RESUMO

Eucalyptus is the most intensively managed tree genus in the world. Different factors, including damage by insect pests, affect its growth and productivity. Among these pests is Glycaspis brimblecombei Moore (Hemiptera: Aphalaridae), an exotic insect of Australian origin. The evolutionary success of this insect depends on symbiotic associations with microorganisms. The influence of these microorganisms on insect pests and their natural enemies is important for integrated management tactics. Within this context, this work aimed to detect Arsenophonus in populations of G. brimblecombei in Brazil. Eucalyptus branches infested with G. brimblecombei nymphs were collected in commercial eucalyptus plantations in six Brazilian states. Specimens of this pest were sampled soon after emergence and frozen for molecular analysis. The genomic DNA of G. brimblecombei adults from each population was extracted and used to detect the endosymbiont Arsenophonus by polymerase chain reaction (PCR) employing specific primers that target its 23 S rRNA gene. This endosymbiont was identified in all of the studied G. brimblecombei populations. This is the first report on the association between Arsenophonus and G. brimblecombei in Brazil.

INTRODUCTION: In this essay, the following themes will be analyzed: Systemic Family Therapy (SFT), sexuality and gender studies, women, and intersectionality. We expose how these themes combine in the family experiences of people who are on the other side or the margins of cisheteronormativity and discuss the diversity that makes up the family, beyond a restrictive view of this term. OBJECTIVE: Make a constructive criticism of TFS, pointing out the erasure that this discipline has in relation to the experiences of LGBTQIAP+ people (lesbians, gays, transsexuals and transvestites, queer, intersex, asexual, pansexual, and other sexual orientations and gender identities). We propose intersectionality as a strategy for analyzing life stories to understand the family arrangements of lesbian, bisexual and pansexuals women. METHODOLOGY: This essay is based on empirical material from biographical interviews/life trajectories. We propose here intersectionality as a strategy for analyzing life stories to understand the family arrangements of lesbian, bisexual and pansexual women. RESULTS: The interviewees demonstrated multiple realities, based on both experiences of violence and acceptance in their families of origin. And focusing their journeys on building supportive bonds with their chosen families, creating sisterhood relationships between friends. CONCLUSIONS: TFS could broaden its view and writing about the LGBTQIAP+ population, using intersectionality as a powerful guide for analyzing family relationships. Since these women's life stories focus on dealing with discrimination and challenges associated with their various social markers of difference.

This work reports the quantification of total mercury in sediments collected in periods with and without rain from the Joanes River, Bahia, Brazil. Determinations were made using Direct Mercury Analysis (DMA), the accuracy of which was confirmed with two certified reference materials. The highest total mercury concentrations were found at the sampling point close to commercial areas and large residential condominiums. On the other hand, the lowest levels were found in the site close to a mangrove region. The geoaccumulation index was applied to the total mercury results, evidencing low contamination in the region studied. The contamination factor showed that of the seven stations investigated, four samples collected in the rainy season showed moderate contamination. The results of the ecological risk assessment were utterly consistent with the contamination factor data. This study showed that the smaller sediment particles concentrate more mercury, corroborating what has been predicted by the adsorption processes.

