RESUMO

Interploidy cross commonly results in complex chromosome number and structural variations. In our previous study, a progeny with segregated ploidy levels was produced by an interploidy cross between diploid female parent Populus tomentosa × Populus bolleana clone TB03 and triploid male parent Populus alba × Populus berolinensis 'Yinzhong'. However, the chromosome compositions of aneuploid genotypes in the progeny were still unclear. In the present study, a microsatellite DNA allele counting-peak ratios (MAC-PR) method was employed to analyze allelic configurations of each genotype to clarify their chromosome compositions, while 45S rDNA fluorescence in situ hybridization (FISH) analysis was used to reveal the mechanism of chromosome number variation. Based on the MAC-PR analysis of 47 polymorphic simple sequence repeat (SSR) markers distributed across all 19 chromosomes of Populus, both chromosomal number and structural variations were detected for the progeny. In the progeny, 26 hypo-triploids, 1 hyper-triploid, 16 hypo-tetraploids, 10 tetraploids, and 5 hyper-tetraploids were found. A total of 13 putative structural variation events (duplications and/or deletions) were detected in 12 genotypes, involved in chromosomes 3, 6, 7, 14, 15, 16, and 18. The 46.2% (six events) structural variation events occurred on chromosome 6, suggesting that there probably is a chromosome breakpoint near the SSR loci of chromosome 6. Based on calculation of the allelic information, the transmission of paternal heterozygosity in the hypo-triploids, hyper-triploid, hypo-tetraploids, tetraploids, and hyper-tetraploids were 0.748, 0.887, 0.830, 0.833, and 0.836, respectively, indicating that the viable pollen gains of the male parent 'Yinzhong' were able to transmit high heterozygosity to progeny. Furthermore, 45S rDNA-FISH analysis showed that specific-chromosome segregation feature during meiosis and chromosome appointment in normal and fused daughter nuclei of telophase II of 'Yinzhong,' which explained that the formation of aneuploids and tetraploids in the progeny could be attributed to imbalanced meiotic chromosomal segregation and division restitution of 'Yinzhong,' The data of chromosomal composition and structural variation of each aneuploid in the full-sib progeny of TB03 × 'Yinzhong' lays a foundation for analyzing mechanisms of trait variation relying on chromosome or gene dosages in Populus.

Objective@#To understand relationship between health literacy and health seeking behavior of undergraduate students in Shandong Province.@*Methods@#A total of 1 460 undergraduate students from three universities in Shandong Province were selected through stratified random sampling method and received a questionnaire survey. Data were analyzed by one-way ANOVA, Chi-square test and regression analysis.@*Results@#The proportion of students with basic health literacy was 64.4% (41.37±10.34). The proportion of students with basic knowledge and concept, healthy lifestyle and behavior, and basic skills was 49.6% (16.28±4.58), 70.3% (17.24±4.99) and 83.7% (7.85±1.58), respectively. About 28.1% of undergraduate students reported delay in seeking medical treatment, 71.9% reported prompt in seeking medical treatment, and 24.2% preferred to seek medical treatment in primary medical institutions, 45.5% secondary hospital and 30.3% tertiary hospital. Grade, major, the ability to gather and understand health information was the main factors affecting health literacy. Major and health information screening ability was the main factors affecting health seeking behavior. Grade, major, origin, the awareness and gathering of health information was the main factors affecting the choose of medical institutions(P<0.05). Basic knowledge and concept, basic skills and health literacy was positively correlated with the timing of health seeking behavior, while basic knowledge and concept, healthy lifestyle and behavior, and basic skills might give information on students’ preference on selecting a medical institution.@*Conclusion@#Attention should be paid to the cultivation of health information literacy. An integrated health education and health promotion model of "family-community-hospital-university" should be established.

RESUMO

The mosquito Aedes albopictus is the primary vector for dengue virus transmission in Fujian Province.Despite that active dengue surveillance has been launched in several sites since 2005,the genetic diversity of A.albopictus from these sites remains exclusive.In this study,mosquito pools collected from dengue surveillance sites during 2015-2016 were randomly selected,the full-length mitochondrial cytochrome C oxidase subunit Ⅰ (mtDNA-COⅠ) were amplified and sequenced.Preliminary sequence alignment of 12 amplicons with the reference sequence indicated 99 % homology at nucleotide level,due to varia tions at 9 nucleotide sites.Three haplotypes,namely H02,H03 and H08,were determined based on phylogenetic analysis with 72 reference sequences of known haplotypes.These observations facilitate surveillance and control of arboviral diseases in Fujian.
