There is an increasing trend toward broiler production systems with higher welfare requirements. Breed and stocking density are considered key factors for broiler welfare that are often specified as criteria for such higher welfare systems. However, it remains unknown how slower-growing broilers respond to a reduction in stocking density with regard to their welfare and performance, and whether this response differs from fast-growing broilers. Therefore, we compared fast- (F) and slower-growing broilers (S) housed at 4 different stocking densities (24, 30, 36, and 42 kg/m2, based on slaughter weight) and measured their welfare scores (i.e., gait, footpad dermatitis, hock burn, skin lesions and cleanliness), litter quality and performance. The experiment had a 2 × 4 factorial design with 4 replicates (pens) per treatment (32 pens in total). Thinning (15%) was done in a 50/50 male/female ratio at 38 (F) and 44 (S) d of age (estimated body weight of 2.2 kg). We hypothesized that breeds would respond differently to a reduction in stocking density. Contrary to our hypothesis, only one interaction between breed and stocking density was found on footpad dermatitis, indicating that fast- and slower-growing broilers generally showed similar responses to a reduction in stocking density. F broilers showed a steeper decline in the prevalence of footpad dermatitis with reducing stocking density compared to S broilers. Broilers housed at lower stocking densities (24 and/or 30 kg/m2) showed improved welfare measures, litter quality and performance compared to those housed at higher stocking densities (36 and/or 42 kg/m2). S broilers had better welfare scores (gait, footpad dermatitis and skin lesions), litter quality and lower performance compared to F broilers. In conclusion, reducing stocking density improved welfare of both F and S broilers, but more for F broilers in case of footpad dermatitis, and using S broilers improved welfare compared to F broilers. Reducing stocking density and using slower-growing broilers benefits broiler welfare, where combining both would further improve broiler welfare.

Improving leg health will support broiler health and welfare. Known factors to improve leg health are: replacing inorganic by organic macro minerals in the diet, providing environmental enrichments and using slower-growing broilers. However, it remains unknown how fast- and slower-growing broilers respond to a combination of providing organic macro minerals and an elevated platform as enrichment with regard to leg health. Therefore, the aim of this study was to identify whether a combined treatment of organic macro minerals and a platform affected leg health, tibia characteristics, behavior and performance of fast- and slower-growing broilers in a semicommercial setting. The experiment had a 2 × 2 factorial arrangement, with 12.800 fast-growing (Ross 308) and 12.800 slower-growing (Hubbard JA757) broilers that were randomly allocated to a control (i.e., inorganic macro minerals without enrichment) or adapted treatment (i.e., organic macro minerals and a platform). Broilers were housed in groups of 800 per pen (47.5 m2), with 8 replicates per treatment (total of 32 pens). Performance was measured weekly and over the total rearing period. Behavior was observed via scan sampling at a target weight of 0.6 and 1.9 kg for both breeds. Walking ability (gait score), footpad dermatitis, and hock burn were assessed in 10 broilers per pen just prior to slaughter weight. Leg disorders and tibia characteristics were assessed in the same broilers at slaughter weight (2.3 kg). Hardly any interaction effects between breed and treatment were found on leg health, tibia characteristics, behavior or performance, suggesting fast- and slower-growing broilers responded to the treatment similarly. The adapted treatment improved tibia characteristics, and increased locomotion and performance, but did not affect leg disorders, walking ability or contact dermatitis in both fast- and slower-growing broilers. The positive effects of the adapted treatment on tibia characteristics in both fast- and slower-growing broilers may improve leg health, although the current study did not confirm this for leg disorders, walking ability or contact dermatitis.

Since the ban in January 2012 of conventional cages for egg production in the European Union (Council Directive 1999/74/EC), alternative systems such as floor, aviary, free-range, and organic systems have become increasingly common, reaching 50% of housing for hens in 2019. Despite the many advantages associated with non-cage systems, the shift to a housing system where laying hens are kept in larger groups and more complex environments has given rise to new challenges related to management, health, and welfare. This review examines the close relationships between damaging behaviours and health in modern husbandry systems for laying hens. These new housing conditions increase social interactions between animals. In cases of suboptimal rearing and/or housing and management conditions, damaging behaviour or infectious diseases are likely to spread to the whole flock. Additionally, health issues, and therefore stimulation of the immune system, may lead to the development of damaging behaviours, which in turn may result in impaired body conditions, leading to health and welfare issues. This raises the need to monitor both behaviour and health of laying hens in order to intervene as quickly as possible to preserve both the welfare and health of the animals.

