RESUMO

The Campos de Palmas Wildlife Refuge (CPWR) (Brazil) is a full protection Conservation Unit (CU) formed by private properties, where the use of land and natural resources are allowed sustainably according to the rules of the CU Management Plan. Inadequate practices of land use and occupation within this CU can affect its quality and the organisms that depend on them. Considering the above, the objective of the present study was to biomonitor different land uses and occupations within the CPWR and its surroundings, during the four seasons of a year, using the bioindicators Allium cepa (cytotoxicity and genotoxicity tests) and Eisenia fetida (leakage test). The soil samples were collected in areas of silviculture, agriculture, and native grasslands within the CU and agriculture outside the limits of the CPWR. The use by silviculture (spring) and native grassland (summer) showed cytotoxic effects for A. cepa. The use by agriculture outside the CPWR (spring) showed genotoxic effect and stimulation of mitotic cell division. For the animal bioindicator, the use by agriculture within the CPWR (winter), native grassland (autumn and spring), silviculture (autumn and spring), and agriculture outside the CU (spring) showed a toxic effect. A PCA analysis showed a correlation between the results of toxicity, cytotoxicity, and genotoxicity with the presence of macronutrients and metals in the evaluated soil samples. Possibly influenced by their soil composition characteristics (silviculture and native grassland) or the management of agricultural areas. In addition, samples from the rainy season (spring) showed higher ecotoxicity. These results show that biomonitoring through different organisms is important for evaluating environmental quality and indicate the need for the implementation of preventive measures in the CPWR to avoid the toxic, cytotoxic, and genotoxic effects found. They also ensure the integrity of this CU and the active protection of the environment and biodiversity.

RESUMO

Salt stress during pre-harvest limits the shelf life and post-harvest quality of produce; however, silicon nutrition can mitigate salt stress in plants. Thus, we evaluated the effects of salinity and fertilization with Si, in pre-harvest, on the morpho-physiological characteristics of onion bulbs during shelf life. The experiment was set up in randomized complete blocks, with treatments arranged in split-split plots. The plots had four levels of electrical conductivity of irrigation water (0.65, 1.7, 2.8, and 4.1 dS m-1). The subplots had five fertilization levels with Si (0, 41.6, 83.2, 124.8, and 166.4 kg ha-1). The sub-sub plots had four shelf times (0, 20, 40, and 60 days after harvest). Irrigation water salinity and shelf time reduced firmness and increased the mass loss of onion bulbs during shelf life. Salt stress reduced the contents of sugars and total soluble solids of onion bulbs during storage; however, Si supply improved the contents of these variables. Salinity, Si supply, and shelf time increased the concentrations of pyruvic and ascorbic acids in onion bulbs during shelf life. Si doses between 121.8 and 127.0 kg ha-1 attenuated the impacts caused by moderate salinity, increasing the synthesis of metabolites and prolonging the onion bulbs' shelf life.

RESUMO

Studies have been developed to improve cultivation conditions through the introduction of beneficial compounds, among which are vitamins. The present work aimed to investigate the effects of the application of vitamins in the onion crop. For that, five treatments were used, composed of different vitamins (ascorbic acid, thiamine, niacin and pyridoxine), in a concentration of 100 mg L-¹ and a control treatment, with water application, via foliar spray. The characteristics of chlorophyll relative index, number of plant leaves, plant height, fresh leaf mass, leaf dry mass, bulb diameter, bulb mass and bulb yield were evaluated. It was found that the application of ascorbic acid and pyridoxine increased the relative levels of chlorophyll concerning the control treatment and those composed by the application of niacin and thiamine. In addition, for liquid photosynthesis, a superior result was shown when niacin and thiamine were applied, concerning the other treatments. It was concluded that the exogenous application of vitamins alters the physiological and developmental behavior of onion plants, but does not positively affect bulb yield. The increases in photosynthetic activity were not reversed in bulb development, but in the elongation of plant leaves. Finally, the use of the vitamins studied for onion cultivation is not recommended, under conditions similar to those of the present study.

