In recent years, the global carbon cycle has garnered significant research attention. However, details of the intricate relationship between planktonic bacteria, hydrochemistry, and dissolved organic matter (DOM) in inland waters remain unclear, especially their effects on lake carbon sequestration. In this study, we analyzed 16S rRNA, chromophoric dissolved organic matter (CDOM), and inorganic nutrients in Erhai Lake, Yunnan Province, China. The results revealed that allochthonous DOM (C3) significantly regulated the microbial community, and that autochthonous DOM, generated via microbial mineralization (C2), was not preferred as a food source by lake bacteria, and neither was allochthonous DOM after microbial mineralization (C4). Specifically, the correlation between the fluorescence index and functional genes (FAPRPTAX) showed that the degree of utilization of DOM was a critical factor in regulating planktonic bacteria associated with the carbon cycle. Further examination of the correlation between environmental factors and planktonic bacteria revealed that Ca2+ had a regulatory influence on the community structure of planktonic bacteria, particularly those linked to the carbon cycle. Consequently, the utilization strategy of DOM by planktonic bacteria was also determined by elevated Ca2+ levels. This in turn influenced the development of specific recalcitrant autochthonous DOM within the high Ca2+ environment of Erhai Lake. These findings are significant for the exploration of the stability of DOM within karst aquatic ecosystems, offering a new perspective for the investigation of terrestrial carbon sinks.

INTRODUCTION: Prostate cancer is a common urology malignant in males, ranking second globally. The disease is especially severe when diagnosed alongside hypertension. MKI67 is an established marker of neoplastic cell proliferation in humans, but the significance of its prognostic value in patients with prostate cancer and hypertension requires further research. METHODS: In this retrospective analysis, we evaluated 296 hypertensive prostate cancer patients between March 2, 2012, and November 1, 2015. We used Cox regression models and prediction analysis to assess overall survival. Furthermore, we created a nomogram and verified its accuracy using a calibration curve. RESULTS: Of all participants, 101 (34.12%) died. Our multi-factor analysis revealed that MKI67 expression was associated with an increased hazard ratio of death (> fivefold) (Hazard Ratio 5.829, 95% CI 3.349-10.138, p value < 0.01) and progression (twofold) (HR 2.059, 95% CI 1.368-3.102, p value < 0.01). Our Lasso analysis model displayed that several factors, including heart failure, smoking, ACS, serum albumin, Gealson score, prognostic nutritional index, MKI67 expression, surgery, and stage were high risks of prostate cancer. To ensure each covariate's contribution to cancer prognosis, we created a Cox model nomogram, which accurately predicted the risk of death (C-statistic of 0.8289) and had a proper calibration plot for risk assessment. CONCLUSION: MKI67 expression predicts poor outcomes for overall mortality in prostate cancer and hypertension patients. Additionally, our cross-validated multivariate score, which includes MKI67, demonstrated accuracy efficacy of predicting prognosis.

The fruits of Rosa roxburghii Tratt. are edible nutritional food with high medicinal value and have been traditionally used as Chinese folk medicine for a long time. In this study, 26 triterpenoids including four new pentacyclic triterpenoids, roxbuterpenes A-D (1, 4, 5, and 24), along with 22 known analogues (2, 3, 6-23, 25, and 26), were isolated from the fruits of R. roxburghii. Their chemical structures were determined on the basis of extensive spectroscopic analyses (including IR, HRESIMS and NMR spectroscopy). The absolute configuration of roxbuterpene A (1) was determined by an X-ray crystallographic analysis. This is the first report of the crystal structure of 5/6/6/6/6-fused system pentacyclic triterpenoid. Notably, roxbuterpenes A and B (1 and 4) possessed the A-ring contracted triterpenoid and nortriterpenoid skeletons with a rare 5/6/6/6/6-fused system, respectively. Compounds 1-7, 11, 13-15, 18-20, 24, and 25 exhibited moderate or potent inhibitory activities against α-glucosidase. Compounds 2, 4, 6, 11, and 14 showed strong activities against α-glucosidase with IC50 values of 8.4 ± 1.6, 7.3 ± 2.2, 13.6 ± 1.4, 0.9 ± 0.4, and 12.5 ± 2.4 µM, respectively (positive control acarbose, 10.1 ± 0.8 µM). Compounds 13, 14, and 16 moderately inhibited the release of NO (nitric oxide) with IC50 values ranging from 25.1 ± 2.0 to 51.4 ± 3.1 µM. Furthermore, the expressions of TNF-α (tumor necrosis factor-α) and IL-6 (interleukin-6) were detected by ELISA (enzyme-linked immunosorbent assay), and compounds 13, 14, and 16 exhibited moderate inhibitory effects on TNF-α and IL-6 release in a dose-dependent manner ranging from 12.5 to 50 µM.