In and around poultry farms, high concentrations of endotoxins are found that have a negative impact on the health of farmers and local residents. However, little is known about the effects of chronic exposure to endotoxins on the health of poultry. The aim of this study was to identify effects of chronic exposure to airborne endotoxins (E. coli LPS) on the immune system, respiratory tract, disease susceptibility and welfare of broilers. Effects of high (HE) and low endotoxin (LE) concentrations on natural antibody titers (NAb), performance and behavior of broilers were determined. After treatment with a respiratory virus infection, infectious bronchitis virus (IBV), mRNA expression of cytokines and Toll-like receptor (TLR) 4 in the lung, tracheal ciliary activity and lesions in the respiratory tract were determined. Endotoxin affected the immune system and respiratory tract, where HE broilers tended to have lower IgM NAb binding Phosphorylcholine-conjugated to Bovine Serum Albumin, and higher interferon (IFN)-α mRNA expression and more lesions in the nasal tissue compared to LE broilers. Furthermore, HE broilers had higher TLR4 mRNA expression compared to LE broilers. However, endotoxin did not affect NAb levels binding Keyhole Limpet Hemocyanin, IFN-ß and interleukin-10 mRNA expression, IBV replication or lesions in the lung and trachea. HE and LE broilers further had similar body weight, but HE broilers showed numerically more passive behavior compared to LE broilers. In conclusion, chronic exposure to high airborne endotoxin concentrations affects components of the immune system and respiratory tract in broilers and could therefore influence disease susceptibility.

Gut microbiota influences host behaviour and physiology, such as anxiety, stress, serotonergic and immune systems. These behavioural and physiological characteristics are related to feather pecking (FP), a damaging behaviour in chickens that reduces animal welfare and productivity. Moreover, high FP (HFP) and low FP (LFP) lines differed in microbiota composition. However, it is unknown whether microbiota can influence the development of FP. For the first time, we identified the effects of microbiota transplantation on FP, and behavioural and physiological characteristics related to FP. HFP and LFP chicks received sterile saline (control), HFP or LFP microbiota transplantation during the first two weeks post-hatch. Microbiota transplantation influenced behavioural responses of the HFP line during treatment and of the LFP line after treatment. In both lines, homologous microbiota transplantation (i.e., receiving microbiota from their line) resulted in more active behavioural responses. Furthermore, microbiota transplantation influenced immune characteristics (natural antibodies) in both lines and peripheral serotonin in the LFP line. However, limited effects on microbiota composition, stress response (corticosterone) and FP were noted. Thus, early-life microbiota transplantation had immediate and long-term effects on behavioural responses and long-term effects on immune characteristics and peripheral serotonin; however, the effects were dependent on host genotype. Since early-life microbiota transplantation influenced behavioural and physiological characteristics that are related to FP, it could thus influence the development of FP later in life.

It is crucial to identify whether relations between immune characteristics and damaging behaviors in production animals exist, as these behaviors reduce animal welfare and productivity. Feather pecking (FP) is a damaging behavior in chickens, which involves hens pecking and pulling at feathers of conspecifics. To further identify relationships between the immune system and FP we characterized high FP (HFP) and low FP (LFP) selection lines with regard to nitric oxide (NO) production by monocytes, specific antibody (SpAb) titers, natural (auto)antibody (N(A)Ab) titers and immune cell subsets. NO production by monocytes was measured as indicator for innate pro-inflammatory immune functioning, SpAb titers were measured as part of the adaptive immune system and N(A)Ab titers were measured as they play an essential role in both innate and adaptive immunity. Immune cell subsets were measured to identify whether differences in immune characteristics were reflected by differences in the relative abundance of immune cell subsets. Divergent selection on FP affected NO production by monocytes, SpAb and N(A)Ab titers, but did not affect immune cell subsets. The HFP line showed higher NO production by monocytes and higher IgG N(A)Ab titers compared to the LFP line. Furthermore the HFP line tended to have lower IgM NAAb titers, but higher IgM and IgG SpAb titers compared to the LFP line. Thus, divergent selection on FP affects the innate and adaptive immune system, where the HFP line seems to have a more responsive immune system compared to the LFP line. Although causation cannot be established in the present study, it is clear that relationships between the immune system and FP exist. Therefore, it is important to take these relationships into account when selecting on behavioral or immunological traits.

