In this study, elastic metamaterial sandwich plates with axially deformed Timoshenko beam cores, considering both the out-of-plane and in-plane deformations of the face plates, are designed and the vibration band-gap properties are explored. The beam cores act as local resonators that can bear axial force, bending moment and shearing force. The finite element method (FEM) and the spectral element method (SEM) are combined to create the finite/spectral element hybrid method (FE-SEHM) for establishing the dynamic model and calculating the frequency response functions (FRFs) of the elastic metamaterial sandwich plate with axially deformed beam cores. It is observed that the metamaterial sandwich plate possesses both the axial and transverse vibration band-gaps of the beams, and the two kinds of band-gaps are independent. Compared with the metamaterial sandwich plates with rod cores, those with axially deformed beam cores have more extensive application ranges for vibration reduction.

Age-related macular degeneration is a common cause of blindless among the aged, which can mainly be attributed to oxidative stress and dysregulated autophagy in retinal pigment epithelium cells. Lactate was reported to act as a signaling molecule and exerted beneficial effect against oxidative stress. This study aims to investigate the protective effect of lactate against oxidative stress-induced retinal degeneration. Here, H2O2-induced oxidative stress cell model and sodium iodate-induced mice retinal degeneration model were established. It was found that H2O2 inhibited cell viability in ARPE-19 cells and sodium iodate induced deterioration of retinal pigment epithelium as well as apoptosis in retina. Pretreatment with lactate alleviated oxidative stress-induced cell death and retinal degeneration. Molecularly, lactate activated autophagy by up-regulating the ratio of LC3II/I, increased formation of LC3 puncta and autophagic vacuole. Further, lactate prevented H2O2-induced mitochondrial fission and maintained mitochondrial function by alleviating H2O2-induced mitochondrial membrane potential disruption and intracellular ROS generation. In contrast, application of 3-methyladenine, an inhibitor of autophagy, effectively weakened the protective effect of lactate against oxidative stress in vivo and in vitro. Taken together, all data in this study indicate that lactate protects against oxidative stress-induced retinal degeneration and preserves mitochondrial function by activating autophagy.

In this study, the effect of the type and content of functional groups on the interfacial shear properties of a functionalized graphene sheet (FGS)/thermoplastic polyurethane (TPU) nanocomposite are investigated by molecular dynamics (MD) simulations. The maximum pull-out force and separation energy were used to characterize the interfacial strength of the FGS/TPU nanocomposite in sliding mode. To find out how the type and content of functional groups affect the interfacial shear properties of the TPU/FGS system from an atomic view, the details of interactions between FGS and TPU were characterized. Based on the results, stronger interfacial shear properties of the TPU/FGS system can be achieved by adding the carboxyl group or hydroxyl group on the surface of graphene than that between TPU and FGS modified by the amine group or epoxy group, because of the strong interaction of electrostatic forces and H-bonds. In addition, interfacial shear properties can also be enhanced by increasing the content of functional groups modified on the surface of graphene.

In the 21st century, with the increasingly urgent requirements for lightweight in the fields of aviation, aerospace, and electronics, especially automobiles, many properties of magnesium alloy materials, especially the low-density performance characteristics, have been widely valued. In order to better use magnesium metal materials, it is very important to evaluate its mechanical properties. This article is based on 196 sets of mechanical performance experimental results and related data of AZ31 and AZ91 2 magnesium alloys. Based on data analysis and sorting, take deformation temperature, deformation rate, deformation coefficient, solid solution temperature, and solid solution time as input and take ultimate tensile strength (UTS), yield strength (YS), and elongation (ELO) as output. The 5-8-1 three-layer BP neural network forecast model optimized by the genetic algorithm is used for data training. The training results show that the prediction model can accurately predict the tensile strength, yield strength, and elongation. Compared with the general BP neural network prediction model, the BP neural network based on the genetic algorithm has small discreteness and high fitness: the average error of UTS and YS of AZ31 magnesium alloy is reduced to 0.88% and 3.3%, respectively. The most obvious is that the elongation of AZ31 ELO is reduced, and the error is reduced to 8.1%.

Cisplatin (DDP) is the first-line chemotherapeutic agent for ovarian cancer. However, the development of DDP resistance seriously influences the chemotherapeutic effect and prognosis of ovarian cancer. It was reported that DDP can directly impinge on the mitochondria and activate the intrinsic apoptotic pathway. Herein, the role of mitochondrial dynamics in DDP chemoresistance in human ovarian cancer SKOV3 cells was investigated. In DDP-resistant SKOV3/DDP cells, mitochondrial fission protein DRP1 was down-regulated, while mitochondrial fusion protein MFN2 was up-regulated. In accordance with the expression of DRP1 and MFN2, the average mitochondrial length was significantly increased in SKOV3/DDP cells. In DDP-sensitive parental SKOV3 cells, downregulation of DRP1 and upregulation of mitochondrial fusion proteins including MFN1,2 and OPA1 occurred at day 2~6 under cisplatin stress. Knockdown of DRP1 or overexpression of MFN2 promoted the resistance of SKOV3 cells to cisplatin. Intriguingly, weaker migration capability and lower ATP level were detected in SKOV3/DDP cells. Respective knockdown of DRP1 in parental SKOV3 cells or MFN2 in SKOV3/DDP cells using siRNA efficiently reversed mitochondrial dynamics, migration capability and ATP level. Moreover, MFN2 siRNA significantly aggravated the DDP-induced ROS production, mitochondrial membrane potential disruption, expression of pro-apoptotic protein BAX and Cleaved Caspase-3/9 in SKOV3/DDP cells. In contrast, DRP1 siRNA alleviated DDP-induced ROS production, mitochondrial membrane potential disruption, expression of pro-apoptotic protein BAX and Cleaved Caspase-3/9 in SKOV3 cells. Thus, these results indicate that mitochondrial dynamics mediated by DRP1 and MFN2 contributes to the development of DDP resistance in ovarian cancer cells, and will also provide a new strategy to prevent chemoresistance in ovarian cancer by targeting mitochondrial dynamics.

Flavonoids are key components of licorice plant that directly affect its medicinal quality. Importantly, the MYB family of transcription factors serves to regulate the synthesis of flavonoids in plants. The MYB transcription factors represent one of the largest families of transcription factors in plants and play important roles in the process of plant growth and development. MYB gene expression is induced by a number of plant hormones, including the lipid-based hormone jasmonate (JA). Methyl jasmonate (MeJA) is an endogenous plant growth regulator that can induce the JA signaling pathway, which functions to regulate the synthesis of secondary metabolites, including flavonoids. In this study, MeJA was added to licorice cell suspensions, and RNA-seq analysis was performed to identify the differentially expressed genes. As a result, the MYB transcription factors GlMYB4 and GlMYB88 were demonstrated to respond significantly to MeJA induction. Subsequently, the GlMYB4 and GlMYB88 protein were shown to localize to the cell nucleus, and it was verified that GlMYB4 and GlMYB88 could positively regulate the synthesis of flavonoids in licorice cells. Overall, this research helps illustrate the molecular regulation of licorice flavonoid biosynthesis induced by MeJA.