The use of microalgae as natural biofertilizer in horticulture has recently been reported, while the use of humic substances is widespread. However, the combined use of microalgae with humic substances applied to plant leaves is still unexploited. Thus, the objective of this work was to combine fulvic acid (FA) with the Scenedesmus subspicatus microalga biomass (SC) as a natural biofertilizer applied via leaf in two onion cultivars in organic system. Four experiments were conducted: i) bioassays to verify the bioactive effect of FA, SC and combinations using the Vigna radiata model plant; ii) greenhouse pot experiment with foliar applications of FA, SC and combination in two onion cultivars; iii) field experiment in organic system with foliar applications of FA, SC and combination in two onion cultivars; and iv) onion bulb storage experiment. The bioactive effect of SC, FA and their combinations was identified by promoting changes in root growth of V. radiata. In pots, treatments containing FA, SC and combination promoted increase in fresh and dry leaf mass. The foliar application of FA, SC and combination promoted an increase in field bulb productivity, reduced mass loss in stored bulbs and increased carbohydrate, amino acid and protein contents in onion bulbs.

RESUMO

The Campos de Palmas Wildlife Refuge (RVS-CP) is a full protection conservation unit (CU) formed by private properties. The present study aimed to use the bioindicators Allium cepa L. (cytotoxicity and mutagenicity tests) and Eisenia fetida (avoidance test) to assess the quality of surface water of the Chopim River within the RVS-CP area and its surroundings during the four seasons of the year. To do so, water samples were collected at five points, four inside the RVS-CP area and a fifth point outside thereof. Samples from all sampling points had cytotoxic effect on A. cepa in at least one season of the year. Such a finding may be related to inadequate management practices (without land-use control) in the areas surrounding the sampling points such as forestry, native fields, pastures, agriculture, and housing areas. As for the animal bioindicator (E. fetida), only points 1 (in the winter) and 5 (in the autumn) were toxic. Concerning mutagenicity, points 1 and 4 (in the spring), 1 and 2 (in the summer), and 3 (in the autumn) showed mutagenic effect on A. cepa meristematic cells, therefore only within the RVS-CP area. Overall, these results show that biomonitoring can be an ally of the residents of the RVS-CP area in controlling management practices, aiming to bring together economic support and conservation of resources, especially water.

RESUMO

Abstract Whereas the effects of the substances found in domestic sewage on live organisms is important to evaluate the use of plants to remove pollutants from the environment. The objective of this study was to assess the phytoremediation activity of Allium cepa L. (onion) roots exposed to raw sewage, as well as the acute toxic activity of this effluent for the bioindicators A. cepa, through a cytotoxicity test, and Artemia salina L., through a mortality/immobility test. The physicochemical assessments of the sewage were conducted in two scenarios: immediately after collection and after being in contact with onion roots (phytoremediation) for 24 hours. The physicochemical data indicate there was a reduction in nitrogen and phosphorus levels and in biochemical oxygen demand in sewage treated. The results from the cytotoxicity test with A. cepa indicated a reduction in the mitotic cell divisions of the onions treated with the raw sewage. The mortality/immobility test with A. salina indicated that the concentrations with 50 and 100% of raw sewage induced the mortality of the nauplii. Thus, the data suggest new studies that seek greater efficiency, efficacy and viability of onion phytoremediation.

Wastewater treatment contributes to environmental sustainable development indicators such as clean water and sanitation, then, is imperative to improve the mechanisms and process of contaminant removals. The sewage and industrial effluents are the major contributors of pollutants in land and water discharges, and are necessary to enrich the available data for having reference parameters for plant designing and optimization. The physical andchemical assays alone could not be considered sufficient to assess properly the plant performance because complex mixtures demand ecological and biological parameters for a holistic evaluation. Hence, the ecotoxicity and the genetic toxicity measurement become an important tool to complement the conventional water quality parameters, but these parameters are not widely reported in the open access literature. Despite of several studies showed ecotoxicity and the genetic toxicity data, these could be considered not sufficient because the resulted information is derived from single compounds. Considering the scarce data mentioned above this article presents data on the genetic an ecological toxicity of an anaerobic effluent post-treated with ozone and ozone/UV generated by Chaparro et al. [1] and Chaparro and Pires [2].