RESUMO

The parasitoid Psyllaephagus bliteus Riek (Hymenoptera: Encyrtidae), entomopathogenic fungi, and chemical insecticides are the main strategies to manage the eucalypts pest Glycaspis brimblecombei Moore. The objective of this study was to isolate and to identify entomopathogenic fungi, collected from Bemisia tabaci Gennadius adults in soybean and tomato crops, and from soil samples in eucalypts, soybean, and native forest areas to evaluate their potential to manage G. brimblecombei. Twelve Beauveria and Cordyceps isolates were selected and compared with the commercial products Boveril Beauveria bassiana Bals. (Hypocreales: Cordycipitaceae), Metarril Metarhizium anisopliae Metschn. (Hypocreales: Clavicipitaceae), and Octane Cordyceps fumosorosea Wize (Hypocreales: Cordycipitaceae) and their respective strains. The fungal isolates were diluted in 0.1% aqueous Tween 80 at a concentration of 1.0 × 108 conidia/ml and sprayed on the G. brimblecombei nymphs with or without lerps. Pest mortality was higher and the TL50 and TL90 lower with the isolates LCBPF 11 C. javanica Frieder. & Bally (Hypocreales: Cordycipitaceae), LCBPF 12 (C. fumosorosea), and LCBPF 67 (C. fumosorosea) from B. tabaci adults. Fungi of the genera Beauveria and Cordyceps developed and caused high mortality of G. brimblecombei nymphs with lerps. The B. bassiana, C. cateniannulata Liang (Hypocreales: Cordycipitaceae), C. fumosorosea, C. javanica, and M. anisopliae isolates showed potential to manage G. brimblecombei. The lerp of this insect enhances entomopathogenic fungus development as a source of inoculum accelerating G. brimblecombei nymph mortality. Entomopathogenic fungi isolated from insects and soils are effective against G. brimblecombei and the presence of the lerp of this insect increases the effectiveness of its control.

RESUMO

Introduction: COVID-19 has greatly affected society by limiting the functioning of sectors of the economy and public services. Considering the essential character of many of these services, especially public security, it is necessary to understand how the disease has affected different groups within the population so that public policies for facing this problem can be implemented. Objectives: To identify and describe the profile of military police officers affected by COVID-19. Methods: This is a descriptive observational study with a quantitative approach, based on secondary data. The electronic medical records of 737 military police officers affected by COVID-19 were accessed; sociodemographic, biological, and professional data were collected, as well as data on disease progression. Data were analyzed using Bioestat® software, v5.3. Results: The peak of the COVID-19 contagion curve happened first among military police officers of the state of Alagoas than in the general population, and a positive effect of social distancing was observed in the containment of disease spread. Moreover, specialized operations units had a higher contagion rate in view of the higher level of exposure linked to their work activities. Conclusions: This study described the profile of military police officers affected by COVID-19, which can substantiate the adoption of public policies and new strategies to fight this disease among officers in Alagoas, thus ensuring the continuity of the service provided to society.

RESUMO

Aquatic ecosystems provide habitats for many organisms. Historically, riverbanks have always been inhabited and exploited for subsistence and navigation. The present study evaluates the contamination and ecological risks caused by potentially toxic elements in surface sediments of the Paraguaçu River, Bahia, Brazil. Seven sediments samples were collected, and eight heavy metals were determined employing inductively coupled plasma spectrometry mass (ICP-MS). The concentrations range as (mg kg-1) found were 6.78-18.68 for lead, 14.21-42.16 for zinc, 27.61-48.63 for nickel, 2.03-6.50 for chromium, 6.06-12.90 for vanadium, 5.99-13.33 for cupper, 1.25-3.19 for cobalt, and 79.52-286.08 for manganese. Nickel showed significant enrichment (EF: 5.75; 7.62, and 14.11), followed by zinc, which showed moderate enrichment (EF: 2.16; 2.19, and 4.52). These enrichment levels are possible of anthropogenic origin. When the pollution index (PI) was evaluated, the elements V, Ni, Zn, Mn, Co, and Cu were strongly polluted (PI ≥3). In general, the pollution index (PI), geoaccumulation index (Igeo), enrichment factor (EF), and potential ecological risk indices (Er and PERI) show that contaminated sediments have adverse effects on aquatic environments, especially for o Mn, Ni, Pb, V, and Zn.

RESUMO

The maintenance of SARS-Cov-2 RNA samples poses a new challenge for laboratories and researchers. In addition, it is a requirement in order to identify what strain of the new coronavirus is predominant in a region, for instance. Therefore, it is a must to keep the quality and viability of stored RNA to respond to this and other valid questions. In other to test the quality of our samples and storing protocols, we randomly checked RNA samples four different times over one year using a second RT-PCR assay after the first test. The virus genes, N1 and N2, showed no significant increase in the media of the CT value between the first assay and subsequent times with p > 0.05. However, the human RP gene showed differences in the first three times analyzed, but within the acceptable sample cut-off, according to the test manufacturer. After one year, the RNA extracted from human nasopharyngeal specimens are viable to detect the virus SARS-CoV-2 genes with minor changes.

