The aim of the study was to determine the effect of carbetocin administration (a long-acting analog of oxytocin) 20 or 10 min before electroejaculation (EE) on the duration of semen collection procedure, quantitative and qualitative characteristics of the ejaculate, and stress biomarkers in rams. Semen was collected from 12 Corriedale rams (age, 2.5-5.5 years old) with EE, in a Latin-square design, administrating carbetocin (0.2 mg/100 kg of body weight i.v.) 20 or 10 min before EE, or without carbetocin administration (CB-20, CB-10, and CON treatments, respectively). Each treatment was applied to different rams every 3-4 days, allowing all the rams to receive all three treatments. Carbetocin administered 20 or 10 min before EE increased the number of sperm ejaculated (P = 0.01), the semen concentration (P = 0.02), the number of insemination doses collected in a single collection (P = 0.01), and the number of insemination doses collected/electrical pulses administered (P = 0.05) compared to control rams. Carbetocin administered 20 or 10 min before semen collection prolonged the time required for EE and the number of pulses administered during EE compared to CON rams (P < 0.03 for both). The CB-10 rams required the administration of more electrical pulses during ejaculation than CON rams (P = 0.001), and CB-20 treatment tended to require more electrical pulses than CON rams (P = 0.06). The volume of the ejaculate was greater in CB-10 than in CON rams (P = 0.01), and that of CB-20 treatment tended to be greater than CON rams (P = 0.08). The percentage of sperm with intact membrane was greater in CB-20 than in CON rams (P = 0.01). Total protein, albumin, and globulin concentrations were lower immediately after carbetocin administration 20 or 10 min before EE. The treatments did not affect cortisol concentration, glycemia, rectal and surface temperatures, heart rate, and facial expressions. Carbetocin administration before EE of rams improved the quantitative and qualitative characteristics of the ejaculate, duplicating the number of insemination doses collected. It can be a promising treatment to obtain a greater quantity of doses to inseminate with a lower frequency of semen collections, reducing the negative impacts of EE on animal welfare.

High circulating progesterone concentrations decrease the responses to stressful situations in farm ruminants. We hypothesized that administering a single dose of long-acting progesterone formulation reduces the stress response of anestrous ewes to weaning. The aim of this study was to compare the behavioral, cortisol, and blood protein responses to weaning of anestrous ewes treated or not, with a single dose of a long-acting progesterone formulation. A complementary aim was to characterize the bioavailability of progesterone after this treatment. Thirty-six multiparous single-lambing Corriedale ewes and their lambs were used in this study. The lambs were weaned at 6:30 h when they were 65 d old (d0), and were taken to another paddock without chemical, visual or auditory contact with their mothers, while the ewes remained in the same paddock. Immediately before weaning, while 19 ewes received oil based long-acting progesterone formulation, other 17 ewes remained as controls, receiving sunflower oil. Twelve ewes from each group were used for behavioral recordings, the other 5 untreated ewes and 7 treated ewes were used to characterize the cortisol response and the progesterone profile. Behaviors were recorded every 10th min for 3 h in the morning and 3 h in the afternoon on day minus 3, d0 and d2, and expressed as percentage of recordings each animal displayed each behavior on each day. Treated ewes were recorded more times standing up than ewes on the day of weaning (P < 0.05), but the reverse result was found 2 d after (P < 0.01). Treated ewes were recorded more times lying down than control ewes on d2 after weaning (P < 0.0001), when treated ewes were observed fewer times walking than control ewes (P = 0.0004), as also happened the day of weaning (P < 0.0001). On the day of weaning, treated ewes grazed and paced less than control ewes (P = 0.001 and P = 0.009). Treated ewes ruminated more than control ewes on the day of weaning (P = 0.02) and 2 d later (P = 0.0002). Control ewes also secreted more cortisol than progesterone-treated ewes (P = 0.03), with no effect of the treatment on the concentration of total proteins, albumin or globulins. While control ewes never had luteal progesterone concentrations (>0.5 ng/mL), treated ewes achieved luteal progesterone concentrations 2 h after its administration, and remained well above luteal levels until 15 h after administration. The administration of a single dose of long-acting progesterone formulation appears to be a viable choice in reducing the stress to weaning in ewes. Ewes treated with progesterone coped better with weaning, returning to their normal basal behavior earlier than untreated ones, and secreting lower cortisol levels.

