RESUMO

Accumulating evidence suggested the participation of long noncoding RNAs (lncRNAs) in regulating various biological processes so as to affecting cancer progression. However, the functional role of most lncRNAs in colorectal carcer (CRC) is still largely covered. In the present study, we disclosed SNHG14 as a carcinogene in CRC development, as it was low-expressed in normal colon tissues but markedly upregulated in CRC cell lines. Besides, SNHG14 contributed to CRC cell proliferation, motility and EMT in vitro, and inhibition of it confined CRC tumor growth and liver metastasis in vivo. Next, the mechanistic investigations confirmed that SNHG14-promoted CRC progression was mediated by EPHA7, which was negatively regulated by SNHG14 in CRC via an EZH2-dependent way. Importantly, EZH2 was proved as a transcription factor of EPHA7 and functioned as a repressor in EPHA7 transcription by enhancing methylation on EPHA7 promoter. Meanwhile, SNHG14 increased EZH2 expression in CRC via stabilizing its mRNA by interacting with FUS, and via freeing its mRNA from miR-186-5p-induced silence. All in all, our observations demonstrated that SNHG14 serves as a facilitator in CRC through targeting EZH2-repressed EPHA7 by enhancing EZH2 via recruiting FUS and absorbing miR-186-5p, indicating a promising new road for CRC diagnosis and treatment.

Objeetive Airway remodeling is an important pathological feature of asthma.This study is to investigate the role of microRNA-21 (miR-21) in airway remodeling in asthmatic mice.Methods A total of 16 female BALB/c mice were randomly divided into control group and asthma model group.Mice were sensitized and challenged by ovalbumin to establish a murine model of asthma.The mice in the normal control group were intraperitoneally injected with phosphate buffered saline for sensitization and given a phosphate buffered saline inhalation for the challenge.Twenty-four hours after the last aerosol inhalation,lung tissues of mice were sampled and subjectd to Western blot for testing expression of TGFβ type Ⅱ receptor,Smad7,and Collagen Type Ⅰ (COL Ⅰ) in lung tissue of mice of each group.Quantitative real-time PCR was used to test miR-21 expression in lung tissue of mice of each group.Target gene prediction software (TargetScan) was used to predict target gene of miR-21.293T cell was used to conduct dual-luciferase reporter gene assay for verification of miR-21 target gene.Results Compared with control group,asthma group had increased expression of COL Ⅰ protein and miR-21 in lung tissue (t =11.94,P < 0.05;t =23.05,P < 0.05).Smad7 and TGF-βR Ⅱ were target genes of miR-21.Conclusion miR-21 down-regulates Smad7,a target gene of miR-21,activates the TGFβ/Smad signaling pathway,and promotes airway remodeling,indicating that miR-21 may be a therapeutic target for the treatment of airway remodeling in asthma.

Objective To observe and compare the clinical efficacy,recurrence rate and safety of differ-ent courses of inhalation fluticasone propionate in treatment of children with cough variant asthma. Methods A total of 101 children with cough variant asthma in our hospital were randomly divided into three treatment groups,all with the same therapy drug -fluticasone propionate and with the treatment time of two months, six months and nine months respectively. Clinical efficacy and treatment compliance were followed up during the treatment. After discontinuation,observation for the relapse of cough was followed up for 6 months. In inhalation therapy before and after,bone mineral density( BMD) and blood cortisol,electrocardiogram were monitored re-spectively to observe its safety. Results Three groups of children with cough were all get effective control,with no significant difference(H=5. 2894,P=0. 0710). The two months treatment group(A group)was more easy to relapse than the six months treatment group( B group) and the nine months treatment group( C group) after drug withdrawal,statistically significant(χ2 =4. 858 1,P=0. 027 5;χ2 =4. 482 6,P=0. 034 2). But after drug with-drawal,there was no significant difference in the rate of relapse in B and C group(χ2 =0. 022 2,P=0. 881 4). There were few changes in laboratory values about morning serum cortosol concentrations. There was no signifi-cant decrease in BMD after treatment. No other adverse events were observed in this study. Conclusion Flutica-sone propionate inhalation treating children cough variant asthma has good effection. A long period of treatment scheme(6 months)has better curative effect and lower recurrence rate after drug withdrawal.