To assess the impact of sulfate mine water on filling material performance, an accelerated sulfate erosion process was used to analyze the effects of various erosion concentrations, aging periods, and cation types on the macroscopic properties of the filling paste. These properties encompassed apparent phenomena, mass changes, and alterations in the uniaxial compressive strength. Observations revealed sulfate erosion, causing the formation of white substances and salt crystals on specimen surfaces. Initially, all solution-eroded specimens exhibited increased mass and strength. Over time, specimens in 5 and 10% MgSO4 solutions displayed the first signs of decline, while variations in other solutions were relatively small. Increasing the erosion concentration led to greater variations in mass and strength during the initial erosion phase. Specimens in 5 and 10% MgSO4 solutions initially peaked in mass and compressive strength, followed by a decline, while other filling paste specimens continued slow increases. Under equivalent conditions, the MgSO4 solution exhibited stronger erosion than the Na2SO4 solution. Composite erosion by Na2SO4 and MgSO4 involved initial strengthening and gel pore filling, intermediate expansion and crystallization, and late-stage substantial degradation, with MgSO4 exhibiting a more pronounced and complex impact. Gray relational analysis ranked factors affecting mass and uniaxial compressive strength variations as erosion concentration > erosion ion type > erosion aging period. Correlation degrees for factors influencing mass variations were 0.8822, 0.8714, and 0.4754, while for factors influencing uniaxial compressive strength variations, the correlation degrees were 0.8336, 0.7943, and 0.6125, respectively.

Intrahepatic cholangiocarcinoma (ICC) is a highly malignant neoplasm prone to metastasis. Whether cancer-associated fibroblasts (CAFs) affect the metastasis of ICC is unclear. Herein, ICC patient-derived CAF lines and related cancerous cell lines were established and the effects of CAFs on the tumor progressive properties of the ICC cancerous cells were analyzed. CAFs could be classified into cancer-restraining or cancer-promoting categories based on distinct tumorigenic effects. The RNA-sequencing analyses of ICC cancerous cell lines identified polycomb group ring finger 4 (PCGF4; alias BMI1) as a potential metastasis regulator. The changes of PCGF4 levels in ICC cells mirrored the restraining or promoting effects of CAFs on ICC migration. Immunohistochemical analyses on the ICC tissue microarrays indicated that PCGF4 was negatively correlated with overall survival of ICC. The promoting effects of PCGF4 on cell migration, drug resistance activity, and stemness properties were confirmed. Mechanistically, cancer-restraining CAFs triggered the proteasome-dependent degradation of PCGF4, whereas cancer-promoting CAFs enhanced the stability of PCGF4 via activating the IL-6/phosphorylated STAT3 pathway. In summary, the current data identified the role of CAFs in ICC metastasis and revealed a new mechanism of the CAFs on ICC progression in which PCGF4 acted as the key effector by both categories of CAFs. These findings shed light on developing comprehensive therapeutic strategies for ICC.

