RESUMO

Objective: To study the significance of Th17 cells in patients with myelodysplastic syndrome (MDS) and iron overload. Methods: A total of 77 patients with MDS admitted to Guangzhou First People's Hospital were enrolled from January 2017 to December 2018,who were divided into iron overload group (37 cases) with serum ferritin (SF) over 1000 µg/L and non-ferrous overload group(40 cases). CD(4)(+)T cells in peripheral blood (PB) and bone marrow (BM) were sorted by flow cytometry. The ratio of Th17 cells and cells with abnormal karyotype were compared. IL-17 and IL-6 protein and RNA expression were detected by ELISA and quantitative real-time PCR(qRT-PCR). Results: The proportions of Th17 cells in PB and BM in iron overload group were significantly higher than those in non-iron overload group [(41.06±0.96)% vs. (26.80±1.21)%; (47.39±1.60)% vs. (34.29±1.03)%; P<0.01]. The Th17 positive cells with abnormal karyotype in iron overload group were more than those in non-iron overload group[(4.96±0.53)% vs. (3.67±0.12)% in PB; (10.06±1.67)% vs. (4.36±0.43)% in BM; P<0.01]. Similarly,the protein levels as well as mRNA expression of IL-6 and IL-17 in patients with iron overload were significantly higher than those in non-iron overload group (P<0.01 both in PB and BM). Conclusions: As hematopoietic regulators secreted by Th17 cells, the expression of IL-6 and IL-17 in MDS patients with iron overload are elevated. This may predict the influence of these factors to the differentiation of Th17 cells.

Objective: To analyze the patterns of female breast cancer and their incidence in Shanghai during 2003-2012, and to compare them with international data. Methods: The data of female breast cancer cases diagnosed between 2003 and 2012 were collected from the Shanghai Cancer Registry. The record of cancer cases included the patient's name, gender, date of birth, permanent and residential address, diagnosis site or name, histological type, date of first diagnosis, diagnostic basis and diagnostic stage, etc. Data source of cancer incidence of World, Developed Countries, Developing Countries, East Asia and China in 2012 sourced from GLOBOCAN 2012 database of the WHO International Agency for Research on Cancer (IARC); and the data of USA during 2008-2012 was collected from SEER 18 database from Surveillance, Epidemiology, and End Results Program of the National Cancer Institute (NCI). Incidence indicators of Shanghai female breast cancer were calculated, including the average cases, median age, average age, proportion, crude rate, age-specific rate and age-standardized rate. Results: During 2003-2012, 41 991 new cases of female breast cancer were diagnosed in Shanghai, with an average of 4 199 cases per year, accounting for 17.23% among total new female cancer cases, ranking first in female cancer incidence in Shanghai during 2003-2012, with a crude incidence rate of 60.90 per 100 000 and an age-standardized incidence rate of 33.90 per 100 000. During 2008-2012, an average 4 528 new cases of female breast cancer was diagnosed per year in Shanghai, with 16.97% among total female cancer cases. The median age was 56 years old and the mean age was 57.73 years old. The crude rate was 64.23 per 100 000 and the age-standardized rate was 35.00 per 100 0000, which was higher than those of China (22.1 per 100 000), Eastern Areas (27.0 per 100 000) and Developing Counties (31.3 per 100 000), and lower than that of World (43.1 per 100 000), and Yellow Race Population of USA (68.77 per 100 000). During 2003-2007, on average, 3 869 new cases of female breast cancer were diagnosed per year in Shanghai, accounting for 17.54% of total female cancer cases. The median age was 53 years old and the mean age was 56.44 years old. The crude rate was 57.42 per 100 000 and the age-standardized rate was 32.69 per 100 000. There was a continuing peak stage spanned 25 years old from 50 to 74 years old, during 2008-2012. Compared with those of 2003-2007, the beginning age group of the continuing peak stage was delayed for 5 years old and the ending age group was delayed for 10 years old. Conclusion: The age-standardized incidence rate in Shanghai was higher than those of China, Eastern Areas and Developing Counties, and lower than that of World, and that of Yellow Race Population of USA; Compared with 2003-2007, the crude rate and age-standardized rate increased, the median age, mean age and the beginning and ending age group of the continuing peak stage were delayed.

PURPOSE: Purpose of the study is to reveal the changes of directional diffusion in cerebral white matter (WM) by normal aging. MATERIALS AND METHODS: Thirty-nine volunteers were recruited to examine the changes in the directional diffusion of cerebral white matter (WM) due to normal aging. RESULTS: No significant difference between the older and younger group (P>.05) was detected in the axial diffusivity (λ(ll)) of any of the regions of interest (ROI), while radial diffusivity (λ(perpendicular)) was significantly higher in the older group (P<.05) except for occipital lobe WM. CONCLUSION: λ(ll) and λ(perpendicular) may be used as in vivo markers that differentially and specifically reflect the WM changes of normal aging.

OBJECTIVE: To examine whether periostin-like factor (PLF) stimulation of the expanded palatal suture would accelerate mineral formation of bone. MATERIALS AND METHODS: Expanded palates of 6-week-old male C57BL/6 mice were maintained in an organ culture system, and tissue was either unstimulated or stimulated with PLF or BMP-7 for 8 days. Bone mineral formation was assessed by Raman microspectroscopy analysis and histological examination. RESULTS: Raman microspectroscopy analysis demonstrated that unstimulated palates maintained their bone mineral concentration within the palatal suture over 8 days (%increase = 11.29 ± 2.34). In comparison, palates exposed to either PLF (%increase = 29.33 ± 1.61, p = 0.012) or BMP-7 (% increase = 25.49 ± 1.09, p = 0.016) formed significantly more bone at the osteogenic fronts of the palatal suture compared with unstimulated samples at day 8. Alkaline phosphatase activity along the bone edges was markedly greater in the PLF and BMP-7 groups compared with that in unstimulated groups at day 0 and day 8. The levels of osteocalcin proteins in the palatal suture tissues of the PLF and BMP-7 groups were significantly higher than those in the unstimulated group. CONCLUSION: PLF can increase bone mineral formation within the expanded palatal suture.