We report on a new, to the best of our knowledge, type of optical memory that allows for the amplification of the optical signal carrying the stored information during its reading process. The memory mechanism is demonstrated in an ensemble of cold cesium atoms and is based on the multiple parametric four-wave mixing exploring the external atomic degrees of freedom via recoil-induced resonances. We have particularly demonstrated the storage of light carrying orbital angular momentum with a fourfold amplifying factor for the retrieved signal during the reading process. Memory lifetimes of the order of hundreds of microseconds have been measured, and possible applications for this self-amplifying memory are discussed.

Modern lifestyle increases the prevalence of obesity and its co-morbidities in the young population. High-salt (HS) diets are associated with hypertension and cardiac remodelling. The present study evaluated the potential effects of cardiometabolic programming induced by HS intake during puberty in lean and obese rats. Additionally, we investigated whether HS could exacerbate the impairment of cardiovascular parameters in adult life due to postnatal early overnutrition (PO). At postnatal day 3 (PN3), twenty-four litters of Wistar rats were divided into two groups: normal litter (NL, nine pups/dam) and small litter (SL, three pups/dam) throughout the lactation period; weaning was at PN21. At PN30, the pups were subdivided into two more groups: NL plus HS (NLHS) and SL plus HS (SLHS). HS intake was from PN30 until PN60. Cardiovascular parameters were evaluated at PN120. SL rats became overweight at adulthood due to persistent hyperphagia; however, HS exposure during puberty reduced the weight gain and food intake of NLHS and SLHS. Both HS and obesity raised the blood pressure, impaired baro- and chemoreflex sensitivity and induced cardiac remodelling but no worsening was observed in the association of these factors, except a little reduction in the angiotensin type-2 receptor in the hearts from SLHS animals. Our results suggest that the response of newborn offspring to PO and juveniles to a HS diet leads to significant changes in cardiovascular parameters in adult rats. This damage may be accompanied by impairment of both angiotensin signalling and antioxidant defence in the heart.

BACKGROUND: Yeasts of the Candida genus are one of the most common causes of bloodstream infections associated with high rates of morbidity and mortality, mainly affecting immunocompromised patients. We aimed to identify yeasts obtained from blood cultures of patients interned at tertiary hospitals in Brazil. METHODS: We evaluated some of the major virulence factors of Candida spp., including the ability to adhere to human buccal epithelial cells, biofilm formation, hemolytic and phospholipase activity. RESULTS: We analyzed 70 isolates of Candida spp. obtained from March 2011 and March 2015. Candida spp. showed different peculiarities in terms of expression of virulence factors evaluated in vitro. C. albicans strains were more adherent to HBEC than all the other Candida species. C. tropicalis strains were considered strong biofilm producers. Strains belonging to the C. parapsilosis species complex were able to produce hemolysins, while C. glabrata was also able to lyse erythrocytes and to produce phospholipase. CONCLUSION: These results suggest that Non-Candida albicans Candida species are also able to express virulence factors which play an important role in bloodstream infectious caused by these yeasts.

Electroejaculation (EE) is stressful and probably painful; thus the administration of anaesthesia is recommended to decrease those negative effects. However, anaesthesia has a direct risk of provoking death, but sedation is less risky than anaesthesia. At the same time, α2-adrenergic agonists may improve semen quality. Therefore, the aim of the present study was to compare the physiological and behavioural responses indicative of stress and possibly pain, and the semen quality in electroejaculated untreated, anaesthetised or sedated goat bucks. Semen was collected from eight bucks using three different procedures in all them (EE in untreated bucks, EE under sedation or EE under general anaesthesia). The number of vocalizations during EE and the behavioural pattern before and after procedures were recorded. Pain visual analogue scale (VAS) score was also determined during EE. Rectal temperature, heart rate, serum cortisol concentration, biochemical and haematological parameters were measured before and after each procedure, and sperm characteristics were determined. Bucks vocalised more often when untreated than sedated or anaesthetised (P<0.02). The pain VAS score was greater when bucks were untreated than sedated or anaesthetised (P<0.002). The rectal temperature, heart rate, total protein, albumin and haemoglobin concentrations were greater when bucks were untreated than anaesthetised or sedated (P<0.02). Serum cortisol increased after EE (P=0.0006), without differences between procedures. The frequency and duration of lying down after EE were greater when bucks were anaesthetised than sedated or untreated (P<0.05), and were also greater when bucks were sedated than untreated (P<0.05). The number of times that the animal tried to stand up after EE was greater when bucks were anaesthetised than sedated or untreated (P<0.02). The sperm mass motility was greater when bucks were anaesthetised or sedated than when they were untreated (P=0.048). When animals were sedated, the ejaculate contained more spermatozoa with functional plasma membrane (P=0.03) and morphologically normal (P=0.05) than when they were untreated. In conclusion, general anaesthesia and sedation decreased the stress and probably the pain response provoked by EE and especially sedation improved the quality of the semen collected.