Feather pecking (FP), a damaging behavior where laying hens peck and pull at feathers of conspecifics, is multifactorial and has been linked to numerous behavioral and physiological characteristics. The gut microbiota has been shown to influence host behavior and physiology in many species, and could therefore affect the development of damaging behaviors, such as FP. Yet, it is unknown whether FP genotypes (high FP [HFP] and low FP [LFP] lines) or FP phenotypes (i.e., individuals differing in FP, feather peckers and neutrals) differ in their gut microbiota composition. Therefore, we identified mucosa-associated microbiota composition of the ileum and cecum at 10 and 30 wk of age. At 30 wk of age, we further identified luminal microbiota composition from combined content of the ileum, ceca, and colon. FP phenotypes could not be distinguished from each other in mucosa-associated or luminal microbiota composition. However, HFP neutrals were characterized by a higher relative abundance of genera of Clostridiales, but lower relative abundance of Lactobacillus for the luminal microbiota composition compared to LFP phenotypes. Furthermore, HFP neutrals had a higher diversity and evenness for the luminal microbiota compared to LFP phenotypes. FP genotypes could not be distinguished from each other in mucosa-associated microbiota composition. Yet, FP genotypes could be distinguished from each other in luminal microbiota composition. HFP birds were characterized by a higher relative abundance of genera of Clostridiales, but lower relative abundance of Staphylococcus and Lactobacillus compared to LFP birds. Furthermore, HFP birds had a higher diversity and evenness for both cecal mucosa-associated and luminal microbiota compared to LFP birds at adult age. In conclusion, we here show that divergent selection on FP can (in)directly affect luminal microbiota composition. Whether differences in microbiota composition are causal to FP or a consequence of FP remains to be elucidated.

Damaging behaviors, like feather pecking (FP), have large economic and welfare consequences in the commercial laying hen industry. Selective breeding can be used to obtain animals that are less likely to perform damaging behavior on their pen-mates. However, with the growing tendency to keep birds in large groups, identifying specific birds that are performing or receiving FP is difficult. With current developments in sensor technologies, it may now be possible to identify laying hens in large groups that show less FP behavior and select them for breeding. We propose using a combination of sensor technology and genomic methods to identify feather peckers and victims in groups. In this review, we will describe the use of "-omics" approaches to understand FP and give an overview of sensor technologies that can be used for animal monitoring, such as ultra-wideband, radio frequency identification, and computer vision. We will then discuss the identification of indicator traits from both sensor technologies and genomics approaches that can be used to select animals for breeding against damaging behavior.

Feather pecking (FP), a serious welfare and economic issue in the egg production industry, has been related to coping style. Proactive and reactive coping styles differ in, among others, the stress response, serotonergic activity and immune activity. Yet, it is unknown whether genetic lines divergently selected on FP (i.e. FP genotypes) or individuals differing in FP (i.e. FP phenotypes) can be categorized into coping styles. Therefore, we determined peripheral serotonin (5-HT) levels, natural antibody (NAb) titers, behavioral and corticosterone (CORT) responses to manual restraint (MR) in FP genotypes (high FP (HFP), low FP (LFP) and unselected control (CON) line) and FP phenotypes (feather pecker, feather pecker-victim, victim and neutral). We further examined the consistency of and relationships between behavioral and physiological measures. FP genotypes differed in behavioral responses to MR, 5-HT levels and NAb titers, but not in CORT levels after MR. HFP birds had less active responses at adolescent age, but more active responses at adult age compared to LFP and CON birds. The CON line had higher 5-HT levels at adolescent age, while the HFP line had lower 5-HT levels than the other lines at adult age. Overall, the HFP line had lower IgM NAb titers, while the LFP line had lower IgG NAb titers compared to the other lines. FP phenotypes differed in behavioral responses to MR and 5-HT levels, but not in CORT levels after MR or NAb titers. Within the HFP line, feather peckers tended to have less active responses compared to neutrals at adolescent age, while victims had more active responses compared to the other phenotypes at adult age. Feather peckers had higher 5-HT levels than neutrals at adult age. Behavioral and CORT responses to MR were not consistent over time, suggesting that responses to MR might not reflect coping style in this study. Furthermore, proactive behavioral responses were correlated with reactive physiological measures and vice versa. Thus, it was not possible to categorize FP genotypes or FP phenotypes into specific coping styles.