RESUMO

BACKGROUND: In a study recently published by our research group, the isoxazoline-acylhydrazone derivatives R-99 and R-123 presented promising antinociceptive activity. However, the mechanism of action of this compound is still unknown. OBJECTIVE: This study aimed to assess the mechanisms involved in the antinociceptive activity of these compounds in chemical models of pain. METHODS: Animals were orally pretreated and evaluated in the acetic acid-, formalin-, capsaicin-, carrageenan- and Complete Freund's Adjuvant (CFA)-induced pain models in mice. The effects of the compounds after pretreatment with naloxone, prazosin, yohimbine, atropine, L-arginine, or glibenclamide were studied, using the acetic acid-induced writhing test to verify the possible involvement of opioid, α1-adrenergic, α2-adrenergic or cholinergic receptors, and nitric oxide or potassium channels pathways, respectively. RESULTS: R-99 and R-123 compounds showed significant antinociceptive activity on pain models induced by acetic acid, formalin, and capsaicin. Both compounds decreased the mechanical hyperalgesia induced by carrageenan or CFA in mice. The antinociceptive effects of R-99 and R-123 on the acetic acid-induced writhing test were significantly attenuated by pretreatment with naloxone, yohimbine or atropine. R-99 also showed an attenuated response after pretreatment with atropine and glibenclamide. However, on the pretreatment with prazosin, there was no change in the animals' response to both compounds. CONCLUSION: R-99 and R-123 showed antinociceptive effects related to mechanisms that involve, at least in part, interaction with the opioid and adrenergic systems and TRPV1 pathways. The compound R-99 also interacts with the cholinergic pathways and potassium channels.

RESUMO

Coronavirus disease 2019 (Covid-19) is an emerging novel respiratory infectious disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) that rapidly spread worldwide. In addition to lung injury, Covid-19 patients may develop extrapulmonary symptoms, including cardiac, liver, kidney, digestive tract, and neurological injuries. Angiotensin converting enzyme 2 is the major receptor for the entry of SARS-CoV-2 into host cells. The specific mechanisms that lead to cell death in different tissues during infection by SARS-CoV-2 remains unknown. Based on data of the previous human coronavirus SARS-CoV together with information about SARS-CoV-2, this review provides a summary of the mechanisms involved in cell death, including apoptosis, autophagy, and necrosis, provoked by severe acute respiratory syndrome coronavirus.

RESUMO

The Joanes River is located in the northeast of Brazil, crosses the Camaçari Petrochemical Complex, the largest integrated industrial complex in the Southern Hemisphere, which has over 90 companies in the chemical and petrochemical industry. The present study aims to evaluate spatial distribution, seasonal variation and identify possible sources of trace metal contamination in surface water samples of the Joanes River. Samples were collected in the dry (December 2018) and rainy (August 2019) seasons. Analysis of trace elements (As, Cd, Cr, Cu, Ni, Mn and Pb) were performed by ICP-MS. A total of 60 water samples were obtained. Samples were analyzed using exploratory techniques such as principal component analysis (PCA) and cluster analysis (CA). It was possible to characterize the samples according to the seasonal variation. The formation of two groups was observed. Among these, samples from the rainy season presented the higher levels of metals in relation to the samples of the dry season. Natural and anthropic sources of metal contamination were identified using CA. Similarity was shown in the relationship between the metals As-Pb and Ni-Cd-Cr-Cu in the dry season, and Cd-Ni and Pb-As-Cr-Cu in the rainy season. Dermal absorption (HQderm) and ingestion hazard quotients (HQing) routes exhibited values of less than one for all the elements analyzed for adults and children, in both rainy and dry seasons. This indicated that the pollutants analyzed posed little or no health risk over a lifetime of exposure. According to international guidelines (US EPA), the values of Cu, Pb and Cr were above the limit established.