In this paper, we report the development of a novel technique in which the magnetoresistance of nanostructures is perturbatively measured by transversally modulating the DC magnetic field. It measures the electrical transport counterpart of the transverse magnetic AC-susceptibility. The technique was developed in a conventional four-probe configuration in which a small DC current flows through the sample and a small transverse AC-field perturbates the equilibrium position of the sample magnetization. Lock-in detection, in-phase with the AC-perturbation, is used to measure the voltage signal between the two inner electrodes of the four-probe station. We successfully demonstrated that this signal is proportional to the product of the first derivative of sample resistance with respect to the equilibrium position of the magnetization times the second derivative of the energy with respect to the equilibrium position of the magnetization. These dependences add new features to the technique investigated here that were not captured by the investigations previously reported on the same subject. To show the effective use of the technique, we discuss its application in measuring magnetic properties of thin magnetic films in the non-saturated regime.

Cervical cancer is a public health problem and the molecular mechanisms underlying radioresistance are still poorly understood. Here, we evaluated the modulation of key molecules involved in cell proliferation, cell cycle and DNA repair in cervical cancer cell lines (CASKI and C33A) and in malignant tissues biopsied from 10 patients before and after radiotherapy. The expression patterns of epidermal growth factor receptor (EGFR), excision repair cross-complementation group 1 (ERCC1) and p53 were evaluated in cancer cell lines by quantitative PCR and western blotting, and in human malignant tissues by immunohistochemistry. The mutation status of TP53 gene was evaluated by direct sequencing. Among cell lines, absent or weak modulations of EGFR, ERCC1 and p53 were observed after exposure to 1.8 Gy. Conversely, increased expressions of p53 (5/10 patients; P=0.0239), ERCC1 (5/10 patients; P=0.0294) and EGFR (4/10 patients; P=0.1773) were observed in malignant tissues after radiotherapy with the same radiation dose. TP53 mutations were found only in one patient. Here we show that a single dose of radiotherapy induced EGFR, ERCC1 and p53 expression in malignant tissues from cervical cancer patients but not in cancer cell lines, highlighting the gap between in vitro and in vivo experimental models. Studies on larger patient cohorts are needed to allow an interpretation that an upregulation of p53, EGFR and ERCC1 may be part of a radioresistance mechanism.

In order to better understand the pathogenesis of VVC, focusing on the role of C. albicans virulence factors in triggering this infirmity; we evaluated four virulence factors of 62 clinical isolates of C. albicans sequentially obtained from the vagina and anus of patients with sporadic and recurrent VVC. Virulence factors were phenotypically evaluated in vitro, including: adhesion capacity to epithelial cells obtained from healthy individuals, morphogenesis in the presence of fetal bovine serum, biofilm formation in polystyrene microtiter plates and proteinase activity using bovine serum albumin. Colonizing anal isolates were as able as infecting vaginal isolates to express the virulence factors evaluated in vitro. It was observed an association between the expression of virulence factors studied and the signs and symptoms of VVC presented by the patients. No statistically significant difference was observed in the expression of virulence factors between vaginal isolates of C. albicans obtained from patients with sporadic VVC and those obtained from patients with recurrent VVC. Our results suggest that the ability to express virulence factors is important for the pathogenesis of VVC, but it seems not to be crucial for the transition from colonization to infection.

Electroejaculation procedures (EEPs) provoke stress; nevertheless, ejaculation produces physiological changes similar as those usually used to measure stress responses. The application of EEP to animals that cannot ejaculate-as ewes-may be useful to discriminate the responses induced by ejaculation from those provoked by EEP. The aim was to determine the stress response to EEP in rams and ewes. The EEPs were applied to 10 rams and 10 ewes during the non-breeding season, and the number of vocalizations, the heart rate, rectal temperature, serum cortisol concentration, biochemical and haematological parameters were measured. Overall, EEP provoked increases in cortisol concentration, glycaemia, rectal temperature and concentration of creatine kinase (all them: p < .0001) as well as relative concentration of granulocytes (p = .003) and absolute granulocyte concentration (p = .0002) in both, rams and ewes. Heart rate, relative concentration of lymphocytes (p = .001), haematocrit (p = .02) and haemoglobin (p = .045) decreased in animals from both genders after EEP. Besides, cortisol (p < .0001), rectal temperature (p = .002) and glycaemia (p = .001) were greater in ewes than rams, and creatine kinase also tended to be greater in ewes than rams (p = .054). On the other hand, the number of animals that vocalized (p = .006), white blood cells (p = .02) and absolute lymphocytes (p = .02) were greater in rams than ewes. The general trends show a similar pattern of stress responses in animals from both genders. Therefore, we concluded that ejaculation does not contribute to the stress response provoked by the EEP. This procedure also provokes muscular damage and probably pain.

