The present study investigated the use of fatty acid (FA) profiling in combination with chemometric modelling to verify claims for cow milk in terms of fresh grass feeding, pasture grazing and organic/biodynamic farming. The FA profile was determined for 113 tank milk samples collected in the Netherlands from 30 farms over four different months, and used to develop classification models based on the PLS-DA algorithm. Milk from cows with daily rations of fresh grass could be successfully distinguished from milk from cows with no fresh grass in their diet. Milk from cows at pasture could easily be distinguished from milk from stabled cows without fresh grass in the diet, but the correct prediction of milk from stabled cows fed fresh grass indoors proved difficult. The FA profile of organic/biodynamic milk was different compared to conventional milk but an unequivocal discrimination was not possible either in summer or in winter.

Consumers' interest in organic foods is increasing and so is the need for robust analytical tools for their authentication. This review focuses on the most promising biomarkers/analytical approaches that are available for the authentication of organic produce. Food products have been subdivided into two groups: foods of plant origin (crops) and foods of animal origin (meat, milk and dairy products, eggs and fish). For each food category the most suitable biomarkers are presented and their potential for authentication is discussed. In the light of current knowledge, it is unlikely that the authentication of organic food products can be attained by the measurement of a single marker. Analytical approaches based on the measurement of multiple markers and/or complex chemical or physical profiles/fingerprints supported by multivariate statistical analysis seem considerably more promising in this respect. For the development of robust classification models, well-designed experimental studies must be performed that rely on data sets that are both well balanced and of sufficient size to ensure that all relevant sources of variation for the target biomarkers are included in the reference database.

Biomarker profiling, as a rapid screening approach for detection of hormone abuse, requires well selected candidate biomarkers and a thorough in vivo biomarker evaluation as previously done for detection of growth hormone doping in athletes. The bovine equivalent of growth hormone, called recombinant bovine somatotropin (rbST) is (il)legally administered to enhance milk production in dairy cows. In this study, first a generic sample pre-treatment and 4-plex flow cytometric immunoassay (FCIA) were developed for simultaneous measurement of four candidate biomarkers selected from literature: insulin-like growth factor 1 (IGF-1), its binding protein 2 (IGFBP2), osteocalcin and endogenously produced antibodies against rbST. Next, bovine serum samples from two extensive controlled rbST animal treatment studies were used for in vivo validation and biomarker evaluation. Finally, advanced statistic tools were tested for the assessment of biomarker combination quality aiming to correctly identify rbST-treated animals. The statistical prediction tool k-nearest neighbours using a combination of the biomarkers osteocalcin and endogenously produced antibodies against rbST proved to be very reliable and correctly predicted 95% of the treated samples starting from the second rbST injection until the end of the treatment period and even thereafter. With the same biomarker combination, only 12% of untreated animals appeared false-positive. This reliability meets the requirements of Commission Decision 2002/657/EC for screening methods in veterinary control. From the results of this multidisciplinary study, it is concluded that the osteocalcin - anti-rbST-antibodies combination represent fit-for-purpose biomarkers for screening of rbST abuse in dairy cattle and can be reliably measured in both the developed 4-plex FCIA as well as in a cost-effective 2-plex microsphere-based binding assay. This screening method can be incorporated in routine veterinary monitoring programmes: in the European Union for detection of rbST abuse and in the control of rbST-free dairy farms in the United States of America and other countries.

Two cultural heritage objects studied with scanning electron microscopy-energy dispersive spectroscopy (EDS) are presented in this article: (1) archeological iron present in a soil sample and (2) a chip from a purple-colored area of an undisclosed 17th century painting. Novel PARC software was used to interpret the data in terms of quantitative distribution of mineral and organo-mineral phases as well as their chemical composition. The study serves to demonstrate the power of PARC rather than solving specific archeological issues. The observations on archeological iron potentially can assist in (1) studing the source of iron-metal and the style of forging, (2) learning about alteration processes of artifacts in the particular soil from which the sample originated, and (3) determining the nature of the fractures in the Fe-oxide envelope (desiccation of the sample after excavation, or as primary feature caused by volume change from oxidation). In the paint chip, 11 consecutive layers can be distinguished using the PARC software. In general, each layer consists of a carrier supporting inorganic fragments. In the basal layer the fragments are dominant; in the superimposed layers the carrier usually is. Both organic and inorganic carriers appear to be present. Organic carriers can contain typically inorganic constituents (e.g., Pb, Al), beyond the chemical spatial resolution of EDS (i.e., <1 µm).
