RESUMO

Pseudosciaena polyactis is an economically important species of marine fish in China that is currently declining due to overexploitation, environmental pollution and related factors. Research in to the genetic structure of Pseudosciaena polyactis populations plays a key role in protecting and promoting sustainable utilization. We collected 53 individuals of Pseudosciaena polyactis from Zhoushan, Zhejiang and sequenced and amplified the mitochondrial DNA (mtDNA) D-loop region using Polymerase Chain Reactions (PCR). The sequence length of the 53 individuals ranged from 795 to 801 bp. The sequences were analyzed by Clustal X1.83, MEGA3.1 and DnaSP4.0. The results showed that the average base content of T, C, A, G was 30.3%, 23.1%, 32.3% and 14.3%, respectively and there were 93 transition or transversion sites, including 53 single nucleotide mutation sites and 40 parsimony informative sites, which accounted for 11.6% of the length of the analyzed sequences. In total, we identified 52 haplotypes and found haplotype diversity (hd) of 0.9993, average number of nucleotide differences were 9.73875 (k), and nucleotide diversity (Π) of 0.01233. The average genetic distance of haplotypes was 0.012, and the average transition/transversion was 4.305. Based on mitochondrial DNA D-loop region sequences, these results indicate that the genetic diversity of the Pseudosciaena polyactis population in Zhoushan is currently at a medium level.

Plant roots move through the soil by elongation. This is vital to their ability to anchor the plant and acquire water and minerals from the soil. In order to identify new genes involved in root elongation in rice, we screened an ethyl methane sulfonate (EMS)-mutagenized rice library, and isolated a short root mutant, Osglu3-1. The map-based cloning results showed that the mutant was due to a point mutation in OsGLU3, which encodes a putative membrane-bound endo-1,4-ß-glucanase. Osglu3-1 displayed less crystalline cellulose content in its root cell wall, shorter root cell length, and a slightly smaller root meristem as visualized by restricted expression of OsCYCB1,1:GUS. Exogenous application of glucose can suppress both the lower root cell wall cellulose content and short root phenotypes of Osglu3-1. Consistently, OsGLU3 is ubiquitously expressed in various tissues with strong expression in root tip, lateral root, and crown root primodia. The fully functional OsGLU3-GFP was detected in plasma membrane, and FM4-64-labeled compartments in the root meristem and elongation zones. We also found that phosphate starvation, an environmental stress, altered cell wall cellulose content to modulate root elongation in a OsGLU3-dependant way.

RESUMO

New short duration time XeCI excimer laser has been generated at high pressure within a large volume in order to apply it to he interaction between laser and material, and material plasma study. The laser spectrum exhibits two laser lines at 307.98 and 308.19 nm, which is realized in the proportion of HCl:Xe: He = 0.1% :1% : 98.9% through UV preionization. Theoretic analysis indicated that the maximum intensity loop is B to X grade. Not only UV preionization, glow discharge and the calculation of dynamic equation, but also the laser spectrum and pulse duration time measurement were carried out. It is shown that the duration time decreases and pulse energy rises with the increase in the pressure and discharge voltage. The minimum duration time exceeds 13 ns, the pulse energy is 450 mJ, and the beam divergence angle is 3 mrad.
