RESUMO

Everyday episodic memories involve linking together related events that are temporally separated. However, the mechanisms of forming this temporal association have remained unclear. Here, using astrocyte-specific manipulations, we show that potentiating astrocyte Ca2+ signaling in the hippocampal cornu ammonis 1 (CA1) enhances the strength of such temporal association, in parallel with long-term potentiation (LTP) enhancement of temporoammonic pathway to CA1, whereas attenuation of astrocyte Ca2+ signaling has the opposite effect. Moreover, we identify that these effects are mediated by astrocytic α4 subunit-containing nicotinic acetylcholine receptors (α4-nAChRs) via mechanisms involving NMDAR co-agonist supply. Finally, astrocytic α4-nAChRs underlie the cognitive enhancer nicotine's physiological effects. Together, these findings highlight the importance of astrocyte Ca2+ signaling in cognitive behavior and reveal a mechanism in governing the temporal association of episodic memory formation that operates through α4-nAChRs on hippocampal astrocytes.

RESUMO

Adeno-associated viruses (AAVs) have become safe and effective tools for therapeutic in vivo gene drug delivery. Among many AAV serotypes, AAV2 is the most well-characterized. Although many studies have been carried out on the engineering of the capsid VR-VIII region, few attempts have been made in the VR-IV region. Here, we targeted amino acid positions 442-469 of the VR-IV region and established an engineering paradigm of computer-aided directed evolution, based on training samples from previous datasets, to obtain a viral vector library with high diversity (~95,089). We further examined two variants selected from the library. The transduction efficiency of these two novel AAV variants, AAV2.A1 and AAV2.A2, in the central nervous system was 10-15 times higher than that of AAV2. This finding provides new vehicles for delivering gene drugs to the brain.

Although the occurrence and distribution of antibiotic resistance genes (ARGs) in various aquatic ecosystems are well explored, understanding of the ecological processes and mechanisms governing the composition and dynamics of bacterial ARGs still remains limited across space and time. Here, we used high-throughput approaches to detect spatial patterns of bacterial ARGs and operational taxonomic units (OTUs) in an urbanizing subtropical watershed, Xiamen, southeast China over a five-year period. At watershed scale, the OTU profiles were undergoing a directional change, but the ARG profiles showed a high stability or stochastic change over time. Compared with the upstream and midstream, the richness, absolute abundance, normalized abundance and diversity of ARGs were significantly higher in the downstream waters. Our results revealed a clear rural-urban disparity in ARG and OTU profiles which were mainly governed by deterministic and stochastic assembly processes, respectively. With the increase of urban building area along the river, the ecological processes of ARG profiles shifted from stochastic to deterministic. In downstream waters, the bacterial ARG profiles were much more stable than bacterial OTUs. Further, our results indicated that both human-dominated environment (e.g., land use) and mobile genetic elements (MGEs) played an important role in shaping the ARG profiles and dynamics. Overall, this was a response to spatially extensive human-landscape interactions that included urban development in the river downstream region, which were common across subtropical coastal cities of China and can alter the ARG profile dynamics along rural-urban gradient. Therefore, watershed management actions aiming at reducing threats posed by ARGs in urbanizing watershed should first consider the surrounding urbanization level and the mode and intensity of human activity. Our findings also imply that due to the decoupling of bacterial function and taxonomy, both aspects should be studied separately.

The Chinese yam peel (CYP) is a by-product of yam processing that is rich in various nutrients and a good source for feed additives. This study investigated the effects of CYP on the intestinal microbiota and gut defence barrier of the common carp (Cyprinus carpio L.). Different groups of experimental fish were fed a normal control diet (NC), a low CYP diet (LYP) and a high CYP diet (HYP) for 8 weeks. After the feeding trial, the fish were assessed for intestinal enzyme activity, intestinal histology, immune-related gene expression, intestinal SCFAs and intestinal microbiota. Our results indicated that the intestinal integrity and antioxidant enzyme (CAT and SOD) activity in the common carp were enhanced following CYP supplementation. The mRNA levels of anti-inflammatory (TGF-ß), tight binding protein (occludin and ZO-1) and pathway factor genes (TLR4 and NF-κB) were significantly upregulated in the HYP group (P<0.05), which was accompanied by an increase in the level of pro-inflammatory IL-1ß in the gut (P<0.05). High-throughput sequencing revealed that Fusobacteria, Proteobacteria, and Bacteroidetes bacteria were most abundant in the microbial community in the gut of the common carp. The relative abundances of Bacteroides, Flavobacterium and Lactobacillus were increased, while the abundances of pathogenic microorganisms such as Enterobacteriaceae, Shewanella, Pseudomonas and Vibrio were reduced after treatment with CYP. Furthermore, the concentrations of acetic acid, propionic acid, butyric acid and total short-chain fatty acids (SCFAs) in the gut were also increased (P<0.05). Finally, our results revealed correlations between gut microbiota, SCFAs, non-specific immunity and antioxidant enzymes in CYP-fed carp. These results suggest that CYP-supplemented feed could improve the immunity of the common carp by modulating the intestinal microflora and enhancing the gut defence barrier and has the potential to be used as an immunostimulating feed additive in aquaculture.