Objective To investigate the role of soluble triggering receptor expressed on myeloid cell-l (sTREM-1) in children with community acquired pneumonia.Methods One hundred and seventy-six children with community acquired pneu-monia, 98 cases were mild and 78 cases were servere, were recruited. Thirty healthy children were recruited as control group. The white blood cell count (WBC), neutrophil percentage (N%), C-reactive protein (CRP), procalcitonin (PCT), interleukin-6 (IL-6), interleukin-10 (IL-10), and sTREM-1 were measured.Results The levels of WBC, N%, CRP, IL-6, IL-10, IL-6/IL-10, PCT, and sTREM-1 were signiifcantly different among children with mild pneumonia, severe pneumonia, and healthy controls. All of the indicators were elevated in children with mild and severe pneumonia than those in healthy controls (P<0.05). IL-6/IL-10 sTREM-1 were further signiifcantly elevated in children with severe pneumonia than children with mild pneumonia (P<0.05). IL-6/IL-10 was positively correlated with sTREM-1 (r=0.42,P<0.05).Conclusions sTREM-1 may help for evaluating the severity and outcome of children with community acquired pneumonia.

How the newly discovered LncRNA expression in colorectal cancer?This review mainly outlines the research de-velopment of LncRNA in colerectal cancer in brief.

Currently ,EphB receptor has been demonstrated as an important factor on the biological be -havior of normal cells .Some studies found that it plays an important role in the regulation of normal cell morpholo -gy,cell adhesion ,migration and angiogenesis .There is a certain relationship between abnormal expression of EphB receptor with a variety of tumor development .Visiblely,EphB receptor is important research direction in cancer studies,understanding the relationship between EphB receptor and tumor can better realize the progress of the tumor,and further to provide more scientific basis for cancer treatment and prevention .

RESUMO

Objective To study the effect of inhalation combined salmeterol plus fluticasone propionate in treatment of asthma in children between 4 ~ 5 years old.Methods Seventy-four children (age range,4 ~ 5 years) were enrolled for our study,they were randomly divided into 2 groups (one group inhaled combined salmeterol plus fluticasone,37 children;the other group inhaled fluticasone,37 children).Comparative studies of clinical efficacy,quantified symptom score,and regular follow-up observations were conducted at the interval of one week,one month,three months.Results In the first week and first month during the treatment,there were no difference between salmeterol plus fluticasone group and fluticasone group (P > 0.05).In the third month during the treatment,the daytime ratings(0.16 ± 0.09 vs 0.32 ± 0.11,P < 0.05) and night ratings(0.04 ± 0.01 vs 0.14 ±0.00,P <0.05) of salmeterol plus fluticasone group were significantly better than those of fluticasone group.After three months,more children in salmeterol plus fluticasone group were responsive to the decreased dose of the drug than those of fluticasone group (x2 =4.57,P < 0.05).Conclusion In the follow-up of exacerbatious,treatment with combined salmeterol and fluticasone propionate was as efficient as that of fluticasone in alleviating symptoms.During remission of asthma,combined salmeterol and fluticasone propionate was probably better than fluticasone for control of symptoms.

Objective To study the clinical application and its impact factors of tidal flow - volume measurement in infants with bronchiolitis and pneumonia. Methods Thirty infants with bronchiolitis and thirty infants with pneumonia were studied. Tidal breathing flow- volume loops were obtained after the patients were sedated. The tidal volume per kilogram(VT/kg) ,the fraction of tidal volume ex-heled at peak tidal expiratory flow (PTEF) to total tidal volume (VPTEF/VE), and the fraction of exhaled time at PTEF to total expirato-ry time (TPTEF/TE) were measured. Results VT/kg was not significant different between the bronehiolitis group and the pneumonia group. VPTEF/VE and TPTEF/TE were lower in the bronehiolitis group than those in the pneumonia group. Conclusion The measure-ment of tidal breathing flow - volume loops is a good technology, which would be helpful to evaluate airway obstruction in wheezing in-fants. The impact factors should be controlled during the measurement. Otherwise, the results would be affected.