RESUMO

Microsporidia are naturally occurring fungal-related parasites that can infect nearly all animal hosts, but their biocontrol potential of insect pests is routinely overlooked in agriculture and forestry. This research brings the first report describing the natural occurrence of a microsporidium causing disease in field-collected populations of the invasive eucalyptus snout beetle, Gonipterus platensis (Coleoptera: Curculionidae), a major destructive pest of eucalyptus plantations in Brazil. Adult beetles were collected during field surveys in commercial eucalyptus plantations in southern Brazil to be examined and dissected with typical symptoms to verify presence of microsporidian spores in haemolymph. From 14 plantations in different sites, the natural infection occurrence in these populations ranged from 0 to 65%, while a lab colony exhibited an infection incidence of 70%. Spore density in haemolymph of symptomatic insects averaged 2.1 (± 0.4) × 107 spores/beetle. Symptoms in infected adults were identified by an abnormal abdomen with malformation of the second pair of wings, impairing their flight activity. Electron transmission microscopy of the pathogen showed morphological features similar to species belonging to the genus Nosema or Vairimorpha. Phylogenetic analysis of the full-length small subunit ribosomal RNA gene suggests this pathogen's placement in the genus Vairimorpha, but with a sequence identity of ~ 94% with the nearest neighbours. The low level of sequence identity suggests this pathogen may represent a novel taxon in the genus and further requires whole genome sequencing for definitive taxonomic resolution. These findings provide insights on the natural occurrence of this novel pathogen of this invasive pest in Eucalyptus plantations in Brazil. Further studies are needed to determine potential of this microsporidium in the design of conservative or augmentative biological control programs for this invasive pest.

RESUMO

The Mayaro virus (MAYV) is an arbovirus that circulates mainly in tropical forests or rural areas in Latin America and is transmitted mainly by Haemagogus mosquitoes. The objective of this study was to evaluate the vector competence, microbiome, and the presence of Wolbachia in three Aedes albopictus populations infected with MAYV. The vector competence was assessed based on viral infection and transmission by RT-qPCR. In addition, the microbiome was evaluated by amplification of the 16S rRNA V4 region and PCR to detect the presence of Wolbachia (strain wAlbA/wAlbB). Our results show that all three populations were susceptible to MAYV infection. The potential transmission of the MAYV was consistent in all populations of naïve mosquitoes injected (more than 50%). The microbiome analysis revealed 118 OTUs (operational taxonomic unit) from the three populations, 8 phyla, 15 classes, 26 orders, 35 families, 65 genera, and 53 species. All populations had Pseudomonas and Wolbachia as predominant genera. There was no difference between the variables for MAYV and Wolbachia (wAlbA or wAlbB) in the abdomen. However, in the head + thorax samples at 14 dpi, there was a difference between the two populations, indicating a possible correlation between the presence of Wolbachia (wAlbB) and infection. Overall, we show evidence that Ae. albopictus displays significant infection and transmission competence for the MAYV in the laboratory, and its bacterial microbiota play an important role in the host, mainly the strains of Wolbachia. The influence of the intestinal microbiota of Ae. albopictus is poorly known, and a better understanding of these interactions would open new perspectives for disease control through the manipulation of microbial communities. The exact contribution of this mosquito species to the transmission of the MAYV in the field remains to be confirmed.

RESUMO

The genomes of two putative new RNA viruses (macula-like virus and bunya-like virus) were identified in total RNA extracted from dead eucalyptus snout beetles (Gonipterus spp.) from a laboratory colony. However, only bunya-like virus was detected in field-collected insects. The macula-like virus has a monopartite single-stranded RNA genome that contains three open reading frames (ORFs) encoding an RNA-dependent RNA polymerase (RdRp), a capsid protein (CP), protein with unknown function. The bunya-like virus genome was predicted to consist of two RNA segments: a large segment (L) encoding a single protein (RdRp) and a small segment (S) encoding a putative nucleocapsid protein.