Low pasture allowance during gestation affects ewes' BW at parturition, the bond with their lamb, lamb development, and thus also may affect their responses to weaning. The objectives were to determine if native pasture allowance from before conception until late pregnancy affects ewe-lamb behaviours at lambing, ewes' milk yield, lambs' BW, and the behavioural and physiological changes of ewes and lambs at weaning. From 23 days before conception until 122 days of pregnancy, 24 ewes grazed on two different native pasture allowances: high (10 to 12 kg of dry matter (DM)/100 kg of BW per day; HPA treatment; n=12) or low (5 to 8 kg of DM/100 kg of BW per day; LPA treatment; n=12). Thereafter, all ewes grazed on Festuca arundinacea and received rice bran and crude glycerine. Ewes' body condition score (BCS) and BW were recorded during pregnancy and postpartum periods. Milk yield was determined on days 32, 41 and 54 after lambing. Lambs' BW was recorded from birth until 72 days after lambing. Latency from parturition until the ewe licked her lamb, maternal behaviour score (a test that evaluates maternal attachment to the lamb) and latency for lamb to stand up and suckle were determined. The behaviour of the lambs and ewes was recorded before and after weaning (at 65 days). The ewes' serum total protein, albumin and globulin concentrations were measured before and after weaning. The HPA ewes presented greater BW (P<0.005) and BCS (P<0.005) than the LPA ewes during pregnancy and postpartum (P<0.04), and had a greater milk yield than the LPA ewes (P<0.03). Treatments did not influence any behaviour at lambing, lambs' BW, neither the ewes' behavioural and physiological changes at weaning. HPA lambs paced and vocalized more than LPA lambs (P<0.0001). The variation of albumin concentration before and after weaning was greater in the HPA lambs than in the LPA lambs (P<0.0001). In conclusion, although ewes' BW, BCS and milk production were affected by pasture allowance until late pregnancy, this did not affect the behaviours that lead to the establishment of the mother-young bond, nor the ewes' behavioural responses at weaning. Lambs reared by ewes that grazed on low pasture allowance during pregnancy presented fewer behavioural changes and a lower decrease of albumin concentration after weaning. Lambs' BW was not affected by the feeding received by their mothers.

BACKGROUND: Antlers are lined by soft velvet tissue during antler growth. Later, the velvet is shed before rut onset. There are no detailed histological descriptions of the growing velvet, nor whether the velvet changes according to stag age. Our aims were to: 1) describe the basic histology of pampas deer antler velvet from adult and yearling males; and 2) determine the influence of age and time of antler growth on velvet's tissues morphometry. MATERIALS AND METHODS: Samples were collected from 10 stags allocated in two groups, either adult (3-5 years old, n = 5) or yearling males (2 years old, n = 5). The day of antler cast was recorded for each animal. In spring, the stags were anaesthetised and velvet samples were collected from the third tine's distal end. Samples were described qualitatively and a restricted morphometrical analysis of the antler velvet was performed. RESULTS: The number of keratinocyte layers and the thicknesses of: total epidermis, corneum, intermediate and basale epidermal strata, total dermis, superficial and deep dermis were determined. Age and days after antler casting positively influenced in conjunction epidermal thickness (p = 0.037), and tended to influence both stratum intermedium (p = 0.076) and stratum corneum (p = 0.1) thicknesses. Age influenced stratum corneum thickness (p = 0.04). The pampas deer antler velvet lacked both sweat glands and arrector pili muscles. CONCLUSIONS: The deep dermis was densely irrigated but displayed abundant and well developed collagen bundles. Both total epidermal and stratum corneum thicknesses related positively to the age of the animals but were not to the time since antler cast.