RESUMO

The emerging pollutants cover a wide range of synthetic chemicals that are indispensable to modern society but with little known effects for aquatic animals and for people who consume polluted waters with these products. Gels manipulated with hormones are widely used for hormone replacement, muscle growth, among other purposes. However, only a small part of these hormones are absorbed into the skin, and so these can be transferred to the domestic sewage during the washing of exposed body regions. Thus, the aim of this study was to evaluate the toxicity levels for the bioindicator Artemia salina L., and cytotoxicity and mutagenicity for the bioindicator Allium cepa L. of gels handled with 1% testosterone and 0.1% estradiol. Data from immobile/dead A. salina numbers after 24 h of exposure showed that the highest concentrations of testosterone (10 and 25 µg/mL) and the highest concentration of estradiol (15 µg/mL) were toxic to this bioindicator. For the bioindicator A. cepa, mitotic indices and chromosomal aberrations did not indicate statistical differences between the groups treated with the testosterone gels (1, 10, 50 µg/mL) and estradiol (0.03, 0.30, 1.5 µg/mL) and the control group. However, all concentrations of the testosterone-containing gel decreased the percentage of cell division in relation to the time 0 h of each treatment and to the time 24 h of the negative control. Therefore, it is concluded that rivers or aquatic environments can be polluted if wastewater with the toxic concentrations found of these hormonal gels is discarded without previous treatment, compromising the life of organisms that live there. And, it encourages the development of techniques for treating sewage and water to reduce/eliminate the hormones present in them.

RESUMO

The production of films and coatings from onion (Allium cepa L.) to be applied as packaging is attractive, due to its high nutritional and therapeutic value. Also, it can collaborate to minimize environmental impacts caused by the improper disposal of products made from plastics. However, despite it being an innovative and novel proposal, onion films for the development of edible packaging should be evaluated before being considered nontoxic and safe for human consumption. Thus, the objective of the present study was to elucidate the cytotoxic and mutagenic profile of eluates of polymer films of Allium cepa L. obtained by the casting process and to verify their safety for commercial purposes. The analysis of cellular viability demonstrated greater cytotoxicity for unwashed hydrothermally treated pulp (HTP) than for films of washed hydrothermally treated pulp (W-HTP). Regarding the mutagenic activity, the HTP and W-HTP films were not able to statistically increase the frequencies of the biomarkers for chromosome damage (micronucleus test) at the tested concentrations. However, the HTP films showed signs of mutagenicity in the Ames test (gene mutations), suggesting caution in their use. The detection of genotoxicity is highly recommended in order to avoid the risk of genotoxic exposure to mutagens and carcinogens. In conclusion, the absence of mutagenicity and cytotoxicity observed in this study is extremely relevant, because it provides support for toxicogenic properties of the Allium cepa films with promising applicability in the food industry. PRACTICAL APPLICATION: The bioplastics made from onion bulbs are multifunctional materials, which requires safety profile assessment. The results of the mutagenicity and cytotoxicity tests suggests that especially the W-HTP films are harmless, supporting at the first level of evidence, its safety potential to be used in the food industry (food films), biodegradable packaging, and biomaterials (substrates for drug delivery system).

RESUMO

ABSTRACT: Microbial biomass is a driving force in the dynamics of soil organic matter, and microbial activity is an indicator of soil quality in agroecosystems, reflecting changes in management practices and environmental conditions. We evaluated the effect of monoculture and intercropped winter cover crops on soil chemical attributes, microbial biomass carbon (MBC), basal respiration (BR), metabolic quotient (qCO2), urease, β-glucosidase, and fluorescein diacetate (FDA) hydrolysis activity, as well as onion yield in a no-tillage system. Soil is a Typic Humudept, and treatments were control with spontaneous vegetation, barley, rye, oilseed radish (OR), OR + rye, and OR + barley. The soil was sampled (0-10 cm) five times between June and December. There were no differences among treatments for MBC and BR, and the highest values for those attributes occurred in June, when cover plants were in their initial stage. Although, qCO2 was not affected by any treatment, it varied among sampling periods, ranging from 0.62 to 10 µg C-CO2 mg-1 MBC h-1, indicating a low- or no stress environment. Cover crops had little influence on enzyme activity, but FDA was lowered in areas with single crops of barley and rye. Average onion yield in cover crops treatments was 13.01 (Mg ha-1), 30-40% higher than in the control treatment.