Evidence for the involvement of N6-Methyladenosine (m6A) modification in the etiology and progression of liver fibrosis has emerged and holds promise as a therapeutic target. Insulin-like growth factor 2 (IGF2) mRNA-binding protein 2 (IGF2BP2) is a newly identified m6A-binding protein that functions to enhance mRNA stability and translation. However, its role as an m6A-binding protein in liver fibrosis remains elusive. Here, we observed that IGF2BP2 is highly expressed in liver fibrosis and activated hepatic stellate cells (HSCs), and inhibition of IGF2BP2 protects against HSCs activation and liver fibrogenesis. Mechanistically, as an m6A-binding protein, IGF2BP2 regulates the expression of Aldolase A (ALDOA), a key target in the glycolytic metabolic pathway, which in turn regulates HSCs activation. Furthermore, we observed that active glycolytic metabolism in activated HSCs generates large amounts of lactate as a substrate for histone lactylation. Importantly, histone lactylation transforms the activation phenotype of HSCs. In conclusion, our findings reveal the essential role of IGF2BP2 in liver fibrosis by regulating glycolytic metabolism and highlight the potential of targeting IGF2BP2 as a therapeutic for liver fibrosis.

Ardisia crenata Sims, an important ethnic medicine, is recorded in the Chinese Pharmacopoeia for treating laryngeal diseases and upper respiratory tract infections. This study aimed to evaluate the antimicrobial effect of extracts and potential antimicrobial compounds of A. crenata Sims. It was found that the roots of A. crenata Sims have a potential inhibitory effect on Candida albicans and Aspergillus flavus, with MICs of 1.56 mg/mL and 0.39 mg/mL, and the leaves of A. crenata Sims have a potential inhibitory effect on Pseudomonas aeruginosa and Staphylococcus aureus, with MICs of 3.12 mg/mL and 6.77 mg/mL, respectively. Meanwhile, five compounds including one catechin and four bergenins were obtained from roots. These components were identified on the fingerprint spectrum, representing chromatographic peaks 16, 21, 22, 23, and 25, respectively. Among these, 11-ß-d-glucopyranosyl-bergenin and (-)-gallocatechin showed potential inhibition for Staphylococcus aureus and Pseudomonas aeruginosa with MIC of 0.26 and 0.33 mg/mL, respectively. The roots, stems, and leaves of A. crenata Sims are very similar in chemical composition, with large differences in content. Principal component analysis (PCA) and Hierarchical cluster analysis (HCA) showed that 16 batches of A. crenata Sims could be divided into four main production areas: Guizhou, Jiangsu, Guangxi, and Jiangxi. Furthermore, molecular docking results showed that 11-ß-d-glucopyranosyl-bergenin had a better affinity for Casein lytic proteinase P (ClpP), and (-)-gallocatechin possessed a strong affinity for LasA hydrolysis protease and LasB elastase. These findings suggest catechin and bergenins from A. crenata Sims can be used as antimicrobial activity molecules.

Due to the large spatiotemporal variability in the processes controlling carbon emissions from lakes, estimates of global lake carbon emission remain uncertain. Identifying the most reliable predictors of CO2 and CH4 concentrations across different hydrological features can enhance the accuracy of carbon emission estimates locally and globally. Here, we used data from 71 lakes in Southwest China varying in surface area (0.01‒702.4 km2), mean depth (< 1‒89.6 m), and climate to analyze differences in CO2 and CH4 concentrations and their driving mechanisms between the dry and rainy seasons and between different mixing regimes. The results showed that the average concentrations of CO2 and CH4 in the rainy season were 23.9 ± 18.8 µmol L-1 and 2.5 ± 4.9 µmol L-1, respectively, which were significantly higher than in the dry season (10.5 ± 10.3 µmol L-1 and 1.8 ± 4.2 µmol L-1, respectively). The average concentrations of CO2 and CH4 at the vertically mixed sites were 24.1 ± 21.8 µmol L-1 and 2.6 ± 5.4 µmol L-1, being higher than those at the stratified sites (14.8 ± 13.4 µmol L-1 and 1.7 ± 3.5 µmol L-1, respectively). Moreover, the environmental factors were divided into four categories, i.e., system productivity (represented by the contents of total nitrogen, total phosphorus, chlorophyll a and dissolved organic matter), physicochemical factors (water temperature, Secchi disk depth, dissolved oxygen and pH value), lake morphology (lake area, water depth and drainage ratio), and geoclimatic factors (altitude, wind speed, precipitation and land-use intensity). In addition to the regression and variance partitioning analyses between the concentrations of CO2 and CH4 and environmental factors, the cascading effects of environmental factors on CO2 and CH4 concentrations were further elucidated under four distinct hydrological scenarios, indicating the different driving mechanisms between the scenarios. Lake morphology and geoclimatic factors were the main direct drivers of the carbon concentrations during the rainy season, while they indirectly affected the carbon concentrations via influencing physicochemical factors and further system productivity during the dry season; although lake morphology and geoclimatic factors directly contributed to the carbon concentrations at the vertically mixed and stratified sites, the direct effect of system productivity was only observed at the stratified sites. Our results emphasize that, when estimating carbon emissions from lakes at broad spatial scales, it is essential to consider the influence of precipitation-related seasons and lake mixing regimes.