RESUMO

Entomopathogenic nematodes (EPNs) can control pests due to mutualistic association with bacteria that reproduce and kill the host from septicemia, making the environment favourable for nematode development and reproduction. The objective of this study was to identify an EPN isolate collected in eucalyptus cultivation and to determine its pathogenicity with regard to Gonipterus platensis Marelli (Coleoptera: Curculionidae). Four steel-mesh traps with two seventh-instar Galleria mellonella larvae were buried 5 cm deep in the soil in a commercial Eucalyptus plantation. After 7 days, the traps were packed in plastic bags and transported to laboratory to isolate the EPNs using White traps. The obtained nematodes were multiplied in G. mellonella larvae and identified by sequencing their D2/D3 expansion of the 28S rDNA region by polymerase chain reaction (PCR) and specific primers for ITS regions. Steinernema diaprepesi was identified and inoculated into G. platensis pupae at doses of 500, 1000 and 5000 infective juveniles (IJs) to determine its pathogenicity to this pest. At 8 days after inoculation, the mortality rate of the G. platensis pupae was 80% with the lowest concentration and 100% with the others. The emergence of nematodes and the rapid degradation of G. platensis pupae were observed in those inoculated with IJs. The pathogenicity to the G. platensis pupae indicates potential for using this nematode in the integrated management of this insect.

RESUMO

Ten surface sediments collected from Joanes River, Bahia, Brazil in rainy and drought periods in 2019 were evaluated according to the enrichment factor (EF), potential ecological risk index (RI), potential contamination index (PCI), pollution load index (PLI), and index of geoaccumulation (Igeo). Initially the dry sediment was subjected to granulometric analysis and determination of the concentration of organic matter. Then, the samples were digested in HNO3 and analyzed by Inductively Coupled Plasma Optical Emission Spectrometry (ICP OES) to determine the metals cadmium (Cd), copper (Cu), chromium (Cr), nickel (Ni), lead (Pb) and zinc (Zn). Zn and Cu were classified in that order as the most contaminated elements in most sediment. Comparison of the total metal concentrations with the threshold (TELs) and probable (PELs) effect levels in sediment quality guidelines suggested a more worrisome situation for Zn (648.83-1415.90 µg g-1; PELZn = 315 µg g-1), of which concentrations were occasionally associated with adverse biological effects in four sediments, followed by Cu in five sediments during dry and rainy periods; while adverse effects were rarely associated with Cd, Cr, Ni, and Pb. In another evaluation, Cd, Cu, Cr, and Zn could be considered the most dangerous in the entire river, as they were classified in the high levels of contamination by the PCI, associated with serious adverse effects in most samples. In an assessment regarding the ecological risks in the study environment, the sediment samples remained below the limit established by the risk index (IR). The Zn presented moderately severe enrichment (6.78-11.83) in all the collection stations in the dry and rainy periods, followed by the Cd that presented moderate enrichment (2.23-4.17), whose values exceeded almost 1000 times the background at one site. Through the PCA it was possible to evidence the existing correlation between metals, organic matter, and silt and clay fraction. The results obtained in the PCA represented more than 80% of the variance between the data. The environmental risk assessment revealed a significant increase in the risk associated with metals during the rainy season. This is probably due to the greater supply of organic matter from the leaching of the margins.