<p><b>OBJECTIVE</b>To study micro morphology and element-mixing distribution of different alloys welded in laser and analyze the feasibility of laser welding different alloys.</p><p><b>METHODS</b>Alloys and titanium were matched into 4 groups: Au-Pt with Ni-Cr; Au-Pt with pure Ti; pure Ti with Ni-Cr; Ni-Cr with Co-Cr. They were welded in laser. Changes in metallography after hybridization of crystalline grain, ranges of heat-affected zone and pores were observed through SEM with ultra-thin windowed X-ray energy atlas. Meanwhile 10 testing points were chosen with area of 300 micro m x 900 micro m along the welding surface from the side A alloy to the side B alloy, than the element mixing distribution and tendency were analyzed with X-ray energy atlas.</p><p><b>RESULTS</b>1. Hybridization of different alloys: (l) in the group of Au-Pt with Ti, there was titanium element mixing into Au-Pt tissue gradually and evenly on the Au-Pt side of the interface without clear boundary and increasing in size of crystalline grain. However, there was titanium crystalline grain increasing in size, irregular morphology and small sacks on the titanium side with clear boundary. (2) in the group of Ni-Cr with Ti, there was mixing regularly, slow transition and interlocks between crystalline grains on the Ni-Cr side of the in terface. Poor transition, clear boundary and small cracks were observed on titanium side. (3) in the group of Co-Cr with Ni-Cr, there was good transition, obscure boundary on both sides resulting from network, cylinder and branch structure growing. 2. Element-mixing distribution of different alloys. In fusion zone, the metal elements in matched groups mixed well and hybridized into new alloys except titanium blocks. The location of wave peak depended on the composition of alloys. Most of elements were from the alloy far from the fusion zone.</p><p><b>CONCLUSION</b>The hybridization between pure titanium and any other alloys is not good The effect of laser welding different alloys is ideal except with pure titanium.</p>

RESUMO

Objective:To investigate the diagnosis and therapy approaches of carotid body tumor (CBT).Method:Retrospective study of diagnosis and treatment of 10 patients with CBT was conducted.Results:Five patients were examined by B ultrasonic technique and all were not diagnosed;eight were examined by color Doppler flow imaging, six of whom were diagnosed as with CBT;six were examined by CT, three of whom were diagnosed;three was examined by digital subtraction angiography(DSA) and all were diagnosed;two were examined by magnetic resonance imaging(MRI) and one was diagnosed.There was no patient died during operation.One patient died 4 days after operation because of anoxemia in brain. Pathological examination after operation confirmed the diagnosis.Conclusion:Synthetical examinations should be taken to confirm the diagnosis of CBT. Color Doppler flow imaging and DSA are significant. Selection of operation methods should be based on the practice.

RESUMO

Objective To study microsatellite instability(MSI) in multiple primary colorectal carcinoma(MPCC) and solitary colorectal tumor(SCT), and to explore the relationship between the expression of mismatch repair(MMR)、p53、Bax、PCNA and MSI. Methods The expression of MMR、p53、Bax、 PCNAwere detected by immunohistochemical staining, and MSI at five microsatellite loci were examined by PCR-SSLP in 51 tumors from 38 MPCC patients and 35 SCT cases. Results The replication errors positive phenotype was observed in 27 of 51(53%) tumor foci from MPCC cases, and in 6 of 35(17%) SCT cases. There was an inverse correlation between replication errors (RER) positive and expression of p53; the PCNA labeling index of RER positive tumors were significantly lower than of RER negative tumors; RER positive related strongly with poor differentiation, the proclivity for proximal colon. Conclusions MSI may play an important role in the development of MPCC and may be used as a tumor marker of MPCC.