Terrestrial inputs and subsequent degradation of dissolved organic matter (DOM) in lake ecosystems can result in rapid depletion of dissolved oxygen (DO). Inputs of terrestrial DOM including organic acids can also lead to decreases in pH. However, to date, few studies have investigated the linkages between terrestrial DOM inputs, DO and pH levels in the water column, and carbon dioxide (CO2) emissions from lake ecosystems. Based on monthly field sampling campaigns across 100 sites in Lake Qiandao, a major man-made drinking water reservoir in China, from May 2020 to April 2021, we estimated an annual CO2 efflux (FCO2) of 37.2 ± 29.0 gC m-2 yr-1, corresponding to 0.02 ± 0.02 TgC yr-1 from this lake. FCO2 increased significantly with decreasing DO, chlorophyll-a (Chl-a) and δ2H-H2O, while FCO2 increased with increasing specific UV absorbance (SUVA254) and a terrestrial humic-like component (C2). We found that DO concentration and pH declined with increasing terrestrial DOM inputs, i.e. increased SUVA254 and terrestrial humic-like C2 levels. Vertical profile sampling revealed that the partial pressure of CO2 (pCO2) increased with increasing terrestrial DOM fluorescence (FDOM), while DO, pH, and δ13C-CO2 declined with increasing terrestrial FDOM. These results highlight the importance of terrestrial DOM inputs in altering physico-chemical environments and fueling CO2 emissions from this lake and potentially other aquatic ecosystems.

OBJECTIVE: To compare the efficacy and safety of relocating the lower pole stones to a favorable pole during flexible ureteroscopy with in situ lithotripsy for the treatment of 10-20 mm lower pole stone (LPS). METHODS: This study was a prospective analysis of patient outcomes who underwent an FURS procedure for the treatment of 10-20 mm lower pole renal stones from January 2020 to November 2022. The patients were randomized into a relocation group or in situ group. The LPSs were relocated into a calyx, during lithotripsy in the relocation group was performed, whereas the in situ group underwent FURS without relocation. All the procedures were performed by the same surgeon. The patients' demographic data, stone characteristics, perioperative parameters and outcomes, stone-free rate (SFR), complications, and overall costs were assessed retrospectively. RESULTS: A total of 90 patients were enrolled and analyzed in this study (45 per group) with no significant differences between the two groups in terms of age, gender, BMI, diabetes, hypertension, stone size, number, laterality, composition, and density. The mean operation time, total energy consumption, postoperative stay, and complications were similar between the groups. Both groups had similar SFR at 1 day postoperative follow-up (p = 0.091), while the relocation group achieved significantly higher SFR 3 months later (97.8% vs 84.4%, p = 0.026). The relocation group also had a significantly higher WisQol score than the in situ group (126.98 vs 110.18, p < 0.001). CONCLUSION: A satisfactory SFR with a relatively low complication rate was achieved by the relocation technique during the FURS procedure.