Chronic social defeat stress (CSDS) exacerbated the development of stress-related psychiatric disorders, and the social recognition dysfunction is the core feature of many psychiatric disorders. However, the effects of CSDS on female social recognition and the underlying neural mechanisms remain unclear. Using highly aggressive adult female mandarin voles (Microtus mandarinus) as animal model, the aim of this work is to investigate the effects of CSDS on social recognition in adult female rodents and the neurobiological mechanisms underlying these effects. Our results indicate the CSDS disrupted the normal social recognition in adult female voles. Meanwhile, defeated voles exhibited increased neural activity in the DG, CA1 and CA3 of the hippocampus. Furthermore, CSDS reduced levels of serotonin (5-HT) and serotonin 1A receptors (5-HT1AR) in the CA3. We also discovered that microinjection of 8-OH-DPAT into the CA3 effectively reversed the social recognition deficits induced by CSDS, and an infusion of WAY-100635 into the CA3 of control female voles impaired social recognition. Moreover, targeted activation of the 5-HT neuron projection from the DRN to CA3 by long-term administration of CNO significantly prevented the CSDS induced social recognition deficits. Taken together, our study demonstrated that CSDS induced social recognition deficits in adult female voles, and these effects were mediated by the action of 5-HT on the 5-HT1AR in the hippocampus CA3. The projection from the DRN to CA3 may be involved in social recognition deficits induced by CSDS.

This study investigated the effects of honeysuckle extract (Lonicera japonica, HE) on the growth performance and lipid metabolism of juvenile grass carp (Ctenopharyngodon idella). HE at doses of 10 g kg-1 (LHE), 20 g kg-1 (MHE), and 40 g kg-1 (HHE) were individually mixed with the basal diet and fed to grass carp for 10 weeks, and ginseng extract (20 g kg-1, GSE) was used as a positive control. The results showed that HE administration exerted no effect on growth performance, but the hepatosomatic index (HSI) and muscle and liver lipid contents were significantly decreased in the LHE and MHE groups. The serum levels of LDL-c, total triglyceride (TG) and total cholesterol (TC) also declined in the HE-treated groups. Moreover, the disordered vacuolization and nucleus migration in the liver were alleviated in the MHE and HHE groups, and mRNA expressions of lipogenesis-related genes, such as acc1, fas, srebp1, and pparγ decreased. Similarly, the expression of genes related to lipolysis, such as cpt1, atgl, lpl, and pparα, was found to be significantly increased in the MHE and HHE groups compared with the control. Taken together, HE can effectively improve the lipid metabolism and ameliorate the lipid deposition of grass carp and thus may be a promising feed additive in aquaculture.

BACKGROUND: Most previous studies have focused on the effects of social defeat in male juvenile individuals. Whether chronic social defeat stress in adulthood affects female emotion and the underlying mechanisms remains unclear. METHODS: Using highly aggressive adult female mandarin voles (Microtus mandarinus), the present study aimed to determine the effects of chronic social defeat stress on anxiety- and depression-like behaviors in adult female rodents and investigate the neurobiological mechanisms underlying these effects. RESULTS: Exposure of adult female voles to social defeat stress for 14 days reduced the time spent in the central area of the open field test and in the open arms of the elevated plus maze and lengthened the immobility time in the tail suspension and forced swimming tests, indicating increased anxiety- and depression-like behaviors. Meanwhile, defeated voles exhibited increased neural activity in the prelimbic cortex of the medial prefrontal cortex. Furthermore, chronic social defeat stress reduced serotonin projections and levels of serotonin 1A receptors in the medial prefrontal cortex-prelimbic cortex. Intra-prelimbic cortex microinjections of the serotonin 1A receptor agonist 8-OH-DPAT reversed the alterations in emotional behaviors, whereas injections of the serotonin 1A receptor antagonist WAY-100635 into the prelimbic cortex of control voles increased the levels of anxiety- and depression-like behaviors. CONCLUSIONS: Taken together, our results demonstrated that chronic social defeat stress increased anxiety- and depression-like behaviors in adult female voles, and these effects were mediated by the action of serotonin on the serotonin 1A receptors in the prelimbic cortex. The serotonin system may be a promising target to treat emotional disorders induced by chronic social defeat stress.