RESUMO

A new biopolymer obtained from onion pulp (Allium cepa L.) was employed to produce a sustainable substrate for flexible organic light-emitting diodes (FOLEDs). Indium tin oxide (ITO) and SiO2 thin films were deposited by rf-magnetron sputtering onto these biosubstrates to obtain flexible, transparent, and conductive anodes, on top of which FOLEDs were produced. This new biomaterial exhibits an optical transparency of 63% at 550 nm. ITO films were optimized by varying rf power during deposition onto the biopolymers, and their electrical properties are comparable to the those of ITO grown on top of rigid substrates: a carrier concentration of -3.63 × 1021 cm-3 and carrier mobility of 7.72 cm2 V-1 s-1 for the optimized film. Consequently, the sheet resistance and resistivity of this ITO film were 8.92 Ω sq-1 and 2.23 × 10-4 Ω cm, respectively, hence allowing the production of FOLEDs. The A. cepa L. based FOLED was fabricated using CuPc, ß-NPB, and Alq3 as organic layers, and it exhibited a maximum luminance of about 2062 cd m-2 at 16.6 V. The current efficiency reached a maximum value of 2.1 cd A-1 at 85.3 mA cm-2. The obtained results suggest the possibility to use these substrates for innovative biocompatible applications in optoelectronics, such as photodynamic therapy.

RESUMO

The genus Azospirillum is recognized as plant growth-promoting bacteria that exert beneficial effects on the host plant and is morphologically converted into cyst-like cells (i.e., c-form) in association with poly-ß-hydroxybutyrate (PHB) accumulation in the cells under stress conditions. We constructed Azospirillum brasilense, labeled with reporter genes (gus/gfp, mCherry) and examined the plant tissue localization along with a morphological conversion into the c-form upon its initial interaction with onion seedlings (Allium cepa L.). The PHB granules in the A. brasilense cells were easily detected under fluorescence as "black holes", rendering it possible to monitor the morphological conversion from vegetative to the c-form cells. The results showed that the A. brasilense cells on the surface of the roots and bulbs (underground stem) began converting at three days following inoculation and that the cell conversion was significantly advanced with time along with the cell population increase. The endophytic infection of A. brasilense into the bulb tissues was also confirmed, although these likely constituted vegetative cells. Moreover, the morphological conversion into the c-form was induced under nitrogen-restricted conditions. Analysis of the biochemical properties of the A. brasilense cells during cell conversion revealed that the acetylene reduction activity correlated positively with the PHB accumulation in the cells converting into the c-form under nitrogen-restricted conditions.

Biological process treatment of landfill leachate produces a significant amount of sludge, characterized by high levels of organic matter from which humic acids are known to activate several enzymes of energy metabolism, stimulating plant growth. This study aimed to characterize humic acids extracted from landfill sludge and assess the effects on plants exposed to different concentrations (0.5, 1, 2 and 4 mM C L-1) by chemical and biological analysis, to elucidate the influence of such organic material and minimize potential risks of using sludge in natura. Landfill humic acids showed high carbon and nitrogen levels, which may represent an important source of nutrients for plants. Biochemical analysis demonstrated an increase of enzyme activity, especially H+-ATPase in 2 mM C L-1 landfill humic acid. Additionally, cytogenetic alterations were observed in meristematic and F1 cells, through nuclear abnormalities and micronuclei. Multivariate statistical analysis provided integration of physical, chemical and biological data. Despite all the nutritional benefits of humic acids and their activation of plant antioxidant systems, the observed biological effects showed concerning levels of mutagenicity.

RESUMO

The present study was carried out in order to evaluate the effects of irrigation depths applied through drip and presence/absence of soil cover in onion yield, under agroecological farming. The experiment was conducted in Seropédica, RJ, Brazil, from May to September 2012. The experimental design was a randomized complete block with split-plot design with 10 treatments and 7 replications, characterized by the presence (+S) and absence (-S) of soil cover and five irrigation depths according to percentages of the crop evapotranspiration (ETc): 0, 22, 45, 75 and 100% (0, 155, 220.6, 320.5 and 372,7mm in +S condition and 0, 145.5, 207.6, 285, 351,4mm in -S condition). The irrigation influenced by second-order polynomial regression the total yield of bulbs, percentage distribution of bulbs in diameter classes and water use efficiency (WUE) in the -S condition, and in the linear regression the WUE in +S condition. The soil cover promoted an increase on the total bulb yield, WUE and the percentage of bulbs classified in classes of greater diameter.(AU)