Stream ecosystem health assessments are crucial for the effective management of aquatic ecosystem service functions and the assessment of anthropogenic and climatic impacts on aquatic ecosystems. However, there is still a lack of comprehensive assessment methods based on complete ecosystem structures. This study improved an ecological health assessment system, which consists of 13 indicators based on ecosystem structure. The assessment system was applied to 30 streams on Changbai Mountain. The results showed that the comprehensive index of stream health (ISH) in Changbai Mountain was relatively high, and the ISH increased with the elevation gradient. The dominant factors controlling the ecosystem health level were water pH, water flow, vegetation coverage, total phosphorus (TP), and vegetation diversity index. All these results indicated that the damage caused by excessive deforestation and farmland occupation in recent years has not been fully reversed and that human disturbance from tourism may be the main reason for the low stream health at lower elevations. Our results suggest that the key to future work is to increase quantitative research on disturbance sensitivity and to develop economically viable restoration measures.

Rivers receive, transport, and are reactors of terrestrial dissolved organic matter (DOM) and are highly influenced by changes in hydrological conditions and anthropogenic disturbances, but the effect of DOM composition on the dynamics of the bacterial community in rivers is poorly understood. We conducted a seasonal field sampling campaign at two eutrophic river mouth sites to examine how DOM composition influences the temporal dynamics of bacterial community networks, assembly processes, and DOM-bacteria associations. DOM composition and seasonal factors explained 34.7% of the variation in bacterial community composition, and 14.4% was explained purely by DOM composition where specific UV absorbance (SUVA254) as an indicator of aromaticity was the most important predictor. Significant correlations were observed between SUVA254 and the topological features of subnetworks of interspecies and DOM-bacteria associations, indicating that high DOM aromaticity results in more complex and connected networks of bacteria. The bipartite networks between bacterial taxa and DOM molecular formulae (identified by ultrahigh-resolution mass spectrometry) further revealed less specialized bacterial processing of DOM molecular formulae under the conditions of high water level and DOM aromaticity in summer than in winter. A shift in community assembly processes from stronger homogeneous selection in summer to higher stochasticity in winter correlated with changes in DOM composition, and more aromatic DOM was associated with greater similarity in bacterial community composition. Our results highlight the importance of DOM aromaticity as a predictor of the temporal dynamics of riverine bacterial community networks and assembly.

The presence of organic matter in lakes profoundly impacts drinking water supplies, yet treatment processes involving coagulants and disinfectants can yield carcinogenic disinfection by-products. Traditional assessments of organic matter, such as chemical oxygen demand (CODMn) and biochemical oxygen demand (BOD5), are often time-consuming. Alternatively, optical measurements of dissolved organic matter (DOM) offer a rapid and reliable means of obtaining organic matter composition data. Here we employed DOM optical measurements in conjunction with parallel factor analysis to scrutinize CODMn and BOD5 variability. Validation was performed using an independent dataset encompassing six lakes on the Yungui Plateau from 2014 to 2016 (n = 256). Leveraging multiple linear regressions (MLRs) applied to DOM absorbance at 254 nm (a254) and fluorescence components C1-C5, we successfully traced CODMn and BOD5 variations across the entire plateau (68 lakes, n = 271, R2 > 0.8, P < 0.0001). Notably, DOM optical indices yielded superior estimates (higher R2) of CODMn and BOD5 during the rainy season compared to the dry season and demonstrated increased accuracy (R2 > 0.9) in mesotrophic lakes compared to oligotrophic and eutrophic lakes. This study underscores the utility of MLR-based DOM indices for inferring CODMn and BOD5 variability in plateau lakes and highlights the potential of integrating in situ and remote sensing platforms for water pollution early warning.

