RESUMO

The aim of this study was to assess the levels of oxidative, inflammatory, and fibrinolytic biomarkers as well as DNA strand breakage in hypercholesterolemic subjects. Fasting glucose, total cholesterol, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, triglycerides, uric acid, total protein, albumin, apolipoprotein (Apo) A, Apo B, advanced oxidation protein products (AOPP), increased ischemia-modified albumin (IMA), -SH, NOx, IL-6, and D-dimer levels were assessed, and DNA strand breakage was evaluated using comet assay in 38 patients with hypercholesterolemia and 20 healthy controls. Total cholesterol, triglycerides, LDL cholesterol, Apo A, Apo B, AOPP, IMA, IL-6, and D-dimer concentrations were significantly higher in subjects with hypercholesterolemia. However, NOx and plasma -SH group concentrations were lower in hypercholesterolemic subjects, while no significant differences were observed with respect to DNA strand breakage between the two groups. Hypercholesterolemia is related to oxidative stress and inflammation. Accordingly, AOPP concentration was higher in subjects with hypercholesterolemia, and we speculate that AOPP can reflect the enhancement of protein oxidation and inflammation.

The aim of this study was to investigate socio-economic, clinical, anthropometric, balance and functional fitness factors present in Amazon riparian older persons that can be associated with a risk of falling. A cross sectional study was performed with 637 riverine elderly residents (≥60 years old) in Maués city Amazonas, Brazil. The elderly were grouped in two categories with and without a history of falls in the past six months. The following variables were compared between these groups: self-reported social and health conditions; biochemical and physiological variables related to the control of metabolic diseases; body composition; hand grip strength; functional fitness evaluation using the Senior Fitness Test (SFT) battery, and balance condition using the Berg Balance Test (BBT). The prevalence of at least one fall in the past six months was 24.6% (n=157) and was similar between the sexes. The mean age between males and females with and without a history of falls was also similar (males with falls=72.67 ± 8.86; males with no falls=73.26 ± 7.58) female falls=71.78 ± 8.18, female with no falls=71.48 ± 8.17). A history of falls was associated with hospitalization in the last year and to self-health perception to both sexes and presence of chronic morbidity and percentage of body fat (BF) to women. However, the other variables including balance and functional fitness, did not present differences between elderly with and without a history of falls. These results suggest that falls experienced by the riparian elderly are strongly associated to accidents due to environmental conditions related to daily life.

The aim of this study was to investigate whether there is a differential response of lymphocytes from healthy MnSOD genotype subjects to oxidative stress. We used UV radiation as a toxic agent due to its genotoxic effects associated with chromosome aberrations caused by breaks in the DNA strands. Cellular growth rate, cell viability, mitotic index, chromosomal instability and biomarkers of oxidative metabolism were analysed in lymphocyte cells from healthy adults with different Ala16Val MnSOD polymorphisms that produce tree genotypes: AA, VV and AV. We found a differential response to UV exposure in cultures of lymphocyte cells from Ala16Val genotype donors. In general, AA cell cultures presented higher viability and mitotic index and lower TBARS levels than VV and AV cells for both the control and UV exposure groups. However, when we compared the DNA damage among the three genotypes, AA lymphocyte cells presented the highest damage from UV exposure. These data suggest that the Ala16Val polymorphism affects the response of cellular oxidative metabolism in different ways.