This experiment was conducted to investigate the effects of extracellular CLA on proliferation and differentiation of cultured adipocytes and its dietary supplementation on backfat and intramuscular fat deposition in finishing pigs. Seven-day-old Duroc x Landrace x Large White pigs were killed to obtain adipocytes for culture. Adding 3 forms of CLA (cis-9, trans-11 CLA, trans-10, cis-12 CLA, or CLA mixture) at concentrations of 50, 100, 150, 200, 250, 300, 350, and 400 microM to culture medium for 10 d increased cell differentiation (P < 0.05). In addition, 3 forms of CLA enhanced cell proliferation (P < 0.05) at 50 to 350 microM and inhibited cell proliferation (P < 0.05) at a concentration of 400 microM. Seventy-two Duroc x Landrace x Large White crossbred gilts weighing 60.7 +/- 2.8 kg were randomly assigned to 1 of the 3 dietary treatments (control, 1.25% CLA, and 2.5% CLA). The pigs were slaughtered at 100.0 +/- 5.7 kg. OBJECTIVE: To observe the effect of Erigeron breviscapus (Vant) Hand Mazz (HEr) in impeding oral leukoplakia carcinogenesis, and to seek effective Chinese herb medicine that can impede precarcinoma of oral mucosas. METHODS: 132 golden hamsters were randomly divided into model group (60 animals), HEr group (60 animals), and control group 12 animals. Salley's leukoplakia carcinogenesis model of golden hamster cheek pouch was used in this study. HEr was injected into the stomach to impede evolution of carcinogenesis. Pathological specimens were observed via naked eye and light microscope between model group and HEr group. Results were compared. RESULTS: Observation via naked-eye showed that leukoplakia rate of HEr group (18.2%) was lower than that of model group (27.3%). Observation via light microscope showed that carcinogenesis rate descended one fold and displasia rate descended 0.4 fold in HEr group. CONCLUSION: HEr has exact effect in impeding leukoplakia carcinogenesis.

OBJECTIVE: To observe dynamically that Erigeron Breviscapus (Vant) Hand-Mazz (HEr) affects the expression of alpha-smooth muscle actin (alpha-SMA). To discuss the probable mechanism of obstructing leukoplakia carcinogenesis of this medicine. METHODS: 120 golden hamsters were randomly divided into model group (48), HEr group (48) and control group (6). HEr was applied to obstruct the evolution of carcinogenesis of golden hamster cheek pouch. Immunohistochemistry was used to detect the expression level of alpha-SMA with cheek pouch specimen that besmears DMBA in 4-9 weeks. Results were compared with model group. RESULTS: Vessel density dyed with alpha-SMA continuously of HEr group was 65.76 significantly higher than that of model group 42.12 (P<0.001). High classification cases in HEr group were much more than model group when cases were divided into five groups as follow: 100%, 50%, 20%, 10%, 3% (P<0.01). CONCLUSION: HEr can raise the expression level of alpha-SMA exactly during the evolution of leukoplakia carcinogenesis of golden hamster, which shows that this medicine obstructs carcinogenesis by keeping the normal physiological function of vascular myoepithelial cell and integrity of vascular basement membrane.

OBJECTIVE: To evaluate the effect of resisting canceration of Herba Erigerontis HEr and that of angiogenesis; seek the mechanism of angiogenesis. METHODS: 150 golden hamsters were randomly divided into model group (66 animals), HEr group (66) and control group (18). Carcinomatous conversions of golden hamster cheek pouches induced by Sally way and interrupted by Herba Erigerontis. Ink perfusion, analysis of images, resin cast form of micro vessel, alpha-SMA detection and histopathological examination were applied to observe their changing patterns. RESULTS: The rate of leukoplakia canceration of Herba Erigerontis group was 50% lower than that of model group. The rate of normal cells was about four times more than that of model. alpha-SMA of Herba Erigerontis 65.76 was higher than that of model (P<0.001). There was no significant difference between angeion vessel area and density (P>0.05). Nevertheless, the resin cast form of micro-vessel showed that its configuration and spacious arrangement was similar to control group. CONCLUSION: Herba Erigerontis has the function of preventing leukoplakia canceration absolutely. Maybe its mechanism is to upregulate the expression of alpha-SMA. It has no obvious effect of angio-hyperplasia and expansion in the course of leukoplakia canceration. However, it has the protecting function to angio-configuration and spacious arrangement and the integrity of angeion vessed wall. Probably its mechanism is promoting blood circulation by removing blood stasis and benign angiogenesis so as to resist canceration.

OBJECTIVE:To analysis post-treatmental reaction and long-term effect of one-visit root canal therapy by microwave instrument. METHODS:Applying heat effect and the other ones of microwave,the changes of dentine tissue of extracted teeth were observated by SEM examination.48hs and 1 year post-treatmental effect of 47 patients of periapical periodontitis treated by one-visit root canal therapy were observated and compared with the results of 54 cases treated with common method.RESULTS:In the cases of acute and chronic periapical periodontitis microwave applied in root canal therapy can close the openings of dentine,enhance the effects of sterilization and decrease times of treatment.CONCLUSION:Microwave applied in one-visit root canal therapy is competent.