The source composition of chromophoric dissolved organic matter (CDOM) in lakes is closely related to regional environmental changes, human activities, and the carbon cycle. The spectral slope ratio (SR) is an important parameter of CDOM optical components, and combined with remote sensing technology, the source composition of CDOM can be tracked comprehensively and efficiently in large regions. Here, we proposed a CDOM source tracking remote sensing model (CDOM-SR) based on the hue angle (α) to assess the spatial pattern and long-term trend of the CDOM source composition in Chinese lakes (surface area ≥ 1 km2) from 1986 to 2021. Validation results show that the CDOM-SR model has a good SR estimation performance with a median absolute percentage difference, root mean square deviation, median ratio, and median deviation of 17.91 %, 0.23, 1.02, and 0.03, respectively. We found that the average SR of Chinese lakes presents an obvious spatial pattern of high in the west and low in the east due to the difference in human activity intensity and the natural geographical environment. Additionally, we found that the average SR of Chinese lakes from 1986 to 2021 decreased at a rate of - 0.06/10 years, of which 64.37 % of lakes decreased significantly, 15.42 % of lakes had no significant change, and only 20.20 % of lakes increased. The widespread decrease in the average SR indicates that the increasing human activity discharge of terrestrial organic matter has had an important impact on the source composition of the CDOM in Chinese lakes. Our results provide a new resource for remote sensing monitoring of CDOM sources and important insights into lake carbon cycling under the influence of ongoing human activities.

STUDY DESIGN: A retrospective study. OBJECTIVE: This study examined the value of neutrophil-to-lymphocyte ratio at admission for early diagnosis, severity assessment, and prognosis of acute traumatic SCI. SETTING: The First People's Hospital of Neijiang, China. METHODS: This was a single-center, retrospective, cohort study of patients treated within 12 h of acute SCI between January 2018 and October 2022. Ninety-four SCI patients were selected as the Observation group, including 26 with complete injury (AIS grade A) and 68 with incomplete injury (AIS grade B-D), while 94 patients with simple spinal fracture were randomly selected as the Control group. Eighty-one observation group patients underwent surgical treatment, of which 33 had a higher AIS grade (Good prognosis subgroup) and 48 a lower or equal grade post-surgery (Poor prognosis subgroup). Univariate and multivariate analyses were performed to assess predictors of early diagnosis, severity, and 6-month outcome. RESULTS: Initial white blood cell count, neutrophil count, monocyte count, and NLR were higher in the Observation group than the Control group, while lymphocyte count was lower in the Observation group. Multivariate logistic regression analysis identified NLR as an independent predictor of early diagnosis. Spinal canal encroachment ≥50%, neutrophil count, and NLR were higher in the complete injury subgroup, and spinal canal encroachment ≥50% was an independent predictor of complete injury, while NLR was not. The NLR was higher in the poor prognosis subgroup and was an independent risk factor. CONCLUSIONS: Peripheral blood NLR is useful for early diagnosis of acute SCI and is predictive of clinical outcome.

One new lactone (1) named Ardisicreolide C, together with three saponin compounds, Ardisiacrispin B (2), Ardisicrenoside A (3), Ardisiacrispin A (4) were isolated and identified from the leaves of Ardisia crenata Sims. The structures of 1-4 were elucidated by 1D, 2D-NMR and HR-MS spectra and together with the published data. In view of structures with lactone moieties showed good anti-inflammatory activity, the anti-inflammatory effects of Ardisicreolide C on LPS-induced RAW264.7 cells were evaluated by enzyme linked immunosorbent assay (ELISA) method. As a result, Ardisicreolide C could reduce release of nitric oxide (NO), tumour necrosis factor α (TNF-α), interleukin 1ß (IL-1ß), interleukin 4 (IL-4) and interleukin 10 (IL-10) of the cell supernatant to exert anti-inflammatory activity. This indicates that the leaves as non-medicinal parts of Ardisia crenata Sims contain compounds with good anti-inflammatory activity, which provides a new direction for the discovery of anti-inflammatory drugs.