The present study was conducted to determine the effects of waterborne copper exposure on the lipid metabolism and intestinal microbiota of juvenile common carp (Cyprinus carpio L.). Common carp were exposed to four waterborne copper (Cu) concentrations (0 (control), 0.07 (low), 0.14 (medium), and 0.28 (high) mg Cu/L) for 8 weeks. Exposure to a high concentration of Cu had a negative effect on growth indices (weight gain rate (WGR) and specific growth rate (SGR)). The biochemical indices measured in serum (low-density lipoprotein (LDL) and triglycerides (TGs)) were significantly affected by exposure to medium concentration levels of Cu. The mRNA levels of lipogenic enzymes (acetyl-CoA carboxylase 1 (ACC-1) and fatty acid synthase (FAS)) and sterol-regulator element-binding protein-1 (SREBP-1) in liver tissue and tight binding protein genes (ZO-1 and occludin) in intestinal epithelial tissue were significantly downregulated in the 0.14 and 0.28 mg/L Cu treatment groups, accompanied by upregulated mRNA levels of lipolysis enzymes (lipoprotein lipase (LPL) and carnitine palmitoyl transferase 1 (CPT-1)) in the liver. The data also showed that the composition of intestinal microbiota was changed following Cu exposure and could alter the α-diversity and ß-diversity. The abundances of few putative short-chain fatty acid (SCFA)-producing bacteria, including Allobaculum, Blautia, Coprococcus, Faecalibacterium, Roseburia, and Ruminococcus, decreased significantly. More specifically, Roseburia sequences were positively associated with lipogenic enzymes, total protein (TP), and TGs and negatively associated with lipolysis enzymes. Other sequences related to probiotics (Lactobacillus, Bacillus and Akkermansia) were also found to decrease, accompanied by an increase in sequences related to pathogens (Pseudomonas and Acinetobacter). To the best of our knowledge, the present study provides the first evidence that waterborne, chronic Cu exposure can disturb the composition of intestinal microbiota related to lipid metabolism and immunity in freshwater fish, thereby increasing the risk of pathogen invasion.

Chronic social defeat affects many aspects of behavior. Most previous studies have focused on effects on males and defeat during adolescence. The extents to which chronic social defeat can impact female social behavior in adulthood and the neural mechanisms of such effects are poorly understood. Using highly social and aggressive female mandarin voles (Microtus mandarinus), the present study found that chronic social defeat reduced social preference in adult females, and that the defeated voles exhibited a high level of freeze, self-grooming and defensive behavior, as well as reduced exploration, intimacy and aggression during social interactions. Furthermore, chronic social defeat reduced levels of oxytocin (OT) and OT receptors (OTR) in the shell region of the nucleus accumbens (NACC). Intra-NACC shell OT microinjections reversed alterations in social behavior induced by chronic social defeat, whereas injections of an OTR antagonist (OTR-A) blocked the effects of OT. Taken together, our data demonstrate that chronic social defeat suppresses measures of sociability, and that these effects are mediated by the action of OT on the OTR in the NACC. NACC OT may be a promising target to treat socio-emotional disorders induced by chronic social stress.

Cellulose xanthogenates, derived from the straw of Eichhornia crassipes, were prepared as adsorbents for heavy metals by CS(2) sulfonation and magnesium substitution after degumming with alkali, self-isolated A(1) strain and pectase, respectively. The effects of three degumming treatments were compared by functional groups analysis, surface morphology and surface element composition and heavy metal (Pb(2+)) adsorption studies. The results demonstrate that bio-degumming treatments by A(1) strain and pectase have weaker degumming effects than alkali treatment. However, the surface characteristics of the bio-degumming products, especially the pectase degumming product, are more beneficial to heavy metal adsorption. In comparison to that of the raw plant materials, the Pb(2+) adsorption performances of the three xanthogenates improved significantly, although no obvious differences being observed among themselves. From an environmental point of view, the two bio-degumming treatments, especially the pectase degumming treatment, are more beneficial to prepare heavy metal adsorbents than the alkali degumming treatment.