A deficient serotonergic system is associated with psychopathological behaviors in various species, among which, feather pecking (FP) in chickens. Deficiency in the serotonergic system can predispose birds to develop FP, while the serotonergic system is affected in birds that feather peck. Serotonin (5-HT) can further influence dopamine (DA) activity. Lines with high FP tendency generally have low central 5-HT and DA turnovers at a young age, but high turnovers at an adult age in brain areas involved in somato-motor regulation and goal-directed behavior. Agonizing 5-HT1A and 5-HT1B receptors increases FP, while antagonizing D2 receptor reduces FP. Genetic associations exist between FP, 5-HT1A and 5-HT1B receptor functioning and metabolism of 5-HT and DA. Birds with deficient functioning of the somatodendritic 5-HT1A autoreceptor and 5-HT metabolism appear predisposed to develop FP. Birds which feather peck often eat feathers, have low whole-blood 5-HT, different gut-microbiota composition and immune competence compared to non-peckers. FP and feather eating likely affect the interaction between gut microbiota, immune system and serotonergic system, but this needs further investigation.

For many animals, long-range signalling is essential to maintain contact with conspecifics. In territorial species, individuals often have to balance signalling towards unfamiliar potential competitors (to solely broadcast territory ownership) with signalling towards familiar immediate neighbours (to also maintain so-called "dear enemy" relations). Hence, to understand how signals evolve due to these multilevel relationships, it is important to understand how general signal traits vary in relation to the overall social environment. For many territorial songbirds dawn is a key signalling period, with several neighbouring individuals singing simultaneously without immediate conflict. In this study we tested whether sharing a territory boundary, rather than spatial proximity, is related to similarity in dawn song traits between territorial great tits (Parus major) in a wild personality-typed population. We collected a large dataset of automatized dawn song recordings from 72 unique male great tits, during the fertile period of their mate, and compared specific song traits between neighbours and non-neighbours. We show here that both song rate and start time of dawn song were repeatable song traits. Moreover, neighbours were significantly more dissimilar in song rate compared to non-neighbours, while there was no effect of proximity on song rate similarity. Additionally, similarity in start time of dawn song was unrelated to sharing a territory boundary, but birds were significantly more similar in start time of dawn song when they were breeding in close proximity of each other. We suggest that the dissimilarity in dawn song rate between neighbours is either the result of neighbouring great tits actively avoiding similar song rates to possibly prevent interference, or a passive consequence of territory settlement preferences relative to the types of neighbours. Neighbourhood structuring is therefore likely to be a relevant selection pressure shaping variation in territorial birdsong.

Rigid endoscopes degrade during clinical use due to sterilization, ionizing radiation and mechanical forces. Despite visual checks on functionality at the department of sterilization, surgeons are still confronted with suboptimal instruments as it is difficult to assess this degradation in an objective manner. To guarantee that endoscopes have sufficient optical quality for minimal invasive surgery, an experimental opto-electronic test bench has been developed in order to be used at the department of sterilization. Transmission of illumination fibres and lens contrast values are stored in a database to enable empirical criteria to reject endoscope for further clinical usage or to accept endoscopes after repair. Results of the test bench are given for an eight month period, where a trained operator performed 1599 measurements on 46 different types. Stability of the system, trends in quality of clinical endoscopes, and effect of repair or replacement were assessed. Although the period was too short to draw firm conclusions, a slow downwards trend in quality of clinically used endoscopes could be observed. Also, endoscopes generally improve in quality after repair or replacement, while endoscope replacement seems to slightly outperform endoscope repair. To optimize the measurement process, a new system is being developed requiring less user interaction and measuring more optical parameters of an endoscope. By commercializing this system, we hope that measurements at different hospitals will give improved insight which acceptance and rejection criteria to use and which factors (usage, cleaning protocol, and brands) determine the economic lifetime of endoscopes.