RESUMO

BACKGROUND: Consuming foods with added fiber may help older adults achieve fiber recommendations; however, many high-fiber ingredients have little effect on laxation and may contribute to unpleasant gastrointestinal side effects. OBJECTIVES: The aim of the study was to determine the effects of consuming snacks fortified with pea hull fiber (PHF) on stool frequency and form, gastrointestinal symptoms, and appetite in older adults. An exploratory aim was to determine if PHF altered the microbiota profile. METHODS: A 10-wk, randomized, blinded, crossover study was carried out. Following a 2-wk baseline period, participants [aged (mean ± SD) 69.7 ± 6.5 y; n = 31; 14 men, 17 women] consumed snacks providing 10 g/d of PHF or a control, each for 2-wk periods followed by 2-wk washouts. Participants used the Bristol Stool Form Scale (BSFS) to record daily stool frequency and gastrointestinal symptoms, and completed the Gastrointestinal Symptom Rating Scale (GSRS) and Simplified Nutritional Appetite Questionnaire (SNAQ) biweekly. One stool was collected per period for 16S ribosomal RNA high-throughput amplicon sequencing of the fecal microbiota profile. RESULTS: Participants reported 1.63 ± 0.05 stools/d and 76.6% normal transit stool form at baseline and no change with PHF. GSRS syndrome scores were similarly unchanged. Daily abdominal noises and bloating were higher for PHF versus control, and flatulence was higher for PHF versus baseline, suggesting fermentation in some individuals. There was no evidence to suggest a common PHF-induced microbiome response for the group as a whole; however, a subgroup of participants (n = 7) who responded with increased flatulence (fermenters), harbored many different taxa than nonfermenters, and demonstrated lower abundance of Clostridiales with PHF. Appetite was unchanged with PHF. CONCLUSIONS: PHF did not modulate stool form or frequency in older adults with normal bowel habits. Because snacks fortified with PHF did not suppress appetite, PHF may be an appropriate fiber source for older adults at nutritional risk. Microbiome profile may be predictive of gastrointestinal symptom response to PHF. This trial was registered at www.clinicaltrials.gov as NCT02778230.

RESUMO

BACKGROUND: Lower limb edema has both systemic and local causes. Using software to differentiate the origin of edema in ultrasound images is an innovation. OBJECTIVE: To determine the parameters for using software to differentiate edema of venous and/or lymphatic origin in ultrasound images of the lower limbs. METHOD: This is a cross-sectional, quantitative, analytical study with non-probabilistic sampling by convenience. Data were collected by patient interview, physical examination, ultrasound examination, and analysis of software for tissue characterization in ultrasound image by means of quantification of echogenicity and Gray Scale Median (GSM). RESULTS: The sample comprised 42 lower limbs with venous edema, 35 with lymphatic edema, 14 with mixed edema, and 11 control limbs. The distributions of pixels in echogenicity intervals by group was as follows. In the venous edema group, 88.31% were distributed from hypoechogenic interval IV to echogenic interval III; in the lymphatic edema group 71.73% were from hypoechogenic interval II to echogenic interval I; in the mixed edema group 76.17% were from hypoechogenic interval III to echogenic interval II; and in the control group 84.87% were distributed from echogenic interval II to hyperechogenic interval I. Mean and standard deviation of GSM values showed statistical differences between groups. CONCLUSION: The CATUS software enabled differentiation of the type of lower limb edema, facilitating diagnosis of edema type and, consequently, choice of the best therapeutic option.

RESUMO

This manuscript reports an evaluation of the effects of simple chemical-heat treatments on the deposition of different ceramic coatings, i.e., TiO2, CaTiO3 and CaP, on commercially pure titanium (cp-Ti) and Ti6Al4V and the influence of the coatings on cells interaction with the surfaces. The ceramic materials were prepared by the sol-gel method and the coating adhesion was analyzed by pull-off bending tests. The wettability of positively or negatively charged surfaces was characterized by contact angle measurements, which also enabled the calculation of the surface free energy through the polar-apolar liquids approach. Both acid and alkaline treatments activated the cp-Ti, whereas Ti6Al4V was only activated by the alkaline treatment. Such treatment led to increased hydrophilicity with inhibition of the fibroblastic response on Ti6Al4V. On the other hand, osteoblastic cells adhered to and proliferated on the positively and negatively charged surfaces. The maximum adhesion strength (~ 3400 N) was obtained with a negative Ti6Al4V-CaTiO3-CaP multilayer surface.