Inland waters are important sources of atmospheric methane (CH4), with a major contribution from the CH4 ebullition pathway. However, there is still a lack of CH4 ebullition flux (eFCH4) and their temperature sensitivity (Q10) in shallow lakes, which might lead to large uncertainties in CH4 emission response from aquatic to climate and environmental change. Herein, the magnitude and regulatory of two CH4 pathways (ebullition and diffusion) were studied in subtropical Lake Chaohu, China, using the real-time portable greenhouse gas (GHG) analyzer-floating chamber method at 18 sites over four seasons. eFCH4 (12.06 ± 4.10 nmol m-2 s-1) was the dominant contributing pathway (73.0 %) to the two CH4 emission pathways in Lake Chaohu. The whole-lake mass balance calculation demonstrated that 56.6 % of the CH4 emitted from the sediment escaped through the ebullition pathway. eFCH4 was significantly higher in the western (WL: 16.54 ± 22.22 nmol m-2 s-1) and eastern lake zones (EL: 11.89 ± 15.43 nmol m-2 s-1) than in the middle lake zone (ML: 8.86 ± 13.78 nmol m-2 s-1; p < 0.05) and were significantly higher in the nearshore lake zone (NL: 15.94 ± 19.58 nmol m-2 s-1) than in the pelagic lake zone (PL: 6.64 ± 12.37 nmol m-2 s-1; p < 0.05). eFCH4 was significantly higher in summer (32.12 ± 13.82 nmol m-2 s-1) than in other seasons (p < 0.05). eFCH4 had a strong temperature dependence. Sediment total organic carbon (STOC) is an important ecosystem level Q10 driver of eFCH4. The meta-analysis also verified that across ecosystems the ecosystem-level Q10 of eFCH4 was significantly positively correlated with STOC and latitude (p < 0.05). This study suggests that eFCH4 will become increasingly crucial in shallow lake ecosystems as climate change and human activities increase. The potential increase in ebullition fluxes in high-latitude lakes is of great importance.

In this paper, quasi-Tamm plasmon polaritons (TPPs)/Fano resonance systems based on metal-dielectric-metal (MDM) waveguides are proposed. TPPs are surface electromagnetic modes formed at the interface between a metal and a one-dimensional dielectric photonic crystal (PhC). A metal plasmonic Bragg reflector (PBR) in a MDM waveguide is equivalent to a dielectric PhC, which is realized by periodic MDM waveguide width modulation and leads to the photonic bandgap. By introducing a thin Ag baffle and a PBR in MDM waveguide core, the quasi-TPPs are excited at the interface between the Ag baffle and the PBR, when the phase-matching condition is met. The proposed structure can be fabricated with focused ion beam or electron beam direct-writing lithography, avoiding complex fabrication procedures of manufacturing dielectric PhC by filling the MDM waveguide core with different dielectric materials. Furthermore, an MDM waveguide side-coupled resonator system is constructed to generate Fano resonance by placing a PBR on the side of the MDM waveguide and an Ag baffle in the waveguide core. The Fano resonance originates from the interference between a broad continuum state provided by the Ag baffle and a discrete state provided by quasi-TPPs. The sensing performance of the Fano resonance system is investigated. In this design, the open PBR structure replaces the traditional closed resonant cavity, which makes it more convenient to contact with analytes. The numerical simulations demonstrate that a high sensitivity of 1500 nm/RIU and figure of merit value of 4.08×105 are achieved.

Gelsegansymines A (1) and B (2), two new indole alkaloids along with six known analogues (3-8) were isolated from the aerial parts of Gelsemium elegans. Their structures were elucidated by means of spectroscopic techniques. Structurally, compounds 1 and 2 possessed the rare cage-like gelsedine skeleton hybrid with bicyclic monoterpenoid. The anti-inflammatory activities of isolated compounds (1-3) were tested on LPS induced RAW264.7 cells. Under the treated concentration without toxicity for cells, the cytokines levels of nitric oxide (NO), tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) were evaluated by Griess method and enzyme-linked immunosorbent assay (ELISA). The results showed that compounds 1-3 exhibited anti-inflammatory activities with dose-dependent manner range from 12.5 to 50 µmol/L. Furthermore, the inhibitory activities of compounds 1 and 2 on receptor activator of NF-κB ligand (RANKL) induced osteoclast formation were tested in vitro. Compounds 1 and 2 at 5 µmol/L exhibited the significant inhibitory effect on the osteoclastogenesis induced by RANKL. This work reported the anti-inflammatory and osteoclast inhibitory activities of new monoterpenoid indole hybrids, which may inspire the further light on the related traditional application research of G. elegans.
