4.64%) and EE (79.68%). The in vitro release profile showed that the 6-GTPMs enhanced the solubility of 6-Gingerol, while the pharmaceutical analysis in rats indicated that 6-GTPMs significantly improved the oral bioavailability of 6-Gingerol (about 3 folds) in circulation. The 6-GTPMs exhibited remarkable brain targetability in the tissue distribution analysis. Collectively, a 6-Gingerol polymeric micelle with enhanced oral bioavailability coupled with excellent brain distribution was successfully developed.

BACKGROUND: Spasticity commonly occurs in hemiplegic patients with stroke, and clinical experience indicates that upper limb spasticity is more apparent when patients are standing than when they are lying down. Few reports have investigated how positional changes impact the spasticity of the upper limb and trigger a combined movement called the "hemiplegic posture". OBJECTIVE: The objective of this study was to explore the influence of different body positions on the spasticity of the upper extremity. METHODS: Eighty-four poststroke patients with upper limb spasticity were recruited for this study. We measured the spasticity of hemiplegic upper limbs involving elbow flexors, wrist flexors and digitorum flexors using the Modified Ashworth Scale (MAS), Modified Tardieu Scale (MTS) and Triple Spasticity Scale (TSS) in three positions, i.e., supine, sitting and standing. The results were obtained by comparing one position with another for each muscle. RESULTS: All the MAS, MTS and TSS scores of all the muscles of the upper limb were significantly lower in the supine position than in the other positions (all P < 0.05), and the MTS and TSS scores were statistically higher while standing than while sitting (P < 0.05). CONCLUSION: High modulation of spasticity in the upper limbs measured using the MAS, MTS and TSS when transitioning to the standing position from either the supine or sitting position demonstrated that posture plays a role in modulating the spasticity of the hemiplegic upper limb in poststroke patients.
