RESUMO

BACKGROUND: Dendrobium nobile and Dendrobium chrysotoxum are important species of the genus Dendrobium and have great economic and medicinal value. However, the medicinal properties of these two plants remain poorly understood. This study aimed to investigate the medical properties of D. nobile and D. chrysotoxum by conducting a comprehensive chemical profiling of the two plants. Additionally, active compounds and predictive targets for anti-hepatoma activity in D. chrysotoxum extracts were identified using Network Pharmacology. RESULTS: Chemical profiling showed that altogether 65 phytochemicals were identified from D. nobile and D. chrysotoxum, with major classes as alkaloids, terpenoids, flavonoids, bibenzyls and phenanthrenes. About 18 compounds were identified as the important differential metabolites in D. nobile and D. chrysotoxum. Furtherly, CCK-8 results showed that the extracts of stems and leaves of D. nobile and D. chrysotoxum could inhibit the growth of Huh-7 cells, and the anti-hepatoma activity of extracts were dose-dependent. Among the extracts, the extract of D. chrysotoxum showed significant anti-hepatoma activity. In order to find the potential mechanism of anti-hepatoma activity of D. chrysotoxum, five key compounds and nine key targets were obtained through constructing and analyzing the compound-target-pathway network. The five key compounds were chrysotobibenzyl, chrysotoxin, moscatilin, gigantol and chrysotoxene. Nine key targets, including GAPDH, EGFR, ESR1, HRAS, SRC, CCND1, HIF1A, ERBB2 and MTOR, could be considered as the core targets of the anti-hepatoma activity of D. chrysotoxum. CONCLUSIONS: In this study, the chemical composition difference and anti-hepatoma activity of stems and leaves of D. nobile and D. chrysotoxum were compared, and the potential anti-hepatoma mechanism of D. chrysotoxum was revealed in a multi-target and multi-pathway manner.

RESUMO

Objective:To explore the implementation of the WeChat platform combined with family participation in the health management of dyslipidemia. Methods:A total of 100 patients with dyslipidemia who underwent physical examination in the Health Management Center of Chongqing General Hospital from March to August 2017 were selected. Participants were divided into an intervention and control group by independent registration, with 50 patients in each group. The control group received routine health guidance and telephone follow-ups. The intervention group underwent the health management mode of the WeChat platform combined with family participation beginning in September for six months. Dyslipidemia was managed through joint participation of family members and medical care, and the professional health management and medical team conducted real-time consultation, monitoring, follow-ups, and answered questions. A t-test was performed to compare total cholesterol, triacylglycerol, and LDL cholesterol in the two groups, and a χ 2 test was used to compare improvements in living habits in the two groups. Results:There were no statistically significant differences between the two groups before intervention ( P>0.05). After 6 months of intervention, fasting total cholesterol (6.18±0.93) mmol/L, low-density lipoprotein cholesterol (2.96±0.67) mmol/L and triglyceride (2.20±0.35) mmol/L were all lower in the intervention group than in the control group (7.17±1.07) mmol/L, (3.26±0.57) mmol/L and (2.41±0.36) mmol/L, respectively, with statistically significant difference ( P<0.05). After 6 months of intervention, the proportion of people in the intervention group (18% irregular diet, 22% lack of exercise, 26% insomnia, 14% smoking and 18% drinking) was significantly lower than that in the control group (58% irregular diet, 74% lack of exercise, 54% insomnia, 38% smoking and 42% drinking), and the difference was statistically significant ( P<0.05). Conclusions:Dyslipidemia is closely related to living habits. Through the help of the WeChat platform, family members and medical staff can participate together and effectively improve the living habits of people with dyslipidemia, so as to improve the indicators of lipid metabolism.

RESUMO

Objective To investigate the effects of leptin on Treg cells and the possible mecha-nism. Methods Leptin-deficient ( ob/ob) mice and homologous wild-type mice were used in this study. The percentages of Treg cells in spleen tissues and peripheral blood samples were measured by flow cytometry ( FCM) . Differences in Treg cell functionality were compared between the two groups. Splenic CD4+T cells, separated from the ob/ob mice and the wild-type mice by magnetic beads, were respectively cultured with leptin and anti-leptin neutralization antibody to evaluate the effects of leptin on Treg cells. Quantitative real-time PCR was performed to analyze the expression of Treg cell-related cytokines at transcriptional level. The levels of IL-10 and TGF-β in the supernatants of CD4+T cell culture were measured with Luminex technolo-gy. Results Compared with the wild-type mice, the ob/ob mice showed higher percentages of Treg cells in both peripheral blood samples and spleen tissues [(11. 56 ± 0. 72)% vs (5. 47 ± 0. 81)%, (10. 16 ± 0.93)% vs (6.29±0. 69)%]. Treg cells isolated from the ob/ob mice had stronger immunosuppressive effects on the proliferation of effector T ( Teff) cells and the secretion of TNF-α and IFN-γ than those from the wild-type mice [TNF-α:(1. 6±0. 2)% vs (2. 4±0. 5)%, IFN-γ:(4. 3±0. 3)% vs (7. 2±1. 2)%]. The percentages of Treg cells were decreased from (12. 2±1. 8)% to (7. 6±0. 9)% upon the in vitro treat-ment of CD4+ T cells from the ob/ob mice with leptin and the immunosuppressive effects of Treg cells were also weakened. However, the percentages of Treg cells were increased from (7. 8±0. 85)% to (13. 1± 1. 5)% upon the in vitro treatment of CD4+T cells from the wild-type mice with anti-leptin antibody and the immunosuppressive effects of Treg cells were improved as well. Moreover, the expression of Foxp3, IL-10 and TGF-β at transcriptional level and the levels of IL-10 and TGF-β in the ob/ob group were higher than those in the wild-type group. Conclusions Leptin deficiency significantly promoted the generation of Treg cells in mice and resulted in an increased expression of Foxp3, IL-10 and TGF-βat mRNA level and elevat-ed levels of IL-10 and TGF-β. The treatment of CD4+T cells with leptin might inhibit the generation of Treg cells through down-regulating the transcription of Foxp3, IL-10 and TGF-β.

RESUMO

Andrographolide (AP) is a diterpenoid separated from Andrographis paniculata with a wide spectrum of biological activities including anti-inflammatory, anticancer, hepatoprotective, and antihyperlipidemic. However, its poor water solubility and instability result in lower bioavailability, which seriously limit its pharmacological function. In this study, the attempt to use regenerated silk fibroin (RSF) as a drug-carrier to encapsulate AP was reported. The AP-loaded RSF nanoparticles were prepared by a facile and clean method without any toxic agents. Moreover, special attention was paid to the optimization of formulation. Finally, the sizes of the AP-loaded RSF nanoparticles ranged from 200 to 1000 nm, and the nanoparticles were spherically shaped, as seen by transmission electron microscopy. The drug loading and encapsulation efficiency were about 25.9% and 87.3%, respectively. Furthermore, the release time of AP-loaded RSF nanoparticles was about 3 days. The particle size and drug release behaviour could be adjusted by treating with glycol amine. The in vitro cytotoxicity studies demonstrated that the RSF nanoparticles showed negligible cytotoxicity to cells, and the anti-proliferative activity of AP-loaded RSF nanoparticles showed that the AP-loaded RSF nanoparticles can adhere to Hela cells and MDA-MB-231 cells easily. All these results imply that this biomacromolecule drug nanocarrier has great potential for chemotherapy in clinical applications.