UNLABELLED: Bee-pollen is an apicultural product with potential for medical and nutritional applications; therefore, its microbiology quality should be monitored. In this context, the objective of this study was to diagnose the microbiological quality of 45 dehydrated bee-pollen samples collected from November 2011 to December 2013 in nine Brazilian States. All the samples were negative for sulphite-reducing Clostridium spores, Salmonella, coagulase-positive Staphylococcus and Escherichia coli, which are micro-organisms of public health concern. Total aerobic mesophilic micro-organism counts ranged from <10 to 1·10 × 10(4) CFU g(-1) , with psychrotroph counts ranging from <10 to 1·12 × 10(3) CFU g(-1) and total coliforms from <10 to 2·80 × 10(3) CFU g(-1) , while the values for yeasts and moulds were between <10 to 7·67 × 10(3) CFU g(-1) . According to the literature, the microbiota observed in this study were typical; however, it is important to consider that these micro-organisms may cause spoilage and diminish shelf life, reason by which quality control programs should be implemented. SIGNIFICANCE AND IMPACT OF THE STUDY: Contamination of bee-pollen can occur during production, collection and processing, but there are few studies of the microbiological quality of this product. Brazil is an important producer of dehydrated bee-pollen, therefore, a diagnosis of the microbiological status is important to ensure the safety of many consumers. Salmonella sp., genus Clostridium, coagulase-positive Staphylococcus, Escherichia coli and even some yeast species are micro-organisms of public health concern and their presence must be monitored. Furthermore, the determination of spoilage micro-organisms indicates whether the production and the processing practices carried out by beekeepers and warehouses were adequate.

We describe the first postnatal diagnosis of a child from Central Brazil with de novo cytogenetic alterations in 13q showing malformations of the brain, eyes, distal limbs, and genitourinary tract, and severe intellectual disability. The karyotype was a constitutive 46,XX,r(13)[77]/45,XX,-13[17]/46,XX,idic r(13)[6]. Interphase and metaphase fluorescence in situ hybridization analyses also showed the absence of 13qter and the presence of 13q14.3 in the cells with r(13), and chromosome microarray analysis detected a 15.39 Mb deletion in chromosome region 13q32.3-q34. This study is intended as the registry of a rare case of chromosomal rearrangement involving chromosome 13 in Central Brazil. Further studies are needed to define whether genetic haploinsufficiency is associated with each major 13q deletion anomaly.

OBJECTIVE To present a case of a patient with clinical and radiological features of reexpansion pulmonary edema, a rare and potentially fatal disease. CASE DESCRIPTION An 11-year-old boy presenting fever, clinical signs and radiological features of large pleural effusion initially treated as a parapneumonic process. Due to clinical deterioration he underwent tube thoracostomy, with evacuation of 3,000 mL of fluid; he shortly presented acute respiratory insufficiency and needed mechanical ventilation. He had an atypical evolution (extubated twice with no satisfactory response). Computerized tomography findings matched those of reexpansion edema. He recovered satisfactorily after intensive care, and pleural tuberculosis was diagnosed afterwards. COMMENTS Despite its rareness in the pediatric population (only five case reports gathered), the knowledge of this pathology and its prevention is very important, due to high mortality rates. It is recommended, among other measures, slow evacuation of the pleural effusion, not removing more than 1,500 mL of fluid at once.

OBJETIVO Relatar caso de paciente con cuadro clínico y radiológico de edema pulmonar de reexpansión, patología rara y potencialmente fatal. DESCRIPCIÓN DEL CASO Niño de 11 años con fiebre y cuadro clínico-radiológico de derrame pleural voluminoso, inicialmente tratado como parapneumónico. Después de la compensación clínica, se realizó drenaje torácico, con salida de 3000mL de líquido. Evolucionó rápidamente con insuficiencia respiratoria aguda, necesitando ventilación mecánica. Tuvo dos extubaciones mal sucedidas y, debido a la evolución atípica, se realizó tomografía computadorizada, cuyos hallazgos fueron compatibles con edema de reexpansión. Después de soporte intensivo, evolucionó satisfactoriamente y, posteriormente, se diagnosticó tuberculosis pleural. COMENTARIOS Es importante el conocimiento de la patología, aunque sea rara en la población pediátrica (encontrados solamente cinco casos descriptos), para prevención, una vez que la tasa de mortalidad es muy alta. Se recomienda, entre otras medidas, que el vaciamiento de un derrame pleural sea lento y que el volumen total retirado no sobrepase 1500mL. .

Após descompensação clínica, realizou-se drenagem torácica, com saída de 3.000mL de líquido. Evoluiu rapidamente com insuficiência respiratória aguda, necessitando de ventilação mecânica. Teve duas extubações malsucedidas e, devido à evolução atípica, realizou-se tomografia computadorizada, cujos achados foram compatíveis com edema de reexpansão. Após suporte intensivo, evoluiu satisfatoriamente e, posteriormente, foi diagnosticado com tuberculose pleural. COMENTÁRIOS É importante o conhecimento da patologia, ainda que seja rara na população pediátrica (encontrados apenas cinco casos descritos), para prevenção, visto que a taxa de mortalidade é muito alta. Recomenda-se, entre outras medidas, que o esvaziamento de um derrame pleural seja lento e que o volume total retirado não ultrapasse 1.500mL. .
